Abstract

External aging factors such as UV exposure, pollution, and lifestyle choices contribute to skin aging, resulting in deep wrinkles, fine lines, and rough skin, which can lower self-confidence. Plant extracts have been widely studied for their antiaging potential, while Salmon DNA has shown promise in stimulating collagen production. This study explores the formulation of a serum combining Centella asiatica, Curcuma longa, Aloe vera, Rosa centifolia, and Salmon DNA for its antiaging effects on injured human fibroblast cells. The serum was formulated using extracts from C. asiatica, C. longa, A. vera, R. centifolia, and Salmon DNA. Antioxidant activity was evaluated with the DPPH method, cytotoxicity using the WST-8 assay, and gene expression through qRT-PCR for COL1A1, TGF-β1, HYAL-1, and FGF-2. The serum exhibited weak antioxidant activity (IC50 = 373.33 µg/ml) and reduced cell viability at high concentrations. Gene expression analysis revealed increased expression of COL1A1, TGF-β1, and FGF-2, along with reduced HYAL-1 expression in injured BJ cells. The formulated serum shows potential as an antiaging agent, promoting collagen production and reducing hyaluronidase activity.

Abstract

Penuaan kulit merupakan proses alami yang tidak dapat dihindari oleh setiap individu. Proses penuaan ini dapat dipengaruhi secara intrinsik maupun ekstrinsik. Faktor penuaan ekstrinsik yang berlebih dapat memberikan pengaruh signifikan pada kesehatan kulit. Di saat bersamaan, sejumlah masyarakat belum mengetahui pentingnya menjaga kualitas kulit. Maka dari itu, bersama dengan Universitas Kristen Maranatha, diselenggarakan penyuluhan untuk meningkatkan kesadaran penjagaan kualitas kulit hingga memperoleh kulit yang optimal. Kegiatan ini juga dilengkapi dengan pemeriksaan kulit menggunakan Skin Analyzer, serta pemberian skincare berbasis ekstrak bahan alam. Penyuluhan disampaikan dengan metode ceramah yang dilengkapi dengan presentasi visual. Kegiatan yang dilaksanakan dievaluasi melalui pengisian formulir evaluasi oleh seluruh peserta. Kegiatan penyuluhan ini dihadiri oleh 129 orang dengan mayoritas peserta berasal dari kelompok usia dewasa. Hasil formulir evaluasi penyuluhan menunjukkan bahwa mayoritas peserta (>50%) menilai bahwa sistematika penyuluhan, isi penyuluhan, organisasi, dan media yang digunakan selama kegiatan tergolong sangat baik. Hasil formulir evaluasi produk menunjukkan lebih dari 60% peserta yang mengikuti pemeriksaan kulit dapat menilai bahwa tekstur, kemasan, kandungan, kualitas, informasi produk, aroma, dan warna dari produk perawatan kulit berada dalam kategori sangat baik. Hasil formulir penilaian menunjukkan tujuan dari penyuluhan telah tercapai, yaitu kemampuan dalam pemahaman terkait karakteristik produk perawatan kulitdan memperoleh respons baik dari peserta. Setelah penelitian ini diharapkan masyarakat dapat lebih selektif dalam menggunakan produk perawatan kulit.

Abstract

Chronic kidney disease (CKD) is a physiological abnormality in the kidneys whose prevalence is expected to continue to increase. On the other hand, Bitter melon (Momordica charantia L.) is known to have the potential to manage CKD. This study explores the compound content of M. charantia ethanol extract (MCEE) and its potential for CKD based on in vitro assays. To model chronic kidney disease (CKD), SV40 MES-13 (mouse glomerular mesangial) cells were exposed for 3 days to 20 mM glucose. After glucose induction, the cells were subjected with different concentrations of MCEE (Momordica charantia L. ethanolic extract). The chemical profile of MCEE was analyzed using LC/MS-MS. Cell viability was examined through the WST assay, while intracellular ROS and apoptosis levels were measured by flowcytometry. Colorimetry was used to analyze SOD, MDA, and CAT levels. ELISA was used to analyze inflammatory proteins (TGF-β 1, IL-6, TNF-α, IL-1β ) levels. Meanwhile, the relative gene expression of SMAD-2, SMAD-3, SMAD-4, SMAD-7 was examined through qRT-PCR. The results exhibited that MCEE contains cucurbitane p-coumaric, ferulic acid, caffeic acid, gallic acid, chlorogenic acid, and epicatechin. MCEE was also known to be non-toxic to SV40 MES-13 cells. In addition, MCEE reduced intracellular ROS levels, MDA, necrosis levels, and inflammatory proteins, while also regulating SMAD-2, SMAD-3, and SMAD-4 gene expression. MCEE increased levels of CAT, and SOD, and regulated SMAD-7 gene expression in the CKD cells model. The most effective MCEE is MCEE 50 μg/mL. MCEE demonstrated potential as a CKD treatment based on in vitro studies through TGF/SMADs signaling activity, antioxidant, anti-inflammatory, and apoptosis inducer.

Abstract

The prevalence of prostate cancer cases in men is expected to continue increase. In 2040, it is estimated that there will be 2.293.818 new cases and a 1.05% increase in the death rate due to prostate cancer. Eurycoma longifolia Jack roots extract (ELE) has potential as an alternative treatment. This study aims to analyze ELE potential as an anti-prostate cancer agent through in silico assay and in vitro assays on the prostate cancer cell line (PC-3). ELE compounds were docked against Casp-3, Casp-8, HAX-1, p27, and PTEN. In vitro assays on PC-3 cells were used, namely cell viability (WST-8), ROS levels; cell cycle; and cell apoptosis (flow cytometry), PC-3 cell senescence (μ-Galactosidase staining), Casp-3; Casp-8; HAX-1; p27; and PTEN gene expression (qRT-PCR). All proteins target were successfully docked with ELE compounds and presented binding interactions. ELE is known to reduce viability, intracellular ROS levels, live cells, necrosis, and reduce HAX-1 gene expression, and inhibit the cell cycle G0/G1 phase. ELE can also increase inhibition, senescence, late and early apoptosis, and Casp-3, Casp-8, p27, and PTEN gene expression. ELE 100 μg/mL is the most effective concentration. ELE has potential as an anti-prostate cancer agent through apoptosis, cell cycle, and antioxidant pathways

Abstract

Liver fibrosis is a pathological state marked by the excessive buildup of extracellular matrix due to persistent liver damage. Despite the potential of traditional medicines, including antiviral medications and lifestyle adjustments, to decelerate fibrosis progression, a completely effective treatment is still lacking. This article examines the function of human-induced pluripotent stem cells (hiPSCs) in mimicking liver fibrosis. HiPSCs can differentiate into multiple liver cell types, such as hepatocytes, hepatic stellate cells, and endothelial cells, facilitating the reconstruction of liver microarchitecture in both two-dimensional cultures and three-dimensional organoids. These technologies offer critical insights into the pathophysiological underpinnings of the disease, facilitate the discovery of therapeutic targets, and aid in the development of innovative antifibrotic drugs. The use of hiPSCs not only enables novel methods for disease modeling but also presents intriguing opportunities for more targeted and effective regenerative therapy for liver fibrosis.

Abstract

Oral health faces significant challenges due to increasing dental infections by pathogenic microbes such as Porphyromonas gingivalis, Streptococcus mutans, and Candida albicans. Topical antioxidants in toothpaste, gel, and mouthwash are often used to treat dental diseases. There is a growing interest in finding natural alternatives for oral hygiene without side effects compared to conventional toothpaste formulations. Nigella sativa (black cumin) has various pharmaceutical efficacy, making it a valuable plant-based source medicinal compound. This study evaluated black cumin ethanol extract's (BCSE) antimicrobial and antioxidant activities. Antibacterial effectiveness was evaluated through Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC), and disc diffusion assays against P. gingivalis, S. mutans, and C. albicans. Antioxidant activity was reported based on DPPH scavenging, ABTS reduction, H2O2 scavenging, and NO scavenging assays. Black cumin seed ethanolic extract (BCSE) showed antibacterial activity of 4.49 mm, 4.33 mm, and 7.12 mm for P. gingivalis, S. mutans, and C. albicans, respectively, with zones of inhibition increasing in a concentration-dependent manner (p<0.05). MIC and MBC evaluations also revealed that MIC for S. mutans was achieved at 12.5% BCSE concentration, while P. gingivalis and C. albicans required 25%, and MBC reached 100%. The IC50 for DPPH, ABTS, NO, and H2O2 were 25.41, 69.93, 87.18, and 95.16 μg/ml, respectively. The antioxidant activity increased in a concentration-dependent manner, especially at 100 µg/ml of BCSE. Based on these results, BCSE could be an optional ingredient in toothpaste.

Abstract

Wharton’s jelly mesenchymal stem cells (WJMSCs) hold promises for treating osteoarthritis (OA) due to their great capacity for regeneration and their accessibility. However, obstacles like poor cell survival and differentiation prevent them from being used in transplants. Thus, this study examined WJMSCs’ Conditioned Medium (CM) (WJMSCs-CM) for OA treatment. In order to determine how well IL1β-induced chondrocyte cells (IL1β-CHON002) heal OA-damaged cells, they were treated with both IGF1-induced and uninduced WJMSCs-CM. Key parameters measured were IL1β, Sox9 and COL2. The STRING database was explored to evaluate the proteins contained in WJMSCs-CM and to see their interactions and CM potential in OA exposure. The STRING database indicates WJMSCs-CM to possess the ability to manage OA because of the growth factors they contain, which raise the expression of cartilage marker genes. The results indicated that 15% WJMSCs-CM with 150 ng/mL IGF1 had the highest Sox9 expression, significant reduction in pro-inflammatory mediators, and an increase in COL2 levels. Additionally, 14 days of treatment resulted in better outcomes compared to 7 days. CM from WJMSCs treated with IGF1 with a concentration of 15% exhibited the highest level for each parameter. Both durations showed difference across parameters, highlighting the potential for OA therapy.

Abstract

Streptococcus mutans merupakan bakteri yang bersifat asidogenik dan kariogenik yang berperan sebagai agen etiologi dalam pembentukan karies. Pemilihan pasta gigi yang mengandung bahan herbal merupakan salah satu upaya dalam pencegahan karies. Kombinasi ekstrak teh hijau dan peppermint dapat digunakan sebagai formulasi sediaan pasta gigi karena memiliki sifat antibakteri. Tujuan penelitian ini adalah mengetahui efek antibakteri sediaan pasta gigi ekstrak teh hijau (Camellia sinensis L.) dan peppermint (Mentha piperita L.) terhadap S. mutans. Penelitian ini menggunakan metode difusi cakram, terdapat empat kelompok perlakuan yang diuji yaitu pasta gigi ekstrak teh hijau dan peppermint dengan konsentrasi 1000 mg/ml, 500 mg/ml, 250 mg/ml, 125 mg/ml, 62,5 mg/ml, dan 31,25 mg/ml, pasta gigi base, pasta gigi herbal sebagai kontrol positif, dan NaCl 0,9% sebagai kontrol negatif. Hasil pengukuran diameter zona hambat terkecil didapatkan pada konsentrasi 125mg/ml dengan diameter zona hambat rata-rata sebesar 6,31 mm sedangkan zona hambat terbesar berada pada konsentrasi 1000 mg/ml dengan diameter zona hambat rata-rata sebesar 11,43 mm. Kesimpulan dari penelitian ini adalah pasta gigi ekstrak teh hijau dan peppermint memiliki efek antibakteri terhadap Streptococcus mutans.

Abstract

Wound healing often requires specialized interventions to accelerate recovery due to prolonged inflammation and limited regenerative factors. Umbilical Cord Mesenchymal Stem Cells secretome (UCMSCs) comprises various cytokines and growth factors that can promote wound healing. This study aims to analyze the potential of a secretome-based hydrogel as a wound-healing agent using BJ fibroblast cells induced by lipopolysaccharide (LPS) and scratch injury as an inflammation model. The secretome hydrogel was formulated using Carbopol, Hydroxypropyl Methylcellulose (HPMC), Hydroxyethyl Cellulose (HEC), and secretome. Cytotoxicity was conducted using the WST-8 assay, while cell migration was evaluated through a scratch assay. Tumor Necrosis Factor-???? (TNF-????), Nuclear Factor kappa-B (NF-????B), and Interleukin-8 (IL-8) gene expression were analyzed via qRT-PCR. Additionally, malondialdehyde (MDA) levels were measured for oxidative stress assessment, whereas Connective Tissue Growth Factor (CTGF) and Transforming Growth Factor-????1 (TGF-????1) levels were quantified using ELISA and colorimetric assays. The secretome hydrogel exhibited no cytotoxic effects on BJ fibroblast cells and significantly enhanced cell migration. Moreover, it reduced the TNF-????, IL-8, and NF-????B expression, indicating anti-inflammatory activity. The hydrogel also decreased MDA levels while increasing TGF-β1 and CTGF expression, suggesting antioxidant properties and enhanced tissue regeneration in the inflammatory model. The secretome-based hydrogel presents a promising therapeutic approach for promoting chronic wound healing by modulating inflammation, reducing oxidative stress, enhancing tissue regeneration, and stimulating fibroblast migration.

Abstract

ABSTRAK Pendahuluan: Plak merupakan lapisan lunak yang terdiri dari kumpulan bakteri yang tumbuh pada permukaan gigi. Pertumbuhan plak dapat berhubungan dengan saliva, sehingga untuk menjaga pH saliva rongga mulut tetap normal perlu dilakukan kontrol plak. Produk herbal seperti pasta gigi yang mengandung teh hijau dan peppermint dapat berpengaruh dalam pencegahan pembentukan plak dan pH saliva. Penelitian ini bertujuan untuk menganalisis efektivitas sediaan pasta gigi ekstrak teh hijau (Camellia sinensis L.) dan peppermint (Mentha piperita L.) terhadap indeks plak dan pH saliva. Metode: Jenis penelitian berupa eksperimental dengan desain pretest dan post test. Subjek sebanyak 32 orang dipilih berdasarkan kriteria inklusi dan kriteria eksklusi yang kemudian dibagi menjadi dua kelompok (n=16) secara random. Kelompok pertama menyikat gigi menggunakan pasta gigi ekstrak teh hijau, peppermint dan kelompok kedua menyikat gigi menggunakan pasta gigi pembanding. pH saliva kedua kelompok diukur menggunakan pH meter dan skor plak gigi menggunakan metode O’Leary. Metode analisis data yang digunakan adalah uji Wilcoxon. Hasil: Kelompok pertama dan kedua menunjukkan tidak terdapat perubahan signifikan pada pH saliva (p=0,068 dan p=0,729) namun terdapat perubahan signifikan pada indeks plak (p=0,000 dan p=0,033). Simpulan: Menyikat gigi menggunakan pasta gigi yang mengandung ekstrak teh hijau (Camellia Sinensis L.) dan peppermint (Mentha piperita L.) memiliki efektivitas dalam menurunkan indeks plak serta menjaga pH saliva tetap dalam keadaan normal. Kata kunci indeks plak, pasta gigi, peppermint, pH saliva, teh hijau  Effectiveness of green tea (Camellia sinensis L.) and peppermint (Mentha piperita L.) toothpaste extracts on plaque index and saliva pH: a laboratory experiment ABSTRACT Introduction: Plaque is a soft layer consisting of a collection of bacteria that grows on the surface of the teeth. Plaque growth can be related to saliva, so to maintain the pH of oral saliva remaining normal, it is necessary to control plaque. Herbal products such as toothpaste containing green tea and peppermint can prevent plaque and salivary pH. This research was conducted to determine the effectiveness of green tea (Camellia sinensis L.) and peppermint (Mentha piperita L.) toothpaste preparations on plaque index and salivary pH. Methods: The research design is experimental with a pretest-posttest design. A total of 32 subjects were recovered based on inclusion and exclusion criteria and then divided into two groups (n=16) randomly. The first group brushed their teeth using green tea extract toothpaste, peppermint and the second group brushed their teeth using CT toothpaste. The saliva pH of both groups was measured using a pH meter and dental plaque scores using the O'Leary method. The data analysis method used is the Wilcoxon test.  Results: The results of the first and second groups of research showed that there was no significant change in saliva pH (p=0.068 and p=0.729) but there was a significant change in the plaque index (p=0.000 and p=0.033). Conclusion: Brushing teeth using toothpaste containing green tea (Camellia Sinensis L.) and peppermint (Mentha piperita L.) extracts is effective in reducing the plaque index and maintaining saliva pH within normal conditions. Keywords green tea, peppermint, plaque index, saliva pH, toothpaste
