Abstract

Free radicals are known as a leading factor in aging. Nitric oxide (NO) and O2•- have been shown to inhibit the synthesis of matrix components and stimulate prolidase activities involved in collagen degradation. Hydroxyl (•OH) radical is a precursor of hyaluronic acid degradation. Antioxidants act as a scavenger for free radicals, creating a possibility to use them to prevent the aging process. Black soybeans (Glycine max (L.) Merr.) contain bioactive compounds that have the ability to scavenge free radicals, including daidzein. Daidzein belongs to the isoflavones group which is the most active compound in black soybean. This study aimed to understand the antioxidant activities of the black soybean extract (BSE) and daidzein compound in the scavenging of •OH and NO radicals and was performed at the Aretha Medika Utama-Biomolecular and Biomedical Research Center (BBRC), Bandung, Indonesia, from September to November 2018. The scavenging activity of •OH was assessed using the deoxyribose method, while the assessment of the scavenging of NO radicals was carried out using the sodium nitroprusside method. BSE and daidzein had an •OH scavenging activity with an IC50 value of 71.07 μg/mL, followed by daidzein with 24.57 μg/mL. A higher NO scavenging activity was seen in BSE with an IC50 of 71.60 μg/mL followed by daidzein with 35.68 μg/mL. Daidzein has a higher antioxidant activity through hydroxyl and nitric oxide scavenging compared to Glycine max (L.) Merr. extract. Hence, daidzein has a higher potential as an anti-aging agent based on the free-radical theory of aging. Aktivitas Antioksidan Ekstrak Kedelai Hitam (Glycine max (L.) Merr.) dan Daidzein sebagai Pemerangkap Hidroksil dan Nitrogen MonoksidaRadikal bebas dikenal sebagai faktor utama penuaan. Nitrogen monoksida (NO) dan O2•- telah terbukti menghambat sintesis komponen matriks dan merangsang aktivitas prolidase yang terlibat dalam degradasi kolagen. Radikal hidroksil (•OH) adalah prekursor dari degradasi asam hialuronat. Antioksidan bertindak sebagai pemerangkap radikal bebas sehingga dapat digunakan untuk mencegah proses penuaan. Kedelai hitam (Glycine max (L.) Merr.) mengandung senyawa bioaktif yang mampu memerangkap radikal bebas, salah satunya adalah daidzein. Daidzein termasuk dalam kelompok isoflavon yang merupakan senyawa paling aktif dalam kedelai hitam. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan, pemerangkapan •OH dan NO dari ekstrak etanol kedelai hitam (BSE) dan senyawa daidzein. Penelitian ini dilakukan selama bulan September sampai dengan November tahun 2018 di Laboratorium Aretha Medika Utama Biomolecular and Biomedical Research Center (BBRC) Bandung. Pemerangkapan •OH dilakukan dengan metode deoksiribosa, sedangkan pemerangkapan radikal NO dilakukan dengan metode menghitung natrium nitroprusida. BSE dan daidzein memiliki aktivitas pemerangkapan •OH dengan nilai IC50 71,07 μg/mL diikuti oleh daidzein 24,57 μg/mL. Aktivitas pemerangkapan NO pada BSE ditunjukkan dengan IC50 71,60 μg/mL sementara daidzein 35,68 μg/mL. Daidzein memiliki aktivitas antioksidan yang lebih tinggi berdasarkan pemerangkapan •OH dan NO dibanding BSE. Oleh karena itu, daidzein memiliki potensi lebih sebagai antiaging menurut teori penuaan akibat radikal bebas.

Abstract

The potential utilization of flavonoid compounds, especially scutellarin, and apigenin, contained in basil (Ocimum basilicum L.) leaf extract to manage the aging effects on the skin, that occurs because of over-activated hyaluronidase enzyme and oxidative stress due to hydrogen peroxide (H2O2) radicals, is not well known. This study was conducted to assess the H2O2 scavenging activity and the inhibition of hyaluronidase from scutellarin and apigenin at various concentrations. The study was conducted by measuring H2O2 scavenging inhibition and hyaluronidase inhibition of scutellarin and apigenin in various concentrations using the spectrophotometry method. The various activity was tested using the One-Way ANOVA test followed by Tukey post hoc test. IC50 values were calculated based on linear regression equations of H2O2 scavenging inhibition and hyaluronidase inhibition. The analysis showed the highest H2O2 scavenging activity was found in scutellarin with IC50 158.76 μg/mL. Scutellarin has greater scavenging activity than apigenin. Hyaluronidase inhibition of scutellarin with IC50 35.25 µg/mL, while apigenin was 162.86 µg/mL. Scutellarin has higher hyaluronidase inhibition activity than apigenin. Antioxidant and antiaging effects of basil leaf extract caused by phytochemical compounds contained, especially scutellarin.

Abstract

Background: Skin aging is a complex natural process caused by both intrinsic or genetically programmed aging and extrinsic aging caused by environmental factors, such as free radical. The use of antioxidant and anti-collagenase to prevent the aging proses has been known. Natural compounds from plants are one of the sources of antioxidant and anti-collagenase that has ability to prevent aging. Genistein and Epicatechin are the major phenolic compound found in G. max. Objectives: This research was to evaluate the antiaging potential of genistein and epicatechin through antioxidant activity assay (ABTS-reducing activity assay) and collagenase inhibitory activity assay. Methods: Antioxidant analysis of Genistein and Epicatechin was performed by 2,2’- azinobis-3-ethylbenzo-thiazoline-6-sulfonic acid (ABTS) reducing activity Assay. Antiaging assay was conducted through inhibitory of collagenase, one of important enzyme in aging process. Results: ABTS- reducing activity assay showed that both compounds had great ABTS-reducing activity in which epicatechin had better activity than genistein. Epicatechin had low value of IC50 ABTS-reducing activity around 14.39 µg/ml better than genistein with IC50 about 43.17 µg/ml. In terms of collagenase inhibitory activity assay, epicatechin had lower value of IC50 (9.08+-3.46 ug/ml), better than genistein (98.74+-4.25 ug/ml). Conclusion: Epicatechin had higher antioxidant and anti- collagenase activity compared to Genistein.

Abstract

Free radicals can cause damage to cells or tissues, autoimmune diseases, degenerative diseases, or cancer. Therefore, the body needs important substances, namely antioxidants that can help protect the body by reducing negative effect from free radicals. Rose flower (Rosa damascena) has anthocyanin pigment which belongs to flavonoid group which has a function as antioxidant or free radical scavenger. This study aims to determine antioxidant and anti-elastase potentials of rose petals and receptacles. The method used in this study was a qualitative phytochemical test to determine the compounds contained in the Rose Petal Extract (RPE) and Rose Receptacle Extract (RRE), ABTS ((2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid))-reducing assay to determine antioxidant activity, and antiaging test with anti-elastase assay. RPE and RRE contained flavonoids, phenols, tannins and alkaloids, but did not contain saponins. RPE contained triterpenoids and terpenoids, while RRE contained steroids, but did not contain terpenoids. The inhibitory concentration (IC50) values in the ABTS reducing assay were 4.46 ± 0.34 μg/mL (RPE) and 15.49 ± 0.23 μg/mL (RRE), while the results of the anti-elastase assay were 17.51 ± 1.47 μg/mL (RPE) and 58.91 ± 2.31 μg/mL (RRE). Both RPE and RRE are potent antioxidant and anti-elastase, and RPE is more active than RRE in these assays.

Abstract

The declining ability of tissues to repair or replace themselves and maintain their structure and normal function is called aging. Aging is caused by free radical that can induce the activation of elastase and collagenase. Ageratum conyzoides (L.) or known as Babadotan is a medicinal plant that has been reported to be a good source of antioxidants that also inhibit aging process. The purpose of this research is to evaluate the antioxidant and antiaging properties of Babadotan Ethanolic Extract (BEE). Franswoth method was used to determine the phytochemical assay of BEE. Measurement of 2,2-diphenyl 1-picrylhydrazyl (DPPH) scavenger was used for antioxidant assay, and inhibitory of elastase and collagenase activities was used for antiaging assay. The analysis of phytochemical compounds exhibited the presence of flavonoids, alkaloids, tannins, polyphenols, steroids and triterpenoids, monoterpenoids and terpenoids, and the absence of saponin. BEE has lower activity on DPPH scavenging activity (IC50 = 80.7 µg/mL) than quercetin (IC50 = 3.25 µg/mL). BEE exhibited lower elastase and collagenase inhibitory activity (IC50 = 45.35 ± 2.2 µg/mL and 55.07 ± 1.1 µg/mL, respectively) compared to quercetin (IC50 = 11.64 ± 0.67 µg/mL and 19.91 ± 0.46 µg/mL). Overall, BEE possesses antioxidant and antiaging activities, although the activities are lower than quercetin.

Abstract

AbstrakStaphylococcus aureus menjadi bakteri penyebab utama terjadinya abses di rongga mulut. Frekuensi dental abses di dunia mulai dari 0,7% sampai 15% dan 14% di Indonesia. Biji pinang (Areca catechu L.) dan Daun sirih (Piper betle L.) memiliki aktivitas antimikroba. Tujuan penelitian ini adalah menilai aktivitas antimikroba dari infusa biji pinang, daun sirih dan kombinasinya terhadap Staphylococcus aureus. Penelitian ini bersifat eksperimental laboratorik metode difusi dengan metode Kirby-Bauer. Penelitian ini dilakukan triplo. Rerata zona inhibisi terbesar terdapat pada ciprofloxacin (25,46±0,19 mm), diikuti infusa biji pinang 6% (11,76±0,96 mm), 8% (11,60±0,64 mm), 10% (11,38±0,96 mm), kombinasi infusa biji pinang 6% dan daun sirih (10,88±0,72mm), infusa daun sirih (9,82±0,81 mm), kombinasi infusa biji pinang 8% dan daun sirih (8,66±0,41 mm), dan kombinasi infusa biji pinang 10% dan daun sirih (8,54±1,23 mm). Simpulan penelitian infusa biji pinang 6%, 8%, 10%, infusa daun sirih, dan kombinasinya memiliki efek antimikroba terhadap S. aureus.Kata Kunci: biji pinang; daun sirih; antimikroba; Staphylococcus aureus AbstractStaphylococcus aureus is the main cause of abscesses in the oral cavity. The frequency of dental abscesses in the world ranges from 0.7% to 15% and 14% in Indonesia. Areca nut (Areca catechu L.) and Betel leaf (Piper betle L.) has antimicrobial activity. The purpose of this study was to measure the inhibition zone from infusion of areca nut, betel leaf and their combination against Staphylococcus aureus and is expected to be a complementary therapy. This research is a laboratory experimental study using the Kirby-Bauer method of diffusion. This research was conducted in triplo. The largest mean zone of inhibition was found in ciprofloxacin (25.46±0.19 mm), followed by areca nut infusion 6% (11.76±0.96 mm), 8% (11.60±0.64 mm), 10% (11.38±0.96 mm), combination of betel leaf infusion and 6% areca nut (10.88±0.72mm), betel leaf infusion (9.82±0.81 mm), combination of betel leaf and 8% areca nut (8.66±0.41 mm), and combination of 10% areca nut infusion and betel leaf (8.54±1.23 mm). Abstract

Herbal teas and plant-based medicines have been widely used worldwide for their therapeutic and healing qualities. Rosella (Hibiscus sabdariffa L.) has various benefits in preventing digestive problems, liver disease, fever, and others. Ginger (Zingiber officinale Rosc.) or known as jahe in Indonesia is believed to benefit numerous clinical conditions linked to oxidative stress, including pancreatitis, hypertension, diabetic kidney disease, Alzheimer’s disease, and tumor development. They have been utilized in traditional medical practice to treat conditions such as fever, nausea, and headaches. In this study, we aimed to determine the antioxidant activities of rosella flower tea, ginger tea, and a combination of both, namely RoJa (Rosella-Jahe) tea. Antioxidant activities were analyzed by the 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydrogen peroxide (H2O2), 2-2′-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) scavenging activities, and Ferric Reducing Antioxidant Power (FRAP) assay, as well as by measuring the amount of total phenolic and flavonoid content. Rosella tea had the greatest activity in the DPPH assay, with an IC50 value of 0.412 mg/ml. Ginger tea had the greatest activity for the ABTS, FRAP, and H2O2 tests, with IC50 values of 12.595 mg/ml, 0.494 mg/ml, and 1.323 mg/ml, respectively. Ginger tea also had the highest amount of total phenolic (3.61 µg GAE/g sample) and flavonoid content (11.81 µg QE/g). In conclusion, ginger tea showed the highest antioxidant activities, as well as total phenolic and flavonoid content, compared to rosella and RoJa tea.

Abstract

Streptococcus mutans and Candida albicans are the main microorganisms that cause dental cavities. It can cause infection, damaged tissue around teeth, abscesses, and focal infection to other organs in the body. Natural products are currently widely used as products or as additives in the prevention of dental caries which have more anti-bacterial and anti-fungal activities than antibiotics that can cause resistance. Citronella (Cymbopogon citratus) is abundant and easy to grow. This study was conducted to determine the effectiveness of citronella (Cymbopogon citratus) oil on the growth of S. mutans and C. albicans microorganisms by Minimum Inhibitory Content (MIC) and Minimum Bactericidal Content (MBC). The method of MIC is broth microdilution by making chlorhexidine concentration levels of 100%, 50%, 25%, 12.5%, 6.25%, 3.125%, and 0.2% with 4 replications each. The MIC value was determined based on absorbance spectrophotometry and the MBC value was determined from the agar plate using the spread method. Biofilm eradication test was conducted by crystal-violet staining and measuring the absorbance. The results of MIC and MBC were obtained on S. mutans, namely at concentrations of 25% and 100%, respectively. Meanwhile, the results of MIC and MBC on C. albicans were obtained at concentrations of 50% and 100%, respectively. Furthermore, the citronella oil has antibacterial and antifungal activities.

Abstract

Abstract: Candida albicans are normal flora of the oral cavity that are opportunistically pathogenic and becomes the main causative agent of oral candidiasis. The addition of herbal ingredients to toothpaste is a solution to resistance and synthetic antifungal side effects. Green tea and peppermint extracts are known to have an antifungal role with various active ingredients such as catechins, tannins, flavonoids, terpenoids, alkaloids, and saponins. This study aimed to determine the antifungal effect of toothpaste preparations of green tea extract and peppermint on C. albicans. This study used the agar diffusion method at several concentrations of green tea and peppermint extracts in toothpaste preparations, namely 1000 mg/ml, 500 mg/ml, 250 mg/ml, 125 mg/ml, 62,5 mg/ml and 31,25 mg/ml. The results showed that there was an antifungal effect of green tea and peppermint extract toothpaste against C. albicans at a concentration of 1000 mg/ml with an average inhibition zone diameter of 7.19 mm. The smallest inhibition zone was 125 mg/ml with a diameter of 1.16 mm. In conclusion, green tea and peppermint extract toothpastes have antifungal effects. The higher the concentration of the extract used in the toothpaste preparation, the greater the antifungal inhibition zone formed. Penambahan bahan herbal pada pasta gigi mikroba merupakan solusi terhadap resistensi dan efek samping antijamur sintetik. Ekstrak teh hijau dan peppermint diketahui memiliki peran sebagai antijamur dengan berbagai kandungan aktifnya seperti katekin, tanin, flavonoid, terpenoid, alkaloid dan saponin. Mengetahui efek antifungi sediaan pasta gigi ekstrak teh hijau dan peppermint terhadap C. albicans. Penelitian ini menggunakan metode difusi agar dengan beberapa konsentrasi ekstrak teh hijau dan peppermint dalam sediaan pasta gigi yaitu 1000 mg/ml, 500 mg/ml, 250 mg/ml, 125 mg/ml, 62,5 mg/ml dan 31,25 mg/ml. Hasil penelitian memperlihatkan adanya efek antifungi pada pasta gigi ekstrak teh hijau dan peppermint terhadap C. albicans pada konsentrasi 1000 mg/ml dengan diameter zona hambat rata-rata sebesar 7,19 mm. Zona hambat terkecil dari konsentrasi 125 mg/ml dengan diameter 1,16 mm. Simpulan penelitian ini ialah pasta gigi ekstrak teh hijau dan peppermint memiliki efek antijamur. Semakin tinggi konsentrasi ekstrak yang digunakan dalam sediaan pasta gigi maka semakin besar zona hambat antijamur yang terbentuk. Abstract

Natural skin aging is indicated by a loss of skin structure and integrity caused by external factors including UV exposure. This exposure causes oxidative stress on skin cells, initiates aging, and degradation of extracellular matrix (ECM) structure which is composed of many proteins, including collagen and elastin. ECM degradation is caused by the increased activity of proteolytic enzymes, one of which is elastase (closely related to wrinkling). To inhibit oxidative stress during the photoaging process, the skin needs antioxidant compounds. Salak (Salacca zalacca (Gaert.)Voss) is a fruit that is rich in antioxidants because it contains flavonoid, phenolic, and polyphenolic compounds, including chlorogenic acid. The purpose of this study is to demonstrate the antioxidant activity and antiaging properties of salak fruit extract (SFE) as a candidate for active ingredients in the prevention of aging. To determine the antioxidant activity of SFE, 2,2-diphenyl-1-picrylhydrazil (DPPH) scavenging and ferric reducing antioxidant power (FRAP) assays were performed, while elastase assays were performed to determine the anti-aging properties of SFE. The IC50 values for DPPH scavenging and FRAP activity of SFE were 107.52 μg/mL and 16.82 μg/mL with the highest activity at 68.79% and 97.96%, at concentrations of 200 and 50 μg/mL, respectively. Meanwhile, the anti-aging properties determined through the elastase assay showed an IC50 value of 19.71 μg/mL with the highest inhibition 72.50% at a concentration of 66.67 μg/mL. SFE has the potential as an active ingredient in preventing aging through its antioxidant activity and anti-aging properties.
