Abstract

The aim of this research was to know the effect of maturity of red coconut water as a diluent to the sperms quality of Boer goats during chilled preservation. The research was carried out at Laboratory Sumber Sekar Faculty of Animal  Husbandry University of Brawijaya Malang, on 11st December 2016 until 31st January 2017. The method used in this research was laboratory experimental using Randomized Block Design with three treatments, P0 (Tris Aminomethane + 20% Egg Yolk), P1 (Young Red Coconut Water + 20% Egg Yolk) and P2 (Mature Red Coconut Water + 20% Egg Yolk) which each treatment using 10 repetitions. The observed variables include the percentage of motility, percentage of viability and percentage of abnormal spermatozoa during chilled preservation The results showed that on the 3ndday the percentage of motility of spermatozoa during chilled preservation gave a significant effect (P<0.05), while on viability gave a very significant effect (P<0.01), and on abnormality did not give a significant effect (P> 0.05). The highest of total motile spermatozoa was P0 (Tris Aminomethan + 20% Egg Yolk). P2 better than P1 on motility, but not on viability and abnormality. The conclusions was the maturity of red coconut water can improve motility, but can not improve viability and abnormality. The suggested was use Mature Red coconut water + 20% Egg Yolk as a diluent  and do further research on coconut water to be able to maintain spermatozoa longer.Keyword : Sperms, egg yolk, red coconut water, tris aminomethane

Abstract

Studi ini bertujuan untuk mengevaluasi kecukupan nilai nutrisi untuk induk sapi potong di Desa Leran Wetan dan Leran Kulon, Kecamatan Palang, Kabupaten Tuban. Data diambil dengan metode survei menggunakan kuisioner kepada 15 responden di masing-masing desa. Sampel pakan diambil di masing-masing peternak responden kemudian dianalisis proksimat dan dilanjutkan uji kecernaan BK dan BO secara in vitro di Laboratorium Nutrisi dan Makanan Ternak, Fakultas Peternakan, Universitas Brawijaya.  Analisis data dilakukan secara deskriptif kuantitatif. Hasil survei menunjukkan bahwa rata-rata kepemilikan induk sapi potong di masing-masing desa antara 2-3 ekor, bobot badan 300-350 kg/ekor, dengan tujuan pemeliharaan untuk menghasilkan pedet. Jenis pakan yang sering diberikan bervariasi meliputi rumput gajah, rumput lapang, limbah pertanian seperti jerami padi dan jerami jagung serta dedak padi sebagai pakan tambahan. Konsumsi BK pakan di Desa Leran Wetan rata-rata adalah 5,853 ± 1,663 kg/ekor/hari dan di Desa Leran Kulon adalah 5,943 ± 1,325 kg/ekor/hari sedangkan kebutuhan berdasarkan bobot badannya adalah 7,4 – 8,3 kg/ekor/hari. Ransum dengan pakan basal limbah pertanian di Desa Leran Kulon memiliki KcBK sebesar 27,844 ± 0,679% sedangkan ransum dengan pakan basal rumput memiliki nilai KcBK lebih tinggi yaitu 44,919 ± 1,800%. Nilai KcBK Ransum berbasis jerami di Desa Leran Wetan adalah 27,918 ± 0,692% dan yang berbasis rumput adalah sebesar 44,434 ± 2,026%. Nilai KcBO di Desa Leran Kulon pada ransum berbasis jerami dan rumput berturut-turut adalah 30,234 ± 46,412% dan 46,412 ± 1,778% sedangkan di Desa Leran Wetan berturut-turut adalah 33,689 ± 1,081% dan 52,989 ± 3,139%. Berdasarkan hasil studi tersebut, dapat disimpulkan bahwa pakan yang dikonsumsi oleh ternak masih belum memenuhi kebutuhannya. Penggunaan limbah pertanian dapat menurunkan nilai kecernaan ransum maka sebaiknya diganti dengan hijauan seperti leguminosa dan rumput. Pakan dengan kualitas yang baik akan meningkatkan performa reproduksi induk sapi potong.

Abstract

ABSTRAK Penyimpanan semen bisa dilakukan dengan pendinginan atau pembekuan. Setelah pendinginan atau pembekuan viabilitas spermatozoa akan menurun. Untuk dapat mempertahankan viabilitas spermatozoa selama pendinginan atau pembekuan diperlukan krioprotektan dengan level tepat. Trehalosa merupakan krioprotektan ekstraseluler yang mampu menstabilkan membrane dan sebagai anti oksidan bagi spermatozoa selama pendinginan dan pembekuan sehingga diharapkan dapat mempertahankan viabilitas spermatozoa. Masalah dalam penelitian ini adalah bagaimanakah peranan trehalosa pada level yang berbeda dalam pengencer dasar tris mampu menekan penurunan viabilitas spermatozoa kambing Boer selama pendinginan dan pembekuan? Tujuan  dari penelitian ini adalah mengkaji pengaruh berbagai level trehalosa dalam pengencer dasar tris terhadap viabilitas spermatozoa kambing Boer setelah  pendinginan dan pembekuan. Metode penelitian yang digunakan adalah eksperimental laboratorium. Penelitian terdiri atas 2 tahap. Tahap I. Pengaruh level trehalosa dalam pengencer tris terhadap viabilitas spermatozoa kambing Boer setelah pendinginan. Tahap 2. Pengaruh level trehalosa dalam pengencer tris terhadap viabilitas spermatozoa kambing Boer setelah pembekuan. Level trehalosa yang dicobakan pada masing-masing tahap adalah: 1,5%; 2,5% dan 3,5%, dan ulangan: 10 kali. Variabel yang diamati adalah viabilitas spermatozoa. Hasil penelitian menunjukkan bahwa level trehalosa berpengaruh terhadap viabilitas spermatozoa, baik setelah pendinginan maupun pembekuan. Dari hasil penelitian dapat disimpulkan bahwa dalam pendinginan dan pembekuan semen kambing Boer, masing-masing penambahan level 1,5% dan 2,5% dalam pengencer dasar tris menghasilkan viabilitas spermatozoa yang optimal. Dari hasil penelitian disarankan bahwa dalam pendinginan dan pembekuan semen kambing Boer sebaiknya ditambahkan trehalosa masing-masing 1,5% dan 2,5% dalam pengencer dasar tris agar mendapatkan viabilitas spermatozoa yang optimal.   Kata kunci: viabilitas spermatozoa, trehalosa, pendinginan, pembekuan, kambing Boer VIABILITY OF BOER GOAT SPERMATOZOA AFTER COOLING AND FREEZING USING TRIS-BASED-SOLUTION SUPPLEMENTED WITH DIFFERENT LEVEL OF TREHALOSE ABSTRACT Semen storage could be conducted in cold or frozen condition. However, during this storage the viability of spermatozoa decreases continuesly. For maintaining viability of spermatozoa during this period, the use of optimal cryoprotectants is sometimes efficient. Trehalose is one of extracellular cryoprotectants that shows capability for stabiling of spermatozoa membrane during cooling and freezing process, and it was expected to be a protectant agent from spermatozoa damage. The problems in this study were: How does the role of trehalose at different level in tris-based-solution minimize the decrease of spermatozoa viability during cooling and freezing? The objectives in this study were to study the role of different levels of trehalose in tris-based medium on the viability of spermatozoa in Boer goat semen after cooling and freezing. This study was designed as an experimental laboratory study. The research consisted 2 steps. While the Step 1, studied the effect of trehalose level in tris-based medium on the viability Boer goat spermatozoa after cooling. Step 2, studied the effect of trehalose level in tris-based medium on the viability Boer goat spermatozoa after freezing. As the levels of trehalose of 1.5%; 2.5% and 3.5% were applied in this research. The results of Step 1 and 2, showed that the level of trehalose statistically affected the spermatozoa viability. It was concluded, that the trehalose levels for resulting optimal spermatozoa viability after cooling and freezing were 1.5% and 2.5%, respectively. In suggestion, for the bettet results in the cooling and freezing of Boer goat spermatozoa, trehalose should be supplemented of 1.5% and 2.5%, respectively, in tris-based medium. Abstract

This research was conducted from 20th Mei to 10th June 2014 at Sumber Sekar Field Laboratory Animal Husbandry Faculty Brawijaya University Malang. The aims of this experiment was to test the effect of semen storage duration diluted with Andromed solution at room temperature on the quality of  Boer goat spermatozoa. The material was 1 boer goat semen aged 3 years old. Methods  used experimental Randomized Design with 6 treatment and 10 replications by used Completely Randomized Design. The interval storage duration were 0, 60, 120, 180, 240 and 300 minutes. Data was analysed by analysis of variance. The result showed that  interval storage duration of Boer goat semen diluted with Andromed solution were highly significant different (P<0,01) on sperm individual motility, sperm viability and sperm abnormality percentage. Optimal time for AI was 300 minutes at room temperature with 40±4,08% of individual motility and 57,94±1,71% of viability percentage.  Optimal time for AI was 300 minutes in room temperature with 13,20±0,20% of sperm abnormality. The conclusion of this result was Boer goat semen diluted with Andromed at room temperature should be used not more than 300 minutes.   Keyword: Boer goat, sperm motility, viability and sperm abnormality

Abstract

Penelitian ini bertujuan untuk mengetahui tampilan reproduksi sapi Rambon betina pada paritas yang berbeda. Variabel dalam penelitian ini adalah nilai Service per Conception, Calving Interval, Conception Rate dan Days Open di Kecamatan Glagah, Kabupaten Banyuwangi. Materi yang digunakan adalah 113 ekor sapi Rambon dipilih secara purposive sampling pada paritas yang berbeda. Data dianalisis dengan analisa deskriptif menggunakan nilai rataan dan standar deviasi pada nilai Service per conception, Calving Interval, Conception Rate, Days Open dan dilanjutkan menggunakan uji Kruskal Wallis. Hasil penelitian ini menunjukkan bahwa nilai Service per Conception pada paritas satu memiliki efisiensi terbaik sebesar 1,1±0,31 kali. Nilai Calving Interval pada paritas tiga memiliki efisiensi terbaik sebesar 374,4±29,02 hari. Nilai Days Open pada paritas tiga memiliki efisiensi terbaik sebesar 96,9±27,93 hari. Nilai Conception Rate pada paritas satu menunjukkan efisiensi terbaik yaitu sebesar 90%. Disimpulkan bahwa tampilan reproduksi sapi Rambon di Kecamatan Glagah Kabupaten Banyuwangi dalam kondisi optimal.

Abstract

Kualitas spermatozoa pada semen beku sexing sangat penting dalam menunjang keberhasilan inseminasi buatan untuk mendapatkan pedat dengan jenis kelamin sesuai harapan. Tujuan penelitian ini bertujuan untuk mengetahui kualitas semen beku semen beku non sexing dan sexing secara makroskopis dan mikroskopis. Materi dalam penelitian menggunakan starw semen beku non sexing dan sexing dengan metode pemisahan Sentrifugasi Gradien Densitas Percoll (SGDP) sapi Limousin yang diproduksi oleh Balai Besar Inseminasi Buatan (BBIB) Singosari, Malang. Metode penelitian ini menggunakan eksperimental dengan percobaan labolatorium (laboratory experiment). Variabel penelitian meliputi keadaan umum meliputi tahun produksi straw, secara makroskopis meliputi pH semen beku dan secara mikroskopis meliputi persentase motilitas, viabilitas, abnormalitas, konsentrasi dan total spermatozoa motil. Data yang diperoleh diolah Microsoft Excel dengan analisis Ragam Analysis of variance (Anova) single factor dianalisis secara deskriptif. Hasil penelitian yaitu semen beku non sexing dan sexing meliputi keadaan umum diproduksi tahun 2012 dan 2018, kualitas secara makroskopis memiliki nilai pH sebesar 6,67 dan 6,60 dan kualitas secara mikroskopis memiliki persentase motilitas sebesar 36,00 % dan 31,40 %, viabilitas sebesar 81,70 % dan 75,89 %, abnormalitas sebesar 6,93 % dan 6,78 %, konsentrasi spermatozoa sebesar 31,67 juta/ mini straw dan 12,125 juta/ mini straw dan total spermatozoa motil sebesar 11,39 juta/mini straw dan 5,10125 juta/mini straw. Kesimpulan penelitian bahwa Kualitas post thawing semen beku non sexing lebih tinggi daripada semen beku sexing menggunakan metode SGDP baik secara makroskopis maupun mikroskopis.

Abstract

ABSTRAKPengencer dasar air kelapa merupakan solusi dari sulit dan mahalnya harga pengadaan bahan baku pembuatan pengencer. Tujuan dari penelitian ini adalah untuk mengetahui kapasitasi spermatozoa sapi Peranakan Ongole dalam berbagai formulasi pengencer air kelapa selama simpan dingin pada suhu 4-5oC. Metodologi yang digunakan adalah eksperimantal laboratorium menggunakan rancangan acak kelompok (RAK). Terdapat tiga perlakuan yaitu, P1 = Pengencer air kelapa + 20% kuning telur, P2 = Pengencer air kelapa + 20% kuning telur + 0,4% putih telur + fruktosa 1000 mg/l. P3 = Pengencer air kelapa + 20% kuning telur + 0,4% putih telur + fruktosa 2000 mg/l, dengan 10 ulangan. Hasil menunjukkan bahwa tidak terdapat perbedaan yang signifikan pada persentase spermatozoa belum kapasitasi, terkapasitasi dan telah rekasi akrosom antara perlakuan formulasi pengencer air kelapa yang berbeda (P1, P2, P3) selama peyimpana suhu 2-5oC. Pengencer berbasis air kelapa mampu mempertahankan kualitas akrosom diatas 50% penyimpanan hari ke lima.Kata kunci : air kelapa, kapasitasi, reaksi akrosom, semen cairABSTRACTCoconut water extender is the solution to the difficulty and high cots of semen diluents. The objective of this study were to examine the capacitation of Ongole Crossbreed sperm in various formulations of coconut water extender during storage in 4-5 oC. The study method used laboratory experimental. The experimental design was Randomized Completely Block Design and the data were analyzed by Analyze of Variance. There are three treatments in this study (P1 = Coconut Water + 20% of yolk, P2 = Coconut Water + 20% of yolk + 0,4% white egg + fructose 1 mg/ml, dan P3 = Coconut Water + 20% of yolk + 0,4% white egg + fructose 2 mg/ml) and ten replications of each treatment. There were not significant differences in percentage of sperm uncapacitation, sperm capacitaion and sperm acrosom reaction between various formulations of coconut water diluents during chilled storage at 4-5 oC. Coconut water diluents are able to maintain quality of sperm acrosom up to day 5th with values above 50%.Keywords: acrosom reaction, capacitation, coconut water, liquid semen

Abstract

The use of appropriate diluent is intended to ensure the physical and chemical requirements ofspermatozoa so that the quality, particularly capacitation can be maintained. This study aimed todetermine the quality of Limousin cattle sperm after sexing with density percoll centrifugation methodusing Andromed diluent and Cauda epididymal Plasma 2 (CEP-2) plus 10% egg yolk. Observations ofspermatozoa quality including : motility, viability, percentage of abnormal, concentrations, membraneintegrity and state of capacitation. The semen used were obtained from Limousin cattle at the Center forArtificial Insemination(BBIB) Singosari Malang, with motility criteria e” 70%. Data were analysed usinga paired T-test in order to determine differences in the use of thinning treatments at each layer. Theresults showed that both diluent, andromed and CEP-2 diluent plus 10% egg yolk were able to maintainthe quality of spermatozoa either in the upper and lower layers

Abstract

Purpose of this research was to investigate the effect of Bovine Serum Albumin substitution by albumen on CEP-2 to semen quality Ongole CrossBred bull stored at 3-5oC. Research was conducted at Research Centre Beef Cattle Laboratory, Grati, Pasuruan on February 2016. Semen diluent was divided into two groups, there were P0 (90% CEP-2 + 10% Egg Yolk (EY)); and P1 (90% CEP + 0.4% albumen + 10% EY). Data of the research were analyzed using paired design t test. The result showed that after eight days chilled preservation, percentage of motility P1 (47.4±10.9%) was higher than P0 (47±9.2%). Percentage of viability P0 (83.1±1.9%) was higher than P1 (81.3±1.5%). Percentage of abnormality P1 (3.6±0.4%) was lower than P0 (3.8±0.3%). Total motile sperm count after six days chilled preservation was significantly higher in all treatments compared to the standard criteria of SNI 40% motile sperm/ml. The conclusion of this research was 0,4% albumen could replace the BSA capability on CEP-2. Keywords: CEP-2, Bovine Serum Albumin, chilled preservation, semen quality

Abstract

Testing of spermatozoa fertility in vivo is needed to find out the extent of the abilityof spermatozoa fertilization in the female reproductive tract. The fertility ability ofspermatozoa in fertilizing the ovum is indicated by the number of fertile eggs fromthe number of eggs incubated resulting from the mating with Artificial Insemination(AI). The aim of this research is to know the percentage of fertility of eggs from AIusing semen which has been stored cold for 8 hours on Ringe's diluent with theaddition of antioxidant glutathione. Collection semen with teaser female method isdone in Poultry Breeder Installation of Sekolah Tinggi Penyuluhan Pertanian (STPP)Malang. The 30 female chickens were divided into 3 groups of treatment P0: AIusing fresh semen with Ringer's diluent, P1: AI using cold cement with Ringer'sDiluent (8 hours duration storage) and P2: AI using cold semen with 0.5 mMaddition in Diluent Ringe's (8 hours duration storage). Experiment design usingComplete Randomized Design (RAL) and  nonparametric data analysis with chi-square.The observed variable was egg fertility calculated based on egg fertility percentage.The results showed that the dilution treatment had a significant effect (P <0,05) onfertility egg, with the best result indicated by P0 that is 76% fertility, followedby P2 with fertility 42% and P1 which showed the lowest fertility was 34%. IB withcold cement has not demonstrated a good percentage of fertility despite 0.05 mMglutathione added as an antioxidant. It is recommended that AI in domestic poultryusing fresh semen with Ringe's diluent, is done as soon as possible to obtain optimumfertility.
