Abstract

Penelitian dilakukan di Balai Inseminasi Buatan Daerah Ungaran Jawa Tengah. Tujuan penelitian untuk mengetahui perbedaan kualitas semen segar dan produksi semen beku sapi Simmental pada umur yang berbeda. Materi yang digunakan dalam penelitian ini adalah catatan produksi semen yang terdiri dari kualitas semen segar dan jumlah straw semen beku dari 8 ekor sapi Simmental. Metode yang digunakan adalah studi kasus dengan Nested Design. Variabel yang diamati meliputi volume semen, pH, motilitas massa, persentase motilitas individu, konsentrasi spermatozoa, total spermatozoa, total spermatozoa motil dan jumlah straw semen beku. Hasil penelitian menunjukkanbahwa kualitas dan kuantitas semen segar pada 4 kelompok umur 3, 4, 7 dan 8 tahun secara berturut-turut adalah volume: 6,9±1,7; 7,3±0,5; 7,8±0,3 dan 9,2±1,6 ml, pH: 6,4±0,1; 6,5±0,1; 6,5±0,1 dan 6,5±0,1, motilitas massa: 2±0,04; 2±0; 2±0,05 dan 1,8±0,22 (+), persentase motilitas individu: 72.1±1.2; 71.3±0.8; 70.3±0.5 dan 66.3±5%, konsentrasi spermatozoa: 1.703,3±167,2; 1.612,2±172,2; 1.450,7±172,2 dan 1.254,6±113,6 (juta/ml), total spermatozoa: 11.586,5±1945,6; 10.468,1±1148,8; 12.593,6±947,8 dan 11.570,2±2642,2 juta sel, total spermatozoa motil: 8.341,8±1.282,5; 7.471,7±845,6; 8.857,6±662,7 dan 7.820,3±2.229,4 juta sel, jumlah straw semen beku: 300±38,2; 263,6±57,8 dan 236,6±180,1 buah straw. Kata kunci: semen beku, motilitas, konsentrasi, spermatozoa motil

Abstract

The purpose of this research wasinvestigated the substitution effect of Bovine Serum Albumin (BSA) with albumen on CEP-2 diluent to semen quality of Boer goat that was stored at 3-50C. Research was conducted at Reproduction Laboratory and Field Laboratory SumberSekarDau Animal Husbandry Faculty of Brawijaya University from December 2016until January 2017. The materials used for this research were fresh semen from 3,5-4,5 years old Boer goat which was collected with artificial vagina. Semen diluent was divided into two groups, those were P0 (90% CEP-2 + 10% Egg Yolk) and P1 (90% CEP-2 (without BSA) + 0,4% albumen + 10% Egg Yolk). Data of the research were analyzed using paired design t test. The result showed that after eight days of chilled preservation, the sperm motility of P0 was not significantly different with P1 (P>0,05). The average percentage of motility during eight days preservation P0 was higher than P1. The viability between P0 and P1 showed highly significantly difference (P<0,01). The average percentage of viability during eight days preservation P0 was higher than P1. The abnormality between P0 and P1 showed highly significant difference (P<0,01). The average percentage of abnormality between eight dayspreservation P1 was higher than P0. Total motile sperms count after seven days chilled preservation was not significantly different with hope value 40 million motile sperm/ml. The conclusion of this research was the substitution of BSA with albumen could maintain Boer goat semen quality. Keywords : CEP-2, Bovine SerumAlbumin, Semen, Cryoprotectant

Abstract

This study was conducted from May to June 2014 at Sumber Sekar Labolatory of Animal Husbandry Faculty, University of Brawijaya, Malang. The purpose of the study was to find out quality of semen Boer goat in room temperature with different storage duration. The result showed that storage treatment of semen with yolk tris aminhomethan diluents had highly signicant defferen as (P<0.01) on individual motility, viability and abnormality of semen. The conclusion of this result was optimal storage time was 240 minutes in room temperature with 53.50±2.42% of individual motility, 61.29±3.15% of viability and 15.15±2.42% of abnormality. Keyword: semen, Boar goat, tris aminomethane yolk, storage

Abstract

Penelitian ini dilaksanakan di Balai Besar Inseminasi Buatan (BBIB) Singosari pada bulan Maret sampai April 2016. Tujuan penelitian untuk mengetahui pengaruh pemberian glutathione pada pegencer Tris Aminomethane kuning telur dalam mempertahankan kualitas semen sapi Limousin selama penyimpanan suhu ruang. Materi yang digunakan adalah semen segar dengan motilitas individu 45-55%. Metode penelitian yang digunakan adalah percobaan. Metode percobaan dengan menggunakan Rancangan Acak Kelompok dan dianalisis dengan menggunakan analisis ragam. Perlakuan pada penelitian ini P0 ( 0 mM glutathione), P1 (0,5 mM glutathione), P2 (1 mM glutathione) dan P3 (1,25 mM glutathione) dengan 10 ulangan. Hasil penelitian menyatakan bahwa penambahan glutathione pada pengencer Tris Aminomethane kuning telur berbeda sangat nyata (P<0,01) persentase motilitas individu selama penyimpanan 3 dan 24 jam, sedangkan viabilitas dan abnormalitas spermatozoa pada semua perlakuan tidak berbeda nyata (P>0,05). Penyimpanan semen 3 jam dan 24 jam disuhu ruang sebaiknya menggunakan penambahan 0,75 mM glutathione dalam Tris Aminomethane kuning telur agar mempertahankan motilitas spermatozoa.Kata kunci : abnormalitas, Motilitas, Pengencer,semen, viabilitas

Abstract

This research aimed to determine the quality of fresh semen and recovery rate (RR) Limousin bulls at different seasons in Indonesia. The research materials were used semen sample and recording of 10 Limousin bulls on November 2015 until September 2016. The analyze was used by descriptive, while seasonal factors were determined by rainfall, they were divided into 2 stage (summer and rainy). The result was showed that the average of individual motility (60±0.07 %) on summer and (57.7±0.09 %) on rainy, Consentration of spermatozoa (1043±208 million/ml) on summer and (1013±175 million/ml) on rainy, post thawing motility (44±0.02 %) on summer and (41±0.05 %) on rainy, RR value (65±0.05 %) on summer and (55±0.09 %) on rainy, the production of frozen semen (1979±644.2 straw) on summer and (1623±379 straw) on rainy. It was concluded that the best quality of fresh until frozen semen was generally on summer with low rainfall. The seasonal effect for quality of semen Limousin bulls were individual motility, concentration, PTM, and RR value will be impacted for frozen semen produce. It was advisable to rejected the Limousin bulls with poor semen quality.Keywords: Seasonality, Summer, Rainy, Quality of Semen

Abstract

Artificial insemination (AI) using liquid semen give an alternative for the areas doesn’t exist a liquit nitrogen.The purpose of the research was to know about the succesfull artificial insemination (AI) using liquid semen with Tris Aminometan + egg yolk diluent of ongole crossbreed cattle indicated by Non Return Rate (NRR) and Conception Rate (CR).  This research was conducted in sub district Tumpang Malang Regency, East Java on June 2017 until November 2017. The material used in this research were 54 ongole crossbreed cows normal reproduction and estrus  condition. The method in this research was experimental using 54 heads of cattle with 27 heads inseminated used frozen semen  (T0) and 27 heads inseminated liquid semen (T1). The parameters observed were NRR1, NRR2, and NRR3 and CR. The data showed the first parameter of AI success was 81.48% of NRR1, 81.48% of NRR2, and 81.48% of NRR3 used frozen semen and 96.30% of NRR1, 88.89% of NRR2and 81.48% of NRR3 used liquid semen. While for CR showed result 62.96% for frozen semen and 59.26% for liquid semen. In conclusion, the value of NRR for all treatments still meet the AI success standards. The result of Conception Rate of Frozen semen treatments is 62.96% and the liquid semen is 59.26%.Keyword: frozen semen, liquid semen, Non Return Rate, and Conception Rate.Inseminasi buatan (IB) menggunakan semen cair memberikan alternatif daerah yang tidak ada nitrogen cair. Tujuan dari penelitian ini adalah untuk mengevaluasi keberhasilan IB menggunakan semen cair dengan pengencer Tris Aminomethan kuning telur pada sapi persilangan ongol yang berdasarkan Non Return Rate (NRR) dan Conception Rate (CR). Penelitian ini dilaksanakan dikecamatan Tumpang Kabupaten Malang, Jawa Timur pada bulan Juni 2017 hingga November 2017. Materi yang digunakan dalam penelitian ini adalah 54 ekor sapi betina dari berbagai bangsa yang normal dan dalam kondisi birahi. Metode dalam penelitian ini adalah percobaan yang menggunakan 54 ekor sapi dengan rincian 27 ekor di IB semen beku (P0) dan 27 ekor di IB semen cair (P1). Parameter yang diamati adalah NRR1, NRR2, dan NRR3 dan CR. Data menunjukan keberhasilan IB adalah NRR1 81,48%, NRR2 81,48%, dan NRR3 81,48% untuk semen beku dan NRR1 96,30%, NRR2 88, 89%, dan NRR3 81,48% untuk semen cair. Sedangkan untuk CR menunjukkan hasil 62,96% untuk semen beku dan 59,26% untuk semen cair. Kesimpulannya, nilai NRR untuk semua perlakuan masih memenuhi standar keberhasilan IB. Hasil konsepsi perlakuan semen beku adalah 62,96% dan semen cair 59,26%. Kata kunci : semen beku, semen cair, Non Return Rate dan Conception Rate.

Abstract

This research was to compare semen quality and quantity of simmental cattle on thevariation of weight. The research used six heads of cattle. The method of this research wassurvey method and analyze data using variety analyzing method, thus would be continuedusing duncan test if there are present significan differences. Simmental cattle at mediumweight (840 and 846 kg) had more quality than other two groups of weight, which showedfrom highest scale of colour, and consistency. Simmental cattle at low weight (764 and 797 kg)showed a highest scale of pH and mass motility rather than other two groups of weight. Theresearch of quantity of Simmental cattle at medium weight (840 and 846 kg) group showedhighest scale of volume average at 8,5 ± 0,92 ml, individual motility average at 60,6 ± 7,95%and amount of motil spermatozoa average at 6.082,6 ± 1.204,06 (106) rather than other groups.Highest scale concentration had showed on low weight (746 and 797 kg) Simmental cattlegroup average at 1.355,7 ± 153,60 (106/ml). The high weight (942 and 952 kg) group ofSimmental cattle showed highest amount of spermatozoa average at 10.826,9 ± 1.777,16 (106).Simmental cattle at medium weight (840-846 kg) had better semen quality and quantity thanothers.Keywords : Simmental cattle, cattle weight, quality and quantity of semen

Abstract

Penelitian ini bertujuan untuk mengkaji dan mengidentifikasi perbedaan kecepatan pubertas calon pejantan sapi PO yang dipelihara pada kelompok sex yang berbeda. Materi yang dipergunakan 28 ekor pedet jantan dan 9 ekor pedet betina dengan umur 8 - 11 bulan, yang dibagi menjadi tiga perlakuan A (kontrol) yaitu satu ekor sapi jantan/kandang ditempatkan dalam kandang  individu; B yaitu empat ekor sapi jantan (sex sejenis ditempat dalam kandang kelompok) dan C yaitu empat ekor sapi jantan dan tiga ekor sapi betina muda (sex beda jenis yang ditempatkan dalam kandang kelompok) dengan tiga ulangan. Kegiatan ini dilaksanakan enam bulan di kandang percobaan Loka Penelitian Sapi Potong, Grati, Pasuruan. Data yang diperoleh dianalisa dengan Two Way Anova menggunakan program Microsoft Excel 2007. Hasil penampungan semen dengan elektroejakulator menunjukkan spermatozoa dengan persentase motilitas individu minimal 10 % dan konsentrasi minimal 50 juta/ml yaitu sapi kandang A = 1 ekor; B= 4 ekor dan C=3 ekor. Hasil analisis ragam terhadap hasil penampungan semen tidak berbeda nyata (P>0,05) dari ketiga perlakuan. Kesimpulan dari penelitian ini menunjukkan bahwa calon pejantan sapi PO yang dipelihara pada kelompok sex sejenis maupun beda jenis memiliki waktu pubertas yang  sama cepatnya (15 bulan) dibandingkan pada kandang individu (16 bulan). Kata Kunci : Calon Pejantan sapi PO, kelompok sex dan pubertas

Abstract

             Tujuan dari penelitian ini yaitu untuk mengevaluasi pengaruh penambahan filtrat kecambah kacang hijau dalam pengencer susu skim terhadap kualitas spermetozoa pejantan sapi Madura pada penyimpanan suhu ruang. Materi dalam penelitian ini adalah semen segar sapi Madura berusia 6-11 tahun dengan persentase motilitas 50-65% diambil dari Balai Besar Inseminasi Buatan Singosari, Malang. Metode yang digunakann dalam penelitian ini adalah percobaan di laboratorium dengan rancangan acak kelompok yang terdiri dari empat level penambahan filtrat kecambah kacang hijau yaitu 0%, 2%, 4%, dan 6% dari jumlah 1ml pengencer susu skim dengan masing-masing 7 pengulangan dan disimpan pada suhu kamar selama 6 jam, setiap 2 jam dilakuakan evaluasi kualitas semen cair yaitu motilitas, viabilitas dan abnormalitas spermatozoa.Hasil penelitian menunjukkan bahwa pengaruh level penambahan filtrat kecambah kacang hijau dalam pengencer susu skim pada penyimpanan suhu ruang tidak memberikan perbedaan yang signifikan (P> 0,05) padamotilitas, viabilitas, dan abnormalitas spermatozoa. Kata kunci: semen cair, susu skim, filtrat kecambah kacang hijau, penyimpanan suhu ruang

Abstract

This study is conducted on December 2016until January 2017 in Sumber Sekar Field Laboratory, Faculty of Animal Husbandry, Brawijaya University. The purpose of this study is to determine the effect of different maturity levels of green coconut water dilution with 10% of egg yolk to the sperm quality of Boer goat during storage. Materials used in the study were male Boer kept in Sumber Sekar Field Laboratory. The semen was collected routinely once in 2 days using artificial vagina with fresh semen quality requirement individual motility ≥70%. The method used in this study is Randomnized Block Design which consists of three treatments, there are P0 as control (CEP-2 + 10% Egg Yolk); P1 (Young Green Coconut Water + 10% Egg Yolk) and P2 (Mature Green Coconut Water + 10% Egg Yolk). If the difference between the treatments showed real influence, then the Duncan’s Multiple Range Test will be done. The grouping is sorted by the semen observation time. The observed variables macroscopically include volume, smell, consistency, color and acidity, and microscopically which include individual motility, mass motility, viability and abnormality.The study result for the mean and standard deviation (SD) of motility in storage until the 4th day. The motility of P0 (36.67 ± 32.96)%, then P1 (20.0 ± 13.78)%, and the last P3 (18.33 ± 19.15). Dilution with the best viabillity average in the 4th day in P0 (58.02 ± 19.93)%, in P1 (26.10 ± 18.47)% and the last is P2 (25,87 ± 20,60)%.the average value of abnormality on the 4th day shows that the lowest average abnormality found in P0 (1.49 ± 0.73)%, then P1 (1.89 ± 1.68)% and the highest average value of abnormality is in P2 (2,87 ± 1.41).Keywords : liquid semen, boer goat, green coconut water, CEP-2, cold storage
