Abstract

Adenostemma platyphyllum is a weed plant containing various secondary metabolites, such as phenolics and flavonoids, making it essential to cultivate properly. This study aims to determine the optimal cow manure dosage for plant growth, focusing on phenolic and flavonoid production in the cultivation of A. platyphyllum. The experiment employed a randomized complete block design (RCBD) with a single factor: cow manure doses of 0.5, 5, 10, 15, and 20 ton/Ha. Phenolic content was measured using the Folin Ciocalteu reagent, while flavonoid content was assessed using the AlCl₃ reagent. Results indicate that the highest phenolic and flavonoid production occurred in plants treated with 20 ton/Ha manure. This dosage also resulted in the best plant growth, yielding the highest crop, number of leaves, and branches compared to other treatments.

Abstract

Essential oils are often also referred to as flying oils produced by plants and usually volatile essential oils at room temperature, one of the essential oils with many uses is clove leaf essential oil (Syzygium aromaticum L). Zein-essential oil nanoparticles were prepared by varying the concentration of clove oil (0, 2, 4, 6, 8, and 20 mg/mL) added to 1.5 mg/mL zein and 1 ml tween 20 and sonicated for 20 minutes. The antioxidant activity of the zein-essential oil nanoparticles formed was tested using the DPPH method. The best antioxidant activity was found in the addition of clove leaf essential oil 8 mg/mL with a percentage of inhibition of 71.13%. Characterization test with a laser beam where the amount of light scattered by the solution. Chemical component analysis using Gas Chromatography Mass Spectrophotometry (GC-MS) showed the presence of 32 compounds with three main compounds, namely trans-?-caryophyllene (46.69%), chavibetol (25.80%) and - humulene (10.84%).

Abstract

Limbah tulang ayam meningkat seiring dengan peningkatan konsumsi ayam. Namun, belum banyak penelitian yang memanfaatkan limbah tulang sebagai sumber kolagen. Penelitian ini bertujuan mengetahui pengaruh ukuran kolagen terhadap aktivitas anti aging berupa aktivitas antioksidan, antiglikasi, dan inhibitor tirosinase  secara in vitro dan mendapatkan teknik isolasi kolagen anti aging optimum dari tulang ayam. Isolasi kolagen dilakukan dengan variasi  konsentrasi NaOH, yaitu 0,05 M; 0,10 M; dan 0,20 M, dilanjutkan dengan perendaman menggunakan asam asetat 1 M. Kolagen yang diisolasi dengan NaOH 0,10 M merupakan kolagen dengan ukuran partikel, rendemen, dan antiglikasi terbesar (berturut-turut 2,34 µm, 12,59%, 61,06%) dan memiliki spektrum inframerah yang paling sesuai dengan kolagen standar. Kolagen ini kemudian diaduk dengan kecepatan 1000 rpm selama 6 dan 8 jam untuk pengecilan ukuran. Kolagen dengan pengadukan 6 jam mempunyai ukuran partikel lebih kecil (1,34 µm) dibandingkan dengan pengadukan 8 jam (1,80 µm). Kolagen dengan ukuran 1,34 µm menunjukkan aktivitas terbaik yaitu aktivitas antioksidan terhadap 2,2-difenil-1-pikrilhidrazil (DPPH) sebesar 24,70% dan inhibitor tirosinase sebesar 26,77%. Berdasarkan aktivitas antioksidan, antiglikasi, dan antitirosinase, kolagen dengan perendaman NaOH 0,10 M dan pengadukan selama 6 jam memiliki sifat anti aging yang paling baik.In Vitro Anti-Aging Activity of Chicken (Gallus gallus domesticus) Bone Waste Collagen. Chicken bone waste increases with increasing chicken compsumtion. However, study on utilizing chicken bone for collagen source has not been widely explored. This study aims to determine the effect of collagen size on their anti aging activity, and to obtain the optimum condition to produce the chicken (Gallus gallus domesticus) collagen in the high yield and the best activity. Collagen isolation was carried out in various NaOH concentrations of 0.05 M, 0.10 M, and 0.20 M, followed by the maceration on acetic acid 1 M. The isolation in NaOH 0.10 M produced the collagen with particle size of 2.34 µm in yield of 12.59% and anti-glycation of 61.06%. The revealed infrared spectrum of the isolated collagen is almost the same with the spectrum of the standart collagen. The collagen in 2.34 µm was further stirred at a 1000 rpm for 6 and 8 hours to reduce the size. Collagen stirred in 6 hours has a smaller particle size (1.34 µm) compared with that of stirred in 8 hours which has a particle size of 1.80 µm. The collagen with size of 1.34 µm showed the best activity, which revealed the antioxidant activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH) of 24.70% and tyrosinase inhibitors of 26.77%. Based on antioxidant activity, anti-glycation, and anti-tyrosinase, the collagen which was isolated in 0.10 M NaOH and was stirred in 6 hours has the best anti-aging property.

Abstract

The Asteraceae family has beneficial medicinal effect as anticancer, such as Chromolaena odorata and Mikaina micrantha. This study we investigated the potency of two Asteraceace plants and determined their active compounds. The plants were extracted using distilled water and then fractionated with chloroform. The phytochemical content, and anticancer potency was determined by literature study. The results showed that both plants contained essential oils, flavonoids, phenolics, terpenoids, and steroids. Toxicity to shrimp larvae through the Brine Shrimp Lethality Test (BLST) showed that both plants were toxic , so they were thought to be anticancer. Ombuin and vanylic acid compounds in C. odorata are reported its bioactive compound has activity anticancer. M. micrantha has less potential as an anticancer and more activity as an anti-inflammatory.

Abstract

Cardamom (Amomum cardamomum), the ‘Queen of spices’, is a native Indonesian spice, a type of potential biopharmaceutical currently prospective because of its high selling value, especially for its fruit, with various benefits and its wide use. So far, cardamom essential oil comes from the utilization of its fruit, but the production is more limited, unlike the sustainably produced leaves, therefore, its leaves have the potential to be developed. This study aims to analyze the essential oil yield and 1.8-cineol content of cardamom leaves essential oil (CLEO) grown in agroforestry systems. The CLEO was obtained by steam-water distillation, while the 1.8-cineol content was analyzed by gas chromatography–mass spectrometry (GC-MS). The experimental design employed was a Randomized Complete Block Design with three cropping patterns as a treatment and three doses of bokashi fertilizer as blocks. The results showed that the highest yield of CLEO was generated in the sengon + cardamom agroforestry pattern of 3.16%, and the highest content of 1,8-cineol in CLEO was generated in the sengon + cardamom + arrowroot agroforestry pattern of 47.23%, while the lowest yield of CLEO and the 1,8-cineol content were found in the monoculture pattern of 2,02% and 43.16% respectively. Agroforestry practices have the potential to increase 1,8-cineol content and the yield of essential oil in cardamom leaf, which will be prospective in forest management to support forestry multi-business and social forestry programs.

Abstract

Breast cancer is a leading cause of cancer-related deaths in Indonesia. However, the drugs that are commonly used for treatment can cause side effects and become resistant over time. A study was conducted to test the cytotoxic activity of broadleaf mahogany (Swietenia macrophylla) seed extract on MCF-7 breast cancer cells in vitro. The study aimed to predict active compounds in the broadleaf mahogany seeds that have the potential to act as anti-breast cancer agents using in silico analysis. Molecular docking, visualization of the interaction between the receptor and the ligands, and physicochemical analysis were used to determine the most promising compounds. The receptors used were fibroblast growth factor receptor 1 (FGFR1), vascular endothelial growth factor receptor 2 (VEGFR2), insulin-like growth factor type 1 receptor (IGF-1R), estrogen receptor (ER-α), and progesterone receptor (PR). The results showed that 12 compounds have the potential to be active as anti-breast cancer agents. Three of these compounds, 3β,6-dihydroxydihydrocarapine, stigmasterol, and 7-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-chroman-4-one, were predicted to have similar mechanisms of inhibition as a comparator drug based on binding site similarity values. These compounds are predicted to be taken orally and are promising for further research.

Abstract

Distillation time can affect the extraction of essential oils such as the rhizome of temu hitam (Curcuma aeruginosa) which has been shown to have antibacterial activity. Therefore, this study aims to determine the optimum distillation time to produce temu hitam rhizome oil with the strongest antibacterial activity. The rhizomes of temu hitam were steam distilled with different length of time every 30 minutes for 4 hours to obtain essential oil and determined the compounds contained in them using gas chromatography-mass spectrometry. Antibacterial activity was determined by disc diffusion against Streptococcus mutans, Escherichia coli, and Staphylococcus aureus. The most dominant compounds present in essential oils are eucalyptol (34.89%), epicurzerenone (20.58%), and camphor (16.90%) based on the average peak of the total distillation time. The strongest antibacterial potency was obtained in the first 30 minutes of distillation for S. mutans and E. coli, and in the seventh 30 minutes of distillation for S. aureus.

Abstract

Fungus comb is a structure made by termite colony Macrotermitinae with lots of potential because it contains secondary metabolites such as steroids, terpenoids, and saponins and compounds containing antioxidant and anti-tyrosinase properties. This study aimed to determine fungus comb extracts' fingerprint pattern, antioxidant capacity, and anti-tyrosinase activity. Thin layer chromatography (TLC) was used to determine fingerprint patterns. The resulting TLC pattern was processed with ImageJ and converted into a densitogram. The fungus comb was extracted with n-hexane and ethyl acetate. TLC results showed that fungus comb extracts contain steroids and terpenoids and have antioxidant properties. The antioxidant capacity assay was carried out with 2,2-diphenyl-1-picrylhydrazyl reagent. N-hexane extract had an antioxidant capacity value of 9.07 mg ascorbic acid equivalent (AAE)/g extract, while ethyl acetate extract had 6.04 mg AAE/g extract. Anti-tyrosinase activity assay was carried out with L-tyrosine and L-3,4-dihydroxyphenylalanine (L-DOPA) as substrates. N-hexane extract had a better anti-tyrosinase activity with IC50 of 1804 µg/mL for L-tyrosine and 6784 µg/mL for L-DOPA.

Abstract

Some Asteraceae plants are reported to contain phytochemicals and to show high antibacterial activity. This study aims to determine the highest antibacterial activity of Wedelia trilobata, Acmella paniculata, Ageratum conyzoides, and Crassocephalum crepidiodes and to predict the chemical compounds that have a role as antibacterial. The samples were extracted with water and further fractionated using chloroform in the laboratory. Extracts determined the phytochemical compounds, antibacterial activity diffusion method, and compounds that have a role as antibacterial through literature review. Acmella paniculata demonstrated high antibacterial activities with the highest diameter of the zones of inhibition with a concentration of just 0.5 mg/disc. The compound that seems to have a role in antibacterial activity is spilanthol.

Abstract

Stroke due to cerebral ischemia is one of the leading causes of disability worldwide. Animal models of cerebral ischemia that is often used is Global Cerebral Ischemia (GCI). In GCI, the hippocampus is the most susceptible to neuronal cell death. Complications that occur after ischemia are due to increased oxidative stress. Some compounds in purwoceng are reported to have antioxidant activity which oxidative stress have not been studied in the GCI model stroke. This study aims to examine the effect of purwoceng extract on a stroke model (GCI) as a neuroprotective agent in the prevention of stroke complications that have not been previously studied. 25 rats with a stroke model were given purwoceng root extract with 3 doses (20, 30, and 40mg/kg BW) orally for three days. The mice were then tested for memory with the Morriz Water Maze (MWM) test; then histopathological analysis of pyramidal cells in the hippocampus and the expression of SOD1 and SOD2 genes was analyzed using RT-PCR. The MWM test showed that the memory results at the dose of 20mg/kg BW were better than that of the GCI group (p=0.0384), and the PCR of SOD2 showed improvement at the dose of 20mg/kg BW (p=0.0171). No significant difference in histopathological analysis and SOD1 mRNA expression across group. The administration of purwoceng root extract at a dose of 20 mg/kg BW had the effect of improving memory and SOD2 expression in GCI model rats
