Abstract

ABSTRACT. The Lamiaceae family which are widely used in traditional medicine in Indonesia. Plants from this family are known to have antioxidant bioactivity. Therefore, in this study, an antioxidant test was carried out first as an initial screening. This study evaluated the antioxidant capacity of four Lamiaceae species: Ocimum americanum L., Ocimum basilicum L., Leucas lavandulifolia Sm., and Perilla frutescens (L.) Britton. The four Lamiaceae species extracts were evaluated for their antioxidant capacity using 2,2-diphenyl-1-picrylhydrazyl (DPPH), cupric-reducing antioxidant capacity (CUPRAC), and ferric-reducing antioxidant power (FRAP). The highest activity extract was fractionated with n-hexane, ethyl acetate, and ethanol-water, and the results were tested for antioxidant capacity. The most active extract and most active fraction were analyzed for metabolites using liquid chromatography-mass spectrometry (LC-MS/MS). The results show that the ethanol 50% leaves extracts of O. americanum L. have the highest antioxidant capacity of about 0.439 (DPPH); 1.517 (CUPRAC); and 1.021 (FRAP) mmol ascorbic acid equivalent (AAE)/g extract. The highest antioxidant capacity from the partition results was possessed by the ethyl acetate fraction with a value of 1.109 (DPPH); 1.540 (CUPRAC); and 1.551 (FRAP) mmol AAE/g fraction. Metabolite analysis using LC-MS/MS succeeded in identifying 18 metabolites consisting of flavonoids, terpenoids, amino acids, phenolic acids, fatty acids, and other carboxylic acids. Abstract

Strobilanthes crispa known as keji beling in Indonesia, belong to the Acanthaceae family. Thin layer chromatography (TLC) fingerprint pattern analysis is an excellent technique for evaluating and controlling the quality of raw materials containing Strobilanthes crispa. TLC fingerprints can be used to analyze the fingerprint profiles of medicinal plants for identification, authentication, and discrimination from related plants. This study aimed to develop a TLC fingerprint analysis method to identify S. crispa for quality control. Eleven bands were effectively separated using a silica gel 60 F254 TLC plate with a mobile phase of dichloromethane, ethyl acetate, and chloroform at 16:3.2:0.8. The derivatization reagent used to detect the separated bands was 10% sulfuric acid at a wavelength of 366 nm. Validation of the TLC fingerprint analysis, which evaluated the robustness, stability, specificity, and precision, met these requirements. S. crispa can be distinguished by its TLC fingerprint from sirih hutan (Piper aduncum), a related plant with similar leaf morphology. This approach could be used to identify and authenticate of S. crispa from P. aduncum.

Abstract

Identifying floral resources used by honey bees and stingless bees is essential for sustainable beekeeping and understanding pollination ecology. This study compared the diversity of floral resources utilized by the honey bee Apis cerana and stingless bees Geniotrigona thoracica and Heterotrigona itama based on pollen in honey. Bee colonies were in the same Flora Nauli Beekeeping area, Pematang Siantar, North Sumatra, Indonesia, ensuring equal access to similar surrounding floral resources. Honey samples were collected, and pollen grains were extracted, acetolysed, and analyzed by counting 1,200 grains to determine frequency classes. Pollen diversity was calculated using the Shannon–Wiener index (H′). Stingless bee honey contained more diverse pollen than A. cerana, which only contained pollen from the Arecaceae family. Heterotrigona itama and G. thoracica honey contained pollen from 20 and 11 plant families, respectively. Each species had a distinct predominant pollen type (>45%): Cocos nucifera (A. cerana), Sapotaceae type (G. thoracica), and Casuarinaceae type (H. itama). A higher pollen diversity index was also observed in H. itama (1.05-1.83) than in G. thoracica (0.34–1.64) and A. cerana (0.66). These results indicate that stingless bees are more generalist than honey bees, highlighting their ecological role in supporting pollination networks in tropical agroecosystems.

Abstract

The increasing demand for faster bioinformatics analysis calls for more efficient approaches for sequence alignment. In this study, we demonstrate that a GPU-based implementation of the Needleman-Wunsch algorithm can achieve up to 14.8× speedup compared to its traditional CPU-based serial counterpart, without compromising alignment accuracy. By leveraging the parallel processing capabilities and shared memory of an NVIDIA GeForce RTX 3060 Laptop GPU, we significantly accelerated global sequence alignment tasks. Using clinically relevant genes such as NRAS, BRCA1, BRCA2, and Saccharomyces cerevisiae from NCBI ensures realistic alignment challenges and biological significance. Performance evaluation across a wide range of sequence lengths demonstrates the scalability and efficiency of the parallel approach. More importantly, this study provides a unique contribution by showing that a commodity GPU, such as the NVIDIA GeForce RTX 3060 Laptop, can serve as a practical alternative when high-performance computing clusters are unavailable or prohibitively expensive, thereby offering an accessible and cost-effective pathway to high-throughput bioinformatics workflows.

Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37 C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory

Abstract

Cajuput is one of plants containing essential oil with cineole as a major component. This aim of study is to separate cineole in cajuput oil and to analyze its potency as slimming aromatherapy through in vivo assay. The essential oil was fractionated by column chromatography resulting 23 fraction (F1-F23). Cajuput oil, cineole, and F9 were analyzed by gas chromatograph-mass spectrometer, and the slimming aromatherapy potency was studied on white adult male Sprague-Dawley rats. Inhalation result of cineole showed that the average body weight of rats after 5 weeks treatment period was lower than that of the normal and the control groups which consumed high cholesterol feed. In conclusion, cineole is a compound that is potential in slimming aromatherapy.

Abstract

Temulawak (Curcuma xanthorrhiza Roxb. Synm. Curcuma javanica) merupakan tanaman obat asli Indonesia yang memiliki khasiat multifungsi. Namun jangka waktu panen temulawak yang cukup lama (sekitar 9-12 bulan) menyebabkan para petani enggan menanam tanaman temulawak. Untuk itu pengembangan metode penanaman perlu dilakukan agar menguntungkan secara ekonomis bagi petani, salah satu metode yang dapat digunakan adalah tumpang sari. Tumpang sari adalah penanaman lebih dari satu tanaman pada waktu yang bersamaan atau selama periode tanam pada satu tempat yang sama. Beberapa keuntungan dari metode tumpang sari antara lain pemanfaatan lahan kosong disela-sela tanaman pokok, penggunaan cahaya, air serta unsur hara yang lebih efektif, mengurangi resiko kegagalan panen, dan menekan pertumbuhan gulma. Penelitian temulawak dilakukan dengan menggunakan rancangan petak-petak terbagi (split plot design). Rancangan acak kelompok (RAK) dibagi menjadi 3 kelompok berdasarkan variabel jarak tanam, jumlah tunas, dan waktu panen. Parameter agronomi yang diamati adalah bobot rimpang basah dan kering yang mengindikasikan produktivitas per tanaman dan jumlah per bagian rimpang. Kandungan bioaktif xanthorrhizol dan kurkuminoid pada temulawak ditentukan dengan metode Kromatografi Cair Kinerja Tinggi (KCKT). Hasil pengukuran kurkuminoid dan xanthorrhizol pada temulawak menunjukkan bahwa tidak ada perbedaan yang signifikan pada kadar kedua metabolit sekunder dari penanaman dengan dan tanpa tumpang sari. Dapat disimpulkan bahwa metode penanaman tumpang sari dapat tidak mempengaruhi kadar metabolit sekunder dari temulawak sebagai tanaman utama, sehingga dapat menjadi pilihan metode pertanian untuk meningkatkan manfaat ekonomi para petani temulawak.

Abstract

Jamu terdiri atas berbagai macam tumbuhan obat yang diolah. Tumbuhan obat memiliki kandungan senyawa metabolit sekunder yang berperan sebagai khasiat. Pengetahuan tumbuhan obat beserta senyawanya perlu dikembangkan lagi menjadi pengetahuan eksplisit yang lebih spesifik dan mudah dimengerti sehingga berguna untuk masyarakat. Maka dari itu perlu mengembangkan sistem untuk digunakan oleh masyarakat sehingga pengobatan dengan tanaman obat lebih populer di kalangan masyarakat luas. Penelitian yang dilakukan mengenai manajemen pengetahuan berbasis ontologi terkait dengan tumbuhan obat dan senyawa. Penelitian ini menggunakan pendekatan proses manajemen pengetahuan model SECI, selain itu pengembangan ontologi menggunakan metode Ontology building life cycle, sedangkan bahasa representasi yang digunakan adalah Resource Description Framework (RDF) dan Web Ontology Language (OWL) dengan toolsProtégé5.0. Pengembangan model ontologi dengan bahasa representasi RDF/OWL dapat menghasilkan pengetahuan dengan melakukan query menggunakan SPARQL. Hasil query tersebut dapat digunakan untuk diimplementasikan pada mobile.

Abstract

Tumbuhan Asteraceae dilaporkan mengandung metabolit sekunder tinggi yang aktif sebagai antioksidan. Senyawa antioksidan juga digunakan untuk mencegah penuaan/reaksi glikasi. Penelitian ini bertujuan menentukan aktivitas antioksidan dan antiglikasi 7 tumbuhan Asteraceae. Daun Adenostema lavenia, Ageratum conyzoides, Dichrocephala integrifolia, Galinsoga parvifolia, Synedrella nodiflora, Mikania micrantha, dan Sphagneticola trilobata masing-masing diekstraksi menggunakan air. Ekstrak kemudian ditentukan kandungan total fenol, flavonoid, dan aktivitas antioksidan menggunakan metode 1,1-difenil-2-pikrilhidrazil (DPPH) dan 2,2’-azino-bis(3 ethylbenzothiazoline-6-sulfonic acid (ABTS), serta aktivitas antiglikasi. Ekstrak dengan kadar total fenolik dan flavonoid tertinggi adalah ekstrak M. micrantha. Ekstrak A. lavenia memiliki aktivitas antioksidan terhadap DPPH tertinggi dengan nilai IC50 sebesar 121,82 μg/mL dan A. conyzoides tertinggi terhadap ABTS dengan kapasitas sebesar 4,51 mg trolox ekivalen (TE)/g ekstrak. Aktivitas antiglikasi tertinggi pada ekstrak G. parvifolia sebesar 90,67% yang tidak berbeda nyata dengan ekstrak A. lavenia (87,87%) dan S. trilobata (87,38%). Secara keseluruhan ekstrak A. lavenia dan S. trilobata baik dikembangkan sebagai antioksidan dan antiglikasi.

Abstract

Sukoharjo is one of the districts in Central Java that has a large number of herbal medicine entrepreneurs, including many small and medium industries. This small and medium industry needs to improve the quality of product safety. Therefore, this study aims to obtain the profile of small and medium industry herbal medicine products in Sukoharjo Regency. A total of 6 types of products (candy (1), mixture (3), extract (1), and tea (1)) were collected and the uniform weights, water content, organoleptic, hazardous metal content and microbiological safety were determined. The results showed that the product had fulfilled the water content, lead, cadmium, and arsenic content, and fulfilled the requirements of total plate count (TPC), coliform, and yeast mold. Product quality still needs to be improved in terms of uniformity of weight and packaging labels. The conclusion of this research is the profile of small and medium industry products in Sukoharjo Regency has met safety requirements but needs to be improved in quality.
