Abstract

Cajuput is one of plants containing essential oil with cineole as a major component. This aim of study is to separate cineole in cajuput oil and to analyze its potency as slimming aromatherapy through in vivo assay. The essential oil was fractionated by column chromatography resulting 23 fraction (F1-F23). Cajuput oil, cineole, and F9 were analyzed by gas chromatograph-mass spectrometer, and the slimming aromatherapy potency was studied on white adult male Sprague-Dawley rats. Inhalation result of cineole showed that the average body weight of rats after 5 weeks treatment period was lower than that of the normal and the control groups which consumed high cholesterol feed. In conclusion, cineole is a compound that is potential in slimming aromatherapy.

Abstract

Temulawak (Curcuma xanthorrhiza Roxb. Synm. Curcuma javanica) merupakan tanaman obat asli Indonesia yang memiliki khasiat multifungsi. Namun jangka waktu panen temulawak yang cukup lama (sekitar 9-12 bulan) menyebabkan para petani enggan menanam tanaman temulawak. Untuk itu pengembangan metode penanaman perlu dilakukan agar menguntungkan secara ekonomis bagi petani, salah satu metode yang dapat digunakan adalah tumpang sari. Tumpang sari adalah penanaman lebih dari satu tanaman pada waktu yang bersamaan atau selama periode tanam pada satu tempat yang sama. Beberapa keuntungan dari metode tumpang sari antara lain pemanfaatan lahan kosong disela-sela tanaman pokok, penggunaan cahaya, air serta unsur hara yang lebih efektif, mengurangi resiko kegagalan panen, dan menekan pertumbuhan gulma. Penelitian temulawak dilakukan dengan menggunakan rancangan petak-petak terbagi (split plot design). Rancangan acak kelompok (RAK) dibagi menjadi 3 kelompok berdasarkan variabel jarak tanam, jumlah tunas, dan waktu panen. Parameter agronomi yang diamati adalah bobot rimpang basah dan kering yang mengindikasikan produktivitas per tanaman dan jumlah per bagian rimpang. Kandungan bioaktif xanthorrhizol dan kurkuminoid pada temulawak ditentukan dengan metode Kromatografi Cair Kinerja Tinggi (KCKT). Hasil pengukuran kurkuminoid dan xanthorrhizol pada temulawak menunjukkan bahwa tidak ada perbedaan yang signifikan pada kadar kedua metabolit sekunder dari penanaman dengan dan tanpa tumpang sari. Dapat disimpulkan bahwa metode penanaman tumpang sari dapat tidak mempengaruhi kadar metabolit sekunder dari temulawak sebagai tanaman utama, sehingga dapat menjadi pilihan metode pertanian untuk meningkatkan manfaat ekonomi para petani temulawak.

Abstract

Jamu terdiri atas berbagai macam tumbuhan obat yang diolah. Tumbuhan obat memiliki kandungan senyawa metabolit sekunder yang berperan sebagai khasiat. Pengetahuan tumbuhan obat beserta senyawanya perlu dikembangkan lagi menjadi pengetahuan eksplisit yang lebih spesifik dan mudah dimengerti sehingga berguna untuk masyarakat. Maka dari itu perlu mengembangkan sistem untuk digunakan oleh masyarakat sehingga pengobatan dengan tanaman obat lebih populer di kalangan masyarakat luas. Penelitian yang dilakukan mengenai manajemen pengetahuan berbasis ontologi terkait dengan tumbuhan obat dan senyawa. Penelitian ini menggunakan pendekatan proses manajemen pengetahuan model SECI, selain itu pengembangan ontologi menggunakan metode Ontology building life cycle, sedangkan bahasa representasi yang digunakan adalah Resource Description Framework (RDF) dan Web Ontology Language (OWL) dengan toolsProtégé5.0. Pengembangan model ontologi dengan bahasa representasi RDF/OWL dapat menghasilkan pengetahuan dengan melakukan query menggunakan SPARQL. Hasil query tersebut dapat digunakan untuk diimplementasikan pada mobile.

Abstract

Tumbuhan Asteraceae dilaporkan mengandung metabolit sekunder tinggi yang aktif sebagai antioksidan. Senyawa antioksidan juga digunakan untuk mencegah penuaan/reaksi glikasi. Penelitian ini bertujuan menentukan aktivitas antioksidan dan antiglikasi 7 tumbuhan Asteraceae. Daun Adenostema lavenia, Ageratum conyzoides, Dichrocephala integrifolia, Galinsoga parvifolia, Synedrella nodiflora, Mikania micrantha, dan Sphagneticola trilobata masing-masing diekstraksi menggunakan air. Ekstrak kemudian ditentukan kandungan total fenol, flavonoid, dan aktivitas antioksidan menggunakan metode 1,1-difenil-2-pikrilhidrazil (DPPH) dan 2,2’-azino-bis(3 ethylbenzothiazoline-6-sulfonic acid (ABTS), serta aktivitas antiglikasi. Ekstrak dengan kadar total fenolik dan flavonoid tertinggi adalah ekstrak M. micrantha. Ekstrak A. lavenia memiliki aktivitas antioksidan terhadap DPPH tertinggi dengan nilai IC50 sebesar 121,82 μg/mL dan A. conyzoides tertinggi terhadap ABTS dengan kapasitas sebesar 4,51 mg trolox ekivalen (TE)/g ekstrak. Aktivitas antiglikasi tertinggi pada ekstrak G. parvifolia sebesar 90,67% yang tidak berbeda nyata dengan ekstrak A. lavenia (87,87%) dan S. trilobata (87,38%). Secara keseluruhan ekstrak A. lavenia dan S. trilobata baik dikembangkan sebagai antioksidan dan antiglikasi.

Abstract

Sukoharjo is one of the districts in Central Java that has a large number of herbal medicine entrepreneurs, including many small and medium industries. This small and medium industry needs to improve the quality of product safety. Therefore, this study aims to obtain the profile of small and medium industry herbal medicine products in Sukoharjo Regency. A total of 6 types of products (candy (1), mixture (3), extract (1), and tea (1)) were collected and the uniform weights, water content, organoleptic, hazardous metal content and microbiological safety were determined. The results showed that the product had fulfilled the water content, lead, cadmium, and arsenic content, and fulfilled the requirements of total plate count (TPC), coliform, and yeast mold. Product quality still needs to be improved in terms of uniformity of weight and packaging labels. The conclusion of this research is the profile of small and medium industry products in Sukoharjo Regency has met safety requirements but needs to be improved in quality.

Abstract

This research aimed to determine the active fraction and profile of active compounds in Manonjaya sSnakefruit skin extract that can be used as antidiabetic and the antidiabetic activity using zebra fish. Manonjaya sSnakefruit skin was extracted by maceration using 70% ethanol and then fractionated using n-hexane, ethyl acetate and methanol. The ethyl acetate fraction showed the highest yield and total phenolic content as well as the strongest α-glucosidase inhibition with an inhibition percentage of 64.43%. The ethyl acetate fraction provided the most active spot as an α-glucosidase inhibitor. The profile of active compounds that act as antidiabetic compound is flavonoid from glycosylflavone and flavone groups. Antidiabetic activity using zebra fish was indicates that Manonjaya Snakefruit skin extract from ethanol extract and ethyl acetate fraction can decrease blood sugar level of zebra fish after induced with alloxan. The crude extract group had 54.51% lower sugar content than the induction group and 41.70% lower than the negative control. Meanwhile, the ethyl acetate fraction group had 65.10% lower sugar content than the induction group and 55.27% lower than the negative control.

Abstract

Some Asteraceae plants have phytochemicals that are commonly used to treat some illness. However, research on the use of Asteraceae plants as tyrosinase inhibitor agents is still rare. Therefore, this study aims to select the most active fraction of Asteraceae species as tyrosinase inhibitors and suspect active compounds that play a role in it. About 12 species from Asteraceae are extracted using water. The resulting filtrate is partially fractionated using chloroform to obtain water extracts, water fractions, and chloroform fractions. Each extract and fraction of each plant determined the activity of its tyrosinase inhibitor using a microplate reader (ELISA) with L-DOPA as the substrate. Thitonia diversipholia water fraction provided the best activity of all the samples tested with the smallest IC50. The active compounds contained are analyzed using LC-MS / MS.  Compounds that act as tyrosinase inhibitors are alkaloid group that dominate the plant, namely trigonelline.

Abstract

Breast cancer is a leading cause of cancer-related deaths in Indonesia. However, the drugs that are commonly used for treatment can cause side effects and become resistant over time. A study was conducted to test the cytotoxic activity of broadleaf mahogany (Swietenia macrophylla) seed extract on MCF-7 breast cancer cells in vitro. The study aimed to predict active compounds in the broadleaf mahogany seeds that have the potential to act as anti-breast cancer agents using in silico analysis. Molecular docking, visualization of the interaction between the receptor and the ligands, and physicochemical analysis were used to determine the most promising compounds. The receptors used were fibroblast growth factor receptor 1 (FGFR1), vascular endothelial growth factor receptor 2 (VEGFR2), insulin-like growth factor type 1 receptor (IGF-1R), estrogen receptor (ER-α), and progesterone receptor (PR). The results showed that 12 compounds have the potential to be active as anti-breast cancer agents. Three of these compounds, 3β,6-dihydroxydihydrocarapine, stigmasterol, and 7-hydroxy-2-(4-hydroxy-3-methoxyphenyl)-chroman-4-one, were predicted to have similar mechanisms of inhibition as a comparator drug based on binding site similarity values. These compounds are predicted to be taken orally and are promising for further research.

Abstract

Adenostemma platyphyllum is a weed plant containing various secondary metabolites, such as phenolics and flavonoids, making it essential to cultivate properly. This study aims to determine the optimal cow manure dosage for plant growth, focusing on phenolic and flavonoid production in the cultivation of A. platyphyllum. The experiment employed a randomized complete block design (RCBD) with a single factor: cow manure doses of 0.5, 5, 10, 15, and 20 ton/Ha. Phenolic content was measured using the Folin Ciocalteu reagent, while flavonoid content was assessed using the AlCl₃ reagent. Results indicate that the highest phenolic and flavonoid production occurred in plants treated with 20 ton/Ha manure. This dosage also resulted in the best plant growth, yielding the highest crop, number of leaves, and branches compared to other treatments.

Abstract

Distillation time can affect the extraction of essential oils such as the rhizome of temu hitam (Curcuma aeruginosa) which has been shown to have antibacterial activity. Therefore, this study aims to determine the optimum distillation time to produce temu hitam rhizome oil with the strongest antibacterial activity. The rhizomes of temu hitam were steam distilled with different length of time every 30 minutes for 4 hours to obtain essential oil and determined the compounds contained in them using gas chromatography-mass spectrometry. Antibacterial activity was determined by disc diffusion against Streptococcus mutans, Escherichia coli, and Staphylococcus aureus. The most dominant compounds present in essential oils are eucalyptol (34.89%), epicurzerenone (20.58%), and camphor (16.90%) based on the average peak of the total distillation time. The strongest antibacterial potency was obtained in the first 30 minutes of distillation for S. mutans and E. coli, and in the seventh 30 minutes of distillation for S. aureus.
