Abstract

Sansevieria trifasciata (also called snake plant or mother in law’s tongue) is predicted to act as a heavy metal biosorbent. S. trifasciata was optimized as a biosorbent by using the response surface method with varying weights of S. trifasciata, pH of Pb (II) solutions, contact times, and temperatures. The ion concentration before and after biosorption was measured with an atomic absorption spectrophotometer (AAS). The optimum biosorption conditions were pH 7, 240 min contact time, and 1.5 g biosorbent with biosorption capacity of Pb (II) ions 0.725 mg/g. The biosorption isotherm analysis showed that the biosorption is consistent with the Freundlich isotherm model. The peak intensity of the FTIR spectrum of S. trifasciata after treatment with Pb (II) was around 2130/cm. The S. trifasciata micrograph showed a porous structure with non-uniform pore sizes. The biosorption of Pb (II) ions on powdered S. trifasciata leaves was found with the SEM EDX analysis. It is concluded from this research that S. trifasciata can be used as a biosorbent to remove Pb ion contamination.

Abstract

Melanin is a crucial amino acid in determining human skin and hair pigmentation. Excessive melanin production can lead to hyperpigmentation and darkening of the skin. This study aims to assess the capability of Adenostemma platyphyllum as a tyrosinase enzyme inhibitor. It predicts its anti-melanogenic activity through molecular docking with proteins involved in the melanogenesis process. The in-vitro approach was conducted by determining the tyrosinase enzyme inhibition capacity, while the in-silico approach involved ligand binding to target proteins from melanogenesis pathways. The highest tyrosinase inhibition capacity was observed in the ethanol extract, with values of 9.74 Kojic Acid Equivalent (KAE)/g extract (L-tyrosine) and 17.91 (KAE)/g extract (L-DOPA). Molecular docking analysis showed that the binding of eriodictyol 7-O-sophoroside (ΔG = -9.7 kcal/mol) has the best energy affinity for the PKC-β protein, genistein (ΔG = -7.5 kcal/mol) for the tyrosinase-related protein-1 (TYRP1) protein, eriodictyol 7-O-sophoroside (ΔG = -10.2 kcal/mol) for the cGMP protein, vincosamide (ΔG = -7.2 kcal/mol) for the microphthalmia-associated transcription factor (MITF) protein, and dicaffeoylquinic acid (ΔG = -7.4 kcal/mol) for the β-catenin protein. Based on a comparison of in-vitro and in-silico studies, melanogenesis inhibition is more potent in the PKC-β and cGMP pathways than direct tyrosinase inhibition because they exhibit lower binding energy.

Abstract

Ubi jalar ungu (Ipomoea batatas var Ayumurasaki) dilaporkan dapat membantu diet harian bagi penderita diabetes melitus tipe 2 karena memiliki kadar glikemik yang rendah. Salah satu mekanisme menurunkan kadar gula darah pada penderita diabetes adalah dengan menghambat enzim α-glukosidase. Potensi fraksi alkaloid dari daun dan fraksi antosianin dari umbi ubi jalar ungu dievaluasi sebagai inhibitor enzim α-glukosidase. Ubi jalar ungu yang digunakan dari Ciampea, Bogor, Jawa Barat. Fraksi alkaloid daun ubi jalar ungu pada konsentrasi 2,00% aktif menghambat kerja enzim sebesar 61,88%, sedangkan fraksi antosianin umbi ubi jalar ungu tidak aktif (0,25%). Fraksi alkaloid daun ubi jalar ungu dipisahkan lebih lanjut menggunakan kromatografi kolom dengan fase diam silika gel dan kloroformsebagai eluen. Fraksi yang mengandung alkaloid dengan semprotan Dragendorf, yakni F10, kemudian dipisahkan kembali dengan kromatografi lapis tipis preparatif. Fraksi F10.15 positif mengandung alkaloid dan dapat menghambat enzim -glukosidase sebesar 18,07% pada konsentrasi 1,25%, yang berarti pemisahan menyebabkan penurunan aktivitas. Spektrum inframerah transformasi Fourier fraksi memperlihatkan gugus fungsi –NH−, –OH, dan –CH2– dan diduga mengandung alkaloid.

Abstract

Kelompok Wanita Tani (KWT) di Kecamatan Nagrak, Sukabumi memiliki kegiatan bertani komoditas tanaman obat termasuk jahe. Jahe hasil budidaya anggota KWT sejauh ini dijual sebagai jahe segar, simplisia dan minuman kesehatan. Harga jual jahe segar dan simplisia yang cenderung fluktuatif menyebabkan pendapatan petani tidak menentu. Diperlukan dukungan inovasi pengolahan jahe untuk mengoptimalkan pemanfaatan jahe dan meningkatkan pendapatan petani. Tujuan kegiatan pengabdian kepada masyarakat ini adalah memberikan edukasi pemanfaatan jahe tanpa limbah (zero waste) kepada anggota KWT di Kecamatan nagrak, Sukabumi, Jawa Barat. Metode kegiatan meliputi pengukuran pengetahuan anggota KWT mengenai pengolahan jahe melalui pengisian kuesioner, pelatihan pengolahan jahe dengan konsep zero waste untuk menghasilkan beberapa produk turunan, dan evaluasi efektivitas pelatihan melalui pengisian post-test dan lomba pembuatan produk olahan jahe.  Kegiatan ini berlangsung pada bulan Juli hingga Oktober 2022 dan diikuti oleh 29 orang anggota KWT yang berasal dari enam KWT di wilayah Kecamatan Nagrak, Kabupaten Sukabumi. Dampak dari kegiatan ini adalah meningkatnya pengetahuan anggota KWT tentang inovasi pengolahan jahe dengan konsep zero waste. Jumlah peserta yang memiliki pengetahuan mengenai pengolahan jahe dengan konsep zero waste meningkat sebanyak 93,22%. Sementara itu, jumlah peserta yang memiliki pengetahuan mengenai diversifikasi produk olahan jahe meningkat sebanyak 62,76%. Sebagai kesimpulan dari kegiatan ini, kemampuan anggota KWT dalam mengolah jahe menjadi produk yang lebih variatif telah mengalami peningkatan, ditunjukkan dengan kemampuan setiap kelompok menyajikan produk olahan jahe saat kegiatan lomba. Meningkatnya pengetahuan dan keterampilan para anggota KWT dalam pengolahan jahe diharapkan dapat menambah peluang peningkatan pendapatan anggota KWT. The group of female farmers (KWT) in Nagrak District, Sukabumi cultivates medicinal plant commodities, including ginger. Up to now, the KWT members sold ginger as fresh ginger, simplicia, and health drink. Since the price of fresh ginger and simplicia tends to fluctuate, the farmers’ income is also varied. A ginger processing innovation is required to optimize ginger utilization and increase farmers' income. The purpose of this community service activity was to provide education of zero waste ginger utilization and processing for KWT members in Nagrak District, Sukabumi, West Java. The methods included measuring the knowledge of KWT members about ginger processing through questionnaires, training in ginger processing to produce several derivative products by applying zero waste concept and evaluating the effectiveness of the training. The activity was attended by 29 KWT members from six KWTs in the Nagrak District, Sukabumi. The impact of this activity was increasing the knowledge of KWT members about zero waste concept in ginger processing. The number of participants who were aware of the zero waste concept for ginger processing was increased by 93.22%. Meanwhile, the number of participants who possess knowledge about the diversification of ginger products increased by 62.76%. As a conclusion from this activity, the ability of KWT members to process ginger into a more varied product has increased, shown by the ability of each group to present processed ginger products during competition activities. The increasing of knowledge and skills in ginger processing are expected to increase the income of KWT members.

Abstract

Adenostemma platyphyllum is considered a weed with various benefits but has not been widely cultivated. This work aims to obtain the optimum concentration of a nutrient solution (AB-mix) to produce optimum phenolic and flavonoid levels, antioxidant capacity and metabolite profiling in A. platyphyllum using a hydroponic wick system. Different AB-mix nutrient solution concentrations were employed in a one-factor randomized block design. The highest total phenolic, flavonoid concentrations and antioxidant capacity were found in plants without additional nutrient solutions. However, the highest phenolic and flavonoid productivity was found in plants with a nutrient solution concentration of 1100 mg/L. The phenolic/flavonoid group compounds were successfully identified by separating the components using thin-layer chromatography. The intensity of the bands produced from each treatment was quite varied. The untreated plant produced thicker phenolic/flavonoid bands than the other treatments. This was supported by the heatmap pattern produced by the untreated ones, which had high color intensity. Therefore, the optimum concentration of nutrient solution to produce optimum phenolic, flavonoid levels, and antioxidant capacity in A. platyphyllum cultivation by hydroponic wick system was the concentration of 1100 mg/L.

Abstract

Glyphosate, which has been documented as a contaminant in tea, promotes the proliferation of human breast cancer cells (MCF-7). Tea, on the other hand, includes various antioxidants, including epigallocatechin gallate (EGCG), which may protect against cancer cell proliferation. The purpose of this research is to determine the preventive effect of concentrated brewed green tea on MCF-7 development caused by glyphosate. The glyphosate concentration that promotes MCF-7 development was determined using a serial concentration of glyphosate. Glyphosate concentrations of up to 64 mg/L were shown to have no effect on MCF-7 cell proliferation. Concentrated brewed tea and EGCG 200 mg/L have the potential to suppress MCF-7 cell proliferation in the presence of glyphosate up to 512 mg/L. The combination of glyphosate and concentrated brewed tea or EGCG protects against glyphosate toxicity via altering the expression of tumor suppressor protein (p53).

Abstract

Emprit and red ginger essential oils (EOs) are natural sources of antioxidants that have the potential to be used in cosmetics, one of which is as an anti-skin-aging. The aim of this study was to determine the component differences and anti-skin-aging potential of the two EOs. The components were determined by GC-MS, while discrimination was done by chemometric. The potential of the components as the anti-skin-aging were evaluated by molecular docking and molecular dynamics (MD) simulations. A total 66 components were identified in both EOs, where eucalyptol (17.92%) and camphene (15.12%) were the main component in emprit and red ginger, respectively. Chemometric analysis revealed two discriminant clusters highlighting their dissimilarity with germacrene D and α-zingiberene are the key markers for differentiation. The docking and MD simulations were demonstrated the four main components of emprit EO, namely α-curcumene, α-zingiberene, β-bisabolene and β-sesquiphellandrene, have the best docking scores and interact with the enzymes with a relatively stable interaction. AdmetSAR evaluation of the four components has shown good bioavailability and declared safe. This study succeeded in revealing two ginger EOs differences based on their components and demonstrated the emprit ginger EO was more promising as a natural anti-skin-aging agent for further research.

Abstract

Tingginya kebutuhan masyarakat terhadap sumber daya hutan, jumlah penduduk yang terus bertambah dan tantangan krisis pangan akibat dampak perubahan iklim, memerlukan upaya peningkatan produktivitas lahan dan jumlah pasokan pangan, termasuk melalui praktik agroforestri. Hasil Survei Status Gizi Indonesia tahun 2022 mencatat angka stunting di Indonesia masih tinggi sebesar 21,6%, melebihi ambang batas WHO. Umbi garut ‘pangan agroforestri’ memiliki beragam manfaat kesehatan, merupakan pangan fungsional potensial mengatasi masalah gizi, stunting pada anak. Pencegahan stunting menjadi penting dengan mengonsumsi gizi seimbang berkualitas dari pangan beraneka ragam diantara melalui diversifikasi pangan dalam agroforestri. Untuk itu diperlukan upaya menyeluruh dan terintegrasi oleh pemangku kebijakan dan masyarakat dari hulu ke hilir, mulai dari peningkatan produktivitas di tingkat tapak, diversifikasi olahan pangan, dukungan pemerintah bagi usaha mikro, kecil, dan menengah (UMKM) dalam pengolahannya, hingga sosialisasi manfaat umbi garut yang saat ini masih belum banyak dikenal melalui berbagai program edukatif dan insentif.

Abstract

Endophytic fungi isolated from red ginger (Zingiber officinale) can inhibit growth of Fusarium oxysporum, but the ability of the fungi to control Candida albicans as human pathogen has not been reported. The aims of this research were to study the mechanism of ten endophytic fungi isolates derived from red ginger to control C. albicans in vitro using dual culture methode and fungal extract, and to determine fungal bioactive chemical groups produced by the fungi. Three out of ten isolates tested, Talaromyces assiutensis JMa 7, T. assiutensis JMbt 3, and Curvularia affinis JMbt 9 inhibited growth of C. albicans with inhibition zones were 4.0 mm, 4.9 mm, and 11.3 mm, respectively. The cultures of the three potential endophytic fungi were extracted by maceration method using 3 solvents i.e ethyl acetate, n-hexane and ethanol. The three isolates were grown in PDB separately for 21 days incubation. At harvest, the culture filtrate was extracted by ethyl acetate and n-hexane, while fungal mycelia were extracted by all the three solvents. Ethyl acetate extracts obtained from culture filtrate of all the three fungal isolates consistently inhibited C. albicans with inhibition zones were 2.0-3.8 mm. For n-hexane extract, however, only Talaromyces assiutensis JMbt 3 that had positive effect with inhibition zone was 2.0 mm. All extracts from mycelia did not have any effects on C. albicans. The ethyl acetate extract of T. assiutensis JMbt 3 was analysed to determine its chemical groups using visible color on thin layer chromatography (TLC). The results showed that the bioactive compounds was terpenoids, and antioxidant.

Abstract

Background: A new approach to marketing an inventive product is needed as the development of a new natural toothpaste product is challenging. When choosing herbal-based products, consumers value taste and flavor, preferring natural, appealing flavors over synthetic flavors. Health advantages are also noteworthy, especially, herbal smells promote well-being and satisfaction.Purpose: The study main objective aimed to examine how the Analytic Hierarchy Process (AHP) was used to create a new natural toothpaste product. The study also aimed to design a strategy for innovative toothpaste commercialization based on nanohydroxyapatite (nanoHAP) and Curcuma aeruginosa.Design/methodology/approach: The AHP technique used to evaluate and prioritize elements of natural ingredient, and consumer preferences. The study framework identified herbal toothpaste benefits with known oral health benefits, such as antibacterial, anti-inflammatory, and desensitizing qualities, as well as consumer preferences for tooth paste. These attributes were ranked according to their performance using the AHP framework.Findings/Result: The results of the study stated that the new natural toothpaste, based on nanoHAP and Curcuma aeruginosa, was designed based on quality natural ingredients and consumer preference for naturalness, influencing formula design and marketing efforts. The strategy to develop new toothpaste aimed to provide a holistic health solution, emphasizing natural components and general health perspectives. This strategy could capture the attention of the target market and outpace competitors, attracting customers interested in natural health products.Conclusion: This study addresses the possibility of using nanoHAP alongside Curcuma aeruginosa in a natural toothpaste product that can enhance the remineralization and antibacterial characteristics of the dental product. The research suggests incorporating nanoHAP with Curcuma aeruginosa in a natural toothpaste product for holistic health solutions. Strategies include targeted marketing, loyalty programs, and strategic partnerships, ensuring a comprehensive and data-driven approach to reach and engage target audiences.Originality/value (State of the art): The AHP facilitates complete review, data-driven planning, and commercial viability for commercialization strategy. AHP aids in balancing functional benefits with quantifying subjective assessments, and incorporating customer feedback to increase product success in the competitive oral care industry.
