Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37° C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory

Abstract

Mango fruit (Mangifera indica L.) is the tropical fruit that grows easily in Indonesia with plenty varieties. This study aimed to determine the varieties of mango fruit and the most potent part of mango as antidiabetic agent through α-glucosidase inhibitory activities. Four types of mango fruit (indramayu, manalagi, harum manis, and budiraja) were used in this study. Each part of the mango fruit:peel, flesh, endosperm, and endocarp were extracted by maceration process with three different solvents (n-hexane, ethyl acetate (EtOAc), and ethanol (EtOH)). An ability of all 46 extracts in inhibiting the α-glucosidase at a concentration of 500 ppm were determined. Then 11 extracts with the high inhibition value were determined their IC50 (concentration to inhibit 50% activity) values. EtOAc extract of manalagi, indramayu, and budi raja endosperm had the lowest IC50 value which was not statistically significantly different (at 95%) with EtOAc extract of budi raja peel. The bioautographic Thin Layer Chromatogram showed that the most active band is characterized by white luminescence under UV 366 nm, yellow color under UV 254 and visible light. The band with Rf 0.93 from EtOAc endosperm extract of indramayu and manalagi and Rf 0.73 from EtOAc budi raja peel extract are the most active band which predicted as a flavonoid. The result adds the value of the peel and seed of mango, as well as an alternative in blood sugar control, which is easy to obtain, relatively cheap, and liked by the community.

Abstract

The Trichoderma hamatum produces various secondary metabolites that can be used as Candida albicans.  This research aimed to isolate T. hamatum from the coffee (Coffea canephora) rhizosphere and analyze the fungal compounds to control the pathogenic fungus C. albicans. T. hamatum was isolated using the dilution method, and the fungal identification was used, combining morphological and molecular characteristics of ITS rDNA. The potency of T. hamatum as C. albicans was determined by antagonist test using the double-layer method, while for culture filtrate, ethyl acetate and n-hexane filtrate extracts were carried out by the agar diffusion method. The compounds in the most active extract were analyzed by Gas Chromatography-Mass Spectrometry (GC-MS). T. hamatum inhibits C. albicans growth in antagonistic and filtrate with 15.53 mm and 14.40 mm of inhibition zone. It indicated that both culture and fungal filtrate had similar activities on C. albicans. The ethyl acetate extract (minimum inhibitory concentration, MIC of 0.50%) showed more potent against C. abicans than n-hexane extract (MIC of 15.00%). The potential active compound in the ethyl acetate extract would be 9-Octadecenoic acid (Z) methyl ester. The 9-Octadecenoic acid (Z) methyl ester could be used as an alternative candidate to control Candida.

Abstract

Jojoba (Simmondsia chinensis (Link)) is a shrub plant that widely used as cosmetic ingredients especially jojoba oil. When making jojoba oil, the residue still reminds and become waste. This study aimed to determine the antioxidant activity of jojoba seed residue (JSR). Jojoba seed residue was collected from Sudan. The JSR extracted by maceration with n-hexane, ethyl acetate, and 70% ethanol solvent. The antioxidant activity was determined using DPPH (2,2-diphenyl-1-picrylhydrazyl) assay. The result showed that 70% ethanol extract of JSR had the highest antioxidant activity with 50% Inhibition Concentration (IC50) value of 219.42 mg/L. The 70% ethanol extract was fractionated by using silica gel column chromatography with gradient elution produced 10 fractions. All fractions showed antioxidant activity (IC50106-447 mg/L). Fraction 3 had the highest antioxidant activity with IC50 value of 106.26 mg/L. Therefore, JSR has a great potential for use as a raw material in cosmetic and pharmaceutical products industry, or as a source of bioactive compounds.

Abstract

Antiobesity medication is available as therapeutic compounds that can reduce fat digestion by the inhibition of pancreatic lipase. Actinobacteria have the potency as source of bioactive compounds with various biological function including as pancreatic lipase inhibitor.  However, the potency of endophytic actinobacteria from Rhododendron spp. as source of pancreatic lipase inhibitor producer has not been reported yet. The aim of this study was to examine the potential of pancreatic lipase inhibitory activity of 23 endophytic actinobacteria from Rhododendron spp.; to characterize their colony based on morphology and molecular analysis. Screening test of pancreatic lipase inhibitor was conducted using the supernatant of endophytic actinobacteria, lipase pancreatic porcine (L3126) and p-nitrophenyl butyrate. The supernatant of selected isolates was extracted using ethyl acetate. The result showed that various inhibitory activities ranging between 0.00 until 91.69%. There were 11 out of 23 isolates that have potential as pancreatic inhibitor. Amongst them, the extract of four selected isolates, i.e. RZP 1.3, RSSB 3.2, RSS 2.1, and RJB F3.2 demonstrated inhibitory percentage of more than 80%. The RJB F3.2 extract showed to have IC50 value by 431.48 µg mL-1 compared to control, i.e. Xenical (89.07 µg mL-1). Phytochemical analysis exhibited that the extract of the selected isolates contained alkaloid which may function as pancreatic lipase inhibitor. Based on the morphological character, the selected isolates have various morphological colonies and 16S rRNA gene sequence revealed the sequence homology to Streptomyces spp. The data clearly indicate that endophytic actinobacteria from Rhododendron spp. have potency as pancreatic lipase inhibitor producer and further studies could be explored for the development of antiobesity agent.

Abstract

The leaves of Zingiberaceae family has not been much explored its potential, especially as a skin lightening. The relationship between total anthocyanins, chlorophyll, carotenoids and tannins contents of Zingiberaceae leaves and tyrosinase activity were determined. Ten species of Zingiberaceae were separated by n-hexane and the residues were extracted with ethyl acetate. The total anthocyanins, chlorophyll, carotenoids, tannins contents and the activities of ethyl acetate extracts were determined by spectrometric method. The tyrosinase inhibition was determined by using L-tyrosine (monophenolase) and L-DOPA (diphenolase) substrates. The most active extract was the leaves extract of Zingiber purpureum which exhibited 82.86% for monophenolase inhibition and the leaves extract of Curcuma zedoaria which exhibited 90.20% for diphenolase inhibition. The correlation between carotenoids content and monophenolase inhibition was 52%, while between tannin content and diphenolase inhibition was only 15%. Therefore, among 10 leaves species of Zingiberaceae, Zingiber purpureum Roscoe and Curcuma zedoaria leaves are the most potential for tyrosinase inhibitors and can be developed as whitening agent.How to CiteBatubara, I., Kartika, Y. Abstract

Xylocarpus granatum is mangrove plant that traditionally used as face powder in Central Sulawesi, Indonesia which related to antioxidant, antiglycation and tyrosinase inhibition activities. This study aimed to evaluate the potency of X. granatum as a tyrosinase inhibitor, antiglycation, and antioxidant. The leaves, stem, stem bark, fruit flesh, fruit peel, and kernel of X. granatum were extracted using ethanol then their tyrosinase inhibition, antiglycation, and antioxidant were evaluated. Tyrosinase inhibition activity was evaluated using in vitro assay with L-tyrosine and L-DOPA as the substrate of monophenolase and diphenolase. Antiglycation activity was studied by measuring the excitation and emission fluorescence from glucose and fructose reaction with Bovine Serum Albumin. Antioxidant activity was measured using the DPPH radical scavenging assay. The result showed that the ethanolic extract of fruit flesh has higher potency as tyrosinase inhibitor (IC50 of 393.8 mg/L and IC50 of 448 mg/L, respectively for monophenolase and diphenolase). Antiglycation assay showed that the ethanolic extract of stem bark provides the strongest antiglycation activity with an IC50 of 118.1 mg/L. Meanwhile, fruit peel provides the strongest antioxidant activity with an IC50 of 5.5 mg/L. Fractionation of ethanolic extracts of each part of X. granatum tree yield fractions with lower bioactivity compared to the crude extract. Moreover, stem extract and fractions from two different locations (Tarakan and Kendari) tend to have different bioactivities strengths.  The stem part of X granatum could be developed as new raw material of cosmetic product in Indonesia, while ethanol as the solvent for extraction, and the different bioactivity of stem extract from different location can be the consideration for the industry to standardize the extract prior to production of final product.

Abstract

The most common human pathogen that colonizes in a third of healthy people around the world are Staphylococcus aureus, and one of the materials allegedly able to overcome the pathogen is resin. Resin has been used in folk medicine for thousands of years to treat diseases. The antimicrobial activity of natural resins can be associated with a variety of organic compounds contained in them such as diterpenoids and triterpenoids. This research aimed to explore the antibacterial and antioxidant activities of Pinus merkusii, P. oocarpa, P. insularis, Agathis loranthifolia resins and essential oil. Resin was separated by distillation process to get essential oil and the residue was extracted using n-hexane, ethyl acetate (EtOAc), and methanol (MeOH). Antioxidant activity was performed by DPPH (1,1diphenyl-2-picryl hydrazyl) radical scavenging method. The antibacterial activity of resins and essential oil of the samples determined using the disc diffusion method against Staphylococcus aureus and Escherichia coli. The results showed that the yield of resin extract was ranging from 8.44 % to 95.56%. All extracts and essential oil could not inhibit E coli growth but inhibit the S. aureus growth. This experiment concluded that resin n-hexane extract from P.oocarpa was the most potent as antibacterial activity against S. aureus. All of the samples used had less potential antioxidant activity compared to positive control ascorbic acid. Result of this study show that pine resin from Indonesia has potential as an antibacterial agent.

Abstract

The aim of the present study was to analyse the peel waste of Xylocarpus granatum fruits as potential source of tyrosinase inhibitors. Dried peel samples were ground to yield a powder (simplicia). Subsequently, they were extracted with organic solvents of distinct polarity levels, namely n-hexane (non-polar), chloroform (semi-polar) and methanol (polar) by use of the single-maceration method. Inhibitory effects on tyrosinase activity (monophenolase) and DOPA auto-oxidation (diphenolase) were determined in bioassays. Assays with the methanol extract revealed IC50 values of 784.87 μg mL-1 (monophenolase) and of 1176.66 μg mL- 1 (diphenolase), respectively. In contrast, n-hexane and chloroform extracts showed no activity. These results indicate that the methanolic fruit peel extract contained tyrosinase-inhibiting compounds, such as flavonoids, tannins and saponins, whereas the nhexane and chloroform extracts yielded alkaloids, steroids and triterpenoids without tyrosinase-inhibiting activity. The phenolic compounds had a strong effect on the tyrosinase enzymes, inhibiting monophenolases by 97% and diphenolases by 96%, with a positive correlation between the total phenolic content and the inhibition rate in both activities. Penelitian ini bertujuan untuk menganalisis kulit buah Xylocarpus granatum sebagai sumber potensial inhibitor tirosinase. Sampel kering digiling untuk menghasilkan (serbuk) simplisia. Berikutnya, simplisia diekstraksi dengan pelarut organik dengan tingkat kepolaran yaitu n-heksana (non polar), kloroform (semi polar) dan metanol (polar) dengan menggunakan metode maserasi tunggal. Pengaruh inhibisi didalam aktivitas tirosinase (monofenolase) dan auto-oksidasi DOPA (difenolase) ditentukan di dalam uji. Uji ekstrak metanol menunjukkan masing-masing nilai IC50: 784,87 μg mL-1 (monofenolase) dan nilai IC50: 1176,66 μg mL- 1 (difenolase). Sebaliknya, ekstrak n-heksana dan kloroform menunjukkan tidak ada aktivitas. Hasil penelitian ini menunjukkan bahwa ekstrak metanol kulit buah X. granatum mengandung senyawa-senyawa yang menghambat aktivitas tirosinase seperti flavonoid, tanin dan saponin sedangkan ekstrak n-heksana dan kloroform menghasilkan senyawa alkaloid, steroid, dan triterpenoid tanpa penghambatan aktivitas tirosinase. Senyawa fenolik mempunyai pengaruh kuat dalam menginhibisi enzim tirosinase baik aktivitas monofenolase sebesar 97% dan aktivitas difenolase sebesar 96% dengan korelasi positif antara kandungan total fenol dan tingkat inhibisi pada kedua aktivitas

Abstract

Daemonorops draco has been reported for its antibacterial activity and empirically used for wound healing by Anak Dalam ethnic at Jambi Province, Sumatera-Indonesia. This study was performed to evaluate antibacterial and antibiofilm activity of D. draco resin collected from Jambi. D. draco resin was extracted using n-hexane, ethyl acetate and methanol, respectively. Antibacterial activity of the extracts was evaluated using agar diffusion method against Staphylococcus aureus and Eschericia coli, whereas the minimum inhibitory concentration (MIC) and minimum bacteriacidal concentration (MBC) was determined by microdilution method. In addition, antibiofilm activity was evaluated by violet crystal method. The result showed that extraction yield of ethyl acetate was higher than methanol and n-hexane. Ethyl acetate and methanol extracts of D. draco exhibited stronger antimicrobial activity against S. aureus compare to n-hexane extract. MIC and MBC of methanol extract and chromatographic fraction (F5.1) of ethyl acetate extract were 0.5 and 1.0 mg/mL, respectively. In addition, antibiofilm assay revealed that all extracts were inhibit initial attachment of bacteria cell in biofilm formation. This result revealed a novel information that  D. draco extracts was potential as inhibitor of biofilm formation. TLC bioautography of D. draco extracts indicated that constituent with Rf of 0.71 performed antimicrobial activity against S. aureus. This finding expected to strengthen the scientific backup for utilization of D. draco by society.
