Abstract

Temulawak (Curcuma xanthorrhiza) merupakan tumbuhan yang banyak dimanfaatkan untuk tujuan kesehatan terutama bagian rimpangnya. Minyak atsiri rimpang temulawak juga telah dilaporkan, namun bunga dan daun temulawak belum banyak dieksplorasi termasuk minyak atsiri dan ekstraknya. Penelitian ini bertujuan menapis minyak atsiri dan ekstrak bunga dan daun C. xanthorrhiza sebagai antioksidan menggunakan metode DPPH (2,2- difenil-1-pikrilhidrasil). Minyak atsiri daun diperoleh menggunakan metode distilasi air. Bunga dan daun diekstraksi menggunakan n-heksana, etil asetat, dan metanol. Ekstrak etil asetat daun mempunyai potensi antioksidan terbaik dengan nilai IC50 41.50±7.80 μg/mL namun tidak sebaik asam askorbat sebagai kontrol positif dengan nilai IC50 3.36±0.29 μg/mL Berdasarkan uji kualitatif fitokimia, didapat bahwa ekstrak etil asetat daun temulawak mengandung kelompok senyawa flavonoid, alkaloid, steroid, dan triterpenoid. Minyak atsiri daun temulawak yang juga mengandung terpenoid tidak mampu menghambat 50% radikal DPPH hingga konsentrasi 167 μg/mL.

Abstract

This research aimed to determine the active fraction and profile of active compounds in Manonjaya sSnakefruit skin extract that can be used as antidiabetic and the antidiabetic activity using zebra fish. Manonjaya sSnakefruit skin was extracted by maceration using 70% ethanol and then fractionated using n-hexane, ethyl acetate and methanol. The ethyl acetate fraction showed the highest yield and total phenolic content as well as the strongest α-glucosidase inhibition with an inhibition percentage of 64.43%. The ethyl acetate fraction provided the most active spot as an α-glucosidase inhibitor. The profile of active compounds that act as antidiabetic compound is flavonoid from glycosylflavone and flavone groups. Antidiabetic activity using zebra fish was indicates that Manonjaya Snakefruit skin extract from ethanol extract and ethyl acetate fraction can decrease blood sugar level of zebra fish after induced with alloxan. The crude extract group had 54.51% lower sugar content than the induction group and 41.70% lower than the negative control. Meanwhile, the ethyl acetate fraction group had 65.10% lower sugar content than the induction group and 55.27% lower than the negative control.

Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37° C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory

Abstract

Metabolites from clove (Syzygium aromaticum L.) have been extensively studied for various pharmacological uses, including quercetin. Endophytic bacteria isolated from plant tissues can produce similar secondary metabolites as their host plants. However, little is known about the antioxidant activity of the endophytic bacterial extract from clove leaves. Amongst the potential bacterial groups, the genera of Bacillus have been widely studied as a source of antioxidants. Thus, this study, we examined the potential antioxidant properties of Bacillus sp., isolated from clove leaves. We have successfully isolated ten endophytic bacteria, of which only one isolate (P1U2) was found non-pathogenic based on a hemolytic assay. Based on the 16S rRNA sequence, P1U2 shared the highest similarity to Fictibacillus phosphorivorans. The extract of extracellular metabolites of P1U2 contains quercetin compounds, based on LC-MS analysis. The results of the antioxidant activity test using the DPPH assay showed that the P1U2 extract had an IC50 value of 161 mg/ml. The supplementation of the extract on culture media reduced the ability of S. cerevisiae to decolorize mutagen malachite green and increased cell viability. Furthermore, the addition of 161 ppm extract was able to counteract UV mutagens effects of UV exposure and toxicity of malachite green, significantly increasing the growth of S. cerevisiae. Suggesting the bioactivity of the extract in combating mutagens. To the best of our knowledge, this is the first study to report a quercetin-rich extract derived from endophytic bacteria, F. phosphorivorans. Thus, further study is required to optimize the synthesis of quercetin from isolate P1U2.

Abstract

Coffee extracts are a rich source of potential compounds with numerous biological activities. This study aimed to investigate the potential of chemical compounds derived from robusta coffee extract (Coffea canephora) and its dominant compound, caffeine, to scavenge free radicals, inhibit bacterial growth and biofilm formation, and cytotoxic properties in human breast adenocarcinoma cell line. Chemical constituents of coffee extract were analysed quantitively for total phenolic, flavonoid, and alkaloid contents, along with Liquid Chromatography Quadrupole-Mass Spectrometry (LC/MS–MS) analysis. Antioxidant activity was measured by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis; 3-ethylbenzothiazoline-6-sulfonic acid (ABTS) methods. Antibacterial pair with antibiofilm properties against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, and Staphylococcus aureus were also determined, as well as cytotoxic activity on the MCF7 cell line model. The LC/MS–MS analysis of coffee extracts revealed high levels of caffeine; thus, the caffeine standard is used in all subsequent assays. Notably, robusta coffee extract showed remarkable antioxidant activity and selective inhibition of the growth against gram-positive and negative bacteria, along with the best inhibition of P. aeruginosa biofilm. However, compared with the caffeine standard, robusta coffee extract had lower cytotoxic activity and different bacterial targets in antibacterial and antibiofilm properties. Our results indicate that Robusta coffee extract is potentially a functional food due to its high alkaloid, phenolic, and flavonoid content and antioxidant activity, besides being used for natural compounds against bacterial infections.

Abstract

The aim of the present study was to analyse the peel waste of Xylocarpus granatum fruits as potential source of tyrosinase inhibitors. Dried peel samples were ground to yield a powder (simplicia). Subsequently, they were extracted with organic solvents of distinct polarity levels, namely n-hexane (non-polar), chloroform (semi-polar) and methanol (polar) by use of the single-maceration method. Inhibitory effects on tyrosinase activity (monophenolase) and DOPA auto-oxidation (diphenolase) were determined in bioassays. Assays with the methanol extract revealed IC50 values of 784.87 μg mL-1 (monophenolase) and of 1176.66 μg mL- 1 (diphenolase), respectively. In contrast, n-hexane and chloroform extracts showed no activity. These results indicate that the methanolic fruit peel extract contained tyrosinase-inhibiting compounds, such as flavonoids, tannins and saponins, whereas the nhexane and chloroform extracts yielded alkaloids, steroids and triterpenoids without tyrosinase-inhibiting activity. The phenolic compounds had a strong effect on the tyrosinase enzymes, inhibiting monophenolases by 97% and diphenolases by 96%, with a positive correlation between the total phenolic content and the inhibition rate in both activities. Penelitian ini bertujuan untuk menganalisis kulit buah Xylocarpus granatum sebagai sumber potensial inhibitor tirosinase. Sampel kering digiling untuk menghasilkan (serbuk) simplisia. Berikutnya, simplisia diekstraksi dengan pelarut organik dengan tingkat kepolaran yaitu n-heksana (non polar), kloroform (semi polar) dan metanol (polar) dengan menggunakan metode maserasi tunggal. Pengaruh inhibisi didalam aktivitas tirosinase (monofenolase) dan auto-oksidasi DOPA (difenolase) ditentukan di dalam uji. Uji ekstrak metanol menunjukkan masing-masing nilai IC50: 784,87 μg mL-1 (monofenolase) dan nilai IC50: 1176,66 μg mL- 1 (difenolase). Sebaliknya, ekstrak n-heksana dan kloroform menunjukkan tidak ada aktivitas. Hasil penelitian ini menunjukkan bahwa ekstrak metanol kulit buah X. granatum mengandung senyawa-senyawa yang menghambat aktivitas tirosinase seperti flavonoid, tanin dan saponin sedangkan ekstrak n-heksana dan kloroform menghasilkan senyawa alkaloid, steroid, dan triterpenoid tanpa penghambatan aktivitas tirosinase. Senyawa fenolik mempunyai pengaruh kuat dalam menginhibisi enzim tirosinase baik aktivitas monofenolase sebesar 97% dan aktivitas difenolase sebesar 96% dengan korelasi positif antara kandungan total fenol dan tingkat inhibisi pada kedua aktivitas

Abstract

Purwoceng (Pimpinella pruatjan) is an Indonesian native plant but difficult to cultivate. Hydroponics can be used as a solution for purwoceng cultivation. Hydroponic types and nutrient concentrations can affect purwoceng’s productivity. This study aimed to determine total phenolic content (TPC) and total flavonoid content (TFC), changes in steroid, and antioxidant capacity due to two hydroponic types (nonrecirculating and recirculating drip) and three nutrient concentrations (1,000, 1,500, and 2,000 ppm). Purwoceng was determined for its moisture content, ash content, and dry weight, and extracted with ethyl acetate for gas chromatography-mass spectrometry analysis and with ethanol for TPC, TFC, and antioxidant capacity. The data were analyzed statistically and grouped using principal component analysis (PCA) and heat map analysis (HMA). Nonrecirculating drip with high nutrient concentration at the aerial part was the best treatment to produce high TPC, TFC, and antioxidant capacity. There were no significant changes in steroid content. PCA showed that purwoceng extracts could be grouped in to two groups, i.e. aerial and root groups. HMA showed that purwoceng extracts could be distinguished from the abundance of palmitic acid and stigmasterol. The antioxidant capacity of purwoceng was directly proportional to the TPC value and also related to the presence of phytol, stigmasterol, and palmitic acid.

Abstract

Optimization extraction of the brown algae Padina australis using the Box-Behnken design has been carried out. Box-Behnken design in relation to Response Surface Methodology analysis was conducted with four experimental factors (i.e., solvent concentration, temperature, extraction time, and sample to solvents ratio) towards the responses of yield antioxidant, anti-tyrosinase, anti-glycation, total phenolic content, and fucoxanthin content, completing with 29 running experiments. P. australis extraction's optimum condition was acquired at 79.99% solvent concentration, 18.48 hours extraction time, 44.50ºC temperature, and 1:9 ratio powders and solvents. The optimum condition provided a 7.30% extraction yield, 43.94% antioxidant activity, 86.83% anti-tyrosinase, 98.06% anti-glycation, 9.53 mg GAE/g total phenolic content, and 347.55 µg/g fucoxanthin content. Respond Surface Methodology analysis with the Box-Behnken design succeeded in making the appropriate model for producing the optimum P. australis extract.

Abstract

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) is the virus that causes Coronavirus 2019 (COVID-19). To date, there has been no proven effective drug for the treatment or prevention of COVID-19. A study on developing inhibitors for this virus is carried out using molecular docking simulation methods. 3CL-Pro, PL-Pro, Helicase, N, E, and M protein were used as protein targets. Autodock Vina, Autodock 4.2, and PSOVina were used in this study. This study aims to obtain a model of ligands interactions of active natural compounds against the receptor protein encoded by the SARS-CoV-2 genome and their free binding energy to propose active compounds from natural products that have potential as a drug for COVID-19. Corilagin (-14,42 kcal/mol), Scutellarein 7-rutinoside (-13,2 kcal/mol), Genistein 7-O-glucuronide (-10,52 kcal/mol), Biflavonoid-flavone base + 3O (-11,88 and -9,61 kcal/mol), and Enoxolone (-6,96 kcal/mol) has the best free energy value at each protein target indicating that the compound has the potential as a viral protein inhibitor for further investigation. This research is limited to computer simulations, where the results obtained are still a prediction.

Abstract

Mango fruit (Mangifera indica L.) is the tropical fruit that grows easily in Indonesia with plenty varieties. This study aimed to determine the varieties of mango fruit and the most potent part of mango as antidiabetic agent through α-glucosidase inhibitory activities. Four types of mango fruit (indramayu, manalagi, harum manis, and budiraja) were used in this study. Each part of the mango fruit:peel, flesh, endosperm, and endocarp were extracted by maceration process with three different solvents (n-hexane, ethyl acetate (EtOAc), and ethanol (EtOH)). An ability of all 46 extracts in inhibiting the α-glucosidase at a concentration of 500 ppm were determined. Then 11 extracts with the high inhibition value were determined their IC50 (concentration to inhibit 50% activity) values. EtOAc extract of manalagi, indramayu, and budi raja endosperm had the lowest IC50 value which was not statistically significantly different (at 95%) with EtOAc extract of budi raja peel. The bioautographic Thin Layer Chromatogram showed that the most active band is characterized by white luminescence under UV 366 nm, yellow color under UV 254 and visible light. The band with Rf 0.93 from EtOAc endosperm extract of indramayu and manalagi and Rf 0.73 from EtOAc budi raja peel extract are the most active band which predicted as a flavonoid. The result adds the value of the peel and seed of mango, as well as an alternative in blood sugar control, which is easy to obtain, relatively cheap, and liked by the community.
