Abstract

Kembang Bulan (Tithonia diversifolia ) is known as an herbal plant that has many useful properties to cure disease, such as diabetes, malaria, and other infections. Instead of much usefulness, T. diversifolia has main biological properties that may induce toxicity in this plant known as sesquiterpene lactones. The aim of this study was to identify the cytotoxicity effect of T. diversifolia leaf extract on limbal mesenchymal stem cells of rabbits using the MTT assay. T. diversifolia leaf extract was divided into 4 concentrations (0.5%, 0.25%, 0.125%, and 0.0625%) and given to limbal mesenchymal stem cells of rabbits in 96-well microplates, which would be incubated for 24 hours. The cytotoxicity result was obtained using the MTT assay method. Every well would be observed to see the cell life level of each concentration. The optical density absorbance was calculated using a microplate reader. T. diversifolia leaf extract with a concentration of 0.0625% has the biggest viability with a level of existence of limbal mesenchymal stem cells of 50%, and the other T. diversifolia leaf extract concentrations of 0.5%, 0.25%, and 0.125% have less than 50% level of existence of limbal mesenchymal stem cells.

Abstract

Subclinical mastitis is an economically important disease in dairy goat farming. Lumajang is a center for dairy goats in East Java. However, routine examination of subclinical mastitis for screening tests has no data. This study aimed to provide prevalence data and identification factors that influenced subclinical mastitis. Therefore, this study was carried out as part of the national surveillance program. The research design is a cross-sectional study with simple random sampling. Samples came from 19 farms with a total individual sample of 224 dairy goats. Detection of subclinical mastitis using an undirect test with the California Mastitis Test (CMT). Identification of management factors and characteristics that can influence subclinical mastitis in dairy goat farms using a structured questionnaire. The results showed that the prevalence of subclinical mastitis in Lumajang District was 36.2%. There was a significant difference in subclinical mastitis in each village (p<0.05). The highest prevalence of subclinical mastitis was in Kandangtepus village. A farmer's characteristic factor that significantly influences subclinical mastitis is the farmer's experience. The aspect of livestock management that significantly affects subclinical mastitis is structured work management. Subclinical mastitis was prevalent in dairy goats in Lumajang District and requires prevention efforts. Prevention approach through education for farmers to increase knowledge, attitude, and socialization to improve livestock management.

Abstract

The aim of this study was to isolate and identify the strain of methicillin-resistant Staphylococcus aureus (MRSA) from cows milk in Surabaya and to determine the fragment Spa-Gene in MRSA strains. There were 50 samples of cows milk obtained from four dairy farms. From 50 tested samples, 19 samples (38%) were positive for Staphylococcus aureus. Antibiotic sensitivity test using oxacillin and erythromycin showed that 6 samples were resistant to the antibiotic oxacillin and 6 samples were resistant to erythromycin. MRSA confirmation tests that were conducted on 6 samples revealed 5 positive results for the MRSA strain. Electrophoresis of Polymerase Chain Reaction (PCR) product showed that 4 out of 5 samples were positive for the presence of the Spa-Gene fragment. The research results showed that there were 3 models of Spa-Gene fragments; the first had a length of 90 bp and 140 bp, the second had a length of 140 bp, and third had a length of 90 bp. This study revealed the nature of Spa-Gene polymorphism of MRSA strains isolated from milk samples. It was concluded that the Spa-Gene can be used as a molecular epidemiological marker of the MRSA strain.

Abstract

Background: Nosocomial infections pose a major challenge not only in human hospitals but also in veterinary settings. These infections are frequently associated with methicillin-resistant Staphylococcus aureus (MRSA) and extended-spectrum β-lactamase-producing Enterobacteriaceae (ESBL-E). Purpose:  This study aimed to detect and characterize MRSA and ESBL-E isolated from environmental samples at the Veterinary Teaching Hospital, Universitas Brawijaya (VTH UB). Method: The study involved four main procedures: (1) sample isolation; (2) bacterial identification of S. aureus and Enterobacteriaceae using MacConkey agar supplemented with 1 mg/L cefotaxime (MCA + 1 mg/L); (3) antibiotic susceptibility testing and MRSA confirmation using Oxacillin Resistance Screening Agar Base (ORSAB); and (4) confirmation of ESBL production through the Double Disk Synergy Test (DDST). Results: Isolation and identification revealed S. aureus in 86.6% (13/15) of samples and Enterobacteriaceae in 26.6% (4/15). Among Enterobacteriaceae, Klebsiella pneumoniae accounted for 6.7% (1/15) and Enterobacter aerogenes for 20% (3/15). Antimicrobial resistance testing of Staphylococcus aureus isolates showed that all (13/13) were resistant to at least one antibiotic class. Resistance rates were 53.8% (7/13) to oxacillin, 92.3% (12/13) to cefoxitin, 23.0% (3/13) to tetracycline, 92.3% (12/13) to erythromycin, and 46.1% (6/13) to gentamicin. The prevalence of MRSA among environmental isolates was 46.6% (7/15), while ESBL-E prevalence was 26.6% (4/15). Conclusion: The detection of MRSA and ESBL-E in the hospital environment indicates potential zoonotic and hospital associated infection risks in veterinary facilities. These findings emphasize the importance of strengthened infection control practices and provide a baseline for broader surveillance across veterinary hospitals.

Abstract

This study aimed to determine the quality of Ettawah crossbred buck semen diluted in skim milk, egg yolk-citrate extenders, or their combination, supplemented with epigallocatechin-3-gallate (EGCG). Semen was collected from Ettawah crossbred bucks using an artificial vagina. Six ejaculates were used for replication and divided into three extender treatments: T1, skim milk; T2, egg yolk-citrate; and T3, a combination of both, each supplemented with 1.5 µg/mL EGCG. The extended semen was stored in a refrigerator at 5 °C. Diluted samples were evaluated every 24 hours until spermatozoa motility declined to 30%, the minimum requirement for artificial insemination. The results showed that spermatozoa motility, viability, plasma membrane integrity, and morphological abnormality decreased during six days of storage at 5 °C. Spermatozoa motility remained above 30% for five days in T1 (36.50±1.22%) and T2 (41.67±2.06%), and for six days in T3 (43.33±1.03%). On day six, semen in the combined extender supplemented with EGCG (T3) showed significantly higher motility, viability, and membrane integrity, and lower morphological abnormalities (p <0.05) than the other treatments. In T2, all spermatozoa quality parameters were higher (p <0.05) than in T1. It can be concluded that the combined skim milk-egg yolk-citrate extender containing 1.5 µg/mL EGCG best maintained the spermatozoa quality of Ettawah crossbred bucks during six days of storage at 5 °C. Based on spermatozoa motility, the semen remained suitable for artificial insemination.
