Abstract

Fluoroquinolone is used as first line drug for Salmonella sp infection, but there were reports of increasing treatment failure with fluoroquinolone in infection caused by Salmonella sp, which in vitro is still succeptible to fluoroquinolone. The identification of nalidixic acid resistance, a first generation quinolone provides a high sensitivity and specifity for the detection of such fluoroquinolone resistance. The researchers aim is to study the prevalence and the minimum ciprofloxacin inhibitory concentration of nalidixic acid resistant but fluoroquinolone sensitive Salmonella sp at Premier Jatinegara Hospital. Blood cultures sent to Premier Jatinegara Hospital Laboratory during 2010 were evaluated according to Clinical and Laboratory Standards Institute guidelines. Identification and MIC succeptibility testing were determined by VITEK® 2 Compact (Biomerieux®) and nalidixic acid succeptibility testing was performed by disc diffusion method according to Kirby Bauer. Thirty eight Salmonella sp isolates were identified, all were succeptible to ciprofloxacin (MIC  0,25 mg/L), but 5 (13,2%) isolates were resistant to nalidixic acid and reported as resistant. This study found that 13,2% of Salmonella sp were resistant to fluoroquinolone but not detected by the recommended CLSI breakpoint values. The researchers recommend that nalidixic acid testing be included in Salmonella sp succeptibility testing in Indonesia and consider 3rd generation cephalosporin as the first line drug before a succeptibility test result is available.

Abstract

Demam Berdarah Dengue (DBD) masih menjadi masalah kesehatan di Kabupaten Grobogan Provinsi Jawa Tengah. Angkakesakitan DBD per 100.000 penduduk di Kabupaten Grobogan dari tahun 2011-2013 semakin meningkat. Penggunaaninsektisida sebagai upaya pengendalian vektor paling banyak dilakukan oleh masyarakat. Penelitian ini bertujuan untuk mengetahui gambaran penggunaan insektisida rumah tangga di Kabupaten Grobogan. Metode yang digunakan adalah surveidan wawancara dengan kuesioner. Hasil penelitian menunjukan sebagian besar masyarakat menggunakan insektisida rumahtangga (86,33%) dengan intensitas penggunaan paling banyak sehari sekali (85,4%) dengan lama penggunaan lebih dari 5tahun (74,51%). Kandungan bahan aktif insektisida yang digunakan merupakan golongan sintetik piretroid. Hal inimerupakan salah satu faktor pendukung terjadinya kerentanan Ae. aegypti terhadap insektisida yang digunakan olehprogram.

Abstract

Pohon ketapang (Terminalia catappa) merupakan tanaman yang sering ditanam di ruang terbuka hijau. Tanaman ini menghasilkan buah dan daun yang merupakan sampah organik yang mengandung lignoselulosa. Tujuan dari penelitian ini adalah untuk meningkatkan nilai ekonomis dari sampah ketapang, menemukan bahan bakar alternatif terbarukan yang memiliki kualitas yang optimal dan nilai kalor yang tinggi dengan penambahan polietilen. Metode yang digunakan dalam penelitian ini adalah metode eksperimen dan studi literatur. Proses karbonisasi dilakukan dengan pirolisis menggunakan suhu 350°C untuk mendapatkan arang yang selanjutnya digerus dan diayak untuk mendapatkan serbuk arang. Serbuk arang selanjutnya dicampur dengan polietilen dengan kadar polietilen 0%, 5%, 10%, dan 15% yang kemudian ditambahkan dengan adonan tepung tapioka sebagai perekat dan diaduk rata. Hasil biobriket yang telah kering kemudian diuji kualitasnya dan dianalisis sesuai SNI 01-6235-2000 tentang Briket Arang Kayu. Hasil penelitian menunjukkan bahwa penambahan polietilen dapat menurunkan kadar air, kadar abu, dan kadar karbon terikat serta meningkatkan kadar zat terbang dan nilai kalor pada biobriket ketapang.Kata Kunci: Biobriket ; Politilen ; Ketapang ; Pirolisis

Abstract

Phytoremediation is one of alternative methods to remove pollutant in leachate. Leachate from Jetis Landfill was applied in this research. The purposes of the study were to analysis the change of BOD and COD concentration using Nymphaea sp. and Pistia stratiotes L. and to observe plants condition.The research was carried out by laboratory experiment. Tools and materials used in this research were reactors with 50 L capacity, 455 L of leachate, 12 pcs of Nymphaea sp., 120 pcs of Pistia stratiotes L., 12 kg of palm fiber, and 120 kg of gravel. There were 25 reactors consisted of 1 control reactor, 12 reactors with 3 day of resident time (H3) and 12 reactors with 7 days of resident time (H7). Each of reactors is filled with 15-20 L of leachate which consisted of only plant reactors (T or A) and palm fiber, gravel and plant reactors (TIK or AIK).  Each reactor was tested the BOD and COD. Plant conditions were observed daily. Changes of BOD and COD levels for each treatment presented in graphs then analysed descriptively.The results showed that BOD and COD levels of each reactor were lower than control. The difference was relatively small. The AIK-H7 reactor was the most optimum in reducing BOD levels, although the percentage of reduction only 24.11%.  In TIK-H7, the COD level was 576 mg/L, the reduction percentage reached 23,6%. The plant observation, found on the 3rd day of treatment, the leaves of the Nymphaea sp. have begun to decay, while the leaves of the Pistia stratiotes L. have started to dry and turn yellow. On the 7th day the whole plant was died. Phytoremediation with Nymphaea sp. and Pistia stratiotes L. in this research was not maximum to remove BOD and COD in leachate of Jetis Landfill.

Abstract

This research aimed to analyze the causal factors on Farmers Low Interest to Implement theEnvironmentally Friendly Red Chili Farming In South Lampung Regency. This research employed a surveymethod by using constructed quisionairs. The research samples were 71 farmers, drawn from nonenvironmentallyfriendly red chilli farmers. The research was conducted in five districts of South LampungRegency, namely Natar, Way Sulan, Candipuro, Kalianda and Penengahan. The data was analyze byanalysis factors of SPSS program. The result showed that there were three factors causing farmers lowinterest to implement environmentally friendly red chilli farming. They were cultivation application and theofficer guidance, means and pest attact, in addition to cultivation result.

Abstract

Indonesia adalah satu negara yang mengandalkan pendapatan negara dari sektor pariwisata. Salah satu lokasi yang sekarang mulai banyak didatangi wisatawan local maupun wisatawan mancanegara adalah Kepulauan Karimunjawa. Kenaikan jumlah wisatawan di Karimunjawa juga akan berdampak terhadap kondisi terumbu karang. Penelitian ini bertujuan untuk mengetahui kondisi tutupan karang terhadap daya dukung wisata bahari di Perairan Kepulauan Karimunjawa. Data tutupan substrat diambil pada dua kedalaman yaitu dangkal (3-6 m) dan dalam (9-12 m), untuk masing-masing lokasi menggunakan metode Underwater Photo Transect  (UPT). Data jumlah wisatawan baik snorkling maupun diving didapat dengan metode wawancara terhadap stakeholder terkait. Hasil penelitian menunjukkan kondisi terumbu karang ke-empat lokasi penelitian masuk dalam kategori sedang (25-49,5 %) hingga baik (50-74,5 %). Jumlah pengunjung rata-rata harian tertinggi terdapat di Perairan Pulau Menjangan Kecil yaitu 194 wisata snorkling dan 216 wisata diving. Sedangkan jumlah wisatawan terendah ada di Perairan Pulau Cemara Kecil yaitu 88 wisatawan per hari yang terdiri dari 36 wisata snorkling dan 52 wisata diving. Berdasarkan hasil yang didapat, dapat disimpulkan bahwa kondisi tutupan karang keras tetap berada pada kategori sedang-baik meskipun persentasenya ada yang mengalami penurunan maupun peningkatan. Jumlah wisatawan snorkling maupun diving yang berkunjung tidak melebihi kapasitas daya dukung lingkungan kecuali wisatawan snorkling di Perairan Cemara Kecil.

Abstract

BA and 2,4-D combination were commonly used for in vitro culture of Shallot (Allium cepa L. var agregatum 2n = 2x = 16) to induce callus, but there was no information for callus induction on shallot seed (TSS) explant. Callus could be utilized for in vitro selection and generating of genetic variation. The aims of the research was to identify the response of TSS (Trisula and Tuk Tuk) as explant  and to obtain the optimum combination of BA and 2,4-D (mg.L-1): (0–0, 2–1, 2–2, 2–3, and 2–4) in callus induction. The research had been carried out in the Tissue Culture Laboratory, Faculty of Agriculture, Universitas Gadjah Mada during the year 2015-2016. Factorial treatments of variety and growth regulators were arranged in Completely Randomized Design with four replications. Data of percentage of germination, shoot height, root length, percentage of callus formation, callus weight, and chromosomes number of callus were recorded. The results showed that combination of 2 mg.L-1 BA + (1– 4) mg.L-1 2,4-D induced callus formation on TSS but inhibits shoots and roots growth. The best callus proliferation was at a concentration of 1 mg.L-1 2,4-D. Tetraploid callus chromosomes (2n = 4x = 32) was detected in Trisula grown in the 2 mg.L-1 BA + 4 mg.L-1 2,4-D, but in the Tuk Tuk callus did not detected the changing of chromosomes number.

Abstract

Lombok upland rice is one of the cultivars that have the potential as a genetic source. However, Lombok upland rice is almost rarely found. Therefore, conservation was carried out through morphological characterization to provide genetic information. This study aimed to describe the characteristics of six Lombok upland rice cultivars, namely, Reket Putek Bulu, Reket Putek Buntung, Reket Bireng Bulu, Reket Bireng Buntung, Pare Beaq Sapit, and Beaq Ganggas. A completely randomized design was used, consisting of these six Lombok upland rice cultivars as treatments. Two superior cultivars were also used for comparison. The study revealed sufficient divergence for various qualitative and quantitative traits. Pare Beaq Sapit and Beaq Ganggas shared common morphological characters, and both were tall plants. The auricle and ligule colors of Reket Bireng Bulu were different from those of the other cultivars. Reket Putek Buntung had the latest flowering and harvesting age. Reket Putek Bulu and Reket Bireng Buntung had a high number of productive tillers. Six cultivars of Lombok upland rice were characterized to have morphological diversity, so that they are expected to be used as genetic material in rice plant breeding, thereby developed to avoid extinction.

Abstract

Phytase enzymes are applied to animal feed to help animals absorb more nutrients. The use of feed raw materials containing phytase enzymes is expected to reduce the cost of animal feed production. Efforts to increase the phytase content in maize were carried out by improving genetics, in the way of assembling transgenic plants containing high phytase content. The 27-kDa γ Zein promoter is a specific promoter that expresses genes in caryopsis, and promoter CaMV 35S is a constitutive promoter that controls gene expression in all tissues and generally does not depend on the growth phase. Transgenic maize was transformed using Agrobacterium tumefacien infection method on maize B104. The reverse transcriptase polymerase chain reaction (RT-PCR) approach was used to examine the expression of phytase genes in leaves, roots, and caryopsis was done 10, 20, and 30 days after pollination (DAP). The phytase enzyme activity test was also carried out by using the colorimetric phosphomolybdate analysis method to see the phytase enzyme activity in unit µg-1. The results showed that the phytase gene in transgenic plants with the 27-kDa γ Zein promoter was highly expressed in maize caryopsis, but in line Z6.10 was also expressed in leaves, while in the CaMV 35S promoter the phytase gene was only expressed on the leaves. Phytase enzyme activity showed that transgenic maize was higher than non-transgenic maize.

Abstract

Humans can survive on the results of utilizing the naturalresources around him. Animals, plants, air, water, minerals,everything in the universe is taken advantage to meet the needsof human life. Disaster after the cause of global warming. Nitrousoxide is produced by vehicle and industrial emissions, whilemethane emissions are caused by industrial and agriculturalactivities. The environmental damage caused by humanintervention in order to take advantage of nature to make endsmeet. But in exploiting the human nature often do not payattention to the balance of natural ecosystems so that the originalharmony be shaken. It is characterized by the loss of one or moreconsumers in certain trophic levels resulting population of certainspecies becomes abundant and others missing. Besides theenvironmental damage in this case the forest is potentiallycreating global warming caused by greenhouse gases in the formof carbon dioxide gas (CO2) resulting from the burning of fossilfuels and the use of CFCs. It is necessary for adaptation to thenew environment in order to survive. So maybe somedayhumans and other organisms evolve to adapt or adjust to a newenvironment where he lives. To prevent damage to the earthnecessary preventive measures friendly to the environment.
