Abstract

Penelitian ini bertujuan untuk mengetahui kualitas semen sapi Rambon Banyuwangi dengan menggunakan dua pengencer yang berbeda, yakni tris kuning telur dan susu skim kuning telur. Metode penelitian ini menggunakan rancangan acak lengkap (RAL). Penelitian ini menggunakan satu ekor pejantan sapi Rambon untuk penampungan semen. Semen ditampung sebanyak sembilan kali untuk dua perlakuan pengencer yaitu P1: semen+tris kuning telur dan P2: semen+susu skim kuning telur. Parameter yang diukur pada penelitian ini adalah motilitas, viabilitas dan abnormalitas spermatozoa. Setiap parameter pada masing-masing perlakuan dibandingkan dengan menggunakan uji T dua sampel. Hasil penelitian ini menunjukkan bahwa P1 lebih mampu mempertahankan motilitas dan viabilitas spermatozoa sapi Rambon Banyuwangi dibanding dengan P2, dimana motilitas P1 yaitu 67,22±4,41% dengan kecepatan 3 dan motilitas P2 yaitu 59,44±8,46% dengan kecepatan 2 berbdea nyata (p<0,05). Viabilitas P1 adalah 77,44±3,21% dan P2 yaitu 73,56±3,32% berbeda nyata (p<0,05). Tidak terdapat perbedaan yang nyata (p>0,05) terhadap abnormalitas spermatozoa baik primer maupun sekunder sapi Rambon Banyuwangi pada kedua pengencer dimana nilai abnormalitas primer P1 yaitu 0,33±0,50% dan P2 0,22±0,44%, abnormalitas sekunder P1 yaitu 8,33±2,17% dan P2 7,56±2,24%. Disimpulkan bahwa media pengencer tris kuing telur dan susu skim kuning telur layak untuk digunakan IB dengan semen segar.

Abstract

Tujuan penelitian ini adalah mengetahui perbedaan kualitas (viabilitas, motilitas dan abnormalitas) spermatozoa Sapi jantan Bali hasil sexing menggunakan alat ESS yang dialiri listrik selama lima menit dan sepuluh menit pada sisi anoda dan katoda dan mengetahui perbedaan efektivitas pemisahan spermatozoa Sapi Bali hasil sexing menggunakan alat ESS yang dialiri listrik selama lima menit dan sepuluh menit pada sisi anoda dan katoda. Penelitian ini menggunakan Sapi Bali yang berumur 4 tahun sampai 7 tahun dengan motilitas di atas 45% dan di bawah 60%. Penelitian ini menggunakan diluter tris kuning telur. Sexing spermatozoa menggunakan alat ESS yang dialiri listrik dengan durasi 5 menit dan 10 menit pada masing-masing anoda dan katoda. Hasil penelitian ini menunjukan kualitas (viabilitas, motilitas dan abnormalitas) mengalami penurunan. Kualitas terbaik pada sexing spermatozoa terdapat pada durasi lima menit.  Efektivitas pemisahan spermatozoa yang paling baik yaitu pada durasi sepuluh menit pada sisi katoda spermatozoa X sebesar 62,17±0,240% dan  pada sisi anoda spermatozoa Y sebesar 67,33± 1,03%.

Abstract

Penelitian ini merupakan penelitian deskriptif dengan menggunakan 20 itik Alabio betina siap bertelur dengan rentang usia 6 bulan dan untuk pemeriksaan kualitas telur untuk masing-masing kelompok 10 sampel. Pengamatan tingkat produksi telur dilakukan selama tiga bulan dengan 10 pengulangan, kualitas telur (ketebalan kulit telur, persentase kulit telur, persentase kulit telur, persentase albumin, persentase kuning telur, huagh unit, warna kuning telur, kadar protein dan lemak kandungan). Data produksi telur dan kualitas telur diuji menggunakan uji Anova. Hasil pengamatan rata-rata produksi harian telur 15.45 ± 0.12, 14.72 ± 0.10, 14.91 ± 0.09; produksi berat harian (kg/hari) 925.27 ± 7.50, 875.40 ± 6.32, 884 ± 5.03; hasil dari ketebalan kulit telur kualitas 0.33 ± 0.01, 0.33 ± 0.12, 0.33 ± 0.12; persentase kulit telur 11.32 ± 0.93, 11.28 ± 0.10, 11.36 ± 0.05; persentase albumin 55.70 ± 1.08, 56.66 ± 1.22, 56.12 ± 1.00; persen kuning telur 32.07 ± 1.24, 31.55 ± 1.48, 31.64 ± 1.20; Haugh unit 88.70 ± 6.21, 91.41 ± 6.70, 94.51 ± 5.06. Warna kuning masing-masing memiliki skor 15. Dapat disimpulkan bahwa produksi dan kualitas telur itik Alabio baik.

Abstract

The aim of this study was to know about genetic profile of Folicle Stimulating Hormone Receptor (rFSH) in Madrasin crossbreed between Madura breed and Limousin breed (Madrasin). The research are using whole blood samples from Madura breed, Limousin breed, and Madrasin crossbreed. Madura breed and Madrasin crossbreed sample were taken from Galis, Pamekasan, East Java and Limousin breed is taken from Balai Embrio Ternak (BET) Cipelang, Bogor, West Java. Samples were used for genetic profile analysis such as DNA amplification with Polymerase Chain Reaction (PCR) method and DNA sequencing in Institute of Tropical Disease, Universitas Airlangga. The result showed that DNA fragment of rFSH is 211bp. In conclusion, this study can detect rFSH gene Madrasin crossbreed which is polymorpism.

Abstract

This study was conducted to find out how much losses the breeders get when their cattle experience a decline in day open (DO), service per conception (S/C), and calving interval (CI).There were 108 lactating dairy cows from the dairy farm, Wagir District, Malang Regency.The data that has been taken is then processed by analysis using linear regression method.The results of the analysis prove that reproductive management has an effect of 82% on farmer expenditure.For each setback DO per day, the farmer pays Rp. 609,05.Meanwhile, for each additional S/C the farmer pays Rp. 99,921.7.For each setback the farmer CI will incur a fee of IDR 228,54 / day.So that the equation found is expendses = 609,05 (DO) + 99,921.7 (s / c) +228,54 (CI).

Abstract

The aim of this study was to determine the potential of honey to the length of small intestine villi of albino rats due to nutritional deficiencies. The experimental animals were used 24 male albino rats with a body weight of 250-300 grams, divided into four groups and each group contained six replication. Negative control (K-) was a group of rat without fasting and without honey. Positive control (K +) was a group of rat with fasting conditions for 5 days and without honey. The treatment group was the rat with fasting conditions for 5 days and given honey therapy at a dose of 30% (P1) and 50% (P2). Data analysis used ANOVA (Analysis of Variants) then continued with the Duncan Multiple Range test with a significance level of 5% it can be seen that the length of small intestine villi was obtained at (K-) which is 531,321 ± 0,64 which is not significantly different (p>0,05) from (P2), which is 519,919 ± 0,49, but is significantly different (p<0,05) with other treatments (K+) 360,564 ± 0,87 and (P1) 434,275 ± 0,40. The conclusion of this study is that giving honey has the potential to increase the length of the intestinal villi of albino rats due nutritional deficiencies.

Abstract

This study was conducted to know the influence of grass to concentrate ratio to milk urea nitrogen (MUN) level, progesterone level and pregnancy status in Friesian Holstein. A total of 18 dairy cows were randomly selected from the population. The cows were grouped based on feed and MUN and pregnancy. Milk sample collection was carried out once on the seventh day after estrus (D7) and tested with the Barthelot method for measuring urea nitrogen levels. Blood samples were taken three times, on the day of estrus (D0), seven days after estrus (D7), and twenty-two days after estrus (D22), and were tested using Immuno-Sorbent Assay (ELISA). Pregnancy examination was carried out at D90 after artificial insemination through rectal palpation. Based on the analysis, grass to concentrate ratio, progesterone level and MUN level showed no significant difference (p>0.05) but it showed significantly different on milk yield (p<0,05). Based on pregnancy status, progesterone level at D22 showed significantly different whereas at D0 and D7 showed insignificantly different. MUN level and grass to concentrate level also showed significantly different. It can be concluded that the ratio of grass and concentrate did not increase MUN level, progesterone level and pregnancy rate.

Abstract

The quality of frozen semen is one of the components of the artificial insemination program. The freezing method, kind of diluent, handling of frozen semen, and thawing process all affect the quality of frozen semen. This study aimed to determine the motility, viability, and plasma membrane integrity of frozen semen of Friesian Holstein cattle at storage periods of 33, 30, 27, and 24 years. The samples used were stored for 33 years (production 1987), 30 years (production 1990), 27 (1993 production), and 24 years (1996 production) stored in Singosari Artificial Insemination Center, Malang. Data were statistically analyzed using One-Way ANOVA followed by Duncan's Multiple Range Test (p < 0,05). Based on the study's findings, it can be concluded that the motility, viability, and plasma membrane integrity of frozen semen that had been preserved for 33, 30, 27, and 24 years were still acceptable and in good condition in comparison to the SNI quality criteria for bull frozen semen in Indonesia.

Abstract

Foot and mouth disease (FMD) virus was reported as an outbreak in Indonesia in April 2022 and belonged to serotype O/ME-SA/Ind-2001e is spread in the country. This study aimed to detect the causative agent based on clinical symptoms in cattle that have the vesicle in the mouth and hooves. A total of 25 samples were collected during August 2022 from Lamongan and Surabaya, Indonesia. FMD was identified in 58% (7/12) using reverse transcription polymerase chain reaction (RT-PCR), respectively. The samples were performed using universal primers with 328 bp length for primary diagnosis of FMD. These findings indicate that the spread of FMD viruses is highly contagious, so rapid and accurate diagnosis is needed as an effort to control and monitor FMD viruses.

Abstract

Etawah crossbreed and Senduro goats as superior local goats and biological genetic resources that must be preserved in Indonesia. Melanocortin receptor type 4 (MC4R) is the primary gene that regulates food intake and energy balance which can be used as a candidate marker for livestock selection genes. This study aimed to analyze the comparison of cement quality and detect the profile of the MC4R gene in Etawah crossbreed and Senduro goat semen. The samples of this study were Etawah crossbreed and Senduro goat semen. Macroscopic examination of cement includes the evaluation of volume, consistency, odor, color, and pH. Microscopic examination of semen includes motility, abnormalities, and spermatozoa concentration. Examination of motility was performed in the condition of fresh semen, after the addition of diluents, and post-thawing of frozen straw. The polymerase chain reaction (PCR) method was performed to amplify specific fragments of the MC4R gene. There was a decrease in spermatozoa motility before freezing and post-thawing frozen semen compared to fresh semen. The quality of frozen straw post-thawing Etawah crossbreed and Senduro goat was still worth using for artificial insemination and in vitro embryo production. The MC4R gene can be detected in liquid and frozen semen from Etawah crossbreed and Senduro goats using a PCR test in the form of a single band along 642 bp.
