Abstract

Biodiversity assessment surveys are necessary for establishing conservation areas. However, such surveys are typically expensive, primarily if they cover a large area and take a long time. The survey difficulty increases when applied to cryptic, sparse, and fast-moving organisms. In addition, it requires expertise in taxonomic-biota classification. The breakthrough environmental DNA (eDNA) metabarcoding technique promises to overcome all the hurdles of assessing the potential for marine biodiversity in a non-invasive, rapid, extensive, and more effective way. We evaluated the ability of the eDNA survey to reveal the potential diversity and character of marine eukaryotes in the Lombok Island Marine Protected Area. A sampling of seawater and sediment eDNA in pore size fractions of 0.4-12 ?m and >12 ?m was carried out in the Core Zone, Non-Core Zone, and Non-Conservation Area, in east, north, and west Lombok, respectively. The detection and classification of eukaryotes using bioinformatics analysis were accomplished following extraction, amplification, and DNA sequencing. We identified 20,478 unique sequences of potential species classified in five kingdoms to 654 marine eukaryotes families. The comparison results show differences in community structure between locations, as well as differences in diversity between media and factions. The eDNA survey can assess marine biodiversity at a macro level and has implications for management in conservation areas.Keywords: Biodiversity, Marine eukaryotes, environmental DNA, the Primary V9-SSU 18S rRNA gene

Abstract

Sponges (Phylum Porifera) are a group of highly diverse, sessile, and filter-feeding basal metazoans, except spongillids. The majority of sponges are marine species that play an important role in benthic habitat by maintaining the stability of marine ecosystems through nutrient cycling, habitat provision, and bioerosion. However, marine sponges are not easily identifiable because of their lack of species-level distinctive morphological features, which limits efforts to monitor actual species biodiversity. Indonesia is home to approximately 850 identified species of marine sponges, and Raja Ampat archipelago of West Papua province is known for its exceptional marine biodiversity. Despite the species abundance, the exact number of sponges in the region is not well-documented due to the lack of specific studies providing comprehensive data on their diversity. Therefore, this study aimed to conduct a DNA barcoding analysis using the mitochondrial cytochrome oxidase I (COI) gene as a marker, combined with morphological analyses of 22 individuals collected in the waters of Waigeo, Mansuar, and Gam Islands in Raja Ampat. The results showed that 3 samples were identified at the species level (Halichondria sp. and Stylissa carteri) with good query cover and percent identity. This showed the possible presence of undescribed or cryptic species, suggesting a severe lack of reference data for both morphology and molecular analyses of marine sponges in the region. Consequently, the analysis showed the presence of a significant gap in the understanding of sponge biodiversity in Raja Ampat's waters.

Abstract

Halophila major is a new species with a wide distribution across various regions in Indonesia. The species was identified in 2020 through a combination of morphology and molecular approaches. Several studies have shown that it has a similar morphology to H. ovalis, which causes significant confusion during identification. Therefore, this study aims to describe the key identification and habitat of H. major and summarize seagrasses study opportunities based on the trend of published articles. A bibliometric analysis was used to summarize the habitat, morphometrics, molecular confirmation, and trend of seagrass topics in Indonesia. Based on the scientific articles, H. major has bigger morphometrics than H. ovalis. In addition, the number of paired and branching cross veins was reported to be an identification key of H. major. The species was also considered a deep ovalis found in 2-4 m depth. Genetically, the Internal Transcribed Spacer (ITS) gene marker was appropriate to showits phylogenetic tree. H. major was also classified into different clades with H. ovalis due to various factors. The results showed that genetics, tourism, and restoration were topics with the potential to be explored in the future. Abstract

The condition of the nest greatly determines the success of turtle egg hatching. The success of turtle egg hatching, both naturally and artificially, is influenced by several factors such as the location of the nest, the depth of the nest, vegetation conditions, environmental temperature, humidity and the surrounding environment. This study aims to see the effect of shade on the length of the incubation period and the success of green turtle egg hatching in semi-natural nests. This study was conducted at Pangumbahan Nesting Beach, Sukabumi. This study used an experimental method with the treatment of semi-natural nests placed in exposed areas and shaded areas with different depths, namely 68 (control), 30, 50, 70 and 90 cm. Measurements and observations of environmental conditions were carried out during the incubation period. Observations of hatching of hatchlings began on the 55th day of incubation and the dismantling of the entire nest was carried out on the 68th day of incubation. The results showed that in exposed areas there was no significant difference given the level of treatment on the length of the incubation period, while in sheltered or protected areas there was at least one difference in the level of treatment that had a different effect on the length of the incubation period. The fastest incubation period in exposed areas was at a depth of 30 cm and in sheltered areas was at a depth of 90 cm. Key words: incubation period, the green turtle, the success of hatching.

Abstract

Lamun memiliki fungsi ekologis penting sebagai penyerap karbon dioksida (CO2), yang dikategorikan sebagai bagian dari blue carbon atau karbon biru. Gas CO2 tersebut diserap oleh tumbuhan lamun dan dikonversi menjadi biomassa melalui proses fotosintesis. Estimasi cadangan karbon dalam biomassa lamun dapat dihitung melalui pendekatan kuantitatif yang melibatkan hubungan antara kerapatan tumbuhan, total biomassa, dan konsentrasi karbon dalam jaringan vegetatifnya. Penelitian ini dilakukan pada ekosistem lamun yang berada di wilayah perairan Desa Kahyapu dan Desa Kaana, Pulau Enggano, Provinsi Bengkulu. Pengumpulan data menggunakan metode purposive sampling dengan teknik transek kuadrat berukuran 1 m × 1 m, sedangkan analisis kandungan karbon dilakukan berdasarkan metode Walkey dan Black (1934). Tujuan utama dari penelitian ini adalah untuk menganalisis besaran cadangan karbon yang tersimpan pada biomassa lamun, baik pada komponen di atas substrat (daun) maupun di bawah substrat (akar dan rimpang). Hasil penelitian mengidentifikasi tiga spesies lamun, yaitu Enhalus acoroides, Thalassia hemprichii, dan Cymodocea spp. Di antara ketiganya, Enhalus acoroides menunjukkan nilai cadangan karbon tertinggi dengan rata-rata 50,75 g C/m2, diikuti oleh Thalassia hemprichii dengan rata-rata 17,53 g C/m2, dan Cymodocea spp. dengan rata-rata 2,99 g C/m2.

Abstract

Pulau Kelapa Dua, bagian dari Kepulauan Seribu, kaya akan keanekaragaman hayati dalam ekosistem mangrove dan lamun. Bivalvia berfungsi sebagai bioindikator yang penting untuk menilai kesehatan ekosistem ini. Penelitian ini bertujuan untuk menganalisis perbedaan keanekaragaman bivalvia antara lingkungan mangrove dan lamun dan mempelajari hubungan bivalvia pada kedua habitat ini. Data dikumpulkan melalui survei kepadatan mangrove dan lamun, pengukuran kualitas air, dan analisis sedimen. Metode transek garis dan plot digunakan untuk pengamatan mangrove, sedangkan metode Seagrass Watch diterapkan untuk pengamatan lamun. Sampel bivalvia dikumpulkan menggunakan pipa PVC (hingga kedalaman 10 cm) untuk individu di bawah permukaan dan survei visual untuk spesies yang terlihat di permukaan. Substrat berpasir di pulau ini mendukung berbagai spesies bivalvia. Sebanyak lima belas spesies bivalvia dari enam famili teridentifikasi: Tellinidae, Cardiidae, Veneridae, Lucinidae, Donacidae, dan Pinnidae. Kepadatan bivalvia secara signifikan lebih tinggi di ekosistem lamun (1.029 ind./100 m²) dibandingkan dengan ekosistem mangrove (366 ind./100 m²), dengan famili Tellinidae menunjukkan kepadatan tertinggi (664 ind./100 m²). Indeks ekologi menunjukkan kondisi yang stabil, dengan komunitas bivalvia yang beragam dan tersebar merata, mencerminkan lingkungan yang sehat. Analisis korespondensi mengungkapkan bahwa spesies bivalvia lebih terkonsentrasi di stasiun yang merepresentasikan ekosistem lamun, menegaskan hubungan yang lebih erat dengan habitat lamun.

Abstract

Terumbu karang merupakan ekosistem laut yang memiliki peran penting dalam menjaga biodiversitas dan ketahanan lingkungan pesisir. Namun, dalam beberapa dekade terakhir, degradasi terumbu karang terus meningkat akibat aktivitas antropogenik dan perubahan iklim. Penelitian ini bertujuan untuk menganalisis Indeks Kesehatan Karang (Coral Health Index / CHI) di Kawasan Konservasi Pulau Pieh dan perairan sekitarnya dalam periode 2021-2024 menggunakan pendekatan biofisik dan statistik. Analisis dilakukan terhadap 16 stasiun pengamatan, menggunakan data parameter bentik (tutupan karang hidup, resiliensi) dan biomassa ikan karang yang diperoleh melalui metode Underwater Photo Transect (UPT) dan Underwater Visual Census (UVC). Hasil penelitian menunjukkan bahwa nilai CHI berkisar antara 3 hingga 10, dengan beberapa stasiun menunjukkan tren peningkatan sementara lainnya mengalami stagnasi atau penurunan. Analisis Spearman correlation mengindikasikan bahwa parameter kualitas air (pH, DO, suhu, dan salinitas) tidak memiliki hubungan signifikan terhadap CHI pada semua tahun pengamatan (p-value > 0,05). Analisis Principal Component Analysis (PCA) menunjukkan bahwa faktor kualitas air bukan penentu utama perubahan CHI, tetapi tekanan ekologis dan aktivitas antropogenik lebih berperan dalam menentukan kondisi kesehatan terumbu karang. Hasil penelitian ini memberikan rekomendasi untuk pengelolaan berbasis ekosistem guna menjaga keberlanjutan ekosistem terumbu karang.Coral reefs are marine ecosystems that play a crucial role in maintaining biodiversity and coastal environmental resilience. However, in recent decades, coral reef degradation has continued to increase due to anthropogenic activities and climate change. This study aims to analyze the Coral Health Index (CHI) in the Pieh Island Conservation Area and its surrounding waters from 2021 to 2024 using biophysical and statistical approaches. The analysis was conducted at 16 observation stations using benthic parameters (live coral cover, resilience) and reef fish biomass data collected through the Underwater Photo Transect (UPT) and Underwater Visual Census (UVC) methods. The results indicate that CHI values range from 3 to 10, with some stations showing an increasing trend while others experience stagnation or decline. Spearman correlation analysis indicates that water quality parameters (pH, DO, temperature, and salinity) do not have a significant relationship with CHI in all observation years (p-value > 0.05). Principal Component Analysis (PCA) shows that water quality factors are not the main determinants of CHI changes; instead, ecological pressure and anthropogenic activities play a more significant role in determining coral reef health. This study provides recommendations for ecosystem-based management to ensure the sustainability of coral reef ecosystems.

Abstract

Terumbu karang adalah ekosistem purba, megah, dan tinggi produktivitas serta keanekaragamannya (Thomas dan Raymond, 2008). Terumbu karang bertumbuh setiap tahun dengan ukuran yang berbeda-beda tergantung pada kondisi lingkungan sekitar (Arman et al., 2013). Penelitian mengenai pertumbuhan terumbu karang sudah banyak dilakukan, namun prediksi pertumbuhan karang menjadi topik yang menarik dan belum banyak diteliti. Penelitian ini bertujuan untuk mengembangkan model pertumbuhan terumbu karang berdasarkan lingkar tahunan dengan metode logika fuzzy. Suhu permukaan laut (SPL) adalah salah satu faktor kunci yang mempengaruhi pertumbuhan terumbu karang (Westmacott et al., 2000; Sterr, 2001; Koontanakulvong, 2008; Thomas dan Raymond, 2008; Arman et al., 2013; Lalang, 2015). Analisis Indian Ocean Dipole (IOD) menemukan lima fenomena, ditandai oleh kenaikan SPL yang cukup signifikan. Lima fenomena tersebut terjadi pada tahun 1966-1967 (+IOD), 1969-1970 (-IOD), 1973-1974 (-IOD), 1983-1984 (-IOD), serta tahun 1987-1988 (+IOD). Hasil penelitian menunjukkan bahwa lingkar tahunan Porites lutea di stasiun sisi utara Pulau Tunda polanya mengikuti fluktuasi SPL. Stasiun di selatan Pulau Tunda yang berhadapan langsung dengan Pulau Jawa, polanya tidak mengikuti pola SPL. Stasiun di utara berbatasan langsung dengan perairan terbuka dan tidak terlindung dari pulau-pulau di sekitar. Simulasi pertumbuhan menunjukkan bahwa terumbu karang akan mampu beradaptasi terhadap perubahan iklim. Pada tahun 2085, terumbu karang bertumbuh dari -0,75 cm dan bergerak naik menjadi 0,1 cm di tahun 2100. Berdasarkan proyeksi SPL pada rentang tahun 1900-2100, anomaly SPL akan terus naik hingga mencapai 0,45 °C.
