Abstract

Latar Belakang: Coronavirus Disease 2019 (COVID-19) merupakan penyakit yang disebabkan oleh severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), yang dilaporkan pertama kali di Wuhan Tiongkok pada 31 Desember 2019. Pada tanggal 11 Maret 2020, peningkatan jumlah kasus dan mortalitas secara drastis diberbagai negara membuat WHO memutuskan COVID-19 sebagai pandemi global. Setiap hari jumlah kasus baru, sembuh, dan meninggal COVID-19 terus diperbarui. Saat ini, jenis kelamin dan usia merupakan standar data epidemiologi COVID-19 yang ditetapkan di dunia dan dijadikan faktor prognosis dari pasien COVID-19. Objektif: Penelitian ini bertujuan untuk menganalisis hubungan jenis kelamin dan usia pasien positif COVID-19 terhadap tingkat kecepatan konversi di Kota Padang pada tahun 2020. Metode: Penelitian ini merupakan penelitian analitik dengan pendekatan cross sectional menggunakan data retrospektif. Teknik pengambilan sampel adalah proportional stratified random sampling. Penelitian ini dilakukan dengan mengambil data pasien terkonfirmasi positif COVID-19 yang telah mendapatkan 2 kali hasil negatif tes RT-PCR di Kota Padang selama tahun 2020 yang diambil dari master tabel Laboratorium Diagnostik dan Riset Terpadu Penyakit Infeksi Fakultas Kedokteran Universitas Andalas. Penelitian ini dilaksanakan pada bulan Juni sampai November 2021 dengan jumlah sampel 382 responden. Analisis data yang digunakan berupa analisis univariat dan analisis bivariat menggunakan uji Chi-square. Hasil: COVID-19 lebih banyak terjadi pada perempuan (52,9%). Kejadian terbanyak COVID-19 dialami oleh kelompok usia 26-45 tahun (43,5%%). Sebagian besar pasien COVID-19 mengalami konversi dalam kurun waktu <15 hari (86,1%). Tidak terdapat hubungan bermakna antara jenis kelamin pasien positif COVID-19 dengan kecepatan konversi (p= 0,454). Tidak terdapat hubungan bermakna antara usia pasien positif COVID-19 dengan kecepatan konversi (p=0,065). Kesimpulan: Jenis kelamin dan usia pada pasien COVID-19 tidak memiliki hubungan bermakna dengan kecepatan konversi.

Abstract

Human Papillomavirus (HPV) berkaitan dengan penyebab terjadinya kanker  serviks. Salah satu HPV yang beresiko tinggi adalah tipe 16. Protein virus yang beresiko tinggi yang menyebabkan karsinogen salah satunya gen E7. Dikarenakan tingginya tingkat kematian yang di sebabkan oleh kanker servik, salah satu cara untuk mengatasinya adalah dengan cara vaksinasi. Protein E7 merupakan target ideal untuk dapat dijadikan sebagai dasar dalam pembuatan vaksin. Dimana hasil ekspresi gen E7 HPV tipe 16 yang selanjutnya dapat dimanfaatkan dalam membuat antigen spesifik dari gen E7. Proses kloning gen E7 HPV tipe 16 dari peneliti sebelumnya, Isolasi plasmid menggunakan kit (Geneaid®), kultur bakteri yang telah ditumbuhkan pada plat yang mengandung antibiotik. Bakteri yang tumbuh merupakan bakteri yang telah mengandung gen sisipan. Selanjutnya lakukan over ekspresi. Hasil over ekspresi di tambah IPTG kemudian hasilnya di visualisasikan dengan SDS PAGE. Over ekspresi dari gen E7 HPV tipe 16 berhasil dilakukan dengan terdapatnya pita dari protein dari gen E7 HPV tipe 16 dari hasil SDS-PAGE. Dari penelitian yang telah dilakukan di dapat  hasil over ekspresi gen E7 HPV tipe 16 pada ukuran protein dengan kisaran 11,0 kDa. Pita yang didapat terlihat tipis dan samar-samar

Abstract

Objective: This study aimed to determine the description of the coagulation marker and how it implicates COVID-19 patients based on clinical outcomes in the form of enhancement severity and death. Method: The subjects were collected from patients with positive COVID-19 RT-PCR with medical records including demographic data, and laboratory results such as hemoglobin, hematocrit, leukocytes, platelets, coagulation markers (D-dimer, PT, and APTT), and patients outcomes post-treatment. Statistical analysis is performed by using chi-square and Mann-Whitney tests were considered statistically difference level of significant between variables. Result: The COVID-19 patients is classified into mild group with 27 subjects  (44.2%) and moderate-severe group 34 subjects (55.8%). Leukocytes, D-dimer, and PT levels significantly differed between mild and moderate-severe groups. All coagulation markers had a symptomatic effect on Covid-19 patients, increased D-dimer and prolonged prothrombin time in adult COVID-19 patients tend to be followed by the enhancement of severe symptoms and death

Abstract

Objective: This study aims to map the distribution of the Aspartate 614 to Glycine (Asp614Gly) mutation in the spike protein of the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) circulating in West Sumatra, to understand the extent of its spread within the local population. Methods: RNA samples were taken from 98 confirmed COVID-19 cases across various area in West Sumatra. Genetic amplification and next-generation sequencing (NGS) were performed to identify the presence of the Asp614Gly mutation. The frequency of mutations was analyzed using bioinformatics software. Results: Out of the 98 samples analyzed, approximately 97% exhibited the Asp614Gly mutation. The geographic distribution of this mutation showed significant variation among the districts of West Sumatra, with the highest prevalence in urban areas. Conclusion: These findings indicate that the Asp614Gly mutation is highly prevalent in West Sumatra. Associated with increased virus transmissibility, this mutation dominates the circulating virus strains in the region, highlighting the need for ongoing genetic surveillance to guide public health interventions and vaccination strategies.

Abstract

Introduction. Coronavirus Disease 2019 (COVID-19) is caused by Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) which in 2020 was declared a global pandemic. In the management of COVID-19, patients are classified according to the severity. Identifying prognostic factors at an early stage helps assess severity risks. Abnormal hematological parameters were present in COVID-19 patients such as the elevation of neutrophil lymphocyte ratio (NLR) and D-dimer patients. This study aimed to determine the NLR and D-dimer levels based on the severity of COVID-19 patients. Methods. This study was an observational analytical study that used a case-control approach using 100 medical records of COVID-19 patients who were treated at Semen Padang Hospital in 2021. Consecutive sampling was used in this study. ANOVA test and the Kruskal-Wallis test were used for bivariate analysis. The results were considered significant if the p-value <0.05. Results. Characteristics of patients treated for COVID-19 were: 51% male and 49% female, mean age 57.79 (SD 13.5) years. NLR values based on the severity (mild, moderate, severe, and critical) were 3.12 (SD 2.04), 3.51 (SD 2.87), 6.89 (SD 3.6), 12.57 (SD 10.34) respectively; p<0.05. The median value of D-dimer (ng/mL) based on the severity (mild, moderate, severe, and critical) were 444, 791, 1,610, 2,135; p<0.05. The results of this study showed that there were significant differences in the NLR values and D-dimer levels based on severity. Conclusion. RNL value and D-dimer levels increase the most in the critical degree group and shows a significant relationship in COVID-19 patients.

Abstract

Background: Polycystic Ovary Syndrome (PCOS) is a disorder characterized by hyperandrogenism, ovulatory dysfunction, and morphological features of polycystic ovaries. It is associated with insulin resistance and hyperinsulinemia. Gut microbial dysbiosis plays a role in PCOS, which is associated with insulin resistance, hyperandrogenism, chronic inflammation, and metabolic syndrome. PCOS treatment with Probiotics maintains the stability and diversity of the gut microbiome. Objective: To determine the comparison of Lactobacillus plantarum, Lactobacillus mucosae, and Lactobacillus farciminis Bacteria Between Polycystic Ovary Syndrome and Non Polycystic Ovary Syndrome Students at the Medical Faculty of Andalas University. Method: This research was an analytic observational study using a cross-sectional method conducted on female students with Polycystic Ovary Syndrome and Non-Polycystic Ovary Syndrome at the Medical Faculty of Andalas University. Results: Bivariate analysis of microbiota comparison of female students with PCOS and without PCOS, Mann-Whitney Test on L.mucosae (p=0.774), while Independent T-Test analysis on L.farciminis (p=0.006). Conclusion: There is a decrease in microbiota L. Mucosae in female students with PCOS, but there is no significant difference. There is a significant comparison in the number of microbiota L.farciminis in female students with and without PCOS at FK Unand.

Abstract

Elderly individuals are the most vulnerable group affected by Covid-19. Anxiety among the elderly regarding Covid-19 is an important mental health factor that needs to be addressed. There are obstacles to meeting the basic needs of the elderly, so they require family assistance. The experience of families in caring for elderly individuals exposed to Covid-19 is very useful for obtaining information related to their care. The purpose of this study is to gain in-depth information about the experiences of family members when accompanying elderly individuals exposed to Covid-19 during isolation in hospitals. This research is qualitative, with a phenomenological approach. Participants consisted of 11 family members selected through purposive sampling, and in-depth interviews were conducted for data collection using five open-ended questions recorded via audio. Data analysis employed content analysis. The findings revealed seven themes: 1) Knowledge of caregivers about the purpose of assisting elderly Covid-19 patients during hospital isolation, 2) Family feelings when their parent is diagnosed with Covid-19, 3) The condition of the elderly after being diagnosed with Covid-19, 4) Activities of caregivers while accompanying elderly Covid-19 patients, 5) Moments when nurses motivate elderly patients with Covid-19, 6) Challenges faced by caregivers during the isolation of the elderly in hospitals, 7) Expectations for caregivers to assist elderly Covid-19 patients during hospital isolation. It is crucial to explore the experiences of families in supporting elderly Covid-19 patients during hospital isolation to provide psychological comfort, which can accelerate their recovery. The presence of family support for elderly Covid-19 patients not only alleviates anxiety and fear but also fosters hope and motivation when they learn of their positive Covid-19 status, especially if they have underlying health conditions. Positive and impactful activities by family members during isolation include continuous communication with the elderly, providing motivation for their recovery, monitoring their nutritional intake, praying, watching shows together, and communicating through phone calls or video calls with other family members at home.

Abstract

ABSTRACT — Background: Laryngopharyngeal reflux (LPR) is the backflow of gastric and or duodenal fluid into the larynx, pharynx, trachea, and bronchi. The prevalence of LPR is difficult to determine due to the limited gold standard and the large variety of LPR symptoms. Damage can occur due to the decrease in pH value and also because of exposure to harmful enzymes in reflux, including bile acid. Purpose : This study was conducted to analyze bile acid levels in the saliva of LPR patient and non LPR subject. Methods:  This study is an observational study with a case-control design. The study was conducted in the ORL-HNS Department of Dr. M. Djamil Hospital, Padang, West Sumatra, Indonesia. The total sample size was 44 people. We enrolled 22 healthy subjects as the control group and 22 patients suspected of having LPR. Result: LPR patients are more common in women than in men, with 12 women and 10 men. Bile acid in the LPR group means of is 25.08±7.67µM, meanwhile, in the healthy group, the mean was 18.99±8.26 µM. There is a statistically significant in the incidence of LPR with the bile acids (p = 0.015) based on t- independent test. Conclusion: Our study confirmed that bile acids in saliva play a major role in diagnosing LPR.

Abstract

Infertilitas adalah sebuah permasalahan sistem reproduksi yang digambarkan dengan kegagalan untuk memperoleh kehamilan setelah 12 bulan atau lebih melakukan hubungan seksual 2-3 kali seminggu secara teratur tanpa menggunakan alat kontrasepsi. Salah satu faktor yang mempengaruhi infertilitas yakni umur dan pendidikan. Penelitian ini bertujuan untuk mengetahui hubungan umur dan pendidikan dengan infertilitas pada wanita pasangan usia subur. Penelitian ini merupakan penelitian studi analitik dengan pendekatan case control dilakukan di labor mikrobiologi Fakultas Kedokteran Universitas Andalas pada bulan Juni hingga November 2019. Sampel pada penelitian ini yakni wanita pasangan usia subur infertilitas dan fertilitas yang berkunjung ke labor mikrobiologi serta dibagi menjadi kelompok kasus dan kontrol sesuai kriteria inklusi sebanyak 33 sampel diambil secara consecutive sampling. Data dikumpulkan menggunakan kuesioner dan hasil pemeriksaan. Hasil penelitian menunjukkan bahwa hubungan umur dan pendidikan dengan kejadian infertilitas pada wanita pasangan usia subur(PUS).

Abstract

The spread of the virus caused by SARS-CoV-2 has been designated as an extraordinary event so that the government to speed up the collection and management of this disease then appoints several hospitals that serve as referral hospitals for people who want to check themselves. One of the hospitals used as a reference by the government in West Sumatra is Andalas University hospital. In just one day, the hospital received thousands of samples for examination. The number of samples to be examined and the limited number of administrations that conduct data collection results in the length of the sample collection process. This article aims to design and implement an information system in the data collection of Swab Covid-19 using waterfall model system development methods. The requirement testing model is used for testing a web-based Covid 19 swab data information system whether it is in accordance with system users' needs analysis and design.
