Abstract

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by cognitive decline, with the accumulation of β-amyloid (Aβ) plaques playing a key role in its progression. Beta-Secretase 1 (BACE1) is a crucial enzyme in Aβ production, making it a prime therapeutic target for AD treatment. However, designing effective BACE1 inhibitors has been challenging due to poor selectivity and limited blood-brain barrier permeability. To address these challenges, we employed a machine learning approach using Support Vector Regression (SVR) in a Quantitative Structure-Activity Relationship (QSAR) model to predict the inhibitory activity of potential BACE1 inhibitors. Our model, trained on a dataset of 7,298 compounds from the ChEMBL database, accurately predicted pIC50 values using molecular descriptors, achieving an R² of 0.690 on the testing set. The model's performance demonstrates its utility in prioritizing drug candidates, potentially accelerating drug discovery. This study highlights the effectiveness of computational approaches in optimizing drug discovery and suggests that further refinement could enhance the model’s predictive power for AD therapeutics.

Abstract

The increased intensity of rainfall is becoming one of the most pressing climate-related issues in many parts of the world. Detecting the factors that affect rainfall intensity requires a combination of modern technologies, such as weather satellites, radar systems, and advanced atmospheric models. Extreme conditions (outliers) often occur. This study aims to model data that is not symmetric or contains outliers. This study examines and models quantile regression on daily rainfall intensity in Jakarta which has extreme rainfall events. The results of the study found that the extreme values in the daily rainfall intensity data in Jakarta are outliers and the assumptions on modeling using linear regression are not satisfied so that the characteristics of the parameter estimator based on OLS do not have BLUE characteristic. In modeling with quantile regression using six quantiles 0.25, 0.50, 0.75, 0.95, 0.99, and 0.9999 with consideration of these quantile values representing all parts of the data distribution including extreme values, it was found that the factors affecting rainfall intensity in Jakarta are different in each rainfall intensity condition. The best model is shown by quantile 0.999 with a coefficient of determination of 58.21%. Based on the best model, it is known that the factors affecting extreme rainfall are maximum temperature, dew point temperature, air humidity, wind speed, air pressure, and length of irradiation. This study indicates that quantile regression can provide a more detailed insight into how these variables affect rainfall intensity in various rainfall conditions ranging from low rainfall to extreme rainfall.

Abstract

Background: Gestational diabetes mellitus increases the risk of a number of short-term and long-term health problems in both mother and baby. Some evidence states that one of the causes of gestational diabetes mellitus is vitamin D deficiency. This study aimed to estimate the magnitude of the influence of vitamin D deficiency on the incidence of gestational diabetes mellitus, through a meta-analysis of previous primary studies. Subjects and Method: This was a systematic review and meta-analysis with the following PICO, population: pregnant women, intervention: vitamin D deficiency, comparison: no vitamin D deficiency, outcome: gestational diabetes mellitus. The articles used in this study were obtained from four databases, namely PubMed, Google Scholar, Science Direct, and Springer Link, using the search keys "Vitamin D Deficiency"[Mesh] OR “Vitamin D Deficiency”[tw] AND "Diabetes, Gestational" [Mesh] OR “Gestational Diabetes Mellitus”[tw] OR “Gestational Diabetes”[tw]. The included article is a full-text cohort study design from 2018 to 2022 and reports the adjusted odds ratio (aOR) in a multivariate analysis. The selection of articles is done using PRISMA flow diagrams. Articles were analyzed using the Review Manager 5.3 application. Results: A total of 9 cohort studies involving 42,972 pregnant women from the continents of Asia, Australia and Europe were used in the meta-analysis. Pregnant women with vitamin D deficiency had a risk of gestational diabetes mellitus 1.87 times compared to those without vitamin D deficiency (aOR= 1.87; 95% CI= 1.27 to 2.75; p= 0.002). Conclusion: Vitamin D deficiency increases the risk of developing gestational diabetes mellitus. Keywords: vitamin D deficiency, gestational diabetes mellitus, pregnancy Correspondence:Dian Handayani. Masters Program in Public Health, Universitas Sebelas Maret. Jl. Ir.
