Abstract

The purpose was to determine the chemical content (water, protein, fat, ash, and carbohydrate) and sensory characteristics of Herbal Chicken Essence (HCE), a functional drink containing chilcken extract, from Indonesia. The experimental design was factorial and completely randomized with two replicates. There are four formulas in this study, namely F1 (broiler chicken and coconut sugar), F2 (broiler chicken and coconut sugar), F3 (IPB D1 chicken and palm sugar), and F4 (IPB D1 chicken and coconut sugar). The chemical content data were analyzed using a two-way ANOVA. Panelists used in the sensory tests included 30 participants. There was a significant effect (p<0.05) of chicken and sugar substitution on water, ash, protein, and carbohydrate contents. Formula 2 (broiler chicken and coconut sugar) is the most preferred in almost all sensory attributes (aroma, taste, mouthfeel, aftertaste, and overall acceptance) because the sour taste found in coconut sugar is able to suppress the fishy taste found in HCE. The substitution of the chicken breed and sugar in the HCE formulations affected the chemical content and sensory characteristics

Abstract

Diare pada Macaca fascicularis sangat mengganggu proses penangkaran, diare mengurangi kualitas hidup kera, E. coli sering dilaporkan sebagai penyebab utama diare pada beberapa primata non-manusia. Tujuan dari penelitian ini adalah untuk mendeteksi faktor virulensi seperti hemolisin, pola hemaglutinin, dan hidrofobisitas permukaan sel. Penelitian ini menggunakan 30 isolat E. coli dari koleksi pusat penelitian primata IPB University yang diisolasi dari kera ekor panjang yang menderita diare. Faktor virulensi seperti hemolisin, hemaglutinasi, dan agregasi garam dideteksi sesuai protokol standar. Sebanyak 30 isolat M. fascicularis yang menderita diare diuji kemampuannya menyebabkan lisis pada eritrosit, di antara 24 (80%) sampel E. coli menunjukkan α -hemolisin pada agar darah dan 2 (6,67%) sampel menunjukkan γ-hemolisis. Sebanyak 21 (70%) sampel E. coli mampu menghemaglutinasi eritrosit sapi dan 27 (90%) sampel mampu menghemaglutinasi eritrosit kelinci. Sebanyak 30 sampel E. coli tidak mampu menghemaglutinasi eritrosit ayam, kucing, anjing, dan manusia. Sebanyak 2 (6,67%) sampel menunjukkan hasil positif pada uji agregasi garam di antara 30 sampel E. coli dari M. fascicularis dan sisanya 28 (93,33%) sampel tidak menunjukkan hasil positif pada uji agregasi garam. Kesimpulan penelitian adalah Sebanyak 30 isolat E. coli menunjukkan variasi faktor virulensi yang berperan dalam mekanisme diare pada Macaca fascicularis. Terdapat 24 isolat (80%) yang menunjukkan α-hemolisin, 2 isolat (6,67%) menunjukkan γ-hemolisin, dan 4 isolat lainnya (13,33%) tidak menunjukkan aktivitas hemolisis. Selain itu, 29 isolat (96,7%) menunjukkan hemaglutinasi terhadap eritrosit sapi dan/atau kelinci, sedangkan 2 isolat (6,7%) bersifat hidrofobik berdasarkan uji salt aggregation.

Abstract

Colistin is the last drug of choice for dealing with carbapenem-resistant Enterobacteriaceae bacteria; hence, this drug is very crucial to human health. The discovery of a plasmid-mediated colistin-resistant gene, the mobilized colistin resistance-1 (mcr-1), signals a significant global health threat. Colistin sulfate is an antimicrobial agent which has been approved for use in broilers in Indonesia. Thus, this study aimed to measure the prevalence of colistin-resistant Escherichia coli and to detect the mcr-1 colistin-resistant gene in E. coli and E. coli O157:H7 in the entire supply chain of broilers in Bogor Regency, West Java Province, Indonesia. Samples were taken from 47 flocks that used colistin sulfate (47 pools of cloacal swabs, 47 pools of drinking water, and 47 pools of litter), seventy fresh meat samples and seven plucker swab samples from seven small-scale poultry slaughterhouses, seventy fresh meat samples from seven traditional markets, and seventy cooked meat samples from seven small restaurants. The isolation of E. coli was done on each of the 358 samples, and 493 isolates were obtained. All the E. coli isolates were then tested for their susceptibility to colistin sulfate by using the agar dilution method. The detection of the mcr-1 gene from the colistin-resistant isolates (minimum inhibitory concentration > 2 µg/mL) was conducted using the polymerase chain reaction (PCR). The prevalence value of colistin-resistant E. coli in all the isolates was 11.76% (CI 95%; CL 9.21–14.91), and the prevalence of the mcr-1 gene was 10.55% (CI 95%; CL 8.13–13.57%). A very good agreement correlation existed between the colistin-resistant phenotype and the mcr-1 gene (κ = 0.939). The mcr-1 gene was found in 89.66% of colistin-resistant E. coli isolates. Two colistin-resistant and mcr-1 carrying isolates were identified as E. coli O157:H7 serotype. This research was the first study attempt on the mcr-1 gene in Indonesia, covering the entire supply chain of broiler meat from farms to consumers. The results indicated the necessity to reduce the use of colistin sulfate in broiler management and to improve biosecurity measures, not only in farms but also in the entire supply chain of broiler meat production.
