Abstract

This research aimed to determine the profiles of total leukocytes and neutrophil -lymphocyte ratio (N/L) in pregnant etawah crossbreed goats. Six etawah crossbreed goats around 3-6 years old and body weight around 30-50 kg were used in this study. Blood samples were taken from jugulars vein in non pregnant (K), and at gestational age 12, 14, 16 and 18 weeks. Blood is inserted into a tube that containsEDTA (Ethylene Diamine Tetraacetic Acid) anticoagulant to obtain whole blood. Whole blood were analyzed to total leukocyte cells and leukocyte types. The results showed that total leukocytes profiles tend to increase during pregnancy. Lymphocytes stable until 16 weeks of pregnancy and tends to decrease at the end of pregnancy. The N/L ratio increase at the end of pregnancy, this condition indicates an increase in the number of neutrophils accompanied by decreased lymphocyte cells and can be found in etawah crossbreedat 18 weeks of gestation.

Abstract

Toll-like Receptor 4 (TLR4) gene is one of the genes that control resistance of chickens againstSalmonella sp. through non-specific immune response. These gene can be used as genetic markers inIndonesian local chickens, if known its diversity. Therefore this research was aimed at evaluating thegenetic polymorphism of TLR4 gene in several types of Indonesian lokal chickens (kampung, kampungpelungcrossbreed, sentul, and tolaki chicken) using PCR-RFLP. The total of samples were 136. There werethree stages of identification : extraction of DNA genom, PCR amplification of TLR4 gene (with size 220 bpon exon 2), and RFLP method using restriction enzym (MscI). The data were analyzed include frequency ofallele, frequency of genotype, heterozygosity values , and value of Polymorphic Informative Content (PIC).The result showed that TLR4|MscI was found polymorphic in all kind of chickens that was genotyped. Itwere found two alleles (A and G), and three genotypes (AA, AG, and GG). The value of x2 showed thatTLR4|MscI was in Hardy-Weinberg equilibrium. The value of Ho and He were 0,11-0,41 and 0,11-0,35. Thevalue of PIC (0,10-0,29) included in the category of low to moderately high. These results declared thatTLR4|MscI gave hope for used as genetic markers in resistance to Salmonella sp. infection in Indonesianlocal chickens.

Abstract

To our knowlege, there is no rapid agglutination test to detect antibodies to viruses which might be due to the small dimension of viral particles. Through complex formation of Staphylococcus aureus bearing protein A-rabbit IgG-anti IgY-IgY anti Marek virus, agglutination of antibodies to viruses can be achieved and visualized. To design the prototype of the test, the bacterial cells of Staphylococcus aureus were coupled to a complex compound consisting of IgG-IgY-Marek antigen. This protocol was able to detect clearly the presence of Marek antibody in chicken sera, showing the rapid, clear and distinct agglutination reaction on the glass objects. No agglutination reaction was observed in the reaction of specified pathogen free chicken sera with this prototype showing the specificity of the test. This finding demonstrates a novel rapid agglutination which can be used for the detection of antibodies to various agents.

Abstract

Latar Belakang. Kandungan zat gizi extra virgin olive oil (EVOO) memiliki banyak manfaat untuk kesehatan manusia. Beberapa manfaat antara lain sebagai imunomodulator, mencegah penyakit jantung dan vaskuler lainnya, mencegah penyakit alergi, memperbaiki fungsi liver, dan mencegah penyakit lainnya. Kandungan zat gizi tiap merek EVOO tidak sama, ditentukan oleh banyak faktor. Tujuan. Penelitian ini bertujuan menganalisis klaster berdasarkan karakteristik kimiawi, kandungan zat gizi, dan senyawa aktif pada tujuh sampel produk EVOO dari supermarket. Metode. Desain deskriptif digunakan dalam penelitian ini. Pengumpulan tujuh sampel produk EVOO di supermarket wilayah Daerah Khusus Ibukota (DKI) Jakarta dilakukan pada bulan Maret 2019. Pemeriksaan sampel produk EVOO dilakukan di Laboratorium Balai Besar Industri Agro (BBIA) Kementerian Perindustrian dan Laboratorium Biofarmaka dan Lembaga Penelitian Pengabdian Masyarakat (LPPM) Institut Pertanian Bogor (IPB). Analisis yang digunakan adalah analisis klaster melalui pendekatan hierarchical method. Hasil. Berdasarkan karakteristik kimiawi, ditemukan bahwa sampel produk EVOO ke-6 memiliki karakteristik kimiawi paling berbeda. Sampel produk EVOO ke-4 memiliki kandungan asam lemak jenuh paling berbeda. Kandungan asam lemak tak jenuh pada sampel produk EVOO ke-1, 3, dan 7 mendekati kesamaan. Kandungan vitamin E pada sampel produk EVOO ke-2 dan kandungan zat besi pada sampel produk EVOO ke-6, berbeda dengan sampel produk EVOO lainnya. Kandungan total flavonoid pada sampel produk EVOO ke-2, 3, dan 4, memiliki kadar mendekati kesamaan. Kandungan total karotenoid pada sampel produk EVOO ke-2 dan ke-6 juga memiliki kadar mendekati kesamaan. Kesimpulan. Karakteristik kimiawi pada semua sampel produk EVOO yang ditemukan dalam penelitian memiliki nilai hampir sama. Sampel produk EVOO di supermarket memiliki perbedaan kandungan asam lemak tak jenuh, total flavonoid, dan total karotenoid.

Abstract

Black Soldier Fly larvae are natural antibiotics. The use of methanol extract in BSF larvae has a dual function, namely high protein content and high antibiotic content to kill Gram-negative bacteria. The biological structure of BSF larvae with antimicrobial peptide (AMP) acts as an inhibitor of pathogenic microorganisms, has a high lauric acid function as a natural antimicrobial and chitin, the polysaccharide plays a role in enhancing the immune response of the animal. This study aims to gain the best type of BSF protein from feed treatments containing live BSF, dry BSF, and BSF extract to increase blood productivity and hematology. The results of this study show that the best P3 treatment for the productivity of laying hens at the age of 18 to 26 weeks with the use of BSF extract since it contains the highest protein compared to other treatments. Productivity variables show the best P3 treatment, each egg weight 57.17 g bird-1, daily egg production 90.88%, egg mass 2408.16 g bird-1 and ration conversion 2.0. The mean hematology of the blood showed the highest P3 treatment, each erythrocyte 2.91±0.13 x 106 mm-3, leucocytes 17.69±3.68 x 103 mm-3, hematocrit 27.8±1.14%, hemoglobin 9.96±1.12%, MCV 95.77±4.41 fl, MCHC 35.99±5.21%, MCH 34.38±4.53 pg, lymphocytes 61.2 ± 4.86%, heterophile 31.2±2%, monocytes 5±0.62% and ratio H/L 0.51±0.04.

Abstract

This study aimed to identify genes encoding tetracycline and plasmid-mediated quinolones resistance to Salmonella spp from Poultry Farm in Bandung and Purwakarta, West Java. A total of 70 samples were collected from poultry farm In Bandung and Purwakarta, West Java. All isolates were test by selective media (SSA) and confirmation Salmonella with PCR. Thirty three isolate positive from selective media Salmonella Shigella Agar (SSA) and 21 isolat was confirmed as Salmonella spp by PCR. Twenty one isolate isolated were tested for tetracycline, doxycicline,, nalidixic acid, oxytetracycline, enrofloxacyn using disk diffusion method. TE-resistant were screened for presence of tet(A) and tet(B) genes by single polymerase chain reaction (PCR). The qnr(A), qnr(B) and qnr(S) genes were detected by multiplex PCR in quinolone resistant Salmonella isolates. The result of antibiotic sensitivity test showed that resistance to ampicillin (95.2%), tetracycline (100%), oxytetracycline (95.2%), nalidixic acid (90.4%), eritromisin (85.7%), enrofloxacin (76.2%), Gentamisin 47.6%, chloramphenicol (38.1%). The distribution of antibiotics-resistance genes in the Salmonella isolates included ampC (95.2%), tet(A)(61.9%), tet(B)(38.1%), qnr(A)(28.5%), qnr(B)(14.3%) and qnr(S)(23.8%).This study shows that a few pathogens of Salmonella are resistant to ampicillin, tetracycline , and quinolone. The tet and qnr genes are responsible for this resistance among Salmonella in Bandung and Purwakarta, West Java Indonesia was high.

Abstract

The aim of this study was to determine the pattern of the spread of antibiotic-resistant E. coli on pig farms in Kupang City by using mapping analysis. Data on E. coli resistance comes from the results of laboratory analysis and as secondary data for spatial analysis. Spatial analysis uses the nearest neighbour index, convex hull and elementary analysis of disease methods. The results of this study have shown a high prevalence of E. coli (85.4%) with clustered distribution patterns and have a wide spread (10920 ha) in the area in Kupang City. MDR E. coli had a moderate prevalence (57.31%) by forming a group spread pattern and a wide spread (7778 Ha) on pig farms in Kupang City. This result encourages prevention and control efforts by all interested parties so that the rate of spread of resistant E. coli can be reduced.

Abstract

Enterococcus faecalis merupakan patogen oportunistik yang membentuk biofilm dengan menghasilkan gen virulen seperti gelE. Penelitian ini bertujuan untuk mengeksplorasi keberadaan gelE dan mengkaji korelasinya terhadap pembentukan biofilm pada isolat E. faecalis asal ayam. Sampel yang digunakan sebanyak 60 sampel arsip usap kloaka ayam. Isolasi dan identifikasi bakteri menggunakan media agar selektif diferensial KF Streptococcus. Konfirmasi molekuler menggunakan gen spesifik Efac untuk bakteri E. faecalis dan gen gelE untuk deteksi gen virulen gelatinase. Uji biofilm menggunakan teknik spektrofotometer pada densitas optik 630 nm. Data dianalisis secara kuantitatif menggunakan uji chi-square dengan nilai P < 0,05 dianggap signifikan secara statistik. Hasil isolasi dan identifikasi bakteri terhadap 60 sampel arsip asal ayam pedaging yang digunakan dalam studi sebanyak 21 isolat positif terkonfirmasi secara molekuler sebagai bakteri E. faecalis dan memiliki gen virulen gelE. Pada uji biofilm terdapat sebanyak 20 isolat (95,23%) positif kuat (OD630 > 0,130) dan 1 (4,76%) positif lemah (0,065 < OD630 ≤ 0,130) dengan nilai P < 0,05 atau memiliki korelasi secara statistik antara keberadaan gen gelE dengan pembentukan biofilm. Berdasarkan hasil penelitian dapat disimpulkan bahwa gen virulen gelE telah ditemukan pada 21 isolat E. faecalis dan 95,23% mampu membentuk biofilm dengan intensitas positif kuat.

Abstract

Beta-lactam antibiotics are important antibiotics that are widely used in the field of human and animal health. Ampicillin resistance has been widely reported. Another increase in resistance is 3rd generation cephalosporins. The purpose of this study was to compare the ampicillin resistance profiles in 2019 and 2021 in the same E. coli isolates and to determine the resistance profiles of ampicillin, ceftazidime, and cefotaxime in live chicken E. coli. The research stages were the preparation of isolates; culture on differential selective media and checking the uniformity of bacterial cell morphology; biochemical test; bacterial DNA extraction; uspA gene amplification; visualization of amplification results; manufacture of bacterial suspensions; Kirby-Bauer disk diffusion resistance test; measurement of inhibition zones and determination of isolate status; and compared the ampicillin resistance test data. All isolates were confirmed positive for E. coli. The uspA gene (884 bp) was detected in all isolates. Ampicillin resistance in 2019 and 2021 in the same E. coli isolates when compared, there was no difference. Resistance test showed E. coli was resistant to ampicillin (100%), ceftazidime (15.4%), and cefotaxime (64.5%). The conclusion of the study was that there was no difference between the ampicillin resistance in 2019 and 2021 in E. coli isolates. Escherichia coli in this study had the highest resistance profile to ampicillin, followed by cefotaxime, and the lowest was ceftazidime.

Abstract

Rabbit Haemmorhagic Disease (RHD) is one of viral diseases in rabbits that is still exotic in Indonesia. The RHD disease is caused by the Rabbit Haemorrhagic Disease Virus (RHDV) which is a calicivirus of the genus Lagovirus, Caliciviridae family. The high genetic variation of the RHDV and the rapid spread have the potential to disrupt rabbit farm (rabbittery) production and trade activities, especially rabbit exports, which require clarity on the status and situation of RHD disease in rabbits in Indonesia. A case of suspected RHD in Indonesia was first detected in the Philippines where rabbits exported from Indonesia were detected to be seropositive to RHD which resulted in rabbit export activities to the country was stopped. There is no data on the presence of RHD disease in rabbits in Indonesia, so the seroprevalence study is useful to provide preliminary information on the presence of this disease in Indonesia. This study was conducted on 163 rabbits samples raised in the Lembang area, Bandung, West Java, which is known as the largest rabbit farming center in Indonesia. Rabbit samples were taken at rabbit farms spread across seven villages namely Lembang, Pagerwangi, Cikahuripan, Cikole, Sukajaya, Gudangkahuripan and Jambudipa. The rabbits serum samples obtained were tested using the Indirect Enzym Linked Immunosorbant Assay (ELISA) method to determine the titer of antibodies against RHD. Based on the results of the analysis, it is known that 120 out of 163 rabbit serum samples showed positive antibody titers against RHD. The presence of antibody titers in rabbits on such farms can be preliminary information to be able to carry out further studies.
