Abstract

This study was conducted to explore the potential of adjuvant for the production of immunoglobulin Y (IgY) as antistreptococcosis in layer chicken with mass production orientation. Enterococcus faecalis which causes streptococcosis in the red tilapia was selected as a candidateantigen. The production of immunoglobulin Y (IgY) was carried out on Isa Brown layer chickens and aged around 20 weeks. Furthermore, thechickens were grouped into four groups (A, B, C, and D groups), each consisting of three chickens based on the type of adjuvant, while twochickens were used as a control group. Each group was treated by giving MONTANIDE™ ISA 71R VG adjuvant (A), Freund's adjuvant (B), aluminum potassium sulphate adjuvant (KAl(SO4)2∙12H2O) concentration of 50 ppm in pH 7 (C), and only antigens without adjuvant (D). Chickens were kept for 35 days and each week was checked for presence the IgY antigen in the serum and egg yolk. Booster was conducted on 14th and 28th days of maintenance. The results showed that IgY in treatment group A was detected on day 28 in the serum and day 35 in the yolk. Whereas the treatment group B could be detected on day 35 in the serum. However, the IgY was not detected in the serum and yolk in C, D, and control groups until the end of the maintenance. Based on the results, it can be concluded that the appearance of IgY in serum and yolk in a relatively fast time is obtained in the combination of Enterococcus faecalis antigen with the emulsion of water-in-oil adjuvant (SEPPICMONTANIDE™ ISA 71R VG) compared to the other types of adjuvant that use in this study.

Abstract

Dalam penelitian ini isolasi dan karakterisasi hemaglutinin Staphylococcus aureus dilakukan dengan teknik afinitas kromatograf Karakterisasi hemaglutinin yang dihasilkan dilajutkan dengan teknik elektroforesis menggunakan metode sodium dodecyl sulphate gel electrophoresis (SDS-PAGE) dan dilanjutkan untuk melihat pengaruh suhu dan enzim terhadap aktivitas hemaglutinin. Hasil penelitia menunjukkan bahwa komponen hemaglutinin Staphylococcus aureus yang telah diisolasi memiliki berat molekul 46 kDa. Aktivit Staphylococcus aureus dalam meghemaglutinasi hilang pada pemanasan 60° C dan pengaruh enzim proteolitik. Hasil ini mengindikasika bahwa hemaglutinin Staphylococcus aureus adalah protein.

Abstract

Antibody to Enteropathogenic Eschericia coli (EPEC) K1.1. can be produced in egg yolk by the application of inactivated bacterial cells intravenously in layer chicken. The presence of specific antibody in sera and egg can bedetected with immunodiffusion techniques (Agar Gel Precipitation Test/AGPT), expressed by the occurrence of specific precipitation reaction between antibody and homolog antigen and no cross reaction of this antibody with antigen of Salmonella sp. and Klebsiella sp. The presence of specific antibody previously can be detected in sera, then 1 week after this the antibody start to be able be detected in egg yolk. The antibody is still present in sera as well as in egg yolk in large amount for 7 weeks and decrease significantly in the 8th week. This results indicated that theegg can be used in the producing specific antibody in the large quantity.

Abstract

Penelitian ini bertujuan mengetahui perbedaan respon antibodi yang ditimbulkan oleh vaksin AI dengan seed virus AI H5N1 Indonesia yang dicampur dengan adjuvant berbeda. Formula vaksin yang dicobakan pada penelitian ini adalah monovalen dan polivalen. Tiga isolat virus HPAI subtipe H5N1 yang digunakan adalah Chicken/Denpasar/Unud-01/2004, Chicken/Klungkung/Unud-12/2006, dan Chicken/Jembrana/Unud-17/2006. Adjuvant yang digunakan yaitu Freund's complete dan incomplete adjuvant, aluminium hidroksida, dan immunostimulating complexs (Iscoms). Vaksin monovalen dibuat dengan cara masing-masing isolat virus AI yang telah diinaktivasi dicampur dengan masing-masing adjuvant. Vaksin campuran (polivalen) dibuat dengan mencampur ketiga isolat dengan masing-masing adjuvant. Vaksin disuntikkan secara subkutan pada ayam layer jenis Isa Brown umur 3 minggu dan diulang pada umur ayam 5 minggu  masing-masing sebanyak 0,5 ml/ekor. Pengambilan serum untuk pengujian titer antibodi dilakukan setiap 1 minggu setelah vaksinasi. Pengujian antibodi poliklonal dilakukan dengan uji hambatan hemaglutinasi (HI). Hasil penelitian menunjukkan bahwa ayam percobaan yang divaksinasi dengan adjuvant aluminium hidroksida mempunyai GMT anti-H5 paling tinggi baik pada vaksin monvalen atau polivalen. Adjuvant aluminium hidroksida adalah adjuvant terbaik untuk pembentukan antibodi anti-AI subtipe H5N1 pada ayam.

Abstract

Tujuan penelitian ini adalah mengetahui kemampuan hemaglutinin pada Staphylococcus aureus (S. aureus) dalam proses adhesi. Uji adhesi pada sel epitel ambing dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian ini menunjukkan bahwa S. aureus yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik (±380 bakteri/20 sel) dibandingkan dengan yang tidak memiliki hemaglutinin (±56 bakteri/20 sel).

Abstract

Red tilapia (Oreochromis hybrid) is Indonesia's leading freshwater fishery commodity susceptible to streptococcal bacterial infection. Many studies have been conducted on various efforts to prevent and treat this disease, one of which uses the immunoglobulin Y (IgY) molecule from chicken egg yolk. This study aimed to observe the biological function of IgY against Enterococcus faecalis as a cause of streptococcal-like infection. The agglutinin function was conducted by observing the growth of Enterococcus faecalis in brain heart infusion (BHI) broth media which was added with IgY suspension. The function of inhibin was performed using a spectrophotometric method to measure the level of turbidity of the bacterial suspension inoculated with IgY suspension. The bactericidal potential through the complementary activation pathway for red tilapia serum was carried out using a scanning electron microscope (SEM) method to evaluate the topography of the bacterial cell wall. The results of the study can be concluded that IgY anti-Enterococcus faecalis has the potential as an agglutinin, inhibin, and bactericidal agent through its putative potential in complement activation in streptococcal bacterial infections in red tilapia commodities.

Abstract

This study was aimed at isolating and identifying Salmonella sp. and then conducting an antibiotics susceptibility test in order to detect resistant genes.  One hundred and five chicken cloaca swab samples were used in this study. 30 samples were taken from a layer farm in Bogor, 45 from a broiler farm in Sukabumi and 30 from a broiler farm in Cianjur. In order to  isolate and identify the bacteria, a tetrathionate broth was used, which was then cultured in a Salmonella-Shigella agar, and finally a Gram stain and biochemical test was conducted. To confirm the presence of Salmonella sp., a pair of primers were used for the polymerase chain reaction (PCR) method to determine the presence of the invA gene.. An antibiotics susceptibility test was used with the Kirby-Bauer disk diffusion method. Nine antibiotics were used in this study. Each primer pair was used for the detection of tetA, blaTEM, aac(3)-IV, gyrA and ermB genes, and for genes encoding antibiotic resistance  a PCR test was used. Eight (7.6%) Salmonella sp. were  isolated in this study. All isolates showed positive results with PCR confirmation. The results of the antibiotics susceptibility test showed that Salmonella sp. isolates were resistant to tetracycline (75%), oxytetracycline (75%), amphicillin (75%), gentamycin (12.5%), nalidixic acid (100%), ciprofloxacin (12.5%), enrofloxacin (0%), erythromycin (100%), and chloramphenicol (0%). The distribution of antibiotic resistance genes in Salmonella sp. were tetA (33.3%), blaTEM (100%), aac(3)-IV (0%), gyrA (100%) and ermB (0%) positive. In conclusion, Salmonella sp. was isolated. All isolates showed positive results in the PCR confirmation. Salmonella sp. isolates were resistant to tetracycline, oxytetracycline, amphicillin, gentamycin, nalidixic acid, ciprofloxacin, and erythromycin. Only the tetA, blaTEM, and gyrA genes were detected in Salmonella sp. isolates.

Abstract

Background: Obesity is a global problem which increasing simultaneously. The high-fat accumulation in the body will result from mild chronic inflammation. Developing a local drink based on seagrapes (Caulerpa racemose) is thought to have a high antioxidant capacity and the potential to become an alternative therapeutic drink for obese people. Seagrapes are easily damaged, so proper drying is needed to maintain antioxidant potential and assisted by adding gum arabic to increase the acceptability. Objectives: To analyze the antioxidant capacity and sensory of sea grapes powder drink (Caulerpa racemose). Methods: The design experiment research was utterly randomized and only used one gum Arabic treatment with a 2.5, 5, and 10% concentration and three replication as sea grapes powder drink samples. The samples were analyzed to seek the antioxidant capacity of DPPH (2,2-diphenyi-1-picrylhydrazyl), CUPRAC (Cupric Ion Reducing Antioxidant Capacity), and FRAP (Ferric Reducing Antioxidant Power). Then research data were analyzed by employing ANOVA  and Duncan's follow-up test. Results: The higher concentration of gum Arabic has significantly increased the antioxidant capacity. DPPH, CUPRAC, and FRAP values in 10% gum Arabic concentration were 13.21±0.1 mg/100 g, 25.26±0.5 mg, and 2.89±0.3mg/g. Based on the sensory test results, the panelists preferred the 10% gum Arabic concentration because the color is lighter, can minimize odor and viscosity, and taste better. Conclusions: Seagrapes powder drink with a concentration of 10% gum Arabic has the potential as a therapeutic obesity with the highest antioxidant capacity and is sensory acceptable.

Abstract

Demam babi afrika atau African swine fever (ASF) adalah penyakit viral lintas batas yang sangat menular pada babi domestik dan babi liar yang disebabkan oleh virus ASF (ASFV). Virus ini memiliki daya tahan hidup dan stabilitas yang tinggi di lingkungan, mampu bertahan lama pada benda mati seperti peralatan dan pakaian yang tercemar atau pada produk daging dari babi yang terinfeksi ASFV. Penelitian ini bertujuan mendeteksi ASFV pada produk daging babi dan olahannya di Indonesia. Sejak tahun 2019, Balai Besar Uji Standar Karantina Pertanian (BBUSKP) yang merupakan salah satu laboratorium rujukan milik pemerintah di bawah naungan Badan Karantina Pertanian (Barantan), memiliki tugas untuk melakukan pengujian deteksi ASFV salah satunya adalah menguji sampel produk daging babi. Prevalensi dihitung berdasarkan data sekunder hasil pengujian qualitative Polymerase Chain Reaction (qPCR) dalam deteksi ASFV pada produk daging babi yang dilakukan di laboratorium BBUSKP, Jakarta Timur tahun 2020-2022. Terdeteksinya ASFV pada produk daging babi sebanyak 8.2% di tahun 2020, 24.8% di tahun 2021 dan 11.6% pada tahun 2022 memperkuat bahwa produk daging babi berpotensi menjadi media pembawa ASF di Indonesia.
