Abstract

Di Indonesia sampai saat ini tidak banyak publikasi data terkait status epidemi Covid-19 yang disebabkan Severe Acute Respiratory Syndrom Coronavirus 2 (SARS-CoV-2) pada anjing dan kucing. Beberapa penelitian di negara lain menunjukkan prevalensi antibodi yang relatif tinggi pada anjing dan kucing dengan riwayat kontak erat dengan manusia yang terinfeksi Covid-19. Hal ini menunjukkan mudahnya penularan dari manusia ke hewan kesayangan. Data dan kajian mengenai kejadian Covid-19 pada anjing dan kucing yang berada di Indonesia maupun yang dilalulintaskan dapat memberikan informasi awal terkait keberadaan penyakit ini pada hewan kesayangan. Studi ini dilakukan untuk melihat keberadaan SARS-CoV-2 pada anjing dan kucing serta responskebal yang terbentuk. Sampel yang digunakan diperoleh dari hewan yang berasal dari berbagai negara dan beberapa klinik di Indonesia. Sebanyak (128 serum kucing dan 53 serum anjing) diuji menggunakan metode Indirect Enzym Linked Immunosorbant Assay (ELISA) sedangkan sampel usap/swab orofaringeal yang terdiri atas 1.499 swab asal kucing dan 958 swab asal anjing diuji menggunakan metode Real Time Reverse Transcription-Polymerase Chain Reaction. Berdasarkan hasil uji terdeteksi antibodi terhadap SARS-CoV-2 terdapat pada dua ekor anjing (3,8 %) dan enam ekor kucing (4,7 %). Uji kuantitatif PCR tidak menunjukkan adanya virus pada semua sampel yang diuji. Hasil studi ini dapat memberikan informasi awal untuk melakukan studi epidemiologi lanjutan terkait infeksi SARS-CoV-2 pada hewan kesayangan.lanjutan terkait infeksi SARS-CoV-2 pada hewan kesayangan.

Abstract

Lactococcus garvieae is a gram-positive ovoid cocci bacterium formerly classified as a member of the Lactococcus genus. This study aims to isolate L. garvieae from catfish rearing pond and characterize it as a potential probiotic candidate. L. garvieae was identified and characterized through phenotypic and genotypic observation, genomic % G~C content analysis, cell surface hydrophobicity assays, acidification test, in vitro antagonism, and a profile of antimicrobial activities. The MT597595.1 accession number corresponds to L. garvieae, as determined by a molecular identification test. Biochemical characterization was performed using API 50 CH kit. The genomics %G~C content of L. garvieae was 51.8. Findings from acidification ability tests, in vitro antagonism tests, and the ability of bacteria to grow in broth medium at pH 4 reveal that L. garvieae can inhibit the growth of Aeromonas hydrophila, Streptococcus agalactiae, Streptococcus iniae, and Edwardsiella ictaluri. However, it does not suppress the growth of L. garvieae  Edwardsiella tarda. Remarkably, L. garvieae has the ability to reduce the pH of neutral broth medium turning it acidic. Furthermore, L. garvieae’s hydrophobic cell surface exhibited an adhesive, hydrophobic, and protein surface cell content with a compact growth pattern consistent with postive SAT and MATH assay. Antimicrobial activity tests, encompassing 11 antibiotics, disclosed resistance to  Nalidixic acid while displaying intermediate sensitivity to Streptomycin and Trimethoprim. In conclusion, L. garvieae demonstrates an inhibitory effect on the growth of pathogenic bacteria, underlining its potential as a probiotic candidate.

Abstract

Gram-positive spore bacteria are widely used as probiotics in general sectors. However, there are still limited bacterial isolates as probiotic candidates available from indigenous isolates, especially in aquaculture. This study aimed to obtain potential spore-forming isolates as probiotic candidate for tilapia. Tilapia fish samples were collected from Sukabumi, Ciamis, Serang, and Papua. Bacterial isolates were isolated from the digestive tract of tilapia. Bacteria were identified based on their morphological, molecular characteristics, complete genome composition, and cell surface identification based on hydrophobic properties. In this study, six bacteria were isolated and identified by molecular characteristics using 16S rRNA sequences. Based on the phylogenetic analysis, the 9 PP isolate was Priestia megaterium basonym: Bacillus megaterium, CMS 16N isolate was Brevibacillus halotolerans, PPN 10 isolate was Bacillus sp., 3.1 SKBM isolate was Bacillus mycoides, CMS 22 N and SRG32 isolate were Bacillus subtilis. Six bacteria had different phenotypicals, ATGC sequence compositions, and a higher proportion of total G~C sequence composition above 50%. The coherent cell surface hydrophobicity test was positive on the SAT, SA, AA, and compact growth patterns in soft-agar media for 9 PP, CMS 22 N, and SRG32 isolates. From our study, the indigenous spore-forming bacteria isolated from tilapia stomachs are enzymatic bacteria, which have a strong attachment to host tissue and high potential as a probiotic candidate for fish. Various hydrophobicity test results from each isolate indicate that the protein composition in the cell surface is different.

Abstract

Virus Rabbit Haemorrhagic Disease (RHDV) adalah virus penyebab Rabbit Haemorrhagic Disease (RHD) pada kelinci Eropa (Oryctolagus cuniculus). Penyakit ini memiliki tingkat kematian dan morbiditas hingga 100%. Penyakit ini pertama kali dilaporkan di Republik Rakyat Tiongkok pada tahun 1984 dan menyebar ke seluruh dunia. Pada tahun 1996, kasus pertama infeksi RHDV pada kelinci tanpa gejala klinis menyebar di peternakan kelinci di Italia. Beberapa studi telah melapaorkan keberadaan variasi genetic RHDV termasuk varian RHDV terbaru. Penyakit ini berpotensi menimbulkan dampak ekologis dan kerugian ekonomi, terutama di peternakan kelinci. Pada studi sebelumnya diketahui bahwa terdapat serokonversi RHDV pada kelinci tanpa gejala klinis di daerah Bandung Barat, Provinsi Jawa barat, Indonesia, meskipun belum ada informasi mengenai keberadaan RHDV di Indonesia. Meskipun Indonesia diketahui berstatus bebas RHDV, namun kelinci yang diimpor berasal dari negara-negara di Eropa, Amerika, dan Australia yang terkenal sebagai negara endemik RHDV. Tulisan ini bertujuan untuk mengkaji penyebaran RHDV di negara asal kelinci impor Indonesia dan dampaknya. Selain itu, tulisan ini diharapkan dapat memberikan informasi tentang penyakit kelinci yang sedang berkembang.

Abstract

ABSTRAK Penelitian tentang pemanfaatan telur sebagai pabrik biologis dilakukan sangat komprehensif di beberapa institusi perguruan tinggi di Inonesia, yakni Fakultas Kedokteran Hewan (FKH) Institut Pertanian Bogor (IPB), FKH Universitas Gadjah Mada (UGM), FKH Universitas Udayana (UNUD), FKH Universitas Syahkuala (Unsyiah), Fakultas Kedokteran Gigi Universitas Indonesia. Jejaring penelitian dibangun dengan baik dan telah menghasilkan beberapa produk, antara lain: telur berkhasiat flu burung H5N1, telur Anti Tetanus Serum, Telur Anti Diare, Telur anti Plaque dan Telur Anti White Spot Syndrome Virus pada udang. Secara ilmiah khasiat IgY spesifik dalam kuning telur sebagai senyawa therapeutic telah diuji dan tinggal memerlukan sentuhan akhir tersendiri untuk dapat disajikan sebagai produk komersial. Peran industri yang relevan sangat dibutuhkan dalam mewujudkan hal ini. THE USE OF EGG AS A BIOLOGICAL PRODUCER FOR SPESIFIC ANTIBODIES ABSTRACTS The use of egg as a biological producer for specific antibodies were done intensively among well known universities in Indonesia, such as Faculty of Veterinary Medicine (FVM) of FVM of IPB, FVM of UGM, FVM of UDAYANA and FVM of UNSYIAH as well as Faculty of Dental Medicine Indonesia University. The collaboration and research networking among those institutions run as expected and succeed to produce some important products such as eggs containing IgY anti avian flu as well as anti tetanus, anti diarrhea, anti plaques and eggs anti WSSV in shrimp. The potency of specific IgY as immunotherapeutic substances had been studied and need the commercial touch from the respective industries until this products in market available.

Abstract

The content of active compounds in natural materials is generally low and highly variable, thus requiring efficient and selective extraction methods. This study aims to evaluate a stepwise extraction process applied to the seeds, oil, and by-products of Nigella sativa (black cumin) to obtain thymoquinone (TQ) and other phytochemicals. The study consisted of four main stages: (1) extraction of HS oil using the cold press method, (2) stepwise extraction using 70% ethanol as solvent, (3) analysis of TQ content and phytochemical compounds in the extracts, and (4) evaluation of the antibacterial activity of habbatussauda oil extracts against pathogenic and non-pathogenic bacteria. Results showed that the cold press method yielded 34.46% oil. Stepwise extraction using 70% ethanol resulted in significant differences (p<0.05) in both yield and TQ content among the seed, oil, and by-product extracts. Among all samples, the ethanol extract from habbatussauda oil exhibited the highest yield and TQ concentration. Furthermore, at a concentration of 100 μg mL⁻¹, this extract inhibited the growth of pathogenic bacteria such as Salmonella typhimurium and Escherichia coli, without affecting non-pathogenic strains like Lactobacillus spp. and Bifidobacteria spp. In conclusion, stepwise extraction using 70% ethanol proved effective in enhancing TQ recovery and demonstrated potential as a phytobiotic agent with selective antibacterial activity against pathogenic bacteria. Abstract

Background and Aim: IPB D1 chicken is a crossbreed developed by crossing local Indonesian chickens-Pelung, Sentul, and Kampung-with commercial broilers. This crossbreeding initiative led by researchers from the Faculty of Animal Science at IPB University, aims to develop a meat-type chicken breed that is well adapted to traditional backyard farming systems commonly found in rural areas.Materials and Methods: The objective of this study was to evaluate the resistance of IPB D1 chickens carrying the Toll-like receptor 4 (TLR4) gene against Salmonella enterica serovar Enteritidis. A total of 11 chickens were used in this experiment. Detection of the TLR4 gene was conducted using Polymerase Chain Reaction (PCR) and sequencing techniques, followed by assessment of the chickens' immune response through total and differential leukocyte counts.Results: The results showed that IPB D1 chickens with the TLR4-positive genotypes (GG and AG) exhibited stronger immune responses and more stable leukocyte profiles compared to TLR4-negative chickens (AA genotype) following exposure to S. Enteritidis. These findings suggest that the TLR4 gene plays an important role in innate immunity and may enhance resistance to bacterial infection.Conclusion: Overall, this study highlights the potential of IPB D1 chickens as a resilient local breed suited for sustainable poultry farming, particularly in resource limited environments.Keywords: IPB D1 chickens, leukocyte profile, non-specific immune response, Salmonella Enteritidis, TLR4 gene

Abstract

Haemorrhagic Septicaemia (HS) is a devastating disease affecting cattle and water buffaloes in Indonesia, causing annual economic losses in livestock industries, particularly in Asia, Africa, and the Middle East. Pasteurella multocida, the causative agent of the disease, has shown increasing antimicrobial resistance, complicating treatment efforts. Therefore, this study aims to provide the first genomic analysis of P. multocida isolates from different Indonesian provinces, focusing on genes conferring resistance to beta-lactam antibiotics based on Whole Genome Sequencing (WGS) results. Genomic data can be used to confirm the results of phenotypic antibiotic resistance testing. P. multocida isolates analyzed in this study were sourced from the Indonesian Research Center for Veterinary Science (IRCVS) collection, which included 2 samples originating from different geographic locations within Indonesia. The samples were subjected to biochemical, molecular, and antimicrobial susceptibility testing. WGS was performed using Oxford Nanopore Technologies (ONT) with subsequent bioinformatics analysis for genome assembly and resistance profiling. Phenotypic analysis showed significant variations between the isolates. The NTT isolate showed resistance to Penicillin, while the Lampung 2952 remained susceptible in the disk antibiotic test. Genome sequencing revealed extensive resistance determinants, including β-lactamase genes (blaZ, blaR1) in the NTT isolate, correlating with its phenotypic resistance. The high-quality genome assemblies (N50: 17,225 bp for NTT; 12,662 bp for Lampung 2952) enabled comprehensive resistome characterization, identifying more than 22 resistance genes in each isolate, including novel variants not previously reported in Indonesian strains. Therefore, this study provides the first genomic and resistance analysis based on Whole Genomic Sequencing data of Indonesian P. multocida isolates. The results show the urgent need for enhanced surveillance and prudent antimicrobial use in livestock management.

Abstract

Luwak Jawa (Paradoxurus hemaphroditus) merupakan hewan yang memiliki kemampuan menghasilkan biji kopi yang berkualitas, sehingga status kesehatannya perlu untuk diperhatikan. Gambaran darah dapat dijadikan sebagai indikator status kesehatan luwak pada kondisi tertentu. Penelitian mengenai luwak telah banyak dilaporkan, namun belum ada penelitian mengenai parameter fisiologis khususnya profil eritrosit dari luwak yang sengaja diberikan buah kopi secara terus menerus selama 3 bulan. Penelitian ini bertujuan untuk menghitung profil eritrosit pada luwak Jawa pemakan pemakan kopi dan tidak pemakan buah kopi. Gambaran darah luwak Jawa pada saat adaptasi dan pemeliharaan berfluktuasi. Rataan jumlah eritrosit luwak pemakan buah kopi dan tidak pemakan buah kopi adalah (8,24±2,40) x 106/mL dan (8,25±2,16) x 106/mL, kadar hemoglobin adalah (6,18±2,21) g/dL dan (6,40±2,21) g/dL dan nilai hematokrit adalah (16,50±2,21) % dan (16,00±2,14) %. Secara umum gambaran darah luwak Jawa pemakan buah kopi memiliki rataan yang lebih rendah dari pada luwak Jawa yang tidak diberi buah kopi.

Abstract

This study aims to determine the prevalence of tetracycline antibiotic resistance to Escherichia coli (E. coli) isolated from farms, pig slaughterhouses, and the environment in Banten Province. This study used 44 samples of solid waste/feces and effluent from pig farms, 10 samples of floor swab and effluent from pig slaughterhouses and 4 samples of the environment (river water). Isolation and identification ofE. coli using Tryptone Bile X-Glucuronide (TBX), MacConcey Agar (MCA) media and confirmed by indole test. Sensitivity test against oxytetracycline and tetracycline antibiotics was carried out using Kirby-Bauer method (disc diffusion). The results showed that oxytetracycline was the most resistant antibiotic. The prevalence of oxytetracycline resistance was highest in farms in solid waste/feces samples (56.82%), pig slaughterhouses in effluent samples (80%) and environment in river samples (upstream, downstream) (100%). Tetracycline showed the highest prevalence in solid waste/feces samples of farm (40.91%), in floor swab samples of pig slaughterhouses (70%), in upstream river samples of environment (100%). This study indicates that waste from farms and pig slaughterhouses could be a source of transmission of E. coli resistant to tetracycline antibiotics to the environment.
