Abstract

Avian Influenza (AI) is well known as Avian flu, Fowl pest, Fowl plaque, or Flu burung, caused by influenza virus type A.  This virus is belonged to Orthomyxoviridae and could infect many kind of species such as bird, pig, horse, cat, as well as human.  Vaccination is applied to control the disease using inactivated vaccine, which induced the specific antibody against H5 antigen.  Passive immunization using specific antisera against H5 antigen is thought to be usefull in controlling the disease especially in the treatment of infected host.  In this experiment the neutralization ability of specific antisera against H5 were studied using various field viral isolates subtype H5N1.  Antisera was developed in Cavia porcellus which vaccinated with AI subtype H5N1 in activated vaccine.  The titre of antisera obtained is 28 used HI test.  Four AI virus subtype H5N1 isolates from 2003 to 2006 agains viral were we as tested virus.  The neutralization test showed that the sera were able to neutralizing 10 4 EID50 AI virus H5N1 with neutralization index range of 1.1-1.3.  The result indicated that the specific antisera had the neutralization potency to the field virus.   Key words: avian influenza, neutralization test, neutralization index

Abstract

The objective of this research was to know the sensitivity of H5N1 clade 2.3.2 AIV from Indonesia to antiviral drug (amantadine) through molecular and in vitro tests. The study was conducted by virus isolation and identification, nucleotide analysis, and susceptibility to the amantadine hydrocloride in MDCK cells. The study result represented that the mean EID50 isolates of H5N1 clade 2.3.2 AIV was determined of >108 EID50/ml. The analysis of phylogenetic tree of M2 gene from six viruses of H5N1 clade 2.3.2 AIV from Indonesia were closed with H5N1 clade 2.3.2 AIV avian influenza viruses from Vietnam, China, Hongkong. The substitution of M2 protein (V27I) was identified in six isolates H5N1 clade 2.3.2 AIV isolated from Indonesia. Avian influenza of clade 2.3.2 H5N1 subtype from Indonesia produced the formation of CPE and the positive HA reaction with non-toxic concentration of amantadine hydrochloride in MDCK cells. The result of genetic analysis of M2 gene for amantadine resistance was related with the results of HA test and the formation of CPE in MDCK cells. These results established that amantadine resistance have been identified in H5N1 clade 2.3.2 AIV viruses isolated from Indonesia

Abstract

Streptococcus agalactiae dan Staphylococcus aureus merupakan dua bakteri utama penyebab mastitis subklinis pada sapi perah di Indonesia. Pada mastitis subklinis kemampuan adesi mempunyai peran lebih penting dari pada kemampuan invasi. Hemaglutinin merupakan faktor yang berperan dalam proses adesi, sebagai langkah awal kolonisasi bakteri pada permukaan set epitel ambing. Tujuan dari penelitian ini adalah mengisolasi dan mengkarakterisasi hemaglutinin dari S. agalactiae dan S. aureu. Penentuan hemaglutinin S. agalactiae dan S. aureus dengan uji hemaglutinasi menggunakan eritrosit sapi perah 1%. Isolasi hemaglutinin dilakukan dengan afinitas kromatogarfi dan analisis dengan Sodium Dodecyl Sulphate-Polyacrilamide Gel Electrophoresis (SDS-PAGE). Hasil penelitian diketahui bahwa hemaglutinin S. agalactiae dan S. aureus dapat ditentukan dengan SDS-PAGE dengan berat molekul 28 kDa pada S. agalactiae dan 27 kDa pada S. aureus.

Abstract

Avian Influenza (AI) virus  is pathogenic agent that can spread from one area to another area through the transportation of infected animals or their products such as feathers. This research was aimed to inactivated AI virus with 775 ppm sodium hypochlorite and 0.1% chloroxylenol to examine the difference of treatment by time (day) for inactivation AI virus on the feathers. AI virus isolate A/chicken/sidrap/ 07160336‐2/2016 used in this research was obtained from Balai Besar Veteriner Maros. The treatment of disinfectants were performed on the first day (the day of disinfectan solutions were prepared), the third day and the seventh day by soaked the feathers in disinfectant solution for 10 minutes. The effectiveness of disinfectans were evaluated by inactivation index. The result show that the average of inactivation index of 775 ppm of sodium hypochlorit was 4.17 for the first day, 5.17 for the third day, and 4.20 for the seventh day, while the average of inactivation index of 0.1% chloroxylenol was 5.50 for the first day, 6.43 for the third day, and 5.77 for the seventh day. Our result indicated that the sodium hypochlorit and chloroxylenol were effective for inactivation of AI virus. The 0.1% of chloroxylenol was more effective for inactivation AI virus than 775 ppm of sodium hypochlorit, whilst the most effective duration for the treatment is the three days.

Abstract

Chronic Respiratory Disease (CRD) is a bacterial infection in chickens caused by Mycoplasma gallisepticum. CRD may result in economic loss in the livestock industry, decreasing of egg production and feed efficiency, increasing of  medication costs, downgrading of carcass and egg qualities. The aim of this study was to develop the Enzyme-Linked Immunosorbent Assay (ELISA) kit for detecting antibodies of M. gallisepticum using freeze-thawing antigens. The cut-off point was determined by S/N method. Chicken antiserum againts M. gallisepticum was collected every week on 1st – 6th week post immunization of whole cell antigen of M. gallisepticum. Optimization of antigen, serum and conjugate were 10 μg/ml protein concentrations, 1:800 serum dilution and 1:10000 conjugate dilution. The validation results on developed ELISA kit (MyGELISA) analyzed  by ROC curve using MedCalc statistical software revealed that developed ELISA kit (MyGELISA) had 100% sensitivity and 86.21% specificity with 95% confidence interval. The results of RSA, MyGELISA and commercial ELISA kit showed antibodies positif againts M. gallisepticum.  In conclusion, MyGELISA using freeze-thawing antigen can be used to detect antibodies of M. gallisepticum.

Abstract

The prevalence of antibiotic resistance is an increasing problem due to the use of false and excessive antibiotics. The use of antibiotics in pig farms is increasing and needs to be aware of side effects, it is also influenced by the knowledge, attitudes and practices of farmers. This study is a cross-sectional study using a structured questionnaire interview to analyze the relationship between knowledge and attitudes towards the practice of using antibiotics by pig farmers in Kupang city. A total of 96 respondents pig farmers spread over 6 districts, District of Maulafa (9 villages) District of Alak (12 villages) District of Oebobo (7 villages), District of Kelapa Lima (5 villages), District of Kota Raja (2 villages) and the district of Kota Lama (1 village). The variables observed were individual characteristics, knowledge, attitudes and practices of the use of antibiotics by pig farmers in relation to the occurrence of antibiotic resistance. The relationship between the variables was tested using the Pearson correlation, while the relationship and magnitude of the direct and indirect impact on the observed variables was determined using path analysis. The results showed knowledge has a very significant impact (p <0.01) and indirectly contributes to the level of antibiotic usage practices preventing the occurrence of antibiotic resistance. Attitudes have a significant effect (p <0.05) and directly affect the practice of antibiotics to prevent the use of antibiotic resistance in Kupang. Conclusion of this study shows that knowledge has a major impact on the practice of using antibiotics by pig farmers, so by increasing farmers' knowledge of the facts incidence of antibiotic resistance, the attitude of farmers will increasingly have a positive significant effect on the level of practice use of antibiotics Prevent antibiotic resistance at a pig farm in the city of Kupang.

Abstract

The aim of this study was to evaluate the in vitro antibacterial activity of Black soldier fly (BSF) larva extract. The BSF larva was extracted using methanol and then tested for antibacterial activity using agar diffusion method (zone growth inhibition). The antibacterial activity was conducted against Salmonella sp. and Escherichia coli, two important bacterial strains in poultry, using six dilution levels (10 mg/ml, 20 mg/ml, 40 mg/ml, 80 mg/ml, 160 mg/ml and 320 mg/ml). All the results were subjected analyze using t-test method. Based on the diameter of the inhibition zone, the BSF larva extract has a strong (P<0.05) antibacterial activity against Salmonella sp. and E. coli when the concentration used 320 mg/ml. In addition, BSF larva extract also contain high amount of lauric acid (49.18%), a saturated fatty acid that has been proven to proposes as antibacterial agent. Therefore, it could be concluded that the BSF larva extract could be used as a candidate for antibacterial substances.

Abstract

To our knowlege, there is no rapid agglutination test to detect antibodies to viruses which might be due to the small dimension of viral particles. Through complex formation of Staphylococcus aureus bearing protein A-rabbit IgG-anti IgY-IgY anti Marek virus, agglutination of antibodies to viruses can be achieved and visualized. To design the prototype of the test, the bacterial cells of Staphylococcus aureus were coupled to a complex compound consisting of IgG-IgY-Marek antigen. This protocol was able to detect clearly the presence of Marek antibody in chicken sera, showing the rapid, clear and distinct agglutination reaction on the glass objects. No agglutination reaction was observed in the reaction of specified pathogen free chicken sera with this prototype showing the specificity of the test. This finding demonstrates a novel rapid agglutination which can be used for the detection of antibodies to various agents.

Abstract

Latar Belakang. Kandungan zat gizi extra virgin olive oil (EVOO) memiliki banyak manfaat untuk kesehatan manusia. Beberapa manfaat antara lain sebagai imunomodulator, mencegah penyakit jantung dan vaskuler lainnya, mencegah penyakit alergi, memperbaiki fungsi liver, dan mencegah penyakit lainnya. Kandungan zat gizi tiap merek EVOO tidak sama, ditentukan oleh banyak faktor. Tujuan. Penelitian ini bertujuan menganalisis klaster berdasarkan karakteristik kimiawi, kandungan zat gizi, dan senyawa aktif pada tujuh sampel produk EVOO dari supermarket. Metode. Desain deskriptif digunakan dalam penelitian ini. Pengumpulan tujuh sampel produk EVOO di supermarket wilayah Daerah Khusus Ibukota (DKI) Jakarta dilakukan pada bulan Maret 2019. Pemeriksaan sampel produk EVOO dilakukan di Laboratorium Balai Besar Industri Agro (BBIA) Kementerian Perindustrian dan Laboratorium Biofarmaka dan Lembaga Penelitian Pengabdian Masyarakat (LPPM) Institut Pertanian Bogor (IPB). Analisis yang digunakan adalah analisis klaster melalui pendekatan hierarchical method. Hasil. Berdasarkan karakteristik kimiawi, ditemukan bahwa sampel produk EVOO ke-6 memiliki karakteristik kimiawi paling berbeda. Sampel produk EVOO ke-4 memiliki kandungan asam lemak jenuh paling berbeda. Kandungan asam lemak tak jenuh pada sampel produk EVOO ke-1, 3, dan 7 mendekati kesamaan. Kandungan vitamin E pada sampel produk EVOO ke-2 dan kandungan zat besi pada sampel produk EVOO ke-6, berbeda dengan sampel produk EVOO lainnya. Kandungan total flavonoid pada sampel produk EVOO ke-2, 3, dan 4, memiliki kadar mendekati kesamaan. Kandungan total karotenoid pada sampel produk EVOO ke-2 dan ke-6 juga memiliki kadar mendekati kesamaan. Kesimpulan. Karakteristik kimiawi pada semua sampel produk EVOO yang ditemukan dalam penelitian memiliki nilai hampir sama. Sampel produk EVOO di supermarket memiliki perbedaan kandungan asam lemak tak jenuh, total flavonoid, dan total karotenoid.

Abstract

Black Soldier Fly larvae are natural antibiotics. The use of methanol extract in BSF larvae has a dual function, namely high protein content and high antibiotic content to kill Gram-negative bacteria. The biological structure of BSF larvae with antimicrobial peptide (AMP) acts as an inhibitor of pathogenic microorganisms, has a high lauric acid function as a natural antimicrobial and chitin, the polysaccharide plays a role in enhancing the immune response of the animal. This study aims to gain the best type of BSF protein from feed treatments containing live BSF, dry BSF, and BSF extract to increase blood productivity and hematology. The results of this study show that the best P3 treatment for the productivity of laying hens at the age of 18 to 26 weeks with the use of BSF extract since it contains the highest protein compared to other treatments. Productivity variables show the best P3 treatment, each egg weight 57.17 g bird-1, daily egg production 90.88%, egg mass 2408.16 g bird-1 and ration conversion 2.0. The mean hematology of the blood showed the highest P3 treatment, each erythrocyte 2.91±0.13 x 106 mm-3, leucocytes 17.69±3.68 x 103 mm-3, hematocrit 27.8±1.14%, hemoglobin 9.96±1.12%, MCV 95.77±4.41 fl, MCHC 35.99±5.21%, MCH 34.38±4.53 pg, lymphocytes 61.2 ± 4.86%, heterophile 31.2±2%, monocytes 5±0.62% and ratio H/L 0.51±0.04.
