Abstract

Eschericia coli contamination was found on egg powder imported to Indonesia during microbialexamination. The study was conducted to describe the presence of E. coli and its resistance to antibioticsin egg powder imported to Indonesia through The port of Tanjung Priok. Samples was determined usingcross sectional study and the sample size was calculated based on assumption of confidence level of 95%with margin of error of 10% and predicted prevalence of 50%. Total of 100 egg powder samples wascollected in August 2014 from two exporting countries, Ukraine (whole egg powder, n=30) and India (wholeegg powder, n=40 and egg yolk powder, n=30). Examination was performed by packaging and label inspectionof the product followed by samples collection and testing for bacterial contamination using rapid test andconventional isolation and identification methods. Detected E. coli was isolated and then tested forantibiotic resistance. Examination results showed that 4 samples were positive to E. coli. E. coli isolatesshowed resistance against 6 types of antibiotics and 75% of the isolates had resistance against minimumof 3 types of antibiotics. These conditions should be taken into consideration since antibiotic resistance inE. coli would cause negative impacts on human, animal and environmental health.

Abstract

This experiment was conducted to study the profile of total protein, albumin, and globulin concentrationson Ettawah crossbreed neonates after consuming various colostrums. Twenty four healthy neonatal kidswere used in this study. The neonates were divided into four groups. Each group received fresh maternal(goat) colostrum, frozen-thawed bovine colostrum, bovine spray dried colostrum, and bovine powdercommercial colostrum, respectively. Colostrums were given at 10% of body weight directly after birth andfollowed by the same amount every 12 hours, for three days. The blood was taken from jugular vein at 0, 12,24, 48, 72, and 168 hours after birth to determine total protein, albumin, and globulin concentrations.Results of this study indicated that the serum total protein and globulin concentration increased andreached the peak at 24 hours after birth. Compared to the concentration at birth, the increase of totalprotein concentration were 62.77%, 59.26%, 48.05%, and 66.67% in fresh maternal (goat), frozen-thawedbovine, bovine spray dried, and commercial bovine colostrum, respectively. Serum globulin concentrationincreased 4.9, 4.4, 4.8, and 14.6 times in fresh matermnal goat, frozen-thawed bovine, spray dried, andcommercial bovine colostrums respectively, compared to the concentration at birth. In conclusion, theconsumption of various colostrums i.e. fresh maternal goat colostrums, bovine colostrums (frozen-thawed,spray dried and commercial colostrums) would increase the concentration of blood total protein and globulin,which both reached the highest concentration at 24 h after birth.

Abstract

H5N1 subtype Avian influenza has harmed for poultry in Indonesia currently. Since clade 2.3.2 Avian influenza has been found in waterfowl from 2012, mortality of waterfowl has been occured until now. Change of extreme weather condition with high rainfall and flood, small poultry that does not adjust stricted biosecurity and without vaccination program, and chain sale of poultry on traders can increase case and transmission of Avian influenza in poultry. The objective of study was to isolate and identify Avian influenza on poultry in Central Java, West Java, East Java and Banten in 2016. The study was conducted by using Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) method to identify H5N1 subtype Avian influenza on poultry. The study was also conducted by isolation of H5N1 subtype Avian influenza from poultry into 9-11 days of embrionated chicken eggs. The study result represented that a number of 24 pool samples were positive reacted on RT-PCR with matrix primer, a total of 15 pool samples was positive reacted on RT-PCR with H5 primer and as many as 11 pool samples were positive reacted on RT-PCR with N1 primer. Two isolates of H5N1 subtype Avian influenza were isolated samples collected from the Serang City and six isolates of H5N1 subtype Avian influenza from Avian influenza case on waterfowl in Lamongan District. EID50 titer isolates of Avian influenza virus subtype H5N1 were > 108. The conclusion of this study was H5N1 subtype Avian influenza detected on samples collected from cloacal, trachea swab and organs of poultry from Pekalongan District, Brebes District, Serang City, and Lamongan District in 2016. H5N1 subtype Avian influenza can be grown and isolated from samples collected of Serang City and Lamongan District in 2016. H5N1 subtype Avian influenza can be detected, isolated, and indentified in poultry in Java Island, Indonesia in 2016.

Abstract

Staphylococcus aureus is known as a major causative agent of mastitis in dairy cattle. In the presentstudy, 104 isolates of Staphylococcus originated from subclinical mastitis cattle characterized for thephenotypic properties and the presence of Staphylococcal protein A (Spa). Some bacteria were resistancesagainst several antibiotics were also studied, such as erythromycin, streptomycin, tetracycline, cefepime,nitrofurantoin, amikacin, chloramphenicol, and ciprofloxacin. About 78% of the isolated were moderatelysensitive to nitrofurantoin, while 89% were highly resistant to cefepime and ciprofloxacin. Using thevarious mammals’ sera, seven isolates out of 104 revealed the presence of Spa.

Abstract

The porpuse of study was to explore the potential use of? anti tetanus IgY from eggs yolk as a substitute for anti tetanus serum raised in ?horses. The eggs were collected from chickens which have previously been immunized with tetanus toxoid. Neutralization potency test of anti tetanus IgY determined by ?Spearman-Karber method.? The highest mean titer of anti tetanus of egg yolk was 80.16 ? 33.55 IU/ml and the lowest was 1.69 ? 0.63 IU/ml. The concentration? of purified IgY was 1.644 ? 0.424 mg/ml. Spearman-Karber value of potency of anti tetanus IgY are 35 IU/ml. ?This research concluded that Chickens was capable of produced of anti tetanus in eggs yolk with value of potency are 35 IU/ml.

Abstract

Colistin sulphate is the ultimate antimicrobial choice for the treatment of multidrug resistance gram negative bacteria infections with in human. The purposes of this study were to detect the presence of colistin resistant E. coli and mcr-1 gene in broiler and to transfer the mcr-1 gene to Salmonella enteritidis ATCC 13076. A total of 54 one day old broilers were divided into three groups that consists of 18 chicks broiler per group and raised up to 40 days old. The first group was used as control. The first treatment group was given colistin sulphate 5 ìg/g feed for 40 days and broilers in second treatment group was given 80.000 IU/kg body weight for first three days. Swab cloaca samples were taken every 10 days from each broiler. At age 40 days all chickens were slaughtered and meat samples were collected. Samples of cloacal swabs, fresh and cooked meat were examined for the presence of colistin resistant E. coli and mcr-1 gene. Susceptibility to colistin sulfate was conducted by agar dilution method, and detection of mcr-1 gene was conducted using polymerase chain reaction. The results showed that no colistin resistant E. coli was detected in the control group. Colistin resistant E. coli (27.78%) and mcr-1 gene (20.00%) were detected in animals in the first treatment group, respectively. Whilst 11.11% colistin resistant E. coli and 5.56% were carriying mcr-1 gene in the second treatment group. Colistin resistant E. coli were found 5.56% from raw meat samples and 3.70% had mcr-1 gene. Transfer of mcr-1 gene from colistin resistant E. coli to Salmonella enteritidis ATCC 13076 was success. These results showed the necessity of limitation usage of colistin sulphate in food animal.

Abstract

Streptococcus agalactiae atau yang juga dikenal dengan Streptokokus grup B (SGB) telah diketahui sebagai agen penyebab pneumonia, septikemia dan meningitis neonatal pada manusia dan hewan. Hialuronidase merupakan produk ekstraseluler dari SGB yang menentukan virulensinya. Penelitian ini dilakukan untuk menguji akitvitas hialuronidase dari 10 isolat SGB yang diisolasi dari kasus komplikase obstetri, serta memurnikan dan mengkarakterisasi hialuronidase yang diisolasi dari SGB SV-14 pada substrat asam hialuronat.

Abstract

The objectives of this study were to characterize the chemical properties and determine the antibacterial activity of S. crassifolium against Salmonella sp. The algae was extracted using water and ethanol. The results showed that the yield of E. spinosum extract using water solvent showed the highest value (6.53%), followed by S. polycystum extract (3.27%) and S. crassifolium (2.34%). The highest concentration of uronic acid was shown in S. crasifolium extraction (0.90%) using water solvent and the lowest in E. spinosum (0.04%) using ethanol solvent. Inhibitory test against Salmonella sp using 10% S. crassifolium extract showed higher value than using 2.5; 5.0 and 7.5% S. crassifolium extract. It is concluded that the extract of S. crasifolium contain 0.90 % uronic acid with pH= 8 of that capable of inhibiting the growth of Salmonella sp.

Abstract

Dengue Haemorrhagic fever (DHF) is one of fearsome diseases in society. Incidence of the disease isincreasing. Dengue fever is caused by dengue virus and transmitted by Aedes aegypti mosquito vector.Various chemical controls have been conducted to prevent the spread of the disease, but active contents ofthe chemical controlling substances are suspected causing many negative effect, in environment, such asvector resistance, death of non target living creatures, and environmental contamination.This researchobjective was to find an alternative solution in order to control the dengue vector by using entomopathogenicfungi as biological control agent. This research was conducted by isolation and identification of fungiinfecting mosquito larvae. Macroscopic observation revealed that one of the nine isolation products wasLagenidium giganteum. The effectiveness test in laboratory showed the zoospore LD50 to Ae.aegypti larvaeof instar 2nd was 2,35 x 106 zoospore/ml, while the LD95 value was 1,35 x 107 zoospore/ml. The oosporeeffectiveness test showed LD50 was 6,7 x 102 oospore/ml and LD95 was 1,94 x 103 oospore/ml. Using LPCBdye and blue tolouidin 2,5%, the infection mechanism of L.giganteum fungi in Ae.aegypti mosquito larvawas detected. The research is concluded that the entomophatogen fungi L. giganteum was very prospectiveto be used as a biological control agent against vector of DHF.
