Abstract

Sensitivity and specificity of nested polymerase chain reaction (nested PCR) to detect Coxiella burnetii(C. burnetii) DNA were studied. The primer system which consists of external primers (OMP1 and OMP2)and internal primers (OMP3 and OMP4), was designed from the nucleotide sequence of the com I geneencoding for 27 kDa outer membrane protein and used to specifically amplify a 501 bp and 438 bp fragment.This nested PCR assay was 50 fold more sensitive than that of using PCR external primer only. TheNested PCR has a detection limit as low as 300 pg/?l. Specificity studies showed that nested PCR onlydetected C. burnetii DNA and did not happened Brucella abortus, Escherichia coli, Pseudomonas aeruginosaand Campylobacter Jejuni DNA. Nested PCR has high senstively and specificaly diagnostic method of C.burnetii as agent of Q fever disease.

Abstract

The objective at this experiment was to study the role of zinc supplementation (Zn) in minimizingstress at parturition. Thirty etawah cross breed age 3-6 years with average body weight 30-50 kg were usedin this study and divided into three groups ; i) animal were given grass + feed concentrate + ZnSO4 40 mg/kg dry matter ( control group) ; (ii) grass + feed concentrate + ZnSO4 60 mg/kg dry matter (treatment 1), and(iii) grass + feed concentrate + ZnSO4 80 mg/kg dry matter (treatment 2), respectively drinking water weregiven ad libitum. To monitor zinc and cortisone level, blood samples were withdrawn from each animalevery two weeks starting at age of pregnancy 3 months up to two months post parturition, in addition, tomonitor the cortisone level blood samples were also collected at 1,2,3 and 7 days post parturition. The Znand cortisone serum were analyzed using atomic absorbent spectro photometric (AAS) andradioimmunoassay (RIA), respectively the result showed that generally pregnancy at the beginning andget serum level at cortisone was relatively similar between the three animal groups and there was atendency to increase and reached peak at parts then gradually decrease until the and of the observation. Atparturition the lowest serum level at cortisone (19.00±18.72 mg/ml) was observed in animals receiving the60/ dry matter zinc supplement compared to animal receiving 80 mg/ dry matter zinc ( 52.65 ± 30.83 mg/ml) and control animal ( 75.92 ± 42.88 mg/ml). in addition serum level at Zn was significantly higher inanimal receiving 60 mg and 80 mg Zn in their diets compared to control animals. The best profiles wereseen in animal given 60 mg/kg dry matter Zn supplement. In conclusion, the addition of Zn in the diets hadsignificant effect in minimizing stress as pictured by the low serum level at cortisone at parturition.

Abstract

Kinetic study on Anti CPV IgY has been performed on six dogs aged 5-10 months. The IgY was injectedintravenously at dose of 21.4mg /10kg body weight. IgY levels in the blood were determined by ELISA. Aresearch was conducted to find out the kinetics of Anti CPV IgY in dogs blood. The kinetics of IgY wascalculated by using regression analysis to determine the association on the levels of IgY in serum againsttime at injection. The results showed that kinetic parameters were calculated based on first order kinetics.The constant elimination rate of IgY was at the range between 0.007 to 0.015 / h. IgY concentration in thedogs blood was from 0.746 to 0.992 mg / mL. The half-life of IgY was from 1.65 to 4.01 / d. Volumedistribution of IgY was between 21.47 to 28,55 / mL. Total IgY in the dog bodies (AUC) was from 42,60 to142,00 mg / mL.h. The duration of the IgY in the dog’s body was 3.08 to 8.51 days. Clearance time of IgY was0.15 to 0.50 mL / h. In conclusion the kinetics of anti CPV IgY in dog’s body follow one compartment andfirst order model, which are only distributed in the blood with the half-life at 2.5 days, and IgY has lesspossibility to accumulate in the body compared to the IgG.

Abstract

This study aimed to figure out the prevalence and risk factors of Mycoplasma gallisepticum (MG) infection in commercial layer farm in Blitar District. Blood samples and questionnaires were taken during December 2014 to February 2015. A total of 264 sera samples were collected from 22 commercial layer farm. Based on serum plate agglutination test, 26 sera samples were MG positive that were indicated an infection prevalence was 9.85%. The highest SRR value for MG infection was occurred in Bakung Subdistrict (SRR = 2.5). Based on Analysis of multivariate logistic regression showed that a very significant influenced risk factors of MG infection have occurred in flocking density more than 3,000 birds per flock (?2= 11.10; p= 0.001; OR= 6.1), flocking density about 1,501 to 3,000 birds per flock (?2 = 11.10; p= 0.001; OR= 6.1), bird feeding once a day (?2= 9.32; p= 0.002; OR= 0.3), house desinfection once in every two weeks (?2 =7.70; p= 0.009; OR= 1.2), house desinfection once a month or only in case (?2= 9.36; p= 0.006; OR= 3.9). It was concluded that seroprevalence of MG infection in studied area was 9,85%. the MG seroprevalence were influenced by flocking density more than 3,000 birds per flock, flocking density about 1,501 to 3,000 birds per flock, bird feeding once a day, house desinfection once every two weeks, and house desinfection once a month or only in case. ABSTRAK Mycoplasmosis merupakan salah satu penyakit paling penting yang dihadapi oleh industri perunggasan di Indonesia. Penelitian ini bertujuan untuk mengetahui prevalensi dan faktor risiko penularan Mycoplasma gallisepticum (MG) pada peternakan ayam petelur komersial di Kabupaten Blitar, Jawa Timur. Contoh darah dan kuisioner diambil selama periode Desember 2014 hingga Februari 2015. Sebanyak 264 contoh serum dikumpulkan dari 22 peternakan ayam petelur komersial. Berdasarkan uji Rapid Serum Agglutination (RSA), 26 contoh ditemukan positif MG dengan prevalensi infeksi MG di Kabupaten Blitar sebesar 9,85%. Nilai rasio risiko terbakukan (standardized risk ratio, SRR) untuk penularan MG paling tinggi adalah Kecamatan Bakung (SRR = 2,5). Hasil analisis regresi logistik multivariate didapatkan bahwa faktor yang berpengaruh sangat nyata (P<0,01) terhadap terjadinya infeksi MG adalah kepadatan flok di atas 3.000 ekor ( ?2= 11,10; p= 0,001; OR= 6,1), kepadatan flok 500-1.500 ekor (?2 = 11,09; p= 0,004; OR= 3,4), pemberian pakan satu kali dalam sehari (?2= 9,32; p= 0,002; OR=0,3), penyemprotan kandang yang dilakukan satu kali dalam dua minggu (?2 = 7,70; p= 0,009; OR= 1,2),dan penyemprotan kandang yang hanya dilakukan satu kali dalam sebulan atau hanya jika terjadi kasus ?2 ( = 9,36; p= 0,006; OR= 3,9). Hasil penelitian dapat disimpulkan bahwa prevalensi infeksi MG di daerah kajian adalah 9,85%. Faktor risiko yang berpengaruh sangat nyata terhadap penularan MG antara lain jumlah ayam yang dipelihara di atas 3.000 ekor per kandang, jumlah ayam yang dipelihara sebanyak 1.501- 3.000 ekor per kandang, pemberian pakan satu kali sehari, penyemprotan kandang satu kali dalam dua minggu, dan penyemprotan kandang satu kali sebulan atau hanya jika ada kasus. Mycoplamosis is one of an important poultry diseases that should be well managed by poultry farmer in Indonesia.

Abstract

Faculty of Animal Science, IPB University has developed new trait of chicken named IPB D1 traits. The chicken is a crossbreed chicken between male F1 PS (Pelung x Sentul) with F1 female (Kampung x parent stock Cobb). The IPB D1 chickens aims to produce the local commercial meat type, which adapted to traditional or semi intensive management (back yard). Salmonellosis is one of the bacterial disease that commonly infected poultry, therefore a study was conducted to determine the immune response of IPB D1 chickens to these disease using genetic markers, namely TLR4 gene (Toll-Like Receptor 4). TLR4 gene is one of the genes that control chicken resistance to Gram negative bacteria infection, through non-specific immune responses, one of Gram negative bacteria is Salmonella enteritidis. The aim of this study was to determine of TLR4 gene and their role in immunity of IPB D1 chickens against S. enteritidis infection. As much as 11 chickens were use in this study. This research was carried out through several stages of i.e determination of the TLR4 gene by PCR and sequencing and observing its immune response through the number of leukocyte, leukocyte differentiation and other hematology profile. Humoral immunity response against S. enteritidis were observe by clearance test before and after challenged with S. enteritidis. The results of this study indicate that IPB D1 chickens which have TLR4 genes (genotype GG and AG) were resistance against S. enteritidis based on the leukocyte, hematology profile and humoral immunity.

Abstract

The aim of this research was to prepare rabbit Immunoglobulin G as anti-idiotype antibody (Ab2) ofAvian Influenza Virus (AIV) H5N1. A polyclonal antibody was collected from guinea pigs immunized withinactivated AI vaccine H5N1of Legok strain. Antibody of H5N1 AI in serum was detected by Agar gelprecipitation test (AGPT) and an Inhibition Hemmaglutination test (IHT). The highest titre of antibodywas obtained one week after the third immunization. Serum of guinea pigs containing IgG was purifiedusing the Montage Antibody purification kit & spin column with Prosep A media (Millipore). The AI H5N1IgG concentration was 8 mg/ml. AI H5N1 IgG, was then digested with pepsin to obtain F(ab)2 fraction andwas called Ab1. The concentration of IgG and F(ab)2 and purity of IgG were determined by UVspectrophotometer which showed Ab1 concentration 1 mg/ml. Molecular weight was estimated by sodiumdodecyl sulfate- polyacrilamide gel electrophoresis (SDS-PAGE). Ab2 was produced by immunization ofrabbit with Ab1. The first immunization was carried out by subcutaneous injection with 500 ?g of Ab1emulsified in Complete Freund Adjuvant. The immunization was repeated with the same dose of Ab1emulsified in Incomplete Freund Adjuvan at 1 week intervals. One week after the second immunization,rabbit’s serum was harvested and IgG was purified using the Montage Antibody purification kit & spincolumn with Prosep A media (Millipore). The rabbit IgG, called Ab2, was an anti-idiotypic antibody againstAIV-H5N1. In AGPT, a precipitation line appeared between Ab1 and Ab2. A partial reaction appearedbetween Ab2 and the AI H5N1 antigen was also detected. The results indicated that Ab2 is a possiblecandidate of imunogen for protection against an AI virus H5N1 infection.

Abstract

This study was conducted to determine the seroprevalence of Brucella canis of imported dog for breederthrough Soekarno Hatta International Airport, Cengkareng Tanggerang, Indonesia. A total of 64 serumsamples were obtained from breeder dogs imported to Indonesia between Mei–September 2014. Theserums were examined by Immunochromatographic Assay. None of the serum samples in this studytested positive. Result of this study indicated that antibodies of B. canis were not detected in 64 dogsimported to Indonesia during the research period. A Knowledge, Attitude and Practises (KAP) Survey wasalso performed. This study used 32 respondents and data were analyzed by using path analysis. Therespondents were breeding kennel owners (breeder) who imported dogs through Soekarno HattaInternational Airport during the research period. The study concluded that the knowledge significantlyinfluenced the attitude of the breeder, and also the attitude significantly influenced the practises ofpreventing and controlling brucellosis. The annually income was also identified as a variable thatsignificantly influenced the practises. Practice prevention and control of canine brucellosis can be improvedby increasing the knowledge of the breeding kennel owner.

Abstract

The effectiveness of homeopathy combination in eliciting nonspecific cellular defense mechanism indairy cows is demonstrated in this study. Two groups of 2-7 years old, in 3-5 months term of pregnancydairy cows which suffering clinical mastitis were used id this study. At 3 weeks and 4 weeks prae partusanimals received their first homeopathy treatment, i.e. Group I: Coenzyme comp®, and Group II: a combinationof Mucosa comp® and Traumeel®, respectively. At one week and two weeks post partus all animals in thetwo groups received their second treatment, i.e. a combination of Lachesis comp® and Traumeel®.Subsequently, at 3 and 4 weeks post partus all animals in the two groups received their third treatment,i.e. a combination of Carduus comp® and Coenzyme comp®, respectively. All homeopathy solutions weregiven at 5 ml subcutaneously. Ten ml of blood samples for haptoglobin analysis were drawn from eachanimal and blood smears for White-Blood-Cell counting and differentiation and phagocytosis activity andcapacity observation were prepared from each animal at 3 and 4 weeks prae partus; and at one week, 3, 5,and 7 weeks post partus, respectively. Our results indicated that although there were non significantdifference in the phagocytosis activity and capacity between animals in Group I and Group II; animals inGroup II showed a marked increase in phagocytosis activity and capacity. Based on haptoglobin analysis,animals in Group II showed a steadily low haptoglobin level in the blood (0.08 mg/ml) up to 3 weeks postpartus and a tendency to further decline until the end of the observation. Based on the WBC counts anddifferentiation, animals in Group II also showed a better response compared to animals in Group I.In conclusion, the use of homeopathy combination is capable in eliciting nonspecific cellular defensemechanism; this phenomena were clearly shown in the combination of Mucosa comp® and Traumeel®. Theuse of this homeopathy combination should be considered at prae partus allowing the recognition of thehost defense mechanism.

Abstract

Salmonella spp. has been recognized as the major cause of food-borne illness in humans worldwidecausing remain relevant to public health. Poultry vaccination is one promising strategy to mitigateSalmonella infection in poultry and, in turn, in humans as well. The objective of this study was to assessthe potential of cross-reaction of serum in Salmonella enteritidis-vaccinated chicken to some serotype ofSalmonella enterica. Four female, Isa Brown layer chickens (20 weeks old), were vaccinated with S. enteritidisstrain Sm24/Rif12/Ssq (intra vena) to induced the production of specific antibodies in serum. Crossreactionof serum in S. enteritidis-vaccinated chicken were assess with agar gel immunodiffusion test(AGID) with S. enteritidis, S. pullorum, S. typhimurium, S. typhi, and Escherichia coli antigens. Serumcould react with S. enteritidis and all types of S enterica used in this study (S. pullorum, S. typhimurium,S. typhi), but could not react with E. coli. The potential of cross-reaction of serum in S. enteritidis-vaccinatedchicken to some serotypes of S. enterica may play a role in reducing the infection caused by that serotype.
