Abstract

Abstract Dayak onion (Eleutherine bulbosa (Mill.) Urb.) has the potential to be an immunostimulant to benefit fish health. This study aimed to evaluate the effectiveness of dietary supplementation with the powder and crude ethanol extracts of Dayak onion on the hematology and immune response parameters of Nile tilapia challenged with Aeromonas hydrophila. The research used a completely randomized design with six treatments and three replications. Nile tilapias were fed with Dayak onion (w/v) consisting of powders of 5% (P5), 10% (P10), and 15% (P15), while crude extract was 0.5% (E05) and given during rearing for 30 days. Positive control (C+) and negative control (C-) were used without Dayak onion. Nile tilapia in treatment C+, P5, P10, and P15 were injected with A. hydrophila 10⁶ CFU mL⁻1, while that in treatment C- was injected with phosphate buffered saline (PBS) intramuscularly. The results showed that the dietary with 15% powder and 0.5% crude extract significantly improved the hematology and immune response parameters compared to the control after the challenge against A. hydrophila (P<0.05). Dayak onion supplementation effectively increased the health status based on hematology and immune response parameters of Nile tilapia against A. hydrophila infection. Highlight Research Dayak onion has significant potential to improve nonspecific immunity in fish. Dayak onion improves hematology and immune response of Nile tilapia. Dayak onion can significantly regulate the immune system against A. hydrophila infection. Dayak onion significantly increases the production of the gene expression of IL-1β and TNF-α.

Abstract

Penelitian ini bertujuan menentukan frekuensi pemberian vaksin DNA koi herpesvirus (KHV) yang menghasilkan kelangsungan hidup tinggi pada ikan mas yang diinfeksi KHV. Frekuensi pemberian vaksin yaitu vaksinasi satu kali seminggu (K1), vaksinasi dua kali seminggu (K2), vaksinasi tiga kali seminggu (K3), ikan tidak divaksin dan diuji tantang KHV (K4), dan ikan diinjeksi dengan larutan phosphate buffered saline (K5). Pemberian pakan dilakukan 2 kali sehari secara satiasi.Mortalitas pada K1; K2; K3; dan K4 masing-masing adalah 33,335,77; 20,0010,00; 6,67 5,77; dan 43,335,77%, sedangkan kelangsungan hidup relatif (RPS) K1; K2; dan K3 masing-masing adalah 23,0713,32; 53,8423,07; 84,6013,32%. Disimpulkan bahwa pemberian pakan mengandung vaksin DNA anti-KHV dengan frekuensi tiga kali dalam satu minggu efektif meningkatkan kelangsungan hidup ikan mas yang diinfeksi KHV.

Abstract

Curcuma spp. is a phytobiotic with potential application in fish farming. This study aimed to evaluate the potential of Curcuma zedoaria, Curcuma aeruginosa and Curcuma mangga extracts in striped catfish infected with Aeromonas hydrophila. The study used a complete randomized design (CRD) consisting of six treatments: KN (negative control), KP (positive control, fish infected without treatment), PE (C. zedoaria extract 6.25 g kg-1), HE (C. aeruginosa extract 6.25 g kg-1), ME (C. mangga extract 6.25 g kg-1) and KE (combination of C. zedoaria extract 2.1 g kg-1 + C. aeruginosa extract 2.1 g kg-1 + C. mangga extract 2.1 g kg-1). A total of 360 striped catfish, 10 ± 0.5 cm were kept in 18 aquariums measuring 50 × 40 × 35 cm3 and fed the treatment for 30 days. The challenge test was conducted on day 31 by injecting A. hydrophila suspension (106 colony-forming units (CFU) mL-1) intramuscularly into fish. The results showed that the treatment administered were able to stimulate the expression of interleukin-1â, interferon-ã 2a, 2b genes, increase the number of red and white blood cells, hematocrit, hemoglobin, phagocytosis activity, respiratory burst, reduce the population of A. hydrophila in the intestine, and reduce tissue damage in striped catfish. Striped catfish treated with the extracts showed significantly higher survival rates (p<0.05) compared to the positive control group. The survival rates were: KN (100.00 %), KP (53.33%), PE (93.33 %), HE (91.67 %), ME (93.33 %), and KE (88.33 %). In conclusion, the treatment administered were able to enhance the immune response and resistance of striped catfish infected with A. hydrophila.

Abstract

Desa Purwasari yang berada di Kabupaten Bogor merupakan salah satu desa yang memiliki potensi lahan sawah yang relatif luas dan pengairan yang sangat baik, namun demikian tingkat pendidikan warganya masih relatif rendah sehingga mengalami keterbatasan dalam mendapatkan pekerjaan yang layak. Salah satu program Desa Purwasari yang dilaksanakan dari Dana Desa adalah program ketahanan pangan. Melihat potensi desa tersebut, maka dilaksanakan program penanaman padi dengan sistem mina padi. Durasi pemeliharaan ikan pada mina padi belum dapat mengakomodir kegiatan pembesaran ikan nila konsumsi, sehingga warga masih belum dapat melakukan pembesaran ikan nila secara maksimal untuk memproduksi protein hewani. Oleh karena itu, tim dosen Fakultas Perikanan dan Ilmu Kelautan IPB University yang dibantu oleh mahasiswanya melaksanakan kegiatan pengabdian pada masyarakat di desa ini. Metode yang dilakukan adalah dengan memberikan pelatihan dasar-dasar budidaya dan budidaya ikan nila secara intensif dengan teknologi bioflok, praktek langsung budidaya ikan nila dengan teknologi bioflok, dan pendampingan, Di akhir kegiatan didapatkan bahwa air kolam sudah mulai membentuk flok. Diharapkan hasil dari kegiatan pengabdian pada masyarakat ini dapat menyebar ke warga desa lainnya sehingga cita-cita Desa Purwasari menjadi desa sentra nila dapat segera terwujud.

Abstract

Penelitian ini bertujuan untuk mengevaluasi pengaruh suplementasi glutamin terhadap pertumbuhan dan aktivitas antioksidan pada ikan Torsoro (Tor soro). Ikan uji yang digunakan adalah juvenil ikan dengan berat awal rata-rata 5,21 ± 0,04 g. Perlakuan yang diberikan terdiri atas suplementasi glutamin bebas dengan dosis yang berbeda (0; 0,5; 1,0; 1,5; dan 2%) dan alanil-glutamin (0,84; 1,67; 2,51; dan 3,35%) pada pakan, setiap perlakuan memiliki lima kali ulangan. Perlakuan pakan diberikan tiga kali sehari sekenyangnya. Ikan uji dipelihara selama 60 hari. Parameter yang diamati meliputi performa pertumbuhan, pemanfaatan nutrisi, indeks biologis, dan aktivitas antioksidan. Hasil penelitian menunjukkan bahwa pakan yang disuplementasi glutamin bebas berpengaruh nyata (p<0,05) terhadap performa pertumbuhan, pemanfaatan nutrisi, indeks biologis, dan aktivitas antioksidan ikan uji. Hasil uji lebih lanjut menunjukkan bahwa suplementasi alanil-glutamin dalam pakan sebesar 2,51% memberikan hasil terbaik terhadap bobot akhir, laju pertumbuhan spesifik, retensi protein, rasio efisiensi protein, konversi pakan, rasio viseral somatik, rasio panjang usus, aktivitas superoksida dismutase, glutation peroksidase, dan katalase pada ikan Torsoro. Berdasarkan evaluasi keseluruhan terhadap parameter yang diamati, penelitian ini menunjukkan bahwa suplementasi alanil-glutamin dalam pakan pada tingkat 2,51% mampu meningkatkan kinerja pertumbuhan, pemanfaatan nutrisi, indeks biologis, dan aktivitas antioksidan pada ikan Torsoro.This study aimed to evaluate the effects of glutamine supplementation on the growth of and antioxidant activity in Torsoro fish (Tor soro). The test fish used were Torsoro fish juveniles with an average initial weight of 5.21 ± 0.04 g. The treatments consisted of different supplementation levels of free glutamine (0; 0.5; 1.0; 1.5; and 2%) and alanyl-glutamine (0.84; 1.67; 2.51; and 3.35%) in feed, where each treatment had five replicates. The feed treatments were given three times a day ad satiation. The experimental fish were reared for 60 days. The parameters observed included growth performance, nutrient utilization, biological indices, and antioxidant activity. The results showed that free glutamine-supplemented feed significantly affects (p<0.05) the growth performance, nutrient utilization, biological indices, and antioxidant activity of the tested fish. Further test results showed that alanyl glutamine supplementation in feed at 2.51% produced the best results on the final weight, specific growth rate, protein retention, protein efficiency ratio, feed conversion, somatic visceral ratio, intestine length ratio, superoxide dismutase, glutathione peroxidase, and catalase activities of Torsoro fish. Based on the overall achievement of the observed parameters, this study determines that alanyl-glutamine supplementation in feed at 2.51% improves the growth performance, nutrient utilization, biological indices, and antioxidant activity of  Torsoro fish.

Tujuan penelitian ini adalah mengevaluasi suplementasi L-karnitin dan tepung kayu manis terhadap kadar lemak dan tekstur daging ikan patin pada fase pembesaran. Ikan patin berukuran 125,4 ± 7,85 g dan panjang 24,71±0,68 cm dipelihara selama 60 hari dalam wadah hapa berukuran 2 x 1 x 1 m3. Ikan diberi pakan tiga kali sehari ad satiation dengan perlakuan, sebagai berikut: kontrol (K), penambahan L-karnitin 1 g kg-1 (LK 1), L-karnitin 2 g kg-1 (LK 2), kayu manis 5 g kg-1 (KM 5), dan kayu manis 10 g kg-1 (KM 10). Sampel diambil pada awal, pertengahan, dan akhir pemeliharaan untuk pengukuran kadar lemak daging. Parameter yang diamati yaitu lemak daging dan tekstur daging. Hasil menunjukkan bahwa pemberian KM 10 menghasilkan kadar lemak yang lebih rendah dari perlakuan lainnya setelah 30 hari dan penurunan yang lebih besar yaitu berkisar 51,06% dan 42,55% pada perlakuan LK 2 dan KM 10 pada hari ke-60. Nilai indeks hepatosomatik juga menurun yang diikuti oleh penurunan kadar lemak hati. Nilai kekerasan daging menunjukkan peningkatan kualitas yang terlihat dari nilai yang semakin rendah. Pemberian tepung kayu manis 10 g kg-1 pada fase pembesaran menunjukkan hasil terbaik pada pemberian selama 60 hari dalam menurunkan lemak daging ikan patin hingga memenuhi standar filet, karena adanya proses lipolisis serta pemanfaatan lemak menjadi energi. Kayu manis ini menjadi sangat potensial untuk dikembangkan sebagai feed additive dalam upaya memperbaiki kualitas daging ikan patin fase pembesaran.Striped catfish meat has a high-fat content which is undesirable to markets and customers alike and needs to be reduced. This study aims to evaluate the effect of dietary L-carnitine and cinnamon powder on reducing the fat content of striped catfish meat in the grow-out stage. Striped catfish measuring 125.4 ± 7.85 g and 24.71 ± 0.68 cm body length were kept for 60 days in a hapa with size 2 x 1 x 1 m3. Thefish were fed three times a day at satiation with the following treatments: control (K), the addition of L-carnitine 1 g kg-1 (LK 1), L-carnitine 2 g kg-1 (LK 2), cinnamon 5 g kg-1 (KM 5) and cinnamon 10 g kg-1 (KM 10). Sampling was conducted on the initial, middle and final day for meat fat content analysis. The parameters observed were meat fat and meat texture. The results showed that dietary KM 10 significantly reduced meat fat content compared to controls on the 30th and it showed a more significant reduction, namely 51.06% and 42.55%, in the treatment LK 2 and KM 10 after 60 days treatment. The hepatosomatic index value also decreased, followed by decreased liver fat levels. The lower values of meat hardness imply an increase in meat quality. It can be concluded that the application of dietary cinnamon powder at 10 g kg-1 (KM 10) is the best level to reduce the fat content of striped catfish meat in meeting the fillet standards. This cinnamon has excellent potential to be developed as a feed additive to improve the quality of striped catfish meat in the rearing phase.

Abstract

ARTICLE HIGLIGHTS- Taurine improves eye development in golden rabbitfish larvae- Enhanced growth performance with taurine-enriched rotifers- Taurine positively impacts fish eye and body growthABSTRACTGolden rabbitfish (Siganus guttatus) is an emerging species for aquaculture industry, despite constrains faced on commercial scale of seed production due to the high mortality during the first-feeding stage. An experiment was conducted to determine the effect of taurine through enrichment of rotifers as live-feed on eye development and growth performance of larval golden rabbitfish. Observation of eye development was carried out by measuring eye diameter of the larva and development of the retina based on histology assessment. Growth performance was measured for absolute growth and fin development of the larvae and survival rate. The results showed that taurine-enriched rotifer generally provided better performances for eye development and growth of larval golden rabbitfish compared with control (without taurine). Increase of taurine dose up to 0.050 g/L resulted in an increase of eye diameter, absolute growth, fin development and survival rate. Further increased increment of the taurine tended to decrease the values of the measured parameters. The eye diameter of larval golden rabbitfish fed with 0.050 g/L taurine-enriched rotifer was significantly wider (106.1±9.8 µm) (P < 0.05) compared with control (58.2±14.3 µm), but did not significantly differ from other doses of taurine (P > 0.05). Body width of larvae fed with 0.050 g/L taurine-enriched rotifer was significantly higher (127.3±14.6 µm) (P < 0.05) compared with control (98.8±18.3 µm). In regard to eye development, growth performances (total length, fin development and survival rate), dose of taurine for rotifer enrichment fed to larval golden rabbitfish Siganus guttatus was 0.050 g/L.

Abstract

An alternative substitution of pollard as an imported feed ingredient is a necessity, and one of the potential ingredients is water hyacinth (Eichhornia crassipes). This study was conducted to evaluate growth performance and enzyme activity in catfish (Pangasianodon hypophthalmus). Diet with five different levels of water hyacinth substitute of pollard (0%, 25%, 50%, 75%, and 100%) were fed to catfish for 60 days. Seventy (70) catfish fry with average initial body weight of 2.45 ± 0.15 g were maintained in 100 cm × 80 cm × 60 cm aquaria. Fish fed at satiation level three times daily at 8 am, 12 pm, and 4 pm. With dietary 25% water hyacinth, growth performance and protease activity similar to 0% treatment. Feed intake, protein digestibility, feed efficiency, protein efficiency ratio, and protease and amylase enzyme activities decreased (p<0.05) in those fed with more than 25% water hyacinth. Catfish fed 25% water hyacinth showed significantly (p<0.05) higher daily growth rate, feed efficiency and protein digestibility than those with other treatments. Based on the growth performance and enzyme activity, we can conclude that the optimum dietary level of water hyacinth substitute pollard for fry catfish is 25%.

Abstract

Although it contains heavy metals, the highly nutritious green mussel Perna viridis is used as a fish meal replacement in fish diets. Fortunately, humic acid (HA) has the ability to chelate heavy metals in animal feeds. Its addition to fish feed formulation is, therefore, needed to prevent the accumulation of heavy metals in the fish's body. Hence, an experiment using a Completely Randomized Design (CRD) with 5 treatments and 3 replications was conducted to evaluate the performance and feed efficiency of Asian seabass Lates calcarifer juveniles with the addition of HA to their green mussel–based diet containing the heavy metal cadmium (Cd). Five experimental diets were formulated with different dosages of HA addition at 0, 400, 800, 1200, and 1600 mg kg⁻¹ of feed as treatments A, B, C, D, and E, respectively. The experimental diets were given to triplicate groups of 15 Asian seabass juveniles with an initial body weight of 4.30 ± 0.60 g over a 70-day cultivation period, feeding until satiation. The results showed that feed consumption, feed digestibility, protein retention, growth performance, and feed efficiency were significantly affected by HA addition in the fish diet. Among all treatments, HA addition of 1600 mg kg⁻¹ produced the best biological response from the Asian seabass. To conclude, the HA addition of 1600 mg kg⁻¹ into the diet was the best dosage, resulting in optimal growth performance and feed efficiency in Asian seabass. The highest dose of HA in Cd-contaminated feeds could reduce Cd content, but had not yet been able to eliminate Cd in fish meat. Therefore, green mussel meal, with HA addition as an alternative protein source, has potential as a feed additive for Cd-contaminated diets of Asian seabass juveniles.
