Abstract

ABSTRACTThis experiment was conducted to evaluate the use of by product of corn ethanol industry, DDGS (distiller dried grain with soluble) and hominy as feed raw material on digestibility and the growth performance of humpback grouper Cromileptes altivelis juvenile. Three experimental diets containing isoprotein (48%) but different in DDGS contain were used. Diet A containing 0% of DDGS and hominy while diet B and C containing 10.13% (6.13% of DDGS and 4% of hominy) and 19.05% (15.05% of DDGS and 4% of hominy). Reduce of fish meal level in the diet was happen as a consequence of balancing amino acid. For digestibility experiment and Cr2O3 at level of 0.05% was used as a tracer. The feces were daily collected 30‒60 minutes after feeding for 14 days. Dry matter and protein were used as digestibility parameters. Completely randomized design with three treatments and two replicates was used in this experiment. Humpback grouper with an average weight of 11‒14 g were reared at floating net cages (FNG) of 3×2×1.5 m3. The stocking density for each cage was 167 fish/FNG and reared for six months. For digestibility measurement 30 fish were held in cylindrical fiber tank filled with 800 L of sea water. Fish were fed three times daily at satiation level. For maintaining net in a clean condition net was changed in every ten days. Feed consumption, digestibility of protein and total, growth performance indicator i,e specific growth rate, feed efficiency protein and lipid retention were used as evaluating parameters. The results showed that the fish fed with diet containing DDGS (6.13%) dan hominy (4%), at the level of 10.13% (diet B), shows similar growth performance indicators with the fish fed diet A (0% of DDGS and hominy) and the growth performance indicator was higher when being compared to those fish fed diet C (containing 19.05% of DDGS (15.05%) and hominy (4%)) (p<0.05). More over the inclusion of DDGS and hominy at the level of 10,13% give no negatif impact to feed palatability (shown by total feed consumption value) and digestibility of protein. The increasing level of DDGS and hominy more than 10% reduced palatability and protein digestibility. Ikan yang digunakan dalam penelitian ini adalah juvenil ikan kerapu bebek berukuran 11‒14 g. Ikan dipelihara dalam karamba jaring apung yang berukuran 3×1,5×2 m3 dengan kepadatan 167 ekor/jaring. Ikan diberi pakan sampai kenyang sebanyak dua kali sehari. Untuk menjaga agar jaring tetap bersih maka dilakukan pergantian jaring setiap satu sampai dua minggu sekali. Ikan dipelihara selama lima bulan dan setiap bulan dilakukan sampling. Tiga macam pakan digunakan dalam penelitian ini dengan kadar protein 47% dan rasio energi protein 10 kkal/g protein. Pakan A adalah pakan yang tidak mengandung DDGS dan homini, pakan B mengandung DDGS 6,13% dan 4% (total 10,13%), dan pakan C mengandung DDGS 15,05% dan Homini 4% (total 19,5%). Untuk mengevaluasi ketercernaan nutrien maka kromium oksida (Cr2O3) digunakan sebagai penelusur. Penelitian ketercernaan dilakukan di laboratorium dengan menggunakan fiber dengan volume 800 L. Rancangan acak lengkap dengan tiga perlakuan dan dua ulangan digunakan dalam penelitian ini. Hasil penelitian menunjukkan bahwa nilai ketercernaan nutrien tidak berbeda sampai total kadar DDGS dan homini 10,13% (pakan B) dan peningkatan DDGS serta homini sampai 19,05% (pakan C) menurunkan nilai ketercernaan. Ikan yang diberi pakan dengan DDGS sebesar 10,13% (pakan B) memiliki nilai efisiensi pakan, sintasan, laju pertumbuhan spesifik, konsumsi pakan dan retensi protein yang sama dengan pakan tanpa DDGS (pakan A), akan tetapi nilai-nilai tersebut lebih tinggi jika dibandingkan dengan ikan yang diberi pakan B (p<0,05). Retensi lemak ikan yang diberi pakan B memiliki nilai yang lebih tinggi jika dibandingkan dengan ikan yang diberi pakan A dan C. Dari penelitian ini dapat disimpulkan bahwa DDGS dan homini dapat digunakan sebagai bahan baku pakan ikan kerapu bebek sampai level 10,13% (DDGS 6,13% dan homini 4%).Kata kunci: pertumbuhan, ketercernaan, DDGS, homini, ikan kerapu bebek, Cromileptes altivelis

Abstract

ABSTRACT The research was conducted to evaluate effect of the administration of Artemia enriched with vitamin C on the survival, total body length, and average body weight of Pangasionodon sp. larvae. Artemia were enriched with either 0, 50, 100, or 150 mg/L vitamin C immersed in the culture medium for 13 hours. Two days old larvae were fed on enriched Artemia 12 times/day for seven days. At the end of feeding experiment, it was found that fish fed on Artemia enriched with 100 mg/L culture medium had the highest survival, total length and body weight, respectively 76.17±12.78%, 1.63±0.34 cm, and 20.6±3.8 mg. Keywords: survival, growth, Artemia, vitamin C, catfish, Pangasionodon sp.  ABSTRAK Penelitian ini dilakukan untuk mengetahui pengaruh Artemia yang diperkaya dengan vitamin C dosis berbeda terhadap kelangsungan hidup, panjang total, dan bobot tubuh larva ikan patin Pangasionodon sp. Pada penelitian ini, Artemia diperkaya dengan vitamin C dosis 0, 50, 100, atau 150 mg/L media pengayaan. Larva yang berumur dua hari diberi pakan Artemia hasil pengayaan sebanyak 12 kali sehari selama tujuh hari masa pemeliharaan. Pada akhir percobaan, didapatkan bahwa larva ikan yang diberi Artemia diperkaya vitamin C dosis 100 mg/L media pengayaan memberikan kelangsungan hidup 76,17±12,78%, panjang total 1,63±0,34 cm, dan bobot tubuh paling tinggi 20,6±3,8 mg. Kata kunci: kelangsungan hidup, pertumbuhan, Artemia, vitamin C, ikan patin, Pangasionodon sp.

Abstract

ABSTRACT This research was aimed to evaluate the role of fish meal from different raw materials in improving growth performance of sangkuriang catfish Clarias sp. The source of raw materials used were derived from trash fish, salted fish, and head fish eal. Fish reared for 44 days and were given feed test three times per day at 08.00, 12.00, and 16.00. Biomass weight of the experimental fish used were 38.49±1.61 g. The fish reared on the aquarium of 60x50x40 cm3 and by using recirculation system. The results showed that fish meal derived from trash fish provide the best result for feed consumption, daily growth rate, absolute length, biomass, retention of protein, retention of fat, feed efficiency, feed conversion (P>0.05). However, it does not significantly different on the survival of fish (P>0.05). The data showed that the use of trash fish meal resulted on the best growth performance of sangkuriang catfish. It was indicated that the trash-fish meal might be used as the main protein source of sangkuriang catfish feed. Keywords: catfish, growth, at satiation, fish meal  ABSTRAK Penelitian ini bertujuan untuk mengevaluasi pakan uji yang menggunakan sumber tepung ikan dari bahan baku berbeda dalam meningkatkan kinerja pertumbuhan lele sangkuriang Clarias sp. Sumber tepung ikan yang digunakan berasal dari ikan rucah, ikan asin, dan kepala ikan, yang memiliki umum digunakan sebagai salah satu bahan baku pakan lokal. Ikan diberi pakan at satiation selama 44 hari dengan frekuensi tiga kali sehari, yaitu pada pukul 08.00, 12.00, dan 16.00. Biomassa rata-rata ikan uji yang digunakan adalah 38,49±1,61 g. Ikan dipelihara dalam akuarium berukuran 60x50x40 cm3 dan dengan menerapkan sistem resirkulasi. Hasil penelitian menunjukkan bahwa tepung ikan yang berasal dari ikan rucah mampu memberikan hasil terbaik dalam meningkatkan kinerja pertumbuhan ikan uji. Hal ini ditunjukkan oleh nilai kinerja pertumbuhan seperti jumlah konsumsi pakan, laju pertumbuhan harian, panjang mutlak, biomassa, retensi protein, retensi lemak, efisiensi pemberian pakan, dan konversi pakan yang menunjukkan perbedaan nyata (P<0,05). Namun, tidak terjadi perbedaan pada kelangsungan hidup ikan (P>0,05). Hasil penelitian menunjukkan bahwa penggunaan tepung ikan rucah menghasilkan pertumbuhan terbaik sebagai sumber protein utama pada pakan ikan lele sangkuriang. Kata kunci : ikan lele, pertumbuhan, at satiation, tepung ikan

Abstract

ABSTRACT This research aimed at determining digestibility coefficient of cassava peel(Manihot utilissima) after immersion in 3% (w/v) NaOH for three days, fermentation using combined fungi of 10% Trichoderma viride and Phanerochaete chrysosporium for seven days, and fermentation using 15% (w/w) Bacillus megaterium for five days as feed ingredients for Nile tilapia Oreochromis niloticus. Total digestibility test was conducted by mixing 30% of cassava peel and 70% of reference diet. Nile tilapia at the average weight of 16.6 g were used as experimental fish. Fish was held for 28 days in aquarium (50x50x50 cm3) at the density of 10 fish/aquarium. Fish were fed twice daily to satiation. Feces collection started after five days of adaptation to chromium oxide diets. The results showed that the three treatments had significant effects compared to control (P<0.05), protein digestibility of were improved 5%, 15%, and 10%, energy digestibilitiy were 20%, 18%, 16%, and total digestibility of test cassava peel were 174%, 151%, and 164%, respectively. Cassava peel fermented with combined 10% mold showed the highest protein digestibility impliying it potency as feed ingredient for Nile tilapia diet. Keywords: Nile tilapia, cassava peel, NaOH, mold, bacteria, digestibility  ABSTRAK Penelitian ini bertujuan untuk mengevaluasi kecernaan kulit ubi kayu (Manihot utilissima) setelah perendaman dengan NaOH 3% (w/v) selama tiga hari, fermentasi kapang Trichoderma viride dan Phanerochaete chrysosporium 10% (w/w) selama tujuh hari, dan fermentasi bakteri Bacillus megaterium 15% (w/w) selama lima hari sebagai bahan baku pakan ikan nila. Uji kecernaan total bahan dilakukan dengan mencampurkan 30% kulit ubi kayu dengan 70% pakan acuan. Ikan uji yang digunakan adalah ikan nila dengan bobot rata-rata 16,6 g. Ikan dipelihara selama 28 hari dengan kepadatan 10 ekor/akuarium berukuran 50x50x50 cm3. Pemberian pakan dilakukan dua kali sehari secara at satiation. Pengumpulan feses dimulai setelah lima hari adaptasi pakan uji yang diberi indikator kromium oksida. Hasil penelitian menunjukkan bahwa ketiga perlakuan memberikan pengaruh yang berbeda nyata terhadap kontrol (P<0,05) dan meningkatkan nilai kecernaan protein pakan berturut-turut sebesar 5%, 15%, dan 10%, nilai kecernaan energi sebesar 20%, 18%, dan 16%, serta nilai kecernaan total bahan sebesar 174%, 151%, dan 164%. Perlakuan kulit ubi kayu yang difermentasi dengan kapang menunjukkan nilai kecernaan protein pakan yang tertinggi sehingga berpotensi sebagai bahan baku pakan ikan nila. Kata kunci: ikan nila, kulit ubi kayu, NaOH, kapang, bakteri, kecernaan

Abstract

ABSTRACT This study was aimed to determine the level of acute toxicity (LC50) and to analyze the effect of Cu on the survival, growth, haematological, and bioaccumulation of Cu in Pangasianodon hypophthalmus juvenile at sublethal conditions. The experiment trial used completely randomized design with five concentration treatments and two replications to determine LC50. The Cu concentration used were 0, 0.2, 0.7, 1.2, and 1.7 ppm. Sublethal treatment used four Cu concentrations with three replicates. The concentrations were 0, 0.167, 0.334 and 0.667 ppm. Experimental fish used had an average total length 11.0±1.7 cm and weight 13.00±1.72 g. Results showed that Cu was highly toxic to P. hypophthalmus with a LC50-96 hours value of 0.667 ppm (0.539–0.805 ppm). At sublethal concentrations, Cu significantly decreased survival and growth, and increased Cu accumulation started from concentration of 0.167 ppm. Bioaccumulation of Cu started from the highest were in liver, gills, skin and flesh, respectively. Decreased of erythrocytes, haemoglobin and hematocrit indicated anemia, while increased of leukocytes indicated infection and physical stress on the body tissues. Keywords: copper toxicity, haematological, bioaccumulation, Siam-catfish juvenile  ABSTRAK Penelitian ini bertujuan untuk menentukan tingkat toksisitas akut (LC50) dan menganalisis pengaruh Cu terhadap sintasan, pertumbuhan, kondisi hematologi, dan akumulasi Cu pada juvenil ikan patin siam (Pangasianodon hypophthalmus) pada kondisi subletal. Rancangan penelitian yang digunakan adalah lima perlakuan konsentrasi dengan dua ulangan untuk menentukan LC50. Konsentrasi Cu yang digunakan adalah 0; 0,2; 0,7; 1,2; dan 1,7 ppm. Pemeliharaan subletal menggunakan empat variasi konsentrasi dengan tiga ulangan. Konsentrasi Cu yang digunakan adalah 0; 0,167; 0,334; dan 0,667 ppm. Ikan uji yang digunakan memiliki panjang total rata-rata 11,0±1,7 cm dan berat 13,00±1,72 g. Hasil menunjukkan bahwa Cu sangat toksik terhadap juvenil P. hypophthalmus dengan nilai LC50-96 jam sebesar 0,667 ppm (0,539–0,805 ppm). Pada konsentrasi subletal, Cu berpengaruh nyata terhadap penurunan sintasan dan pertumbuhan serta menyebabkan peningkatan akumulasi Cu mulai pada konsentrasi 0,167 ppm. Bioakumulasi Cu secara berurutan mulai dari yang tertinggi yaitu pada hati, insang, kulit dan daging. Abstract

ABSTRACT This research was performed to evaluate growth performance and meat quality at different feeding period of striped catfish Pangasianodon hypopthalmus fed on cinnamon leaves Cinnamomum burmanni enriched diet. Twenty fishes at the average body weight of 21.24±2.39 g were distributed  into 100×40×50 cm3 of aquarium. Fish were fed on the diet contained C. burmanni leaves of either 0.0%, 0.5%, 1.0%, or 1.5% respectively for 60 days. Fish were fed on those diets three times a day at satiation. Fish meat quality was evaluated by organoleptic test and proximate analysis at day-20, 40, and  60. The result showed that until day-40, fish fed on the diet contained 0.0%, 0.5%, and 1.0% cinnamon leaves had no significant difference on growth, hence still showed higher growth than fish fed at the dose of 1,5%. Meanwhile at day-60, fish fed on the diets contained either 0.5%, 1.0%, and 1.5% cinnamon leaves significantly had lower growth than that fed on 0.0% cinnamon leaves. In accordance to the growth response, diet contained 0.5% cinnamon leaves given for 40 days has shown the best texture and the highest protein content of fish meat.               Keywords: cinnamon leaves, striped catfish, growth, meat texture  ABSTRAK Penelitian ini dilakukan untuk mengevaluasi pertumbuhan dan kualitas daging ikan patin Pangasianodon hypopthalmus yang diberi pakan mengandung tepung daun kayu manis Cinnamomum burmanni dengan dosis berbeda. Sebanyak 20 ekor ikan patin berukuran 21,24±2,39 g dipelihara dalam akuarium berukuran 100×40×50 cm3. Ikan diberi empat jenis pakan yang mengandung daun kayu manis masing-masing sebesar 0,0%, 0,5%, 1,0%, dan 1,5% selama 60 hari. Pemberian pakan dilakukan sebanyak tiga kali sehari secara at satiation. Kualitas daging ikan diuji melalui uji organoleptik dan analisis proksimat pada hari ke-20, 40, dan 60. Hasil menunjukkan bahwa sampai hari ke-40 pemeliharaan, pertumbuhan ikan yang diberi pakan mengandung 0,0%, 0,5%, dan 1,5% daun kayu manis menunjukkan kinerja pertumbuhan yang tidak berbeda nyata, tetapi bernilai lebih tinggi daripada ikan yang diberi  dosis 1,5%. Berbeda halnya dengan kinerja pertumbuhan ikan pada hari ke-60, ikan yang diberi pakan 0,5%, 1,0%, dan 1,5% secara signifikan memiliki pertumbuhan yang lebih rendah dari ikan yang diberi pakan 0,0%. Sesuai dengan respons pertumbuhan, ikan yang diberi pakan 0,5% daun kayu manis selama 40 hari memiliki tekstur daging yang terbaik dan protein tertinggi. Kata kunci: daun kayu manis, ikan patin, pertumbuhan, tekstur

Abstract

ABSTRACT The aim of this study was to determine the effect of various types of honey (longan honey, lychee honey, and cottonwoods honey) on sperm quality of Siamese catfish Pangasianodon hypopthalmus with different dilution ratios after the storage period which includes sperm viability, fertilization rate, and hatching rate. The best treatment was obtained on the aplication of longan honey as a sperm diluent at 1:50 dilution ratio, with sperm viability 96.33±0.58%, fertilization rate 97.10±0.70%, and hatching rate 93.44±2.39%. Keywords: Siamese catfish, Pangasianodon hypophthalmus, honey, sperm dilution ratio  ABSTRAK Tujuan penelitian ini adalah mengevaluasi pengaruh pemberian berbagai jenis madu (madu kelengkeng, madu leci, dan madu randu) terhadap kualitas sperma ikan patin siam Pangasianodon hypopthalmus dengan rasio pengenceran berbeda setelah masa penyimpanan, yang meliputi viabilitas sperma, tingkat fertilisasi, dan derajat penetasan. Hasil penelitian menunjukkan bahwa perlakuan terbaik didapatkan pada pemberian madu kelengkeng dengan rasio pengenceran 1:50 dengan viabilitas sperma 96,33±0,58%, tingkat fertilisasi 97,10±0,70%, dan derajat penetasan 93,44±2,39%. Kata kunci: ikan patin, Pangasianodon hypopthalmus, madu, rasio pengenceran sperma

Abstract

ABSTRACT This study aimed to evaluate the effects of cinnamon Cinnamomum burmannii leaves extract addition with different doses in diet for the lipid muscle content of catfish Pangsianodon hypophthalmus. The cinnamon leaves extract was mixed in to the diet with five doses i.e: 0 (control); 0.5; 1; 2; and 4 g/kg diet. Catfish (7.43±0.01 g) were reared in 15 aquariums (160 L volume) with density of 30 fishes in each aquarium for 60 days. Fish were fed until apparent satiation three times daily at 08.00, 12.00, 16.00 WIB. The addition of cinnamon leaves extract at 1 g/kg of diet showed the optimal dose because it could lower flesh fat content, cholesterol, and triglycerides of catfish. Keywords: Pangasianodon hypopthalmus, Cinnamomum burmannii, fat, flesh  ABSTRAK Penelitian ini bertujuan untuk mengevaluasi pengaruh penambahan ekstrak daun kayu manis Cinnamomum burmannii dengan dosis berbeda pada pakan terhadap kandungan lemak daging ikan patin Pangsianodon hypopthalmus. Ikan diberi pakan secara at satiation sebanyak tiga kali sehari pada pukul 08.00, 12.00, dan 16.00 WIB. Hasil penelitian menunjukkan bahwa penambahan ekstrak daun kayu manis sebanyak 1 g/kg pakan memberikan hasil yang optimal karena dapat menurunkan kandungan lemak daging, kolesterol, dan trigliserida ikan patin. Kata kunci: Pangasianodon hypopthalmus, Cinnamomum burmannii, lemak, daging

Abstract

  Tilapia fish farming using monosex male population has been reported to have 10% higher productivity compared to mix-sex culture. This study aimed to determine immersion dose of 17α-metiltestosterone (MT) that allowed higher male percentage, growth, survival, and measure testosterone level in fish body. The experiment was designed as three immersion doses, namely: 0; 1.8; and 5.4 mg/L MT, each with three replications. Immersion was conducted to 14-day-old larvae for four hours, with density of 100 fish/L of water. Testosterone levels in fish was measured using ELISA method, and sex identification was performed by histology method. The result showed that percentage of male fish was the same in the treatment of 1.8 mg/L and 5.4 mg/L, and 53–65% higher than the control without MT treatment (54% male). Growth and survival of fish until day 56 post immersion were the same. By ELISA analysis, the levels of testosterone in larvae just after immersion was similar in 1.8 mg/L and 5.4 mg/L treatments, decreased drastically on day 14 after immersion, and then the levels of testosterone to be similar with the control on day 28, i.e. about 2 ng/g. By PCR method with specific primer, sex reversed and normal males could be distinguished, and on day 71 the testosterone levels in sex reversed and normal males fish was also the same, 0.3 ng/g (P>0.05). Thus, sex reversal by immersion at a dose of 1.8 mg/L can be consider as a standard protocol for monosex tilapia production. Testosterone level in the body of MT-treated fish becomes the same to the control of less than one month post immersion

Abstract

ABSTRACT  Immersion of undifferentiated larval tilapia in high temperature and 17α-methyltestosterone (MT) cab increase the male ratio. However, the effectiveness of immersion in high temperature of water containing MT remains to be evaluated. The purposes of this study were: 1) evaluate the male ratio, growth, and survival of tilapia, and 2) analyze the aromatase brain-type gene expression level in tilapia after immersing in high temperature (36 °C) containing MT at 2 mg/L for four hour with single and double immersion. Aromatase gene expression was analyzed by semi-quantitative RT-PCR (sqRT-PCR) method. The result showed that higher monosex male ratio was obtained by single immersion of MT at 36 °C at room temperature. Gene expression level of aromatase brain-type was lower on single immersion and increased significantly at second immersion compared to control (immersion at room temperature without MT). Immersion using MT and high temperature had no significant effect on fish survival. However the specific growth rate and fish biomass were higher than control. Thus, monosex male tilapia can be produced by single immersion of undifferentiated larvae at 36 °C temperature containing MT. Keywords: male ratio, aromatase, Oreochromis niloticus, temperature, 17α-methyltestosterone  ABSTRAK  Perendaman larva ikan nila yang belum terdeferensiasi kelaminnya dengan suhu tinggi dan hormon 17α-metiltestosteron (MT) dapat meningkatkan nisbah kelamin jantan. Tetapi, efektivitas perendaman menggunakan MT pada suhu tinggi belum diteliti. Tujuan dari penelitian ini adalah 1) mengevaluasi nisbah kelamin jantan, pertumbuhan, dan kelangsungan hidup ikan nila, dan 2) menganalisis ekspresi gen aromatase tipe-otak pada ikan direndam menggunakan MT dengan dosis 2 mg/L selama empat jam sebanyak satu dan dua kali perendaman pada suhu 36 °C. Ekspresi gen aromatase dianalisis menggunakan metode RT-PCR semi-kuantitatif (sqRT-PCR). Hasil penelitian menunjukkan bahwa kombinasi perendaman MT satu kali pada suhu 36 °C lebih tinggi menghasilkan ikan nila jantan monoseks dibandingkan perendaman MT satu kali pada suhu ruang. Tingkat ekspresi gen aromatase tipe otak pada perendaman satu kali lebih rendah, dan meningkat secara signifikan pada perendaman kedua dibandingkan dengan kontrol (perendaman pada suhu ruang tanpa MT). Perendaman larva menggunakan MT dan suhu 36 °C tidak berpengaruh nyata terhadap kelangsungan hidup, tetapi laju pertumbuhan spesifik dan biomassa ikan perlakuan tersebut lebih tinggi daripada kontrol. Dengan demikian, ikan nila jantan monoseks dapat diproduksi dengan perendaman satu kali pada larva yang belum terdeferensiasi jenis kelaminnya menggunakan MT pada suhu 36 °C. Kata kunci: rasio jantan, aromatase, Oreochromis niloticus, suhu, 17α-metiltestosteron
