Abstract

Dental caries is a chronic disease exclusive to humans and remains one of the most prevalent oral health problems worldwide. The increasing emergence of antibiotic resistance and the adverse effects associated with conventional antimicrobials have encouraged the exploration of natural plant-derived compounds for caries prevention. Murraya koenigii leaves are widely used as a culinary ingredient and are known to possess various therapeutic properties, including antioxidant, antibacterial, anti-inflammatory, and hepatoprotective activities. This study aimed to evaluate the total flavonoid content, antibacterial activity, and in silico toxicity profile of the ethanolic extract of M. koenigii leaves against Streptococcus mutans, a primary cariogenic pathogen. Total flavonoid content was determined using the aluminum chloride colorimetric method, while antibacterial activity was assessed using the agar disk diffusion assay. The toxicity profile of major flavonoid constituents was further analyzed using in silico ADMET prediction tools. The results showed that the ethanolic extract of M. koenigii leaves contained a high total flavonoid content (281.83 ± 10.23 mg QE/g extract) and exhibited strong antibacterial activity against Streptococcus mutans, producing inhibition zones ranging from 14.4 to 19.4 mm in a concentration-dependent manner. In silico analysis indicated that the major flavonoid constituents possessed favorable pharmacokinetic characteristics with low predicted toxicity risks. These findings suggest that the ethanolic extract of M. koenigii leaves is rich in flavonoids and demonstrates significant antibacterial potential, supporting its promise as a natural antibacterial agent for oral health applications. Further studies focusing on compound isolation, formulation development, and in vivo validation are recommended.

Abstract

Skin diseases are the most common type of skin disease in tropical countries like Indonesia. One of the main causes is bacterial infection. Sintrong leaves (Crassocephalum crepidioides (Benth.) S. Moore) and scallion (Allium fistulosum L.) contain secondary metabolites with antibacterial potential. The objective of the research was to determine the antibacterial activity of sintrong leaf extracts (Crassocephalum crepidioides (Benth.) S. Moore) and scallion (Allium fistulosum L.) formulated in gel form. This research was experimental in nature with the preparation of gel preparations using variations in the ratio of sintrong leaf extract (Crassocephalum crepidioides (Benth.) S.Moore) and spring onion (Allium fistulosum L.) (2:3, 2:2, 3:2). Evaluation parameters included organoleptic tests, homogeneity, pH, viscosity, spreadability, and adhesiveness. Antibacterial activity was tested by the disc diffusion method on Staphylococcus aureus and Staphylococcus epidermidis using clindamycin as a positive control and gel base as a negative control. The results showed that all formulas met the physical criteria of gel preparations with a pH of 5.1–5.8, viscosity of 23726–38580 cPs, spreadability in the range of 5–7 cm, and adhesiveness of >4 seconds. Antibacterial activity showed a zone of inhibition in all formulas, with the largest diameter in formula F3 (3:2), with an average inhibition diameter of 18.23 ± 0.67 mm on Staphylococcus aureus and 17.51± 0.30 mm on Staphylococcus epidermidis, which is categorized as strong.

Abstract

Background: Bacterial resistance to antibiotics has become a global concern, prompting the exploration of alternative antibacterial agents derived from natural sources. Enterococcus faecalis is a pathogenic bacterium frequently associated with root canal treatment failure due to its high resistance level. Seminyak leaves (Champereia manillana (Blume) Merr.) are known to contain secondary metabolites with potential antibacterial activity; however, studies based on solvent fractionation remain limited. Objective: This study aimed to evaluate the antibacterial activity of the ethanol extract, n-hexane fraction, and ethyl acetate fraction of seminyak leaves against Enterococcus faecalis. Methods: An experimental study was conducted through plant identification, preparation of simplicia, extraction using the maceration method with 96% ethanol, and liquid–liquid fractionation using n-hexane and ethyl acetate. Phytochemical screening was performed qualitatively and confirmed by thin-layer chromatography (TLC). Antibacterial activity was assessed using the disc diffusion method at concentrations of 15%, 30%, and 45% with three replications. Dimethyl sulfoxide (DMSO) 10% was used as the negative control, while chlorhexidine (standard concentration) served as the positive control. Data were analyzed using One-Way ANOVA followed by Tukey HSD post hoc test. Results: Phytochemical screening revealed that the ethanol extract contained alkaloids, flavonoids, tannins, saponins, and steroids. The n-hexane fraction predominantly contained nonpolar compounds (alkaloids and steroids), while the ethyl acetate fraction contained more diverse semi-polar compounds. Antibacterial testing showed that all samples exhibited activity against Enterococcus faecalis. The highest inhibition zone diameters at 45% concentration were 14.1 ± 0.21 mm (ethanol extract), 10.3 ± 0.17 mm (ethyl acetate fraction), and 8.5 ± 0.14 mm (n-hexane fraction). Statistical analysis indicated significant differences among treatments (p < 0.05). Conclusion: The ethanol extract, ethyl acetate fraction, and n-hexane fraction of seminyak leaves exhibit antibacterial activity against Enterococcus faecalis. The ethanol extract showed the highest activity, followed by the ethyl acetate and n-hexane fractions. These findings suggest that antibacterial compounds are more dominant in polar to semi-polar fractions.

Abstract

Krim kosmetik pencerah mengandung bahan berbahaya yang dapat membahayakan kulit. Tujuan penelitian ini untuk pemeriksan kandungan merkuri dan hidrokuinon pada krim kosmetik yang dijual di kota Medan. Hasil pemeriksaan menunjukkan bahwa tidak ada merkuri dalam semua sampelyang diuji, tetapi hidrokuinon ditemukan dalam salah satu sampel dalam jumlah yang melebihi batas yang diizinkan. Menurut pemeriksaan ini, masih ada krim kosmetik yang tidak memenuhi persyaratan keamanan. Untuk melindungi konsumen, pengawasan perederan kosmetik harus terus ditingkatkan.

Abstract

Background: Pagagan is a plant that has antioxidant properties. The benefits of antioxidants are essential for everyone because they can prevent cell damage due to the bad influence of free radicals. Formulating gotu kola in the form of candy is one effort so that the antioxidant effects of gotu kola can always be obtained easily because candy is a snack everyone likes and can be obtained easily anywhere. Objective: Formulate, physical characterization and antioxidant activity of hard candy immuno candies. Method: Hard candy formulation with varying concentrations of gotu kola herbal essence (0%, 1%, 5%, 10%, and 15%), physical characterization using organoleptic tests, water content, weight uniformity and stability, and antioxidant activity using the DPPH method. Results: Hard candy made with concentrations of 0%, 1%, 5%, 10%, and 15% has a sweet and slightly bitter taste (typical of gotu kola) at a concentration of 5%-15%, has a typical weak sugar aroma, and highwater content. respectively 0.72%, 0.77%, 0.84%, 0.8%, 0.89%, 0.95%, and is stable at a storage temperature of 8ºC-15ºC with closed packaging, has good weight uniformity, and IC50 values ​​were 32552.69 µg/mL, 12992.589 µg/mL, 12077.09 µg/mL 9669.15 µg/mL, 9484.519 µg/mL Conclusion: Gotu kola herbal juice can be formulated into hard candy up to a concentration of 15 %, has good physical characteristics as well as inactive antioxidant activity.

Abstract

The complexity of treatment with the disease in patients with chronic kidney disease (CKD) can increase the incidence of Drug Related Problems (DRPs). Analysis of DRPs is very useful to reduce the duration of illness, death, and treatment costs. The condition of patients who require hemodialysis therapy can cause changes such as physical, psychological, lifestyle and social that have an impact on the quality of life. This study aims to determine correlation between DRPs and the quality of life of CKD patients undergoing hemodialysis at Dr. Pirngadi Hospital, Medan, Indonesia. This study uses an observational method with a retrospective and prospective cross sectional research design on 72 patients who met the inclusion criteria in February-March 2024. DRPs were analyzed applying Cipolle and quality of life was obtained using the EQ-5D-5L questionnaire. The data was analyzed using the Spearman Rho test in the SPSS program. The results showed that there were DRPs in 64 patients with the categories of needing additional drugs (3.38%), unnecessary therapy (15.54%), drug interactions (35.13%) and ineffective drugs (45.95%). Quality of life with categories of good (36.1%), adequate (41.7%), and poor (22.2%). The patient's average quality of life score was 0.657. There was a correlation between the number of DRPs and the patient's quality of life (r=-0.802) with a significance value of p=0.032 (p<0.05), which shows that the higher the incidence rate of DRPs, the lower the patient's quality of life. Based on the results of the study, it was concluded that there was a significant relationship between the number of DRPs events and the quality-of-life value of CKD patients undergoing hemodialysis.

Abstract

Xerosis is a skin condition characterized by decreased water content in the stratum corneum, leading to dryness, scaling, roughness, and pruritus, which may cause discomfort and increase the risk of infection. Purified used cooking oil has potential as an alternative emollient that is economical and environmentally friendly. This study aimed to evaluate the quality characteristics of purified used cooking oil, formulate a skin balm preparation, and assess its effectiveness in treating heel xerosis. This study employed a quantitative experimental method, including purification of used cooking oil, quality characterization based on national standards, toxicity testing using the Brine Shrimp Lethality Test(BSLT), formulation of skin balm with concentrations of 0%, 9%, 18%, and 27%, as well as physical stability and effectiveness evaluation using a skin analyzer. The results showed that the purified used cooking oil met quality standards. The skin balm preparation demonstrated good stability in terms of organoleptic properties, homogeneity, pH, spreadability, and adhesiveness during storage and cycling tests. Effectiveness testing indicated an increase in skin moisture corresponding to higher oil concentrations, with the highest improvement observed at 27% concentration (73.9% water content and 69.2% oil content). In conclusion, the skin balm formulation based on purified used cooking oil is effective in improving skin moisture and treating heel xerosis.

Abstract

Acne is a common skin issue among teenagers, primarily caused by bacterial infections from Propionibacterium acnes and Staphylococcus epidermidis. Pineapple peel has been found to contain antibacterial properties that can help treat acne. This study aims to evaluate the antibacterial effectiveness of an anti-acne patch made from pineapple peel extract (Ananas comosus (L.) Merr.) against these two bacteria. The pineapple peel extract was obtained through a reflux extraction method, followed by phytochemical screening, which identified the presence of flavonoids, saponins, tannins, and triterpenoids. The extract was then formulated into patches with concentrations of 5%, 10%, and 15% and tested for antibacterial activity. Physical evaluations showed that the patches had an appropriate pH (4.5-5.5), uniform weight (CV < 5%), acceptable thickness (0.1-0.3 mm), moisture content below 10%, and did not cause irritation in volunteers. Antibacterial testing using the agar diffusion method showed the highest inhibition zones for P. acnes at 21.1 mm and S. epidermidis at 18.2 mm at a 15% concentration. Meanwhile, the well-diffusion method showed inhibition zones ranging from 12 to 21.8 mm for P. acnes and 12.7 to 24.7 mm for S. epidermidis across various concentrations. These results suggest that pineapple peel extract and its patch formulation have strong antibacterial activity, indicating potential as an acne treatment. Abstract

The use of conventional cigarettes and e-cigarettes (vapes) among adolescents is a public health issue that requires serious attention. Poor understanding of the dangers of nicotine and the perception that vapes are safer than cigarettes are factors driving the increased use of nicotine products among students. This community service activity aims to increase the knowledge and awareness of students at SMKS Al Washliyah 3 Medan regarding the dangers of cigarettes and vapes, their nicotine content, and their impact on health through scientific evidence-based education. The activity was implemented through a participatory educational approach, including health education, presentations, interactive discussions, and refusal skills training for students who are tempted to use cigarettes and vapes. The target group of the activity was adolescent students at SMKS Al Washliyah 3 Medan. Educational materials covered the differences between cigarettes and vapes, the harmful substances contained in nicotine products, the impact of nicotine on adolescent brain development, the risks of short-term and long-term health problems, and strategies for developing a healthy lifestyle without nicotine. The results of the activity showed an increase in students' understanding of the risks of cigarette and vape use, particularly regarding nicotine content and the potential for dependence. Through discussion sessions, students also began to understand that vaping is not a safe product, but rather carries health risks and can be a gateway to conventional smoking. This activity also encouraged students to develop positive attitudes toward rejecting the use of nicotine products and supporting the creation of a healthy, smoke-free school environment. This community service activity contributed to improving adolescent health literacy through a promotive and preventative approach based on evidence-based education. Continuous education about the dangers of smoking and vaping in the school environment is expected to foster a more health-conscious, productive, and nicotine-free young generation.

Abstract

Latar belakang: Setiap penyakit ada obatnya, tapi ada kalanya penggunaan obat yang tidak tepat justru menyebabkan bakteri penyakit tersebut menjadi kebal terhadap antibiotik sehingga menjadi sulit untuk diobati. Antibiotik merupakan obat untuk mengobati infeksi yang disebabkan oleh bakteri. Pemberian antibiotik pada penderita penyakit infeksi bertujuan untuk menghambat pertumbuhan atau membunuh mikroorganisme, terutama bakteri penyebab penyakit. Penggunaan antibiotik akan memberikan keberhasilan terapi jika digunakan secara rasional. Tujuan: untuk mengetahui gambaran tingkat pengetahuan dan sikap serta pengaruh edukasi terhadap pengetahuan dan sikap penggunaan antibiotik pada siswa SMAN di Kota Medan Sumatera Utara. Metode: menggunakan desain Quasi-experimental dengan jenis rangcangan one grup pretest-posttest. Populasi dalam penelitian ini adalah 6 SMAN di Kota Medan Sumatera Utara dengan sampel sebanyak 460 siswa. Alat ukur yang digunakan dalam penelitian ini adalah kuesioner pretest dan kuesioner posttest. Data diolah dan dianalisa mulai dari analisis univariat dan analisis paired T-Test menggunakan aplikasi komputer SPSS versi IBM SPSS Statistics 18.0 Hasil: Berdasarkan analisis data diperoleh adanya pengaruh edukasi terhadap tingkat pengetahuan dan sikap penggunaan antibiotik dengan hasil analisa data diperoleh nilai signifikan (2-tailed) yaitu 0,000 (p<0,05). Kesimpulanhipotesis alternatif/hipotesis kerja (Ha) diterima dan hipotesis nol (Ho) ditolak. Berdasarkan hasil data yang diperoleh terdapat adanya pengaruh edukasi terhadap tingkat pengetahuan dan sikap penggunaan antibiotik pada siswa SMAN di Kota Medan Sumatera Utara.
