Abstract

The Double–Spotted Gecko (Gekko monarchus) is one of the reptiles on the list of wild animals given an annual utilization quota for their capture in the wild. Bengkulu Province, as one of the distribution place of G. monarchus does not yet have data on its population. The research was conducted on 13 November to 2 December 2021, using two methods: interviews with the community and a Visual Encountered Survey at night. The research area in the five districts is 61.49 ha, the number of houses visited is 433, and the number of people interviewed is 386. The number of geckos counted in the interviews was 315, while from direct observation, there were 275. Geckos were most found at house and dominated (41.2%) by hiding between wooden walls. Based on the results of extrapolation calculations, the population estimates for the Double–Spotted Gecko are North Bengkulu 43,720–54,427 individuals; Mukomuko 32,997–51,810 tails; Central Bengkulu 6,248–9,187 individuals; Total 39,883–72,720 individuals; and South Bengkulu 11,469–11,592 individuals.

Abstract

Ikan lele (Clarias batrachus L.) merupakan salah satu spesies penting sebagai komoditas air tawar di Indonesia dengan produksi yang terus meningkat setiap tahunnya. Namun, ikan lele rentan terhadap beberapa infeksi bakteri, terutama bila dipelihara dalam kondisi kepadatan tinggi, salah satunya oleh bakteri patogen Bacillus cereus. Banyak sumber bahan baku lokal yang dapat dimanfaatkan sebagai pakan ikan untuk mengatasi masalah tersebut dan mengatasi penyakit yang muncul pada budidaya ikan, salah satunya adalah tanaman tradisional ketapang (Terminalia catappa L.) yang merupakan tanaman yang memiliki potensi sebagai zat antibakteri. Tujuan penelitian ini untuk mengetahui dan menguji aktivitas antibakteri ekstrak daun ketapang (T. catappa L.) terhadap bakteri patogen B. cereus dari isolat hati ikan lele (Clarias batrachus L.) penyebab penyakit pada budidaya. Metode yang digunakan yaitu maserasi dengan pelarut metanol perbandingan 1:10 yang dimaserasi 7 x 24 jam. Uji aktivitas antibakteri dengan metode cakram difusi dengan konsentrasi 45%, 52,5%, 60%, 67,5%, dan 75%. Hasil uji aktivitas antibakteri ekstrak daun ketapang mampu menghambat pertumbuhan bakteri B. cereus pada konsentrasi 45% dengan rata-rata diameter zona hambat sebesar 13,51 mm (kategori kuat). Dari hasil yang diperoleh, dapat disimpulkan bahwa ekstrak daun ketapang memiliki kemampuan dalam menghambat pertumbuhan bakteri patogen B. cereus  penyebab penyakit pada hati ikan lele (C. batrachus L.)

Abstract

Sungkai (Peronema canescens Jack) was used traditionally as a medicine to help relieve fever, malaria and flu. In general, people was used sungkai leaves in the form as infusion. This study was aimed to screening of secondary metabolite of Sungkai leaf infusion using the color reaction method. The sample in this study was sungkai leaf that obtained from Bengkulu city. The infusion was obtained by boiling 10 gram of sungkai leaf simplicia with 100 ml of distilled water for 15 minutes at 90 0C until a concentration of 10% (w/v) was obtained. Secondary metabolite screening was carried out using the color reaction method to identify alkaloids, flavonoids, saponins, tannins and steroids. Phytochemical screening using specific reagents with the test was repeated three times. The results were showed that sungkai leaves infusion was positive in alkaloid test as evidenced by the presence of white precipitate with Mayer reagent, brown precipitate with Wagner reagent, and orange precipitate with Dragendorff reagent. Positive results were also shown in flavonoid, saponin and tannin tests. While, negative result was seen in steroid test using Liebermann Burchard reagent. In this study it can be concluded that sungkai leaf infusion positively contains alkaloids, flavonoids, saponins and tannins.

Abstract

Abstract: The mangrove plants from Enggano Island are found in a well-preserved ecosystem. Mangroves contain secondary metabolite compounds that result from the interaction between plants and endophytic bacteria. This study aims to obtain endophytic bacterial isolates from the mangrove plant Sonneratia alba Sm. from Enggano Island and to assess bacterial diversity. Endophytic bacteria were isolated using grinding and then diluted with serial dilutions of 10-1, 10-3, and 10-5. The growing bacteria were purified using the quadrant streak method. They were then identified by observing colony morphology, Gram staining, and biochemical tests including catalase, motility, citrate, urea, and sugar fermentation tests. Bacterial identification was carried out using Bergey's Manual of Determinative Bacteriology. A total of 22 endophytic bacterial isolates were collected from the mangrove plant Sonneratia alba from Enggano Island, consisting of 4 genera: Bacillus, Marinococcus, Micrococcus, and Pseudomonas. There were 17 isolates closely related to the genus of Bacillus, 1 isolate closely related to the Marinococcus, 2 isolates closely related to the Micrococcus, and 2 isolates closely related to the Pseudomonas.Abstrak: Tumbuhan mangrove asal Pulau Enggano berada pada ekosistem yang masih terjaga. Mangrove memiliki kandungan senyawa metabolit sekunder yang merupakan hasil interaksi antara tumbuhan dan bakteri endofit. Penelitian ini bertujuan untuk memperoleh isolat bakteri endofit tumbuhan mangrove Sonneratia alba Sm. asal Pulau Enggano dan mendapatkan keanekaragaman bakteri. Isolasi bakteri endofit dilakukan dengan metode gerus, kemudian diencerkan dengan pengenceran berseri dari 10-1, 10-3, dan 10-5. Bakteri yang tumbuh dimurnikan dengan metode gores kuadran. Kemudian diidentifikasi dengan mengamati morfologi koloni, pewarnaan Gram, dan uji biokimia meliputi uji katalase, uji motilitas, uji sitrat, uji urea, uji gula-gula. Kemudian dilakukan identifikasi bakteri dengan buku Bergey’s Manual of Determinative Bacteriology. Total isolat bakteri endofit yang berhasil dikoleksi dari tumbuhan mangrove Sonneratia alba asal Pulau Enggano adalah sebanyak 22 isolat dengan keanekaragaman bakteri endofit yang terdiri dari 4 genus yaitu Bacillus, Marinococcus, Micrococcus, dan Pseudomonas. Isolat yang memiliki kedekatan dengan genus Bacilus adalah sebanyak 17, yang memiliki kedekatan genus Marinococcus hanya 1 isolat, yang memiliki kedekatan dengan genus Micrococcus adalah 2 isolat, dan 2 isolat memiliki kedekatan dengan genus Pseudomonas.

Abstract

Staphylococcus aureus is a pathogenic bacterium commonly associated with various infections. An initial step in preventing and addressing infections is the use of antibacterial soaps. The purslane extract can serve as an antibacterial agent that inhibits the growth of Staphylococcus aureus, while VCO offers additional protection through its antimicrobial properties and ability to maintain skin moisture. This study aimed to formulate a solid soap using 96% ethanol extract of purslane combined with VCO. Purslane powder was extracted using a maceration method. The physical properties of the soap were evaluated, including organoleptic evaluation, PH, homogeneity, foaming capacity, foam stability, and antibacterial activity against Staphylococcus aureus. Based on the results of the physical property evaluations, the solid soap met the required standards. The antibacterial effectiveness test showed inhibition zones for F1 at 3.21 ± 0,023 (weak category), F2 at 4,26 ± 0,102 (weak category), and F3 at 5,2 ± 0,070 (weak category). The results indicate that the purslane extract, in combination with varying amounts of VCO, can be formulated into solid soap that meets the physical property evaluation criteria and possesses antibacterial activity against the growth of Staphylococcus aureus.  Keywords: Solid Soap; Antibacterial; Purslane Plant; VCO; Staphylococcus aureus

Abstract

Excessive exposure to sunlight can adversely affect the skin and cause various kinds of skin problems. Agarwood leaf extract (Aquilaria malaccensis Lam) contains flavonoid compounds that can protect the skin from the harmful effects of UV rays. This study aims to formulate a cream from the ethanol extract of agarwood leaves (Aquilaria malaccensis Lam) which can be used as a sunscreen cream. In this study, a cream formula was prepared from ethanol extract of agarwood leaves with 3 variations of different concentrations of the extract, namely F1 2% extract of agarwood leaves, F2 4% extract of agarwood leaves and F3 8% extract of agarwood leaves. Homogeneity tests, pH tests, spreadability tests, adhesion tests, and SPF tests. The results showed that F2 with an extract concentration of 4% was the best formula with a pH of 7.11; spreadability of 6.0 cm; adhesive power of 4.9 seconds and SPF value of 39.22 which is the maximum category as a sunscreen.

Abstract

Patikan Kebo (Euphorbia hirta L.) is a plant that lives scattered in the yard, has potential as an antibacterial, one way to extract plants is by isolating endophytic bacteria. Endophytes are bacteria found in plant tissue. The purpose of this study was to isolate and identify the morphology of endophytic bacteria from  plant Patikan Kebo (Euphorbia hirta L.). The method used is the Total Plate Count (TPC) method by taking samples, isolating bacteria, pure culture, morphological identification, and Gram staining. Endophytic bacteria were obtained by isolating parts of the plant Patikan Kebo (E. hirta L.), namely the roots of patikan kebo (PRE), Patikan kebo stem endophyte (PSE) and Patikan kebo leaves endophyte (PLE). The results of the study found 22 isolates, endophytic bacterial isolates have diverse morphological characteristics including appearance, elevation, edges and color. There are 20 isolates of endophytic bacteria that belong to the Gram positive group and 2 isolates of endophytic bacteria that belong to the Gram negative group. It can be concluded that endophytic bacterial isolates from Patikan Kebo (Euphorbia hirta L.) plants have different morphologies

Abstract

Beuveria is a genus of entomopathogenic fungi that plays an important role in controlling agricultural pests. Pest control with entomopathogenic fungi is an alternative to reduce influence the use of chemical insecticides. The diversity of entomopathogenic fungi intraspecies is by the genetic diversity of isolates, a high diversity of isolates can result diversity of fungus virulence. The purpose of this study was to perform the characterization molecular of entomopathogenic fungi Beauveria spp. Bengkulu isolate. Polymerase chain reaction (PCR) was performed for molecular identification Beauveria isolates were obtained from infected larvae in the field (1. Isolate from Coleoptera in Kepahiang, Bengkulu (BBC), 2. Isolate from Hemiptera: Pentatomidae in Rejang Lebong, Bengkulu (BBL), and 3. Isolate from Hemiptera: Alydidae in Taba Mulan, Merigi District, Kepahiang, Bengkulu (MT). The results of the amplification of the entomopathogen Beauveria samples produce DNA bands around ±580 bp. The DNA sequence analysis of ITS 5F and ITS 4R primers showed that the isolates BBC, MT and BBL have fairly high similarity with the Beauveria bassiana isolate 1397, Beauveria bassiana isolate SBI TNSPI, Beauveria bassiana strain B-Bug, Beauveria bassiana voucher TSJBB, Beauveria bassiana isolate SASRI C2, Beauveria bassiana isolate IMI 382764, Beauveria bassiana BCRC:FU31669, Beauveria bassiana isolate B098, Beauveria bassiana isolate INRS-CFL, Beauveria sp. JS-2009a isolate B4B, Beauveria bassiana isolate KACC with the homology of 99.83%, 99.83%, and 98.61%.

Abstract

[TUNGRO DISEASE ON RICE PLANT (Oryza sativa) IN TABA PENANJUNG DISTRICT: DISEASE INCIDENCE AND MOLECULAR DETECTION OF VIRUSES]. Tungro is one of the important diseases of rice plants. Tungro disease caused by Rice tungro bacilliform virus (RTBV) and Rice tungro spherical virus (RTSV). This research aims to obtain information on the spread of the disease, vector population, and molecular detection by polymerase chain reaction method. This research was conducted from December 2019 to April 2020 in rice fields in Taba Penanjung sub-district, Bengkulu Tengah district at ± 540 meters above sea level and virus detection in the Laboratory of Biotechnology and Genetics Department of Biology Faculty of Mathematics and Natural Sciences (MIPA) and Laboratory of Plant Protection, Agriculture Faculty, Bengkulu University. Data analysis used descriptive analysis. The results showed that the highest disease incidence on the seven weeks after planting (wap) is 1.7% and the highest population of the vector on the 7 wap with an average of 7.7 vector/100m2 in the village of Taba Penanjung. Detection of viruses with the PCR method with the specific primer DAF/DAR succeeded in amplification of DNA band measuring 1,400 bp. RTSV was not detected by RT- PCR using a specific primer or RTSV. The result of this detection shows that the symptoms of tungro on rice plants in the cultivation of Taba Penanjung district associated with RTBV infection. RTSV unable to detect can be caused by many factors in the detection process, including total RNA extraction, cDNA synthesis, DNA amplification, and DNA visualization of PCR products.

Abstract

This study aims to identify types of macrofungi that are resistant to the presence of Hg in their environment. The method used is survey research by exploring mushrooms in the area around the Small Scale Gold Mining (ASGM). Tambang Sawah Village, Lebong Regency, Bengkulu Data findings were analyzed by descriptive qualitative. The results showed that the types of macrofungi that were able to survive Hg stress were Ganoderma applanatum, Dacryopinax spathularia, Daldinia concentrica, Ganoderma lucidum and Polyporus alveolaris. In conclusion, five types of macrofungi that are adaptive to mercury (Hg) were found. The highest Hg accumulation was found in D. concentrica.
