Abstract

Catfish farming has been increasingly threatened by outbreaks of diseases caused by bacteria which are major problems for Indonesia’s catfish industry. This can lead to a decrease in the food quality of freshwater fishery products, especially catfish. In catfish farming, the occurrence of diseases can cause severe financial losses. This study aimed to identify bacteria that infected catfish and discover new potential antibacterial agents from Bacillus spp. isolated from a marine sponge to inhibit several bacteria from the infected catfish. Catfish samples were obtained from catfish farming with clinical observation of ulcerative lesions on the outside of the body. Several bacteria were isolated from the catfish and used sponge-associated bacterium to inhibit them in which using two methods namely spot assay and disc diffusion assay. Bacterial supernatant and pellet were used to performed antibacterial assay using disc diffusion method. These bacterial isolates were identified through their colony morphology, Gram staining, biochemical assays, and 16S rRNA gene sequence. Bacterial identification based on the 16S rRNA gene sequence showed that GL1 was 99.92% closely related to Aeromonas jandaei, HL1 was 100% closely to Bacillus amyloliquefaciens, and GL2 and HL2 was closely related to Bacillus cereus. The results of antibacterial assay indicated that sponge-associated Bacillus spp. successfully inhibited some bacteria that associated with infected catfish. In addition, Bacillus velezensis APD10 exhibited the most potential strain with antibacterial property which could inhibit all bacteria from the diseased catfish.

Abstract

Xylanase is an important hydrolytic enzymes with many application in several industries, but to obtain enzyme derived products is not easy. Thus, the optimization of efficient xylanases production is a great interest for biotechnological application. This study aims to determine the type of substrate, medium composition, and optimum conditions of xylanase production by S. costaricanus 45I-3. Determination of substrate type was done by growing the tested bacteria on birchwood xylan, beechwood xylan, oat spelled xylan, corn cobs xylan, and tobacco xylan substrate, meanwhile the determination of medium composition and enzyme production were done by measuring xylanase activity at various substrate concentration and replacing the carbon, nitrogen, phosphate and surfactants source. The results showed that the highest enzymatic index (EI) produced from corn cob xylan substrate at 3.60 meanwhile the second highest was beechwood xylan substrate at 2.87 EI, however this substrate is purer, thus this substrate was selected and used as xylan sources for further optimization measurement. The best xylanase activity (2.29 U/mL) obtained on eighth day after inoculation on rotary incubator at 120 rpm in 28 ºC. Arabinose as the source of carbon generate the highest activity at 3.161 U/mL meanwhile the most preferred source of phosphate is Na2HPO4 (2.37 U/mL). Both source of nitrogen i.e. nitrogen ammonium sulphate (NH4)2SO4 and yeast extract were able to produce xylanase at 2.57 and 2.36 U/mL. The addition of surfactant in production medium showed addition of SDS surfactant (0.146 U/mL) and Tween 80 (0.438 U/mL) showed a negative response by decreasing the activity. The conclusion showed that the xylanase activity was increased after optimization at various C, N, and P sources, and the use of nitrogen source (NH4)2SO4), become a more economical alternative to replacing a nitrogen source yeast extract so it can lower the production costs of xylanase enzyme.

Abstract

The majority of thalassemia patients in Rejang Lebong Bengkulu had dietary issues. Due to their parents' insufficient understanding of thalassemia, some of them do not undergo routine blood transfusions. With the goal to improve parents' understanding of thalassemia and its problems, this community service project was carried out at the Indonesian Thalassemia Parents Association (POPTI) organization in the Rejang Lebong Regency of Bengkulu. It included measuring oxygen saturation and introducing nutritious foods for people with thalassemia. In order to explain thalassemia, the inheritance of thalassemia features, and the introduction of significant nutritious foods for persons with thalassemia, this activity was carried out utilizing the telling method. On 10 thalassemia patients with ages ranging from 2 to 20 years old, the oxygen saturation was assessed using the direct survey method and an oximeter. According to the oximeter, all individuals with thalassemia have normal oxygen saturation levels. Both parents and kids are aware of the value of blood transfusions and a healthy diet for thalassemia. This activity has a good impact on parents and kids to improve parents' knowledge about thalassemia and its issues, the causes of thalassemia and its heredity, children's oxygen saturation, and its meaning. Parents may reduce the risk of health issues in people with halassemia by understanding the reasons for food that needs to be taken and needs to be avoided. Sebagian besar anak penyandang talasemia yang ada di Rejang Lebong Bengkulu mengalami permasalahan gizi. Beberapa diantaranya tidak melakukan transfusi darah rutin akibat pemahaman orang tua yang masih rendah mengenai talasemia. Kegiatan pengabdian kepada masyarakat ini dilakukan pada komunitas Persatuan Orang Tua Penderita Thalassemia Indonesia (POPTI) cabang Bengkulu yang ada di wilayah Kabupaten Rejang Lebong Bengkulu dengan tujuan untuk meningkatkan pemahaman orang tua tentang penyakit talasemia dan permasalahannya melalui pemeriksaan saturasi oksigen dan pengenalan makanan bernutrisi bagi penyandang talasemia. Kegiatan ini dilaksanakan dengan metode ceramah untuk menyampaikan penjelasan tentang talasemia, pewarisan sifat talasemia, pengenalan makanan bernutrisi penting bagi penyandang talasemia. Pemeriksaan saturasi oksigen dilakukan dengan metode survei langsung menggunakan alat oximeter pada 10 penyandang talasemia dengan rentang usia 2-20 tahun. Oximeter menunjukkan bahwa, semua penyandang talasemia memiliki saturasi oksigen dalam kadar normal. Orang tua dan anak telah memahami pentingnya transfusi darah dan makanan bernutrisi yang tepat bagi talasemia. Kegiatan ini telah memberikan manfaat yang baik bagi orang tua dan anak dalam upaya peningkatan wawasan dan pemahaman orang tua tentang penyakit talasemia dan permasalahannya, penyebab munculnya talasemia dan pewarisannya, saturasi oksigen anak serta maknanya. Orang tua dapat memahami alasan suatu makanan perlu dikonsumsi dan perlu dihindari bagi anak penyandang talasemia sehingga dapat mengurangi kemungkinan permasalahan kesehatan yang muncul pada anak penyandang talasemia.

Abstract

This study aims to conduct a field study on the need to develop Augmented Reality (AR) assisted practicum guidelines. Data collection instruments were in the form of questionnaires and interview guidelines. The subjects in this field study were students who became practicum assistants, physics laboratory staff, and lecturers of basic physics courses. Questionnaire data and interview results were analyzed qualitatively to see the real conditions of the practicum. Based on the results of data analysis, it is known that practicum manuals are still dominant in text, and practicum activities can be directed to practice problem-solving skills. The phenomenon of coastal areas can also be associated with physics practicum material. Based on the results of the practicum assistant's questionnaire, it is known that AR can facilitate physics practicum, so it is necessary to develop an AR-assisted practicum guide that presents audio-visual presentations. Based on the results of the interviews, it is also known that the constraints on practicum activities during this pandemic were that not all practicum courses were available at PheT to carry out virtual practicums, counter-dependence with assistant explanations in carrying out practicum procedures, so strict procedures could not be carried out, so it could be interpreted that a solution was needed an alternative practicum guide that is equipped with a video tutorial about practicum procedures. AR technology can be used to visualize tutorial videos that can be accessed offline.

Abstract

The research aims to determine the number of individuals, age structure and sex ratio in the Simpai population or sub-population in the Bukit Daun Protected Forest. The research used the direct sencus method or direct counting of individual hoops (P. m. melalophos) in a sub-population in the field. Observations were carried out at 06.00-18.00 WIB and increased observations were carried out four times on different days. The research results show that there are three sub-populations of Simpai in the Bukit Daun Protected Forest Area. The number of individuals in the first sub-population is 10 individuals, the second sub-population is 13 individuals, and the third sub-population is 15 individuals. Based on the age structure, the number of young individuals, namely teenagers and children, including babies, in the third sub-population is greater (24 individuals) compared to adults (14 individuals). The sex ratio of adult male and female hoops, namely (1:3) in subpopulations I and II, is higher than subpopulation III (1:2), whereas in the juvenile group, the sex ratio in subpopulation III (1:4) is higher than subpopulation I and II (1:2). Abstract

Staphylococcus aureus dan Propianibacterium acnes adalah bakteri yang menjadi salah satu penyebab masalah jerawat pada kulit. Penelitian ini bertujuan untuk menentukan daya hambat dari ekstrak etanol 96% tali putri (Cassytha filiformis L.) terhadap S.aureus dan P. acnes. Uji aktivitas antibakteri yang dilakukan pada peneltian ini menggunakan metode difusi cakram. Ekstrak etanol 96% tanaman tali putri diuji pada konsentrasi 0,75%, 1,5%, 3%, 6%, 9%, 12%. Hasil uji aktivitas ekstrak etanol 96 % tanaman tali putri terhadap S. aureus pada konsentrasi ekstrak 0,75% , 1,5%, 3%, 6%, 9%, 12% masing-masing memiliki diameter hambat (clear zone) rata-rata  adalah 6,3 mm, 6 mm, 5 mm, 4,3 mm, 3,9 mm, dan 3,1 mm. Sedangkan hasil uji aktivitas ekstrak etanol 96 % tanaman tali putri terhadap P. acnes pada konsentrasi ekstrak 0,75% , 1,5%, 3%, 6%, 9%, 12% masing-masing memiliki diameter daya hambat (clear zone) rata-rata  adalah 1,1 mm, 8,2 mm, 2,6 mm, 2,95 mm, 2,87 mm, dan 5,72 mm. Aktivitas hambatan ekstrak etanol 96% tali putri terhadap bakteri S. aureus dan P. Acne terlihat pada konsentrasi 0.75% dengan diameter daya hambat masing-masingnya adalah 6,3 mm dan 1,1 mm.

Abstract

Environmental and genetic variables can exert an influence on alterations in morphological traits. Within fish species inhabiting diverse aquatic settings, there can be observed variations in morphological traits. Genetically, variations in fish morphological characteristics can occur through mating and gene flow. To date, there has been a lack of research conducted on the variability in morphological traits and genetic relationships between Lutjanus timoriensis, L. malabaricus, and L. erythropterus. Thus, the current research aimed to identify variations in the morphological characteristics as well as in the intra- and inter-specific relationships between three red snapper species from the genus Lutjanus.  Cytochrome oxidase I (COI) gene was used to study the molecular relationship among species of red snapper. The results showed that L. timoriensis had high intraspecific morphological variation in young individuals. Morphologically, L. timoriensis, L. malabaricus, and L. erythropterus are very similar. Unlike L. malabaricus and L. erythropterus, young and adult L. timoriensis have black patches in the pectoral fin axils. The adult L. erythropterus has a comparatively small mouth, no hump on its head, and no black saddle at the base of its tail. Meanwhile, L. malabaricus has a comparatively large mouth, a head with a hump, and a black saddle at the base of its tail. In terms of body size, L. erythropterus is larger than L. timoriensis and L. malabaricus. Based on NCBI and Bold System data, molecular analyses determined that the observed fish were L. timoriensis, L. malabaricus, L. erythropterus with a similarity of between 99.85 and 100%. The phylogenetic tree construction demonstrated that L. malabaricus, L. timoriensis and L. erythropterus were closely related.

Abstract

Background: Irrational use of antibiotics has various side effects to resistance. Utilization of secondary metabolites of Andaliman plant endophytic bacteria (Z. acanthopodium DC) is an alternative known to act as an antibacterial to overcome this problem.Objective: This study aimed to determine the antimicrobial activity of the crude extract of Andaliman endophytic bacteria against Escherichia coli and Staphylococcus aureus.  Methods: This research is an experimental laboratory with qualitative data collection methods. The initial stages will be the isolation of endophytic bacteria from Andaliman, the antagonist test of Andaliman endophyte isolates against E. coli and S. aureus, and the extraction of endophytic bacterial isolates using ethyl acetate as solvent. An antagonistic test of bacterial isolate extract was performed using the disc diffusion method against pathogenic bacteria E. coli and S. aureus and observed by an inhibition zone. The final stage is the minimum inhibition test of endophytic isolates by dilution method on three potential endophytic isolates.Results: The results of the extraction of isolates EAA22, EAA28, EAB5, EAB6, and EAB7 Andaliman endophytic bacteria have inhibitory activity against S.aureus, and the results of the extraction of isolates EAB5, EAB6, EAA22 have inhibitory activity against E. coli.Conclusion: Endophyte bacteria isolates from Andaliman produce antibacterial compounds against E. coli and S. aureus.

Abstract

The Rejang tribe is one of the indigenous communities in Bengkulu. They are dispersed throughout several areas of Bengkulu, however, the majority of them live in the Rejang Lebong district. The Rejang tribe is classified as a Malayan Mongoloid race. Generally, every tribe has distinctive characteristics. These characteristics include language, culture, religion, and physical characteristics. The purpose of this study was to analyze the characteristics of the Rejang tribal community residing in Pematang Sapang village, North Bengkulu, with a focus on their fingerprint and palm patterns. Data was gathered from 100 individuals of the Rejang tribe, who fulfilled the inclusion criteria. The data was analyzed quantitatively. The study determined that there are three fingerprint patterns in the Rejang community, with the loop pattern being most prevalent at 59.8%. There are only two palmar patterns; normal and simian, with the normal pattern being dominant at 94.5%. Statistical analysis revealed no significant correlation between fingerprint or palmar patterns and sex. Furthermore, there was no observable relationship between fingerprint and palmar patterns.  The Rejang tribe is characterized by the prevalence of the loop patterns for fingerprints and the normal patterns for palmar.

Abstract

Penyakit yang dapat disebabkan akibat penggunaan alat selam secara bersama yang tidak aseptik yaitu Infeksi Saluran Pernapasan Akut (ISPA). ISPA merupakan penyakit yang menginfeksi saluran pernapasan atas dan menginfeksi seluruh bagian pernapasan bawah (alveoli). ISPA dapat menular melalui droplet mengandung virus atau bakteri terhirup oleh orang sehat. Penggunaan alat selam secara langsung akan bersentuhan dengan bagian mulut dan hidung, apabila perawatan alat penyelaman kurang tepat akan menimbulkan adanya risiko penularan ISPA. Kelompok penyelam di Pulau Enggano menggunakan alat selam bersama untuk aktivitas mencari ikan dan menyewakan untuk para wisatawan. Namun para penyelam masih minim pengetahuan terkait dampak risiko penyakit dari penggunaan alat selam bersama. Kegiatan dihadiri oleh perwakilan 5 kelompok penyelam yang ada di Pulau Enggano. Kegiatan pengabdian meliputi pembukaan, sosialisasi, pelatihan mengenai cara membersihkan alat selam yang benar, tanya jawab, evaluasi kegiatan, dan penutupan. Hasil evaluasi dari kegiatan ini, kelompok penyelam menilai sangat bermanfaat dan membantu meningkatkan pengetahuan peserta. Para penyelam menjadi sadar akan pentingnya kebersihan dan perawatan alat selamnya. Secara umum, berdasarkan hasil evaluasi, menunjukkan bahwa masyarakat puas dengan pelaksanaan kegiatan ini dan mengucapkan terima kasih atas penyerahan alat dan bahan pembersih alat selam untuk kelompok penyelam. Pemahaman dan penerapan aseptik pada alat selam ini perlu dilakukan secara berkelanjutan agar menjadi nilai lebih untuk kelompok penyelam. Tidak hanya untuk kesehatan para penyelam yang mencari ikan namun juga untuk wisatawan yang hendak menggunakan alat selam. Diseases that can be caused by sharing diving equipment that is not aseptic are Acute Respiratory Infections (ARI). ARI is a disease that infects the upper respiratory tract and infects the entire lower respiratory tract (alveoli). ARI can be transmitted through droplets containing viruses or bacteria inhaled by healthy people. Using diving equipment will directly come into contact with the mouth and nose, if the diving equipment is not maintained properly it will pose a risk of transmitting ARI. Groups of divers on Enggano Island use diving equipment together for fishing activities and rent them out to tourists. However, divers still lack knowledge regarding the impact of disease risks from using shared diving equipment. The activity was attended by representatives of 5 diving groups on Enggano Island. Service activities include opening, socialization, training on how to properly clean diving equipment, questions and answers, activity evaluation, and closing. As a result of the evaluation of this activity, the diving group considered it very useful and helped increase the participants' knowledge. Divers become aware of the importance of cleanliness and maintenance of their diving equipment. In general, based on the evaluation results, it showed that the community was satisfied with the implementation of this activity and expressed their gratitude for the delivery of diving equipment and cleaning materials to the diving group. The understanding and application of aseptics in diving equipment needs to be carried out on an ongoing basis so that it becomes an added value for groups of divers. Not only for the health of divers looking for fish but also for tourists who want to use diving equipment.
