Abstract

Streptomyces costaricanus 45I-3 is a bacterial strain belongs to actinomycetes group isolated from peat soil. Thebacterium is known to produce extracellular xylanase. The aims of this study were to analyze DNA sequence andsub-clone gene involved in the synthesis of extracellular xylanase. Complete DNA sequence predicted to encodexylanase genes was isolated from bacterial genome using Inverse Polymerase Chain Reaction (I-PCR). Total DNAsequence of 1664 bp in size obtained from I-PCR consisted of two open reading frames (ORF) in opposite direction.ORF1 was 1029 bp and ORF2 (partial sequence) was 309 bp. Analysis sequence using BlastX indicated that ORF1was homologous with xylanase bacterium enrichment culture clone Xyl8B8 (GenBank accession No. AFH35005.1),i.e. 95% in identity and 99% in similarity. In addition, ORF2 was homologous with glyoxalase bacterium enrichmentculture clone Xyl8B8 (GenBank accession No. AFH35007.1), i.e. 95% in identity and 98% in similarity. Analysis ofamino acid sequence revealed that ORF1 consisted of 2 domains, i.e. glyco-hydrolase 11 (GH11) and CarbohydrateBinding Type 2 (CBM2). Active site was found at 130th amino acid on GH11 domain. Visualization of 3-dimensionstructure showed that 1029 bp fragment is of 19 areas.

Abstract

Extraintestinal pathogenic Escherichia coli (ExPEC) is a group of pathogens that can colonize the outside of the intestine, such as the kidney, urinary tract, or bloodstream. This study analyzed more about the genetic background of β-lactamase genes among Indonesian ExPEC ST131. Whole-genome sequencing (WGS) sequences from the National Center for Biotechnology Information (NCBI) of Indonesian ExPEC ST131 were taken, then analyzed. Circular genomic mapping and genomic comparison of surrounding genes of β-lactamase in these isolates were generated. blaOXA-1 and blaDHA-1 were analyzed deeply due to their presence in a relatively long contig, making them available for analysis. Indonesian ExPEC ST131 isolates had blaOXA-1 with an identical genetic background of E. coli originating from China and Austria with aac (6’)-1b-cr5 in the downstream and cab83 in the upstream. The blaOXA-1 was also detected in other species, including Klebsiella pneumoniae INF142 originating from Australia and Salmonella enterica S146 from China. While, blaDHA-1 in EC_0406 had an identical genetic background to E. coli 142 and other species such as Shigella sonei FC1428 from India and S. flexneri M2901 from Australia, with Globulin-encoded gene in the upstream and lysR in the downstream. Our findings demonstrate the global spread of β-lactamase genes detected in Indonesian ExPEC ST131. These genes were identical to those isolated from some countries around the world.

Abstract

Abstract: Chitinolytic actinobacteria are currently more widely used because of their ability as the biological control agents to the pathogenic fungi, especially in horticultural and plantation crops. This research was conducted to obtain isolates of chitinolytic soil actinobacteria from the rhizosphere of the rubber plant (Hevea brasiliensis) area in IPB University. Antifungal activities from these actinobacteria hopely able to inhibit the growth of plant pathogenic fungi in Vitro on chitin agar media. Fusarium oxysporum and Sclerotium rolfsii are used in the inhibition test of chitinolytic actinobacteria. The results successfully obtained 16 isolates of actinobacteria were grew on  Humic Acid Vitamin (HV) agar. It showed that six of 16 actinobacteria isolates were able to produce inhibition zones to the growth of hyphae of pathogenic fungi on potato dextrose agar (PDA). KK-15 and KK-07 isolates were able to produce the largest inhibition percentages in F. oxysporum and S. rolfsii. Based on the chitinolytic index (CI) values, KK-15 and KK-07 isolates produced CI values of 1.25 and 1.5, respectively. The morphological characteristics and Gram staining of both KK-15 and KK-07 isolates are closely relative with Streptomyces sp. Abstrak: Bakteri kitinolitik saat ini banyak diteliti karena kemampuannya sebagai agens pengendali hayati jamur patogen khususnya pada tanaman hortikultura dan perkebunan. Penelitian ini dilakukan untuk mendapatkan isolat-isolat bakteri kitinolitik asal tanah dari bagian perakaran tanaman karet (Hevea brasiliensis) di daerah perkebunan karet Institut Pertanian Bogor (IPB). Aktivitas antifungi dari bakteri ini diharapkan  mampu menghambat pertumbuhan jamur patogen tanaman secara in Vitro pada media agar-agar kitin. Fusarium oxysporum dan Sclerotium rolfsii digunakan dalam uji hambat aktinobakteri kitinolitik. Hasil penelitian memperoleh 16 isolat aktinobakteri berhasil diisolasi dengan menggunakan media agar-agar Humic Acid Vitamin (HV), dan dari 16 isolat tersebut, enam isolat mampu menghasilkan zona hambat terhadap pertumbuhan hifa dari kedua jamur patogen pada media Potato Dextrose Agar (PDA) secara in Vitro. Isolat KK-15 dan KK-07 mampu menghasilkan diameter hambatan terbesar pada jamur F. oxysporum dan S. rolfsii. Berdasarkan nilai index kitinolitik (IK), isolat KK-15 dan KK-07 menghasilkan nilai IK sebesar 1,25 dan 1,5 secara berurutan. Karakteristik morfologi dan pewarnaan Gram dari kedua isolat yaitu KK-15 dan KK-07 memiliki kedekatan dengan kelompok Streptomyces spp.

Abstract

ABSTRACT[Development of Visual Audio-Based Microbiology Research Materials for Lipolithic Bacteria to Measure The Ability of Creative Thinking]. The purpose of this research was to develop the design of audio visual teaching materials to measure the creative thinking ability of biology students of FMIPA University of Bengkulu, based on research of lipolytic bacteria degrading palm oil liquid waste. The development model used is the 4D model (define, design, develop, disseminate). In the defining phase, an analysis of the depth of the material, from learning indicators in syllabus, units of lectures agenda and a great line of lesson plan. The analysis is also done by direct interviews to lecturers of microbiology subjects, and participate in learning activities both in the classroom and laboratory. The results of this material analysis serve as guidance in making the design of audio visual teaching materials. The next stage of activity is the design of the preparation of teaching materials developed from the existing learning syllabus. The design of audio visual teaching materials is carried out by feasibility measurement by media experts, IT experts and education practitioners. The results of the measurement of audio-visual material by media experts as 80%, by IT experts as 91.2%, and by education practitioners 75.2%. The deployment stage was only tested on 72 students of Biology Strata one math and science faculty University of Bengkulu, to see the level of creative thinking ability. Measurements to the level of creative thinking ability of students obtained a value of 12.5% in the high category, 70.8% medium category, and 16.7% low category. Keywords: Audio visual; Creative Thinking; 4D Model(Received August 13, 2018; Accepted January 12, 2019; Published February 25, 2019)                                                    ABSTRAKTujuan penelitian ini adalah mengembangkan desain bahan ajar audio visual untuk mengukur kemampuan berpikir kreatif mahasiswa biologi Fakultas Matematika dan Ilmu Pengetahuan Alam  Universitas Bengkulu, yang berbasis penelitian bakteri lipolitik pendegradasi limbah cair kelapa sawit. Model pengembangan  yang digunakan yaitu model 4D (define, design, develop, desseminate). Pada tahap pendefinisian dilakukan analisis terhadap kedalaman  materi, dari indikator pembelajaran yang ada di silabus, satuan acara perkuliahan dan garis besar rencana pembelajaran. Analisis juga dilakukan dengan wawancara langsung kepada dosen pemangku mata kuliah mikrobiologi, dan ikut serta dalam kegiatan pembelajaran baik di kelas maupun praktikum di laboratorium. Hasil dari analisa materi ini dijadikan pedoman dalam membuat desain bahan ajar audio visual. Tahapan kegiatan selanjutnya adalah  rancangan desain penyusunan bahan ajar yang dikembangkan dari silabus pembelajaran yang sudah ada. Desain bahan ajar audio visual dilakukan pengukuran kelayakan oleh ahli media, ahli IT dan praktisi pendidikan. Hasil pengukuran kelayakan bahan ajar audio visual oleh ahli media  80 %, oleh ahli IT  91,2 %, dan oleh praktisi pendidikan 75,2 %. Tahap penyebaran hanya diuji cobakan pada 72 orang  mahasiswa S-1 Biologi Fakultas Matematika dan  Ilmu Pengetahuan Alam Universitas Bengkulu, untuk melihat tingkat kemampuan berpikir kreatifnya. Pengukuran terhadap tingkat kemampuan berpikir kreatif mahasiswa didapatkan nilai 12,5% dalam kategori tinggi, 70,8 % kategori sedang, dan 16,7 % kategori rendah. Kata kunci : Audio visual; Berpikir kreatif; Model 4D.

Abstract

Pulau Enggano merupakan salah satu pulau terluar di Indonesia yang memiliki keragaman biota yang cukup tinggi, salah satunya adalah hewan spons. Spons adalah sekelompok hewan multiseluler, memiliki ciri tubuh berpori, dan organisme invertebrata berguna dalam memproduksi senyawa bioaktif. Bakteri yang terkait dengan spons dapat menjadi sumber alternatif senyawa bioaktif baru, terutama antimikrob. Penelitian ini bertujuan untuk mengetahui potensi bakteri yang terkait dengan Aplysina sp. asal Pulau Enggano sebagai penghasil senyawa antimikrob melawan mikrob patogen. Total 263 isolat berhasil diisolasi pada media agar-agar Sea water Complete (SWC). Isolat bakteri potensial selanjutnya diidentifikasi dengan pengamatan morfologi, pewarnaan Gram, serta dilanjutkan dengan uji biokimia. Hasil uji aktivitas antimikrob dari 16 isolat terpilih melalui uji antagonis, supernatan dan pelet terhadap mikrob patogen Escherichia coli, Staphylococcus aureus, Pseudomonas aeroginosa dan Candida albicans menunjukkan bahwa isolat ENG-4 memiliki kemampuanmenghambat pada dua mikrob pathogen yaitu E. coli, S. aureus. Isolat ENG-4 termasuk ke dalam bakteri gram positif dan merujuk pada genus Bacillus berdasarkan uji biokimia.POTENTIAL OF ISOLATE Bacillus sp. ENG-4 ASSOCIATED WITH MARINE SPONGE Aplysina sp. PRODUCING ANTIMICROBIAL COMPOUNDS FROM ENGGANO ISLAND. Enggano Island is one of the outer islands in Indonesia that has a high diversity of marine biota, one of them are sponges. Sponge is a group of multicellular animals, has a characteristic porous body, and invertebrate organisms are useful in producing bioactive compounds. Bacteria associated with sponges can be an alternative source of new bioactive compounds, especially antimicrobial. This study aims to determine the potential of bacteria associated with Aplysina sp. from Enggano island as a producer of antimicrobial compounds against pathogenic microbes. Total 263 isolates were successfully isolated in Sea water Complete (SWC) agar media. Potential isolates were further identified morphologically, Gram staining, and also biochemical tests. The results of the antimicrobial activity test of 16 selected isolates through antagonist, supernatant and pellet tests on pathogenic microbes of Escherichia coli, Staphylococcus aureus, Pseudomonas aeroginosa and Candida albicans showed that the ENG-4 isolate had the ability to inhibit two spesies of pathogens which are E. coli and S. aureus. ENG-4 isolate belong to gram-positive bacteria and refer to the genus Bacillus based on biochemical tests.

Abstract

Hand is the main transmission pathogen entry into the body which is the part of the body that is most often in direct contact with other limbs and surrounding objects. The importance of maintaining hand hygiene by always washing hands with soap is the first step in preventing transmission of COVID-19. However, during a pandemic like this, it will be difficult to find soap available everywhere, and tends to be hard to carry all the time. Therefore, this service aims to invite the people of Pematang Gubernur, Bengkulu City, to use paper soap because it is easier to use its use and flexible to carry anywhere besides being an effort to prevent the COVID-19 outbreak, it can also add to new information and skills for residents in making attractive soaps. The feedback response given by the community service implementation team to the community service participants resulted in a positive response to this activity, who initially did not know about making and use of paper soap and became aware of this.

Abstract

Limbah organik berupa sayuran dan buah-buahan di Desa Sukasari, Kabupaten  Kepahiang, Kota Bengkulu yang melimpah dapat dijadikan sebagai produk Eco-enzyme sebagai bahan alternatif pembuatan Bio-handsanitizer dan Biofertilizer. Dari hasil kegiatan pengabdian kepada masyarakat mengenai aplikasi pemanfaatan ecoenzyme dari limbah organik sebagai bahan alternatif pembuatan Bio-handsanitizer dan Bio-fertilizer, para kelompok tani  telah mengetahui dan memahami cara pembuatan dan manfaat ecoenzyme. Hal ini menunjukkan kegiatan pengabdian sukses dan mencapai sasaran. Sosialisasi dan demonstrasi pelatihan yang dilakukan memberikan hasil persentase yang baik ketika pemberian umpan balik kepada peserta. Maka dari itu kegiatan pengabdian ini diharapkan untuk dapat memberi solusi terhadap kasus  pengolahan serta penanganan limbah organik melalui pembuatan produk ecoenzyme sebagai bentuk pengolahan limbah organik, seperti limbah sisa buah dan sayur dan diharapkan dapat memberi manfaat berupa wawasan untuk dimanfaatkan dalam realisasi produk ecoenzyme di Desa Suka Sari, Kecamatan Kabawetan, Kabupaten Kepahiang. Sehingga kedepannya kelompok tani dapat mengelola limbah organik secara tepat sehingga menghasilkan produk bermanfaat seperti biohandsanitizer dan biofertilizer dengan nilai jual yang tinggi dan diharapkan bisa mengurangi penggunaan pupuk kimia.

Abstract

This study aims to identify the VEGF-A gene polymorphism (rs2010963) in the first generation of patients with coronary heart disease (CHD) and non-CHD in Bengkulu City. The method used is molecular analysis of polymorphism through Polymerase Chain Reaction Restriction Fragment Length Polymorphism (PCR-RFLP). This type of research is descriptive observational with a cross-sectional research design. The results of this study indicate that the genotype frequency of the VEGF-A gene polymorphism (rs2010963) in the first generation of patients with CHD is genotype CC = 11 (52.4 %), GG = 7 (33.3%) and CG = 3 (14.3%). ). The distribution of the VEGF-A genotype (rs2010963) in the first non-CHD offspring, namely GG = 9 (42.9%), CC = 7 (33.3%) and GC = 5 (23.8%). The statistical analysis results showed that the p-value obtained was 0.460 for the VEGF-A gene polymorphism genotype (rs2010963) in the first CHD and Non-CHD derived samples. In conclusion, there was no significant difference in the VEGF-A gene polymorphism in the first generation of CHD and non-CHD groups in Bengkulu City. Abstract

This study aimed to determine the antibacterial activity of the ethanol extract of the leaves of Tapak Kuda (Ipomoea pes-caprae) against Staphylococcus epidermidis. The method used was Kirby-Bauer for the antibacterial test with various concentrations of ethanolic extract of I. pes-caprae leaf extract: 5 mg/mL, 20 mg/mL, 35 mg/mL, and 50 mg/mL, with five replications. The antibiotic clindamycin was used as a positive control, while DMSO was a negative control. The results showed that after 24 hours of incubation, the zone of inhibition of bacterial growth was demonstrated by the experimental and the positive control groups, while the negative control did not show the zone of inhibition. The diameter of the largest inhibition zone (11.40 mm) was at a 50 mg/mL test concentration, which was indicated as a strong category. In conclusion, the ethanolic extract of the leaves of I. pes-caprae can inhibit the growth of S. epidermidis at all test concentrations. Abstract

AQUAPONIC APPLICATION OF ORGANIC VEGETABLES-CATFISH IN BUCKETS (ASOILELEDAMBER) IN BENGKULU CITY. Bengkulu City is the capital of Bengkulu Province with the highest population in Bengkulu Province so it requires the availability of high food consumption as well. Required foodstuffs such as vegetables, meat, and fish, not all are available from Bengkulu City, so it is not uncommon the price on the market becomes more expensive. To fulfill the needs of foodstuffs, some people of Bengkulu City make effort to cultivate vegetables and maintain fish in ponds, but this method is also difficult to do because of the limited land area. Based on the description, it is necessary to apply the Appropriate Technology (TTG) for the people of Bengkulu City to fulfill family food security, one of which is the application of aquaponic technology: Asoileledamber (Organic Vegetable-Catfish Aquaponics in Buckets). This community service activity aims to provide information and training to the group of service partners in Bengkulu City about the benefits of Asoileledamber. Community service activities include Asoileledamber counseling activities, the provision of guidebooks, provision of tools and materials for cultivation such as fish seedlings, vegetable seeds and pellet feed. The results perceived by the service partners were harvesting Catfish from the second month of maintenance to the fourth month of maintenance. Organic vegetables are harvested starting from 2 weeks after planting time (SMT) until the 4th month of maintenance with an average harvest of 1-2 bunches per bucket.
