Abstract

Accessible tourism has become an increasingly urgent need as awareness of the rights of persons with disabilities, the elderly, and other travelers with special needs grows. Although Indonesia, particularly Bali, has enormous potential as a tourism destination, implementing inclusive principles in travel services still faces numerous challenges. This study aims to design a tailor-made travel package based on accessible tourism as a destination management company innovation to address accessibility issues and realize inclusive tourism in Bali. This research applied a qualitative descriptive method using a case study approach. Data were collected through observation, in-depth interviews with tourism stakeholders, including destination management companies, travelers with special needs, and local partners, as well as field documentation. The research was conducted in Bali as the tourist destination region and the Netherlands as the traveler-generating region, with a focus on the destination company management, Jewel Indonesia Tour and Travel. Findings show that destination management companies still face challenges such as inaccessible infrastructure, limited staff knowledge, and a lack of understanding of universal design concepts. However, there are significant opportunities through niche market demands, national regulatory support, and collaboration with disability communities and international partners. The developed travel package includes inclusive accommodations, disability-friendly activities, accessible transportation, and digital information adapted to user needs. This research significantly contributes to the existing literature gap regarding the role of destination management companies in accessible tourism. Furthermore, the designed model is expected to serve as a best practice framework for developing inclusive tourism products in other destinations across Indonesia. The study concludes that a customer-centered approach, cross-sector collaboration, and sustainable innovation are crucial in building an equitable tourism ecosystem. It is recommended that tourism stakeholders enhance accessibility education and integrate universal design principles into all tourism services.

Abstract

CLOCK merupakan protein penting yang  berperan dalam adaptasi ekologi dan fisiologi organisme. Studi sebelumnya menjelaskan bahwa CLOCK memiliki pengaruh terhadap perubahan perilaku hewan. Penelitian ini bertujuan untuk menganalisis protein CLOCK pada burung Patagioenas fasciata monilis. Metode yang digunakan pada penelitian ini adalah metode homologi. Hasil struktur model berdasarkan assessment menunjukkan akurasi model yang mendekati struktur sebenarnya. Struktur model yang diperoleh memiliki berat 96200,04 Daltons, tiga daerah struktur beta-sheet, lima daerah struktur alpha-helix, 1,47 % daerah outliers,  dua residu C-Beta Deviations, nilai identity 94,43 %, 34 sudut buruk, nilai QMEAN  -1,69 dan nilai GMQE 0,31. Kata Kunci : CLOCK, Patagioenas fasciata monilis, in silico, homologi

Abstract

Penelitian ini bertujuan untuk menganalisis dan mendeskripsikan hubungan kekerabatan varietas ubi banggai (Dioscorea sp.) berdasarkan karakter fenotipik. Penelitian ini merupakan jenis penelitian dekskriptif kuantitatif. Sampel penelitian  diperoleh dari di Tinangkung Utara Kabupaten Banggai Kepulauan yaitu Desa Palam, Desa Luksagu dan Desa Bangpanga. Karakterisasi mengacu pada 15 karakter morfometrik, 2 karakter meristik dan 27 karakter kualitatif. Data diolah menggunakan Program File Editor (PFE), dianalisis menggunakanan analisis klaster dengan program Multivariat Statistical Package (MVSP). Indeks similaritas dihitung menggunakan metode Simple Matching Coefficient, rekonstruksi dendogram menggunakan UPGMA alogarithm. Hasil penelitian menunjukkan bahwa 13 varietas ubi banggai di Kabupaten Banggai Kepulauan, Sulawesi Tengah membentuk 2 klaster dan memiliki hubungan kekerabatan yang dekat dengan indeks similaritas antara 0,766-1,00%. Hasil penelitian ini dapat menjadi informasi dasar yang sangat diperlukan dalam upaya pelestarian dan pengembangan Ubi Banggai sebagai sumber pangan alternatif. Selain itu, informasi ini juga diharapkan dapat dimanfaatkan dalam melengkapi referensi ilmiah terkait penelitian Ubi Banggai.

Abstract

Apis cerana is a member of the Insecta class grouped in the order Hymenoptera. Phylogenetic analysis is used to describe the kinship of living things through phylogeny trees. This study aims to describe the phylogenetic relationship of cryptic species in Apis cerana based on the 16S rRNA gene. Sampling use the cruising method in various regions, the DNA isolation, DNA amplification, sequencing, followed by bioinformatics analysis using GeneStudio, DNASTAR, MESQUITE, and MEGA 11. The result showed that phylogenetic analysis with the Neighbor-Joining (NJ) and Maximum Likelihood (ML) methods produced a phylogenetic tree that illustrates clear evolutionary relationship. A. cerana from Central Sulawesi is more closely related to other A. cerana species in the same clade, while other species are separated. The result of this study show a clear pattern of kinship between A. cerana populations.

Abstract

Apis nigrocincta is an endemic honey bee that is only found in two regions in Indonesia, namely the Sangihe Islands and Sulawesi Islands. Phylogenetic analysis is used to describe the phylogenetic relationship of Sulawesi endemic honey bee A. nigrocincta based on the 16S rRNA gene. Abstract

Xylocopa appendiculata  is an insect from the order Hymenotera and has bright yellow morphological characteristics on the thorax, while the rest of the body is black. Phylogenetic analysis is used to describe the kinship of living things throughh phylogeny trees. This study aims to describe the phylogenetic relationship of carpenter bees Xylocopa appendiculata based on the cytochrome oxidase 1 (CO1). DNA isolation, DNA amplification, sequencing, followed by bioinformatics analysis using GeneStudio, DNASTAR, MESQUITE, and MEGA 11. The results showed that phylogenetic analysis using Neighbor-Joining (NJ) and Maximum Likelihood (ML) methods produced a phylogenetic tree that illustrates a clear evalutionary relationship X. appendiculata from Central Sulawesi is more closely related to other X. appendiculata species in the same clade. The results of this study show a clear kinship pattern between populations of X. Appendiculata.

Abstract

This study aims to identify the presence of Escherichia coli and Salmonella sp. in "ayam geprek" products sold at the canteens of Tadulako University. The type of research used is descriptive-exploratory with quantitative examination methods using the Most Probable Number (MPN), Total Plate Count (TPC), and further identification using the Microbact System and Microbact System 2000 Software. Samples were taken from three different faculty canteens using a stratified random sampling technique. The results showed that (1) Test using the Most Probable Number (MPN) method obtained the highest value of 150x102 CFU / ml came from the FMIPA canteen sample, 28x102 CFU / ml samples from the FKIP canteen and the lowest value of 27x102 CFU / ml samples from the FKIP canteen. (2) Test using the Total PlateCount (TPC) method obtained the highest value of 105x103 CFU / ml came from the FMIPA canteen sample, 217x102 CFU / ml FKIP canteen sample, and the lowest value of 120x102 CFU / ml FKIP canteen sample. (3) Validation identification of the percentage of samples showing contamination by Escherichia coli and Salmonella sp., with a validation level above 90%. Thus, all samples in this study were positive for the presence of Escherichia coli and Salmonella sp. bacteria.

Abstract

This study aims to determine the effect of Roselle flower extract (Hibiscus sabdariffa L.) on cholesterol levels in White Rats (Rattus norvegicus) induced by LDL. The research method used is a Laboratory experimental method with a Completely Randomized Design (CRD) pattern. The number of samples used was 24 White Rats consisting of 6 treatment groups and 4 replications. The normal control group (KN) is a group that is only given food and drink, the negative control group (K-) is given LDL induction for 30 days, the positive control group (K+) is given LDL induction for 30 days and simvastatin, groups PI, PII and PIII are given LDL induction for 30 days and Roselle flower extract doses of 50 mg/kg BW, 100 mg/kg BW and 150 mg/kg BW for 21 days. The data obtained were tabulated and analyzed by the one-way ANOVA statistical test using SPSS version-30. The results showed that Roselle flower extract positively contains bioactive compounds such as flavonoids and tannins and has a significant effect (p<0.05) on reducing cholesterol levels in White Rats induced by LDL. The effective dose to reduce cholesterol levels is a dose of 100 mg/kg BW and 150 mg/kg BW.

Abstract

This research aimed to isolate and identify the types of bacteria present on the surface of biology education students' mobile phone screens. This was an exploratory study using a bacterial isolation method.Bacteria found on the mobile phone screens were cultivated using MacConkey agar, a selective medium specifically designed to detect and identify Gram-negative bacteria, particularly Enterobacteriaceae like Escherichia coli. The accuracy of the Escherichia coli data from the phone screen samples, after processing with the Microbact System, was 96.39%.Malt Extract Agar (MEA) was also used as a selective medium to detect and identify Gram-positive fungi, such as Candida albicans.Based on these findings, the developed module was deemed highly feasible. A learning medium is considered highly feasible for use if it achieves a presentation rate between 60-100%. With an average score of 82.39%, the module meets this criterion and can be effectively used in learning.

Abstract

This study aims to analyze the phylogenetic relationships among six species of fiddler crabs (Uca spp.) found in Lalombi Village, Banawa Selatan Subdistrict, Donggala Regency, based on morphological characters. A descriptive-exploratory approach was employed, with purposive sampling to select representative specimens. Eighteen morphometric characters were measured from three individuals of each species: U. triangularis, U. perplexa, U. dussumieri, U. annulipes, U. tetragonon, and U. bellator. Data were analyzed using Principal Component Analysis (PCA) and UPGMA clustering with the aid of PAST software version 4.03. The PCA results revealed that carapace length (CL), anterior carapace width (ACW), and posterior carapace width (PCW) contributed significantly to total morphological variation. Cluster analysis indicated a close phylogenetic relationship between U. tetragonon and U. triangularis, as well as between U. perplexa and U. bellator. U. annulipes appeared intermediate between the two groups, suggesting a moderate level of relatedness. In contrast, U. dussumieri formed a distinct branch, likely due to its markedly larger body size compared to the other species. These findings affirm that morphometric characters are valuable in elucidating phylogenetic relationships within the genus Uca and support taxonomy based on morphological traits.
