Abstract

This study aims to determine the effect of rosella flower extract on reducing blood glucose levels in alloxan-induced white rats. The research method used is an experiment conducted in the laboratory with a complete randomized design (RAL) pattern. The number of samples used was 24 white rats consisting of 6 treatment groups and 4 replicates. Normal control group (Kn) is a group that is only given food and drink, negative control group      (K-) is a group given alloxan induction, positive control group (K+) is a group given alloxan induction and metformin synthesized drug, treatment groups I, II, and III are groups given alloxan induction and rosella flower extract at a dose of 50 mg/kg BB, 100 mg/kg BB and 150 mg/kg BB. The data obtained were analyzed by ANOVA statistical test. The results showed that rosella flower extract had an effect on reducing blood glucose levels in white rats induced by alloxan. The most effective dose in reducing blood glucose levels in white rats is a dose of 150 mg/kg.

Abstract

Silau Laut Mangrove Tourism in Silo Baru Village, Asahan Regency has developed into a community-based ecotourism destination. This study aims to assess the balance between ecological conservation and tourism development at the site. Using qualitative methods, this study employs field observations, stakeholder interviews, and policy analysis. Findings show that although tourism boosts the local economy, significant challenges exist in preserving mangrove ecosystems, particularly from increased tourism activities and oil palm expansion. Strategic recommendations include strict regulations, community empowerment, environmental education, and eco-friendly infrastructure development. Integration of government programs like BRGM reinforces sustainability. The study contributes to developing a sustainable ecotourism management model integrating ecological, social, and economic perspectives.

Abstract

This study aims to describe the interspecies phylogenetic relationships of parrotfish (Scarus spp.) in Labean Village, Donggala Regency. The research employed an exploratory descriptive method conducted from October 2024 to March 2025. Samples were collected using a purposive sampling technique, with fishing lines and nets as the primary capture tools. The collected specimens were then identified and subjected to morphological measurements, including morphometric, meristic, and qualitative characters. Phylogenetic analysis was carried out using MVSP software with the simple matching coefficient method and UPGMA algorithm. The results revealed six identified parrotfish species: Scarus rivulatus, Scarus ovifrons, Scarus niger, Scarus tricolor, Scarus sordidus, and Scarus quoyi. Phylogenetic analysis indicated that Scarus rivulatus and Scarus quoyi exhibited the closest relationship (similarity index 0.912), whereas the most distant relationship was observed between node 4 and Scarus sordidus (similarity index 0.376). Morphological characters proved to be relatively stable in determining interspecies phylogenetic relationships among parrotfish. Therefore, the findings of this study are expected to contribute to parrotfish conservation efforts and serve as a reference for future research.

Abstract

This research aims to describe the kinship relationship of baronang fish species, based on morphological character analysis. The approach used is descriptive explorative. Research samples were obtained from the sea waters of Labean Village, Donggala Regency using purposive sampling techniques. The obtained baronang fish were then identified and measured based on 21 morphometric characters, 5 meristic characters, and 4 qualitative characters in each of the six species found. Data is processed using Microsoft Excel then analyzed using cluster analysis on MVSP (Multivariate Statistical Package) software. The similarity index uses the Simple Matching Coefficient method, the reconstruction of the dendogram using the UPGMA algorithm. The results of this research found six species of baronang fish, namely: Siganus canaliculatus, Siganus doliatus, Siganus vulpinus, Siganus vermiculatus, Siganus guttatus, and Siganus puellus. The kinship relationship between six types of baronang fish is formed into five clusters with various kinship categories, namely very close, close, and not close. The first cluster shows a very close kinship occupied by Siganus canaliculatus with Siganus doliatus with a similarity index of 0.93%. Meanwhile, the fifth cluster that shows not close kinship is the grouping between fish species on Node 4 with fish species on Node 3 with a similarity index of 0.43%. The results of this research, are expected to be the basis for further research related to kinship or the development of baronang fish species in Labean Village, Donggala Regency.

Abstract

This study aims to describe the obstacles faced by biology teachers at the senior high school (SMA) level in South Palu District in implementing the Independent Curriculum. The method used was descriptive qualitative, with biology teachers from two schools in South Palu District as subjects. The research instruments were interview sheets and questionnaires. The questionnaires were analyzed using an average score calculation to determine the category of obstacles, while the interview data were analyzed qualitatively through data reduction, data presentation, and conclusion drawing. The results of the questionnaire analysis showed that teacher obstacles ranged from low to very low. Among the three aspects, implementation had a relatively greater level of obstacles than planning and assessment, but remained in the low category, while planning and assessment were both in the very low category. Interviews supported these findings, namely that teachers still relied on training modules and experienced difficulties in adapting assessments to student characteristics, as well as facing limitations in facilities and technology. In general, it can be concluded that the obstacles faced by Biology teachers in implementing the Independent Curriculum in South Palu District are in the low and very low categories, but mentoring, competency strengthening, and infrastructure support are still needed so that curriculum implementation can run optimally.

Abstract

This study aims to identify the types of fungi and bacteria that play a role in the fermentation process of cocoa beans. This type of research uses an exploratory laboratory method. The samples in this study were fungi and bacteria during the fermentation process of cocoa beans that had been fermented for approximately 3-5 days. The tools used in this study were digital scales, clear plastic boxes, autoclaves, scissors, micropipettes, spoons, label paper, Petri dishes, incubators, glass covers, Bunsen burners, Erlenmeyer flasks, inoculation needles, stirring rods, microscopes, beakers, measuring cups, scales, hot plates, test tubes, spirit lamp, colony counter, aluminum foil, camera, and writing instruments. The materials used in this study were fermented cocoa bean waste samples colonized by fungi and bacteria, Nutrient Agar (NA) medium, Triple Sugar Iron Agar (TSIA) medium, Bacillus Agar Base, distilled water, crystal violet, eosin dye, iodine, alcohol, and safranin. There are five stages in this research procedure, namely sample collection and preparation, media preparation and sample dilution, sample cultivation on general and selective media, Gram staining, and biochemical testing. The results of this study show that the fungi Saccharomyces cerevisiae and Mucor Sp, as well as the bacteria Bacilllus Sp, are involved in the fermentation process of cacao beans.

Abstract

This study aims to describe the variation in haplotype distribution and phylogenetic relationships of the Philippine scrub fowl based on the EEF2 gene. Eggshell samples were obtained from Tolitoli and Luwuk, Central Sulawesi. DNA isolation followed the gSYNCTM Extraction Kit protocol, and the amplification process was carried out in 38 cycles, including the pre-denaturation stage (95°C, 5 minutes), denaturation stage (95°C, 45 seconds), annealing stage (53°C, 40 seconds), elongation stage (72°C, 1 minute), and post-elongation stage (72°C, 10 minutes), followed by sequencing. Sequence editing was performed using GeneStudio, sequence alignment and phylogenetic tree reconstruction using MEGA 11, and haplotype network reconstruction using DnaSP and PopART. The results showed that the haplotype network based on the EEF2 gene formed three haplotypes, with the population from Tolitoli having a widespread haplotype, while the population from Luwuk had a specific haplotype. In addition, based on the results of phylogenetic tree reconstruction, it was found that the Philippine white-eye birds from Tolitoli and Luwuk were in the same clade but there was genetic variation between populations.

Abstract

Accessible tourism has become an increasingly urgent need as awareness of the rights of persons with disabilities, the elderly, and other travelers with special needs grows. Although Indonesia, particularly Bali, has enormous potential as a tourism destination, implementing inclusive principles in travel services still faces numerous challenges. This study aims to design a tailor-made travel package based on accessible tourism as a destination management company innovation to address accessibility issues and realize inclusive tourism in Bali. This research applied a qualitative descriptive method using a case study approach. Data were collected through observation, in-depth interviews with tourism stakeholders, including destination management companies, travelers with special needs, and local partners, as well as field documentation. The research was conducted in Bali as the tourist destination region and the Netherlands as the traveler-generating region, with a focus on the destination company management, Jewel Indonesia Tour and Travel. Findings show that destination management companies still face challenges such as inaccessible infrastructure, limited staff knowledge, and a lack of understanding of universal design concepts. However, there are significant opportunities through niche market demands, national regulatory support, and collaboration with disability communities and international partners. The developed travel package includes inclusive accommodations, disability-friendly activities, accessible transportation, and digital information adapted to user needs. This research significantly contributes to the existing literature gap regarding the role of destination management companies in accessible tourism. Furthermore, the designed model is expected to serve as a best practice framework for developing inclusive tourism products in other destinations across Indonesia. The study concludes that a customer-centered approach, cross-sector collaboration, and sustainable innovation are crucial in building an equitable tourism ecosystem. It is recommended that tourism stakeholders enhance accessibility education and integrate universal design principles into all tourism services.

Abstract

CLOCK merupakan protein penting yang  berperan dalam adaptasi ekologi dan fisiologi organisme. Studi sebelumnya menjelaskan bahwa CLOCK memiliki pengaruh terhadap perubahan perilaku hewan. Penelitian ini bertujuan untuk menganalisis protein CLOCK pada burung Patagioenas fasciata monilis. Metode yang digunakan pada penelitian ini adalah metode homologi. Hasil struktur model berdasarkan assessment menunjukkan akurasi model yang mendekati struktur sebenarnya. Struktur model yang diperoleh memiliki berat 96200,04 Daltons, tiga daerah struktur beta-sheet, lima daerah struktur alpha-helix, 1,47 % daerah outliers,  dua residu C-Beta Deviations, nilai identity 94,43 %, 34 sudut buruk, nilai QMEAN  -1,69 dan nilai GMQE 0,31. Kata Kunci : CLOCK, Patagioenas fasciata monilis, in silico, homologi

Abstract

Penelitian ini bertujuan untuk menganalisis dan mendeskripsikan hubungan kekerabatan varietas ubi banggai (Dioscorea sp.) berdasarkan karakter fenotipik. Penelitian ini merupakan jenis penelitian dekskriptif kuantitatif. Sampel penelitian  diperoleh dari di Tinangkung Utara Kabupaten Banggai Kepulauan yaitu Desa Palam, Desa Luksagu dan Desa Bangpanga. Karakterisasi mengacu pada 15 karakter morfometrik, 2 karakter meristik dan 27 karakter kualitatif. Data diolah menggunakan Program File Editor (PFE), dianalisis menggunakanan analisis klaster dengan program Multivariat Statistical Package (MVSP). Indeks similaritas dihitung menggunakan metode Simple Matching Coefficient, rekonstruksi dendogram menggunakan UPGMA alogarithm. Hasil penelitian menunjukkan bahwa 13 varietas ubi banggai di Kabupaten Banggai Kepulauan, Sulawesi Tengah membentuk 2 klaster dan memiliki hubungan kekerabatan yang dekat dengan indeks similaritas antara 0,766-1,00%. Hasil penelitian ini dapat menjadi informasi dasar yang sangat diperlukan dalam upaya pelestarian dan pengembangan Ubi Banggai sebagai sumber pangan alternatif. Selain itu, informasi ini juga diharapkan dapat dimanfaatkan dalam melengkapi referensi ilmiah terkait penelitian Ubi Banggai.
