Abstract

Gama Melon Parfum (GMP) cultivar is a result of crossbreeding between the Natsuno Omoide and Miyamauri melon varieties in 2011. GMP exhibits a unique phenotypic characteristic, including a bitter taste of the fruit flesh and produces a stronger aroma. The objective of this study was to investigate the physiological responses and productivity of GMP under varying water conditions. The experiment was conducted using a single factor consisting of different levels of water availability with field capacity at 50%, 75%, and 100%, as well as submergence at 2 cm, 4 cm, and 8 cm above the soil surface. Each treatment was replicated three times. The plot size for each replication was 2 m x 2 m, ensuring consistent conditions for all treatments. The results of the study showed that treatment with 100% field capacity increased stem diameter, while treatment with 50% field capacity increased the root-to-shoot ratio of GMP. Submergence treatment at 8 cm decreased stem diameter and the root-to-shoot ratio of GMP. The 50% field capacity treatment reduced the total chlorophyll levels in GMP leaves. Submergence treatment at 8 cm increased the total chlorophyll levels in GMP leaves. The 50% field capacity treatment increased fruit fresh weight, while submergence treatment at 2 cm decreased fruit fresh weight. Submergence treatment at 2 cm reduced fruit water content, whereas submergence treatment at 8 cm increased water content in GMP fruits. Abstract

Pesisir Pantai Bocor yang terletak di selatan Kabupaten Kebumen, merupakan salah satu lokasi yang berpotensi sebagai lahan pertanian. Sepanjang Pantai Bocor terbentang gumuk pasir yang berfungsi sebagai penghalang angin dan gelombang. Wilayah tersebut dimanfaatkan sebagai lokasi menanam melon yang menjadi komoditas unggulan daerah. Oleh sebab itu dikembangkan melon ramah lingkungan untuk menjaga kelestarian gumuk pasir. Upaya pengembangan melon ramah lingkungan termasuk dalam Education for Sustainable Development (ESD).  yang didanai UGM. Pogram ini bertujuan untuk pengembangan masyarakat Desa Setrojenar, Kabupaten Kebumen berbasis ESD. Kegiatan program ini antara lain sosialisasi tentang pelestarian gumuk pasir, pengenalan alternatif tanaman pangan yaitu umbi-umbian, penanaman tanaman Casuarina equisetifolia Linn. sebagai barier, dan pendampingan budidaya melon menggunakan potensi lokal oleh petani. Hasil kegiatan antara lain partisipasi masyarakat tinggi, gumuk pasir tidak rusak, tanaman barier dapat tumbuh baik dan petani dapat panen melon dengan kualitas baik. Kegiatan ini diharapkan dapat menjadi role model bagi masyarakat untuk terus mengembangkan budidaya melon dan tetap menjaga kelestarian gumuk pasir.

Abstract

Melon ‘Tacapa Gold’ merupakan varietas melon hasil pemuliaan yang dilakukan oleh Laboratorium Genetika dan Pemuliaan yang memiliki karakter berupa bentuk globular, kulit muda hingga tua berwarna dasar kuning dengan net pada kulit buahnya serta warna daging buah hijau. Tujuan penelitian ini adalah untuk mengetahui keseragaman dan kestabilan karakter fenotip berdasarkan karakter morfologis dan molekuler. Metode yang yaitu pengamatan karakter fenotip dan amplifikasi DNA dengan PCR-ISSR. Analisis karakter kuantitatif menggunakan analisis sidik ragam menggunakan PKBT-STAT 3.1. dengan metode Turkey (5%). Pengambilan data keseragaman secara molekuler dilakukan dengan menggunakan metode PCR-ISSR dengan primer UBC-807, UBC-808, UBC- 810, dan UBC-824. Hasil analisis fenotip kualitatif menunjukkan karakter melon ‘Tacapa Gold’ seragam. Karakter fenotip secara kuantitatif cenderung masih bervariasi dan belum seragam apabila dibandingkan dengan melon 'Tacapa Gold’ generasi sebelumnya. Hasil keseragaman fenotip melon ‘Tacapa Gold’ berdasarkan penanda ISSR memiliki tingkat keseragaman yang tinggi (indeks similaritas ≥70%). Rata-rata polimorfisme pada melon ‘Tacapa Gold’ rendah yaitu 24,25%.

Abstract

Bawang merah (Allium cepa L. var. ascalonicum) termasuk dalam tujuh komoditas pangan yang harganya berfluktuasi dan berkontribusi terhadap inflasi. Komoditas bawang merah ini memiliki banyak manfaat dan potensial dalam memenuhi kebutuhan pangan nasional. Pengembangan varietas bawang merah baru memerlukan ketersediaan variasi genetik dalam plasma nutfah. Oleh karena itu, diperlukan pengembangan sumber daya genetik untuk meningkatkan produktivitas bawang merah di Indonesia. Penelitian ini bertujuan untuk mengidentifikasi variasi 21 kultivar bawang merah di Indonesia berdasarkan karakter morfologis sebagai informasi potensi sumber daya genetik. Penelitian dilaksanakan pada bulan Oktober 2021-Oktober 2023 di Laboratorium Kebun Percobaan Sawitsari, Fakultas Biologi, UGM. Sampel penelitian terdiri dari 21 kultivar bawang merah yang dilepas oleh Balai Penelitian Tanaman Sayuran (BALITSA) pada tahun 1984-2018. Penelitian ini merupakan penelitian deskriptif mengikuti metode pengamatan. Prosedur kerja menggunakan teknik penanaman mengikuti standar operasional prosedur dari Departemen Pertanian yang dimodifikasi media tanam. Data penelitian berupa data deskriptif dari pengamatan morfologis 55 karakter bawang merah, terdiri dari daun, umbi, akar, bunga, biji, serta masa tanam, panen dan pascapanen. Hasil penelitian mengungkapkan 21 kultivar bawang merah terdapat karakter morfologis yang beragam pada daun dan umbi. Adanya variasi tersebut kemungkinan disebabkan oleh faktor genetik yang dihasilkan dari adaptasi tanaman terhadap lingkungan yang berbeda. Hasil yang diperoleh menunjukkan pentingnya karakter morfologis sebagai bahan informasi sumber daya genetik komoditas bawang merah Indonesia.

Abstract

Stunting dan anemia merupakan permasalahan gizi kronis yang masih menjadi tantangan kesehatan masyarakat di Indonesia. Kekurangan zat gizi mikro seperti zat besi, asam folat, dan β-karoten menjadi faktor utama yang berkontribusi terhadap kedua kondisi tersebut. Salah satu upaya pencegahannya adalah melalui pemanfaatan pangan lokal bergizi tinggi, seperti labu susu (Cucurbita moschata ‘Citra Laga’), yang kaya akan vitamin, mineral, dan senyawa bioaktif. Penelitian ini bertujuan untuk mengembangkan inovasi budidaya labu susu pada lahan terbatas sebagai bentuk intervensi gizi masyarakat dan peningkatan ketahanan pangan keluarga di Kelurahan Bener, Kemantren Tegalrejo, Kota Yogyakarta. Kegiatan dilaksanakan selama enam bulan melalui tahapan sosialisasi, budidaya di greenhouse dan pot di lahan terbuka, pendampingan budidaya, uji kandungan gizi, serta sosialisasi pengolahan pangan berbasis labu susu. Pendekatan yang digunakan bersifat deskriptif kualitatif dengan analisis berdasarkan observasi lapangan dan hasil uji laboratorium. Hasil menunjukkan bahwa sistem budidaya di greenhouse menghasilkan pertumbuhan tanaman yang lebih baik dan kandungan β-karoten, asam folat, serta zat besi yang lebih tinggi dibandingkan sistem pot terbuka. Masyarakat menunjukkan partisipasi dan penerimaan positif terhadap kegiatan ini, serta memahami potensi labu susu sebagai pangan fungsional untuk pencegahan anemia dan stunting. Program ini berkontribusi terhadap peningkatan pengetahuan gizi, pemanfaatan pekarangan produktif, dan pemberdayaan ekonomi keluarga di wilayah perkotaan.

Abstract

Phosphorus (P) is an essential element for oil palm growth and development. Acid phosphatase (Apase) and Pti‐interacting serine/threonine kinase are two enzymes which enzymes confirmed to be related to P‐uptake in oil palm, therefore their activities in oil palm treated with P‐limitation need to be quantified. Acid phosphatase is believed to be induced by P‐deprivation. Conversely, the Pto‐interaction (Pti) serine/threonine kinase activity is associated with abiotic stress. The aim of this study was to quantify of activities of two selected enzymes and phytohormone content in oil palm‐clones in the P‐limitation condition. Two oil palms genotypes were treated with three P dosages i.e. 0% (v/v), 4.67% (v/v), and 14.02% (v/v) represented as starvation, deficiency, and optimum condition, respectively. The activity of these two enzymes was quantified in mitochondria and cytoplasm using spectrophotometry and modified dot‐blot methods, while abscisic acid, indole acetic acid and gibberellic acid content was quantified using ultra performance liquid chromatography (UPLC). The result showed that the Apase activity in P‐optimum was higher than starvation and deficiency in leaf and root tissues in both genotypes, whereas Pti serine/threonine kinase activity was higher in prolific than non‐prolific genotypes in P‐deficient dosage. Furthermore, abscisic acid content was higher in prolific than non‐prolific genotypes in starvation and deficient, whereas other hormone contents were similar. Association study showed that prolific was separated with non‐prolific ones at different doses of P. Finally, the prolific genotype is more adaptable with P deficiency.

Abstract

The application of Fusarium‐antagonistic endophytic bacteria with plant growth‐promoting traits offers an effective method to enhance the success of banana plantlet tissue culture while combating Fusarium wilt disease caused by Fusarium oxysporum f.sp. cubense Tropical Race 4 (FocTR4) (VCG 01213). This study evaluates the endophytic bacterium AP3311, isolated from healthy banana roots in direct association with orchid roots. AP3311 exhibited strong antagonism toward FocTR4, hyphal colonization ability, and multiple growth‐promoting activities, including phosphate solubilization, nitrogen fixation and auxin production. 16S rRNA gene sequencing identified that AP3311 belongs to the genus Bacillus, while metabarcoding analysis revealed that Bacillus species dominate the root microbiomes of both bananas and orchids. The bacterial supernatants stimulated root development and leaf growth in vitro. Metabolomic profiling indicated that antimicrobial compounds, together with plant growth regulators, promoted both root and shoot growth. Overall, the research demonstrates that Bacillus sp. AP3311 and its supernatants are valuable components in banana tissue culture, providing the dual benefits of plant growth promotion and effective disease control.

Abstract

Anemia and stunting remain a major challenge in public health issues, especially in pregnant women and toddlers. Cucurbita moscatta ‘Butternut’ (Labu susu), which is rich in essential micronutrients uch as Beta Carotene (provitamin A), folic acid, iron, and zinc, can be processed into highly potent functional foods. Aside from being an excellent source of alternative carbohydrates, it can also meet the micronutrient needs to support the prevention of Anemia and stunting. This activity was carried out at the Community-Friendly Green Open Space (RTH - Ruang Terbuka Hijau) in the Bener sub-district, Tegalrejo district, Yogyakarta City, with the aim of analyzing the taste acceptability of processed bottle gourd products among the target groups. The implementation methods included socialization, taste testing, and acceptability analysis of three processed products based on Bottle Gourd: Nutrient-Rich Baby Porridge, High-Protein Mud Cake (Kue Lumpur), and Compote for Pregnant Women (Kolak Ibu Hamil). The activity involved 50 panelists who assessed the color, aroma, taste, texture, and overall acceptance of the three processed products using a six-level hedonic scale. The results showed that all three products were well-received. For all indicators ( Color, Aroma, Taste, and Texture), all products received scores above 4.6, and the overall acceptance was above 5. The overall acceptance score for the High-Protein Mud Cake (5.22 ± 0.6), the Kolak for Pregnant Women (5.18 ± 0.7), and Nutrient-Rich Baby Porridge (5.00 ± 0.8). The conclusion of this research is that panellists “like” and “accept / receive” all three products. This activity had a positive impact on increasing public knowledge regarding the diversification of local food and served as a strategic rocessed products; step in the effort to prevent anemia and stunting at the family level through the fulfillment of essential micronutrients.

Abstract

The cultivation of arrowroot (Maranta arundinacea L.) in Indonesia, particularly in Yogyakarta Province has a great potential to be developed. This study aims to determine the genetic variability and analyze the intraspecific relation of arrowroot using Inter Simple Sequence Repeats (ISSR) markers as basic informations for considerations in the character selection of arrowroot cultivation in Yogyakarta. Exploratory survey method was occured in Gunungkidul, Kulon Progo, Sleman, and Bantul Districts to collect cultivar accessions. Accessions were replanted in Sawitsari Research Station. DNA isolation from the leaves of 7 moths old accessions was done using CTAB buffer solution. The DNA fingerprinting analysis was carried out using the result of DNA amplification with 4 ISSR markers. The polymorphism data then used for phenetic analysis using UPGMA algorithm and Baroni-Urbani Busser similarity coefficient to form dendrogram. Total of 5 local cultivars were found, identified as ‘Sili’, ‘Sembowo’,’ Sugo’, ‘Kebo’, and ‘Teropng’, each showed distinct rhizome morphological characters. The ISSR-PCR analysis resulted high polymorphism with 68,17% polymorphism mean and mean of polymorphic band was 6,75. Dendrogram resulted 4 clusters in 80% similarity index. Cluster A, B.I., and B.II.b consisted of ‘Sili’, ‘Teropong’, and ‘Kebo’ cultivars, respectively, while the cluster B.II.a gathered ‘Sugo’and ‘Sembowo’ cultivars.

Abstract

CmBGI is the enzymatic genes encoding β-glucosidase that involved in Abscisic Acid (ABA) metabolism of Cucumis melo L. β-glucosidase promotes the accumulation of glucose, fructose, and sucrose, and it might act as a regulator that mediates melon fruit ripening both climacteric and nonclimacteric. ABA mediates adaptive responses to abiotic and biotic stresses. Agricultural Balitbang in 1997 showed that there were approximately 158.600 ha of degraded land scattered in three zones of agroecosystems in Yogyakarta (DIY). One of them is Dlingo Bantul area which has a karst type critical land area. Karst provides stress to the certain plant growth. One way to conserve critical land is making this area for agriculture. Cultivar TACAPA and TA were superior melons that have been developed by Genetic Laboratory of Biology Faculty UGM. This preliminary research was conducted to examine molecular characterization of CmBGI gene expression in cultivar TACAPA and TA which are planted in normal condition medium and in critical land medium treatment. Total RNA was extracted from leaf tissue then Reversed Transcriptase (RT-PCR) to collect cDNA library. cDNA was amplified using specific primer. Spectrophotometry was conducted in λ260 nm and electrophoresis run in 1.5% agarose gel. Control of band chosen was Cm-Actin. CmBGI gene concentration of TACAPA and TA in normal condition medium are in succession 578.5 and 579.4 μg/ml then for critical land medium treatment 743.4 and 773.5 μg/ml. CmBGI band was showed both of TACAPA and TA as ± 1258 bp. Cm-actin was showed band of DNA as ± 445 bp. CmBGI gene concentration in critical land medium treatment which is given greater stress on melons are higher than normal condition.
