Abstract

Alocasia longiloba Miq. is a member of the Araceae family, which was complex and has diverse leaf shapes, color, and venation. Studies on the diversity of A. longiloba focused mostly on morphological characteristics, while the genetic diversity of this species has never been published. This research aims to determine the genetic diversity and phylogeny of the Indonesian accession of A. longiloba based on molecular approaches using trnL-F IGS region. A total of 20 accesses of A. longiloba from Indonesia were sequenced then together with 11 DNA sequences of A. longiloba registered at the NCBI, were aligned with the Mesquite software. Genetic diversity was examined using DnaSP ver. 6.12.03. The phylogenetic tree was reconstructed by MEGA 11 using the Maximum Parsimony method and the Tree-Bisection-Regrafting (TBR) algorithm. Among 389 sites recorded, 356 site were invariable, 5 site were variable and 28 site were gaps or missing data. This genetic variation in the form of insertion, deletion, transition, and transversion and the deletion event has received the most contributions. Reconstruction of the phylogenetic tree resulted in A. longiloba being polyphyletic. The use of cpDNA with the trnL-F IGS region could not show the relationship between the Indonesian accession of A. longiloba, so further research is needed in other regions of chloroplast and nuclear DNA.

Abstract

Abstract Euglena sp. is a unicellular, flagellated microalga considered one of the most promising microalgal feedstock species for biofuels. Reducing  the  level  of  liquid waste pollutants  can be  done  biologically  by  using  microalgal  organisms. Its metabolites, including lipids, proteins, carbohydrates, and pigments, are appropriate for producing biorefinery products such as biodiesel and jet fuels. They can be isolated from extreme environments, such as highly acidic and ammonia-rich environments, that are not conducive to their proliferation. This study sought to determine the effect of the photoperiod or (light: dark) cycle (24 L:0 D, 12 L:12 D, 14 L:10 D, and 16 L:8 D) on the growth, biomass, metabolite content consisting of lipids, carbohydrates, and proteins, and the rate of CO­2 uptake by Euglena sp. As stated previously, the study was conducted by cultivating Euglena sp. on a laboratory scale with four photoperiod regimens. The results indicated that optimal growth, biomass content, and metabolite content were obtained with a 24 D:0 L lighting cycle. The control treatment (24 L: 0 D) had the highest biomass productivity (0.032 g.L-1.day-1 ± 0.004), lipid content (0.387 g.L-1 ± 0.031), protein content (0.542 mg.Ml-1 ± 0.007), carbohydrate content (0.409 x104 g.L-1), chlorophyll a (6.237 g.L-1 ± 0.184), chlorophyll b (2.838 g.L-1 ± 0.253), and total carotenoid (1.566 g.L-1 ± 0.105). Full light illumination (24 L:0 D) was significantly producing carotenoid content, including phaeophytin a, phaeophytin b, violaxanthin, 9'-cis-neoxanthin, dino xanthin, and fucoxanthin. Highlight Research The growth rate was higher when Euglena was cultivated under continuous illumination. The biomass productivity of Euglena increased significantly under continuous illumination. The metabolite content of Euglena (lipids, proteins, and pigments) was higher in continuous illumination.

Abstract

Pandemi Covid-19 menyebabkan perubahan kondisi sosial ekonomi masyarakat Indonesia. Sekitar 40% pekerja atau pelaku usaha mengalami perubahan pendapatan. Hal ini sedikit banyak mempengaruhi kemampuan masyarakat dalam memenuhi kebutuhan pangan keluarga. Pengabdian masyarakat ini bertujuan untuk memberikan edukasi kepada masyarakat untuk mengoptimalkan pemanfaatan pekarangan dan limbah rumah tangga untuk memenuhi kebutuhan pangan keluarga melalui budidaya lele. Pelaksanaan program dibagi ke dalam tiga tahap, yaitu pelatihan warga, pendampingan, dan evaluasi.Pelatihan warga meliputi pelatihan budidaya lele dan kangkung secara hidroponik menggunakan pot dan pelatihan pembuatan pakan lele dari limbah rumah tangga. Pendampingan dilakukan secara berkala setiap minggu hingga lele bisa dipanen. Evaluasi dilakukan di akhir kegiatan untuk mengetahui efektivitas dan potensi teknologi untuk dikomersilkan. Hasil dari program ini menunjukkan bahwa budidaya lele dan kangkung menggunakan pot dapat digunakan untuk memenuhi kebutuhan pangan keluarga dengan harga yang murah. Desain teknologi kolam portabel dari pot ini juga berpotensi untuk dikembangkan sebagai industri rumah tangga. Pakan lele dari limbah rumah tangga mampu menekan biaya pakan dalam budidaya lele hingga 20% dengan nilai rasio konversi pakan (FCR) sebesar 1.11 atau efisiens pakan hingga  90%. Pakan lele ini membantu pengurangan sampah sisa makanan dan sampah dapur hingga 100%.

Abstract

Indonesia memiliki beragam tumbuhan yang kaya akan manfaat salah satunya sebagai bahan dalam industri kosmetik. Bahan alami dipilih karena memiliki keuntungan yang ramah lingkungan dan rendah iritasi. Salah satu bahan alami yang saat ini sedang dikembangkan untuk dijadikan sebagai bahan kosmetik adalah dengan menggunakan ekstrak melon kultivar Gama Melon Parfum (GMP) dari Universitas Gadjah Mada. Tujuan penelitian ini untuk mengetahui proses pembuatan sediaan lotion dari ekstrak melon kultivar Gama Melon Parfum di Laboratorium Farmasi, Universitas Gadjah Mada. Metode yang digunakan adalah penelitian eksperimental, yaitu penelitian yang terfokus pada percobaan pembuatan sediaan lotion dengan ekstrak melon diperoleh melalui metode decoct (pemanasan) dengan beeswax dan cetyl alcohol dimasukkan ke dalam paraffin liquid dan diaduk hingga membentuk lotion. Hasil yang diperoleh yaitu tidak didapatkan konsistensi yang sesuai pada lotion atau mengalami kegagalan disebabkan oleh basis yang tidak cocok dan beberapa faktor lainnya.

Abstract

Kosmetika bahan alam saat ini menjadi penggunaan yang sangat esensial misalnya penggunaan Gama Melon Parfum sebagai bahan baku pembuatan gel, karena penggunaan tumbuhan sebagai bahan dasar kosmetika memiliki bahan baku yang aman untuk digunakan dan memiliki efek samping yang lebih kecil. Gama Melon Parfum merupakan melon yang saat ini dikembangkan oleh Universitas Gadjah Mada yang dapat dijadikan sebagai bahan baku kosmetik alami karena memiliki metabolit sekunder berupa flavonoid, saponin, terpenoid dan cucurbitacin. Penelitian ini bertujuan untuk mengetahui proses pembuatan gel dari ekstrak Gama Melon Parfum. Metode yang digunakan yaitu metode eksperimental, yang terdiri atas pembuatan ekstrak melon melalui metode dekokta (pemanasan) dan pembuatan gel. Adapun hasil yang didapatkan yaitu sediaan gel dengan banyak gelembung udara di dalamnya disebabkan karena proses pengadukan yang terlalu cepat dan banyaknya udara yang masuk pada saat pembuatan sediaan gel. Hal ini menunjukkan proses pembuatan gel harus diperhatikan agar mendapatkan sediaan gel yang sesuai.

Abstract

Kosmetik menurut Food and Drug Administration merupakan produk yang digunakan pada kulit dengan tujuan untuk membersihkan, mempercantik, memperbaiki diri ataupun untuk meningkatkan daya tarik. Berdasarkan Peraturan Kepala BPOM nomor 18 tahun 2015 tentang persyaratan teknis bahan kosmetik, bahan kimia pada kosmetik dibedakan menjadi dua, yaitu bahan kimia berbahaya yang dilarang dan bahan kimia berbahaya yang diperbolehkan digunakan dengan pembatasan dalam penggunaannya pada produk kosmetik. Namun pada dasarnya, kosmetik tidak hanya terbuat dari bahan kimia melainkan dapat pula terbuat dari bahan alami seperti ekstrak buah yang dimanfaatkan. Salah satu tumbuhan yang dapat dijadikan bahan alami dalam pembuatan kosmetik yaitu buah melon. Jenis penelitian merupakan penelitian kualitatif yang terfokus pada proses pembuatan sediaan serum dari ekstrak melon kultivar Gama Melon Parfum yang diperoleh melalui metode dekokta (pemanasan). Hasil yang diperoleh yaitu diketahui adanya batasan-batasan dalam pembuatan kosmetik terkhusus pada penambahan bahan dan perlakuan yang membantu keberhasilan pembuatan serum, seperti pengadukan campuran harus dilakukan secara konstan, jika tidak maka akan menimbulkan buih atau gelembung yang menyebabkan pH serum tidak stabil.

Abstract

This research purpose was to study the phenotype characters of body weight and pectoralis thoracicus muscle performance of hybrid chicken (F1) derived from crossbreed between parent stock broiler strain Cobb-500 and Pelung (Gallus gallus gallus). The chicks were divided into 3 groups and each group consists of 5 days-old chicks (DOC). Group 1 (T1) was used as control involving DOC derived from crossbreed between female and male of broiler Cobb-500 strain. Group 2 (T2) involving DOC derived from crossbreed between female and male Pelung, while Group 3 (T3) was DOC derived from crossbreed between female broiler strain Cobb-500 and male Pelung. The chicken was grown up to 49 days to evaluate the body weight, the pectoralis thoracicus weight, the pectoralis thoracicus area, and myofiber area. Data were statistically analyzed using one way Anova. The results showed that the average of body weight in T1; T2; and T3 were 2,17086.02; 506.220.02; and 1,23868.25 g, respectively (P0.05). The average of pectoralis thoracicus muscle weight in T1; T2; and T3 were 165.0411.97; 21.262.44; and 68.741.94 g, respectively (P0.05). The average of pectoralis thoracicus muscle area in T1; T2; and T3 were 97.09,7; 616,4; and 63,41,8 cm2 , respectively (P0,05). The average of pectoralis thoracicus myofiber area in T1; T2; and T3 were 14.721.04; 4.662.1; and 13.131.3 m2 , respectively (P0.05). It was concluded that crossbreed between broiler strain Cobb-500 and Pelung improved the body weight and pectoralis thoracicus muscle performance of hybrid chicken (F1).

Abstract

Sorghum is a strategic commodity with good nutritional and high antioxidant content that can be used as a source of food, feed, and industrial raw materials. Sorghum is a relatively low and less intensive crop than other staple food crops. Sorghum yield can be increased by establishing plant population through a mulching planting system. The purpose of this study was to determine the yield power of several sorghum genotypes, namely three new superior varieties (Bioguma, Kawali, and Samurai) and three local varieties (Plonco, Hitam Wareng, and Ketan Merah), planted with a plastic mulch planting system. The research was conducted on farmers' land in Karangmojo, Gunungkidul, Daerah Istimewa Yogyakarta, which is a dryland soil type. The experimental design used a randomized complete block design (RCBD) with 5 replications. Observation parameters included growth components and sorghum production. The results showed that sorghum planting with a mulching planting system was able to provide yields for the Bioguma variety of 7.70 tons/ha, Plonco variety of 8.41 tons/ha, Samurai variety of 5.38 tons/ha, Kawali variety of 8.21 tons/ha, Hitam Wareng variety of 3.91 tons/ha, and Ketan Merah variety of 4.00 tons/ha. Based on the results of the study, it was found that the genotypes of local varieties were able to compete with new superior varieties and had the best growth response and yield power in the rainy season planting.

Abstract

The field of identification of molecular markers has seen significant advancements, with the Inter-Simple Sequence Repeat (ISSR) emerging as a prominent tool for characterizing genetic variation within species. This study focuses on the new Indonesian melon cultivar “Melona,” developed through selective breeding of commercial melons. This study aimed to assess the utility of ISSR markers in evaluating the genetic variation and stability of the “Melona” cultivar. Comparisons were made with established cultivars—‘Inthanon,” “Rich Sweetness,” “Chamoe,” and “Hikapel”—using five ISSR primers: UBC 807, UBC 808, UBC 810, UBC 812, and UBC 826. A total of 70 DNA fragments with a high polymorphism rate were produced, thereby providing insight into phenetic relationships among the cultivars with simi-larity indices <70%. These genetic distinctions were corroborated through morphological evaluations. These findings indicate that ISSR markers effectively reveal genetic variation within species, offering a practical approach for devel-oping new plant varieties in the Cucurbitaceae family.

Abstract

Kunir asem is one of the traditional drinks that still survive in Javanese society. The young generation needs to be introduced to the heritage of traditional herbal medicine, so that its existence is maintained. Some people judge that herbal medicine is an ancient traditional drink and does not keep up with the times. The objectives of this study are: (1) Explain the study of etnobotany which make up turmeric acid; (2) Explain how the principle of the benefits of kunir asem is based on the understanding of the seller of herbs in the Province of Yogyakarta. This study uses a qualitative method. The research data was taken with documentation, interviews, and documentation. The research data were analyzed by qualitative descriptive. The conclusion of this study is that biopharmaca plants in kunir asem are turmeric and tamarind, and some are added spices in the form of brown sugar, lemongrass, cardamom, cloves, kedawung, lime, and cinnamon. Herb sellers reveal that kunir asem can promote haids, facilitate digestion, reduce stomach acid, as an antibiotic, fresh body, and eliminate acne.
