Abstract

Pesisir Pantai Bocor yang terletak di selatan Kabupaten Kebumen, merupakan salah satu lokasi yang berpotensi sebagai lahan pertanian. Sepanjang Pantai Bocor terbentang gumuk pasir yang berfungsi sebagai penghalang angin dan gelombang. Wilayah tersebut dimanfaatkan sebagai lokasi menanam melon yang menjadi komoditas unggulan daerah. Oleh sebab itu dikembangkan melon ramah lingkungan untuk menjaga kelestarian gumuk pasir. Upaya pengembangan melon ramah lingkungan termasuk dalam Education for Sustainable Development (ESD).  yang didanai UGM. Pogram ini bertujuan untuk pengembangan masyarakat Desa Setrojenar, Kabupaten Kebumen berbasis ESD. Kegiatan program ini antara lain sosialisasi tentang pelestarian gumuk pasir, pengenalan alternatif tanaman pangan yaitu umbi-umbian, penanaman tanaman Casuarina equisetifolia Linn. sebagai barier, dan pendampingan budidaya melon menggunakan potensi lokal oleh petani. Hasil kegiatan antara lain partisipasi masyarakat tinggi, gumuk pasir tidak rusak, tanaman barier dapat tumbuh baik dan petani dapat panen melon dengan kualitas baik. Kegiatan ini diharapkan dapat menjadi role model bagi masyarakat untuk terus mengembangkan budidaya melon dan tetap menjaga kelestarian gumuk pasir.

Abstract

ABSTRACTSpecies Identification of Scarlet gourd (Coccinia grandis (L.) Voigt) in Three Population inYogyakarta. Papasan is a dioecious plant belongs to the family Cucurbitaceae. This plant iscommonly used as vegetable, anti diabetic, anti bacterial, and anti diarrhea. In Daerah IstimewaYogyakarta, there were two varians of Papasan (Papasan I and II), found in three population(Ngebel, Berbah and Gajah Wong Riverbank). They differ in phenotype, especially in shapeand taste of fruit. Genotype observation using squash method on the root tips with modificationin the duration of maceration were used in this research indicated that cells devided of PapasanI at about 8-11.30 a.m, while Papasan II at about 08.30-09.30 a.m, 11 a.m-00.30 p.m and 2-2.30p.m. The chromosome number of both Papasan is 2n=24, contains of 22 autosomes and 2 sexchromosomes. The karyotype formulas of Papasan I and II were 2n=24=20m+2sm+XX(m).Based on the statistic test, significant difference on chromosomes character between PapasanI and II was only in short arm of autosome pair number 5. The difference R value betweenPapasan I and II was smaller than 0.25. It revealed that the both of Papasan is closely relatedand belongs to the same species of Coccinia grandis L. Abstract

Soybean is an important food crop. Productivity of local soybean needs to beimproved to support community food security. Research in the framework of Education forsustainable development has implemented since April until September 2011 in the village ofBeji, District Ngawen, Gunung Kidul Regency, Yogyakarta Special Region. Counseling andtraining of soybean processing was done to educate the public. Soybeans seed areprocessed into soybean milk and added to the manufacture of ‘tiwul’ to enhance protein inthese traditional foods. Communities are trained to process tempe and tofu into nuggets tocreate a variety of flavors. After the training they are expected to increase nutritional intake,especially for the Beji village children and rural communities become healthier.Keywords: soybean, welfare, community

Abstract

This study conducted in Laboratory of Molecular Biology, Balai Besar Bioteknologi dan Sumberdaya Genetik Pertanian (BB- Biogen) Bogor. The aims of this study are to determine and comparing the quantity,  quality and the efficiency of DNA isolation result using detergent method and CTAB method.  The parameters observed in this study are the value of DNA concentration, purity, and visualization result using gel electrophoresis. The samples are the leaves of Salak ‘Pondoh’ (Salacca zalacca (Gaert.) Voss.). Detergent method is a method which was developed by Faculty of Biology UGM, it has simple method and relatively affordable cost. Meanwhile, CTAB method is one of the commonly used methods of DNA isolation protocol with relatively expensive cost.  Detergent method used detergent in the cell wall separation and protein removal in the sample. The CTAB method used Cetyltrimethyl ammonium bromide (CTAB) for cell membrane separation in the sample. The research methods included DNA isolation with detergent and CTAB methods, PCR analysis and electrophoresis. Data analysis was done quantitatively  using spectrophotometric method and qualitative used electrophoresis method. The result of the study  showed that DNA isolation using  CTAB method showed higher purity compared with detergent method with the purity values ranging from 1,3- 1,4 . Meanwhile, the concentration of DNA in the detergent method was higher than that of CTAB with the highest concentration of 1730 µg/ml. There is no difference between the  quality of genomic DNA isolated by CTAB and detergent methods.

Abstract

Borneo has a a wealth of various flora, including the Aroids (Araceae). Taro is one of the Aroids that cultivated and used as food crop since a long time ago by the people in Borneo. This study aimed to determine the utilization of Taros traditionally by several ethnic communities in Borneo. The research used Survey Explorative Method with Direct Interview Technique in the field. Taro samples were taken from various habitats of Banjar, Dayak, Kutai, Malay, Bugis, Toraja and China ethnic. The results showed that Taro species widely used are : Colocasia esculenta var. esculenta and var. antiquorum, Colocasia affinis and wild of Colocasiaesculenta. C. esculenta var. esculenta and var. antiquorum is used as subsistence food crop and vegetables. Besides, it is used as medicine such as high blood pressure lowering and for consumption of diabetics. Parts of the plants consumed include leaves, petiole, corm and stolon. C. affinis is used as ornamental plants of the home garden, while wild C. esculenta is used as animal fodder. Taro in Borneo have a considerable variation of traditional cultivars and vernacular names. Fifty eight traditional cultivars from 5 different habitat to be used by some ethnics.

Abstract

Advances in technology have encouraged the growth of a variety of biodiversity processing methods. These conditions encourage efforts to use natural resources more broadly. Ethnobotany can be used to document the local knowledge of the plants and their utilization. This study is aimed to describe the various forms of bamboo utilization by Sangirese. The method used was survey with purposive sampling. The data were analyzed descriptively based on exploration results and interviews with the people. The results showed there were 11 species of bamboo that are directly related to the life of local community in Sangirese, consisting of seven species of Bambusa, three species of Schizostachyum, and each one species of Dendrocalamus and Gigantochloa. Local community use bamboo for a variety of daily necessities; as building materials, handicraft, furniture, ceremonies, musical instruments, transportation, medicine, ornamental plants, as well as food. These exploration results are expected to complement the data of bamboo ethnobotany in Indonesia. Applicatively, this research is expected to be a reference in the national strategy of bamboo germplasm resource conservation.

Abstract

Synedrella nodiflora (L.) Gaertn has taxonomically been the only species of genus Synedrella, which spreads over many tropical countries. In spite of its wide range of distribution, the genus remains monospecific. This leads to assumption of the very low genetic diversity among S. nodiflora populations worldwide. It may also be the case in Java Island, though rapid changes in ecosystem condition occurs. Here we report our study on S. nodiflora population genetics in Java Island using intergenic spacer (IGS) atpB –  rbcL as a molecular marker, since it has been well known as one of the most variable chloroplast genome regions in a wide range of plant species so far. As many as 58 individuals were collected randomly from ten different locations in the island. Based on IGS atpB – rbcL sequences of 860 bp length, only two haplotypes were observed. Both show only one polymorphic site (0.12%) and one transversion, where T is substituted by G at position 790, indicating that high connectivity among populations of S. nodiflora in Java Island is observed. This results in a low genetic differences among the populations, which at the same time provides a fact of nearly no variation among the IGS atpB – rbcL sequences.

Abstract

Perakitan melon tahan penyakit gummy stem blight (Gsb-4) merupakan upaya yang dapat dilakukan untuk menanggulangi penyakit gummy stem blight. Untuk itu harus diketahui ada tidaknya gen ketahanan terhadap gummy stem blight. Tujuan penelitian ini adalah mendeteksi adanya gen ketahanan terhadap gummy stem blight pada 18 kultivar melon dan 1 kultivar mentimun. Hasil amplifikasi menggunakan penanda molekular Simple Sequence Repeat (SSR) dengan primer spesifik yaitu CMTA170a menunjukkan pita berukuran 120 bp sedangkan dengan primer CMCT160a+b menunjukkan pita berukuran 212 bp. Hasil yang diuji menunjukkan bahwa semua kultivar melon memiliki gen ketahanan terhadap gummy stem blight baik homozigot maupun heterozigot.

Abstract

Powdery mildews are plant diseases caused by fungi, including Sphaerotheca fuliginea Poll. They cause a decrease on leaves health. Plant resistance to Sphaerotheca fuliginea Poll. might have a relationship to leaf anatomical structure, especially to epidermis. Therefore, the aim of this research was to characterize epidermal leaf structure of resistant and susceptible cultivars. This study used two resistant cultivars (PI 371795 and PMAR), one susceptible cultivar (Action 434) and two other cultivars (Fresh 848 and Sakata 144) which are not known their characteristic to powdery mildews. The fifth leaves (counted from the cotyledon) were collected from melon plants which reached early generative development. Leaf epidermal slides were prepared semi permanently. The results showed that the morphological leaves of resistant cultivars have dendatus leaf edges, while the susceptible one has repandus leaf edges. Anatomically, the leaves of PI 371795 had upper epidermal cells with wavy edges, but not in the others. Both PI 371795 and PMAR 5 had irregular shape of lower epidermal cells with curved edges, while leaf epidermal cells from susceptible cultivar tended to have irregular polygonal upper and lower epidermal cells with straight edges and shorter non-glandular trichomes. PMAR 5 had long glandular trichomes. There was significant difference between cultivars in the average of stomata number, the average of stomata index and trichome number on the lower epidermal tissue, but not on the lower epidermal tissue. In the lower epidermis, PI 371 and PMAR 5 had lower average of stomata number and stomata index compared to Action 434. PMAR 5 had higher number of trichomes than susceptible cultivar (Action 434). Meanwhile, Fresh 848 cultivar tends to have similar characters to resistant ones in terms of leaf edges, lower epidermal shape, stomata number and index. Sakata 144 had similar characters to those the susceptible one in terms on leaf edges, lower epidermal cells shape, stomata number and index, but not in trichome number. It may be assumed that the first protection of melon against S. fuliginea Poll. is located on lower epidermal tissue.
