Abstract

This study aims to design automatic school bell hardware based on the ATmega32 microcontroller, design automatic school bell software based on the ATmega32 microcontroller using Basic programming language, and test the performance of the automatic school bell based on the ATmega32 microcontroller. This automatic school bell uses a minimum system of ATmega32 microcontrollers which equipped with data input from the push button, LCD to display character output, ISD2560 to output sound, and a timer taken from DS1307 RTC. This research is a Research or Development (R & D) research. This research is carried out through two stages of design. The first stage is the stage of hardware design. The second stage is the stage of software design. Programs are made using the Basic programming language using the Bascom-AVR application. The results of the product were tested using the black box method with two stages. In the first stage, software testing uses a series of simulations on the Proteus 7 Professional application. The second stage, testing hardware.

Abstract

Chrysanthemum (Dendranthema grandiflorum) is well known as a commercially valuable flower. However, the plant cultivation has not been well developed. Poliploidization is one of cultivation technique using colchicines as mutagenic agent to produce high quality of flowers. The aims of this research were to study phenotypic characters of chrysanthemum treated by colchicines and to determine the optimum of soaking time to obtain the highly phenotypic characters.Colchicines concentration used in this research was 0.01% with soaking times were 6, 12 and 24 hours. Phenotypic characters observed in this experiment were flower diameter, plant height, stem diameter, long and wide of 5th and 10th leaf from flower base with 10 replications for each characters.The result showed that soaking times treatment with 0.01% colchicines have real influence on flower diameter, plant height, stem diameter, long and wide of 5th and 10th leaf from flower base. The result of colchicines with soaking treatment of 12 hours showed the highly phenotypic characters. In addition, chrysanthemum with colchicines treatment has bigger phenotypic characters than control. Key words: Chrysanthemum, Dendranthema grandiflorum, colchicines, phenotypic characters

Abstract

Species of Bambusa had widespread in Celebes, especially for Bambusa striata and Bambusa vulgaris. As an effect of the lacking of flowering, species identification mainly depends on vegetative features, and leaf epidermal micromorphology has proven to be useful in bamboo taxonomy. The objective of this research was to describe the classification of Bambusa from Celebes based on the micromorphological characters of leaf epidermis. The specimens were collected from wild population. The samples of leaf were collected from five member of Bambusa i.e.: Bambusa blumeana, Bambusa maculata, B. striata, B. vulgaris and Bambusa sp. Micromorphological characters were identified using Scanning Electron Microscopy (SEM). Leaf epidermis characters separated B. blumeana from other species of Bambusa. Furthermore, B. striata were closely related to B. maculata in variation of bulliform cells. As an invention, we release peltate hair as a new type of trichome in Bambusa. The presence of various prickles, trichomes and bulliform cells may be especially useful in delimiting species.

Abstract

Jamusan village is one of the areas that becomes the center of melon production at Prambanan subdistrict, Sleman, Yogyakarta. However, at this moment, the villagers of Jamusan still encountered many obstacles in melon farming. This is caused by the poor quality of the melon, crops that have not been handled well and the lack of farmer’s knowledge about the management after harvesting, melon production that is not absorbed by the market. LANTPYCATION Program (Plant, Apply, and Diversification) aims to provide knowledge about quality and good management of the melons in the form of processed product diversification melon. The method applied in the implementation of this program which begins with socialization, counseling, training in management and cultivation of melons, good governance, advocacy and marketing of melon processed product. The result of the implementation of this method showed the increase of farmer’s knowledge about good quality melon, farm management and procedures for the proper cultivation of melons in creating diversificated processed melon to improve the welfare of villagers Jamusan.It could be revealed that LANTPYCATION Program is a new method on sustainable agriculture of melon cultivation in Indonesia. Abstract

ABSTRACTBartek is one of local melon varieties which mainly cultivated in Pemalang, Central Java. Bartek has three shapes of fruit variation; Long-Green, Ellipse-Green, and Yellow. Chromosome characterization of the Bartek was investigated to determine the genetic variation. The main purpose of this research was to determine the genetic characters of Bartek including chromosome number, mitosis, cell cycle, and karyotype. Squash method was used for chromosome preparation. The results showed that all of Bartek observed in this study have similar diploid (2n) chromosome number = 24. According to total number of chromosome, Bartek is more related to melon. The mitotic analysis exhibited that the Bartek has same Karyotype formula, 2n = 2x = 24m. According to the R value of the three kind of Bartek (R< 0.27), it has indicated that three kind of Bartek were considered to be originated from same species and one of melon varieties (Cucumis melo L. var. Abstract

Melon plants (Cucumis melo L.) are susceptible to disease attacks primarily caused by viruses. One of the viruses often attacking melon plants is Cucumber mosaic virus (CMV). Previous studies have shown that melon callus produced from in vitro culture is detected to have CMV resistance gene (Creb-2). However, the trait of resistance to CMV in that melon (F2) is still segregated. To know the stability of the Creb-2 gene, further research is needed to detect the gene in the differentiated callus. This study aims to  callus induce and callus differentiate melon. Firstly, melon seeds were grown on ¼ MS medium with addition of 2 mg / L 2.4-Dichlorophenoxyacetate and 1 mg / L BAP (Benzyl Amino Purine) to grow callus. Secondly, the callus was differentiated in MS medium with the addition of 2,4-D , BAP and IAA in different doses. 0 mg / L; 0.01 mg / L 2.4-D and 0.1 mg / L BAP, and MS medium with 0.01 mg / L; 0.1 mg / L; 1 mg / L IAA (Iodole Acetic Acid) and 0.1 mg / L BAP. The results showed that callus was successfully differentiated into root and it grew optimally in MS medium with 0.01 mg / L 2.4-D: 0.1 mg / L BAP and 1 mg / L IAA: 0.1 mg / L BAP . Therefore the roots can be used as isolation samples and DNA amplification to detect stability of the Creb-2 gene.

Abstract

Lahan kritis berkapur, karst, memberikan cekaman abiotik pada tanaman karena hambatan hidrasi dan nutrisi. Asam absisat (ABA) adalah hormon yang diekspresikan tumbuhan pada kondisi cekaman abiotik. CmBG1 merupakan salah satu gen peregulasi hormon ABA pada melon yang akan terakumulasi saat tumbuhan mengalami cekaman. Tujuan penelitian ini adalah mendeskripsikan pengaruh lahan kritis karst terhadap ekspresi gen CmBG1 secara kualitatif dan kuantitatif melon hasil turunan kultivar TACAPA, yaitu kultivar PT dan AT yang ditanam pada medium tanah karst dari wilayah Agroekosistem II dan III Yogyakarta (Gunungsewu, Dlingo, maupun Sentolo). cDNA library diperoleh dari reverse transcription isolat RNA. cDNA diamplifi kasi dengan primer spesifi k, kemudian dispektrofotometri pada λ260 nm untuk mengetahui konsentrasi gen CmBG1. Ekspresi gen dianalisis dengan real time PCR dengan gen referensi Cm-actin. Uji kualitatif dilakukan dengan elektroforesis gel agarosa 1,5%. Hasil penelitian menunjukkan gen CmBG1 terdeteksi dengan ukuran ±1258 bp pada kultivar PT dan AT. Konsentrasi gen CmBG1 melalui spektrofotometri menunjukkan semua kultivar yang ditanam pada media kontrol memiliki konsentrasi yang lebih rendah bila dibandingkan media tanam dengan perlakuan lahan kritis baik Gunungsewu, Dlingo, maupun Sentolo. Hasil ini sama dengan uji ekspresi gen CmBG1 menggunakan analisis kuantitatif real time PCR. Karsts critical land gives abiotic stresses in plants because of hydration and nutrition disturbances. Abscisic acid (ABA) is a hormone that expressed in the plant in abiotic stress conditions. CmBG1 is one of the regulatory genes encoding hormone ABA in melon plants which is accumulated in stress condition. The purpose of this study was to describe the infl uence of karsts critical land on the genes expression of CmBG1 melon cultivars PT and AT qualitatively and quantitatively. The plants were grown in medium karst land of Agroecosystems II and III of Yogyakarta (Gunungsewu, Dlingo, and Sentolo). Total RNA was extracted from leaf tissue then Reversed Transcriptase (RT-PCR) to collect cDNA library. cDNA was amplifi ed using specifi c primer. Spectrophotometry was conducted in λ260 nm and electrophoresis run in 1.5% agarose gel. Control band and reference gene chosen in Real Time PCR was Cm-Actin. CmBGI band (± 1258 bp) was showed both on PT and AT. Cm-actin was showed band of DNA as ± 445 bp. CmBGI gene concentration in critical land medium treatment which is given greater stress on melons are higher than normal condition. This suggests that the CmBGI gene is expressed more in cultivar PT and AT melons when they are grown under stress condition. This result show similarly when using real time PCR.

Abstract

Tanaman melon (Cucumis melo  L.) merupakan tanaman pertanian yang banyak dibudidayakan untuk konsumsi buahnya. Salah satu permasalahan dalam budidaya melon adalah benih yang diperoleh secara impor sehingga harganya relatif mahal. Perakitan kultivar MelodiGama 3 (MG3) merupakan strategi untuk mengurangi ketergantungan terhadap benih impor. Kultivar MG3 merupakan hasil persilangan antara kultivar ♀ MG1 dengan ♂ La-3 (tahun 2011). Tujuan penelitian ini untuk mengetahui kemampuan adaptasi kultivar MG3 terhadap kondisi lingkungan pada musim kemarau (tahun 2012) dan musim penghujan (tahun 2013) dan mengamati kestabilan kultivar MG3 di kedua musim tersebut di demplot milik Kebun Pendidikan Penelitian dan Pengembangan Pertanian (KP4) Universitas Gadjah Mada (UGM), Yogyakarta. Hasil yang diperoleh menunjukkan bahwa karakter melon MG3 dari penanaman musim penghujan umumnya lebih unggul daripada penanaman musim kemarau. Melon MG3 penanaman musim penghujan unggul pada karakter berat buah, keliling buah, ukuran buah, dan berat biji. Namun terdapat penurunan karakter unggul yaitu tingkat kemanisan (brix) dari 11,53% menjadi 9,98%. Hasil tersebut terjadi karena musim penghujan yang mempengaruhi kadar air dalam buah sehingga tingkat kemanisanya menurun. Kata kunci: MG3, Karakter Fenotip, Pemuliaan Tanaman
