Abstract

ABSTRAK Penelitian ini adalah penelitian tindakan kelas. Terdiri beberapa aspekperlakuan dan pengamatan utama yaitu peningkatan kraetivitas anak. Permasalahan dalam penelitian ini adalah apakah kreativitas anak dapat ditingkatkan melalui kegiatan menggambar di kelompok B TK PGRI Taripa? Dan tujuan penelitian ini adalah untuk meningkatkan kraetivitas anak melalui kegiatan menggambar.Penelitian dilaksanakan di TK PGRI Taripa, melibatkan 15orang anak terdiri atas 8 orang anak laki-laki dan 7 orang anak perempuanyang terdaftar pada tahun ajaran 2012/2013. Penelitian ini menggunakan desain penelitian Kemmis dan Mc. Taggart yang terdiri atas dua siklus. Di mana pada setiap siklus dilaksanakan satu kali pertemuan di kelas dan setiap siklus terdiri empat tahap yaitu perencanaan, pelaksanaan, observasi, dan refleksi. Data yang dikumpulkan melalui teknik obsevasi, wawancara, dan pemberian tugas kemudian dianalisis secara deskriptif dari data kualitatif dan kuantitatif. Data yang dikumpulkan sebelum tindakan pengamatan kreativitas anak dalam menggambar di kelas kategori Berkembang Berkembang sangat baik6,66%, Berkembang Sesuai Harapan 13,33%, Mulai Berkembang 26,66%, dan Belum Berkembang 53,33%, kemudian kreativitas anak mewarnai gambar dengan kategori Berkembang Berkembang sangat baik 0%, Berkembang Sesuai Harapan 13,33%, Mulai Berkembang 20%, Belum Berkembang 66,66%, dan pengamatan anak memberi judul gambar dengan kategori Berkembang Berkembang sangat baik6,66%, Berkembang Sesuai Harapan 6,66%, Mulai Berkembang 13,33%, Belum Berkembang 73,33%. Setelah dilakukan tindakan maka hasil penelitian ini dapat disimpulkan bahwa melalui kegiatan menggambar dapat meningkatkan kraetivitas anak, terbukti ada peningkatan kraetivitas dari siklus I ke siklus II yang diamati dalam menggambar kategori berkembang sangat baik dan baik dari 53,33% menjadi 79,99% (26,66%), kraetivitas dalam mewarnai gambar kategori berkembang sangat baik dan baik dari 46,66% menjadi 80% (33,34%), kemudian yang terakhir pengamatan anak dalam memberi judul gambar kategori berkembang sangat baik dan baik dari 53,33% menjadi 86,66% (33,33%). Secara umum terjadi peningkatan dari semua kemampuan yang diukur. Sehingga dapat disimpulkan bahwa pembelajaran dengan menerapkan kegiatan menggambardapat meningkatkan kreativitas anak di TK PGRI Taripa. Kata Kunci : Kreativitas,Kegiatan Menggambar

Abstract

Abstract: This Study is aimed to find out: (1) the influence of time budget pressure and dysfunctional behavior to audit quality simultaneously, and (2) the influence of time budget pressure and dysfunctional behavior to audit quality partially. Population in this study are all auditors of Offices of BAWASDA in Gorontalo Province. The study has used a census method, whereas its data collection techniques were done through spreading and filling questionnaires and direct interviews. Hypotheses in this study have been tested using a path analysis. The results of the study have shown that time budget pressure and dysfunctional behavior have simultaneously and partially affected the audit quality.

Abstract

ABSTRACTBartek is one of local melon varieties which mainly cultivated in Pemalang, Central Java. Bartek has three shapes of fruit variation; Long-Green, Ellipse-Green, and Yellow. Chromosome characterization of the Bartek was investigated to determine the genetic variation. The main purpose of this research was to determine the genetic characters of Bartek including chromosome number, mitosis, cell cycle, and karyotype. Squash method was used for chromosome preparation. The results showed that all of Bartek observed in this study have similar diploid (2n) chromosome number = 24. According to total number of chromosome, Bartek is more related to melon. The mitotic analysis exhibited that the Bartek has same Karyotype formula, 2n = 2x = 24m. According to the R value of the three kind of Bartek (R< 0.27), it has indicated that three kind of Bartek were considered to be originated from same species and one of melon varieties (Cucumis melo L. var. Abstract

Dioscorea spp. were cultivated as alternative food in the garden and yard on dry season, and it’s planted by peoples in the villages. The objectives of the research are to identify the species, the tuber use, and the local wisdom to support the conservation of Dioscorea around Wonosadi forest. Samples and data collection was conducted in 15 villages around Wonosadi forest. Interview survey was done on peoples who are cultivate yam species (Dioscorea spp.) in their garden. Research were emphasized to tuber characteristic, traditional uses, and local wisdom to supporting yam conservation. Data was analyzed descriptively to illustrate the kind of local usage and local wisdom of connect with Dioscorea spp. The results showed that there were found species of gembili (D. esculenta (Lour.) Burk.), uwi (D. alata L.), and gadung (D. hispida dennst.) as cultivated plants, and tomboreso (D. pentaphylla L.), gembolo (D. bulbifera L.), and jebubuk (D. numularia Lamk.) as wild species in the forest. Tuber is raw material for many kind of food based on carbohydrate content. Gadung were used as fickle food with high diversification and insecticide for rice and corn. Peoples more consume gembili and uwi than gembolo and others species as rice substitute at dry season. The local wisdom were indicated that peoples still consume gembili and uwi as rice substitute, while tuber also used as sesaji in traditional concept jajan pasar or tukon pasar, and tuber of uwi was used as diet for diabetes complaint, and row material of uwi ungu tuber was used effectively for diarrhea complaint in the villages.

Abstract

Tanaman melon (Cucumis melo  L.) merupakan tanaman pertanian yang banyak dibudidayakan untuk konsumsi buahnya. Salah satu permasalahan dalam budidaya melon adalah benih yang diperoleh secara impor sehingga harganya relatif mahal. Perakitan kultivar MelodiGama 3 (MG3) merupakan strategi untuk mengurangi ketergantungan terhadap benih impor. Kultivar MG3 merupakan hasil persilangan antara kultivar ? MG1 dengan ? La-3 (tahun 2011). Tujuan penelitian ini untuk mengetahui kemampuan adaptasi kultivar MG3 terhadap kondisi lingkungan pada musim kemarau (tahun 2012) dan musim penghujan (tahun 2013) dan mengamati kestabilan kultivar MG3 di kedua musim tersebut di demplot milik Kebun Pendidikan Penelitian dan Pengembangan Pertanian (KP4) Universitas Gadjah Mada (UGM), Yogyakarta. Hasil yang diperoleh menunjukkan bahwa karakter melon MG3 dari penanaman musim penghujan umumnya lebih unggul daripada penanaman musim kemarau. Melon MG3 penanaman musim penghujan unggul pada karakter berat buah, keliling buah, ukuran buah, dan berat biji. Namun terdapat penurunan karakter unggul yaitu tingkat kemanisan (brix) dari 11,53% menjadi 9,98%. Hasil tersebut terjadi karena musim penghujan yang mempengaruhi kadar air dalam buah sehingga tingkat kemanisanya menurun. Kata kunci: MG3, Karakter Fenotip, Pemuliaan Tanaman

Abstract

Coastal area around sand dune on the Bocor Beach potentially used as agricultural land especially melon cultivation. It was located at Buluspesantren Subdistric , Kebumen Distric, Central Java. Cultivation techniques in that land were not green cultivation .The use of pesticides, non-organic material, and plant cutting in the area of sand dune have a potential to damage the environment. This event was aimed to introduce the technique of green melon cultivation so that melon cultivation around sand dune can develop and sustainable. The technique of melon cultivation used in this activity was production of melon superior seed and fruit selection. Melodi Gama3 (MG3) was one of the superior seed which used in this activity. Superior seed was able to reduce the use of pesticides and fruit selection can produce fruit in a high quality. Then the group of farmers trained to use local potential as a substitute for a common material used by other farmers. Palm midrib instead of bamboo, pandanus fiber as a rope replacement, and some other potential materials can be used. Cultivation techniques are transfered to farmers group by training and farmers models. The results of economic analysis indicates that the green melon cultivation techniques are able to increase the income of farmers in the area melon around sand dune. After this event, the farmers was expected to develop environmentally welfare farming techniques in the area around sand dune in Bocor Beach. Keywords:     Sustainable development, melon cultivation, sand dune

Abstract

ABSTRACTSpecies Identification of Scarlet gourd (Coccinia grandis (L.) Voigt) in Three Population inYogyakarta. Papasan is a dioecious plant belongs to the family Cucurbitaceae. This plant iscommonly used as vegetable, anti diabetic, anti bacterial, and anti diarrhea. In Daerah IstimewaYogyakarta, there were two varians of Papasan (Papasan I and II), found in three population(Ngebel, Berbah and Gajah Wong Riverbank). They differ in phenotype, especially in shapeand taste of fruit. Genotype observation using squash method on the root tips with modificationin the duration of maceration were used in this research indicated that cells devided of PapasanI at about 8-11.30 a.m, while Papasan II at about 08.30-09.30 a.m, 11 a.m-00.30 p.m and 2-2.30p.m. The chromosome number of both Papasan is 2n=24, contains of 22 autosomes and 2 sexchromosomes. The karyotype formulas of Papasan I and II were 2n=24=20m+2sm+XX(m).Based on the statistic test, significant difference on chromosomes character between PapasanI and II was only in short arm of autosome pair number 5. The difference R value betweenPapasan I and II was smaller than 0.25. It revealed that the both of Papasan is closely relatedand belongs to the same species of Coccinia grandis L. Abstract

Powdery mildew is one of the diseases caused by fungal infections that can reduce the production of melon fruit worldwide including in Indonesia. A powdery mildew-resistant cultivar of melon is needed to increase melon yield crops. This study aimed to detect resistance gene linked to powdery mildew using a sequence characterized amplified region (SCAR) markers. The melon cultivar Meloni was used in this study. SL-3, PI 371795, and Aramis cultivar were used to compare. Amplification of the marker was performed employing a pair of primers. The result showed that Meloni had a powdery mildew resistance gene by the presence of a DNA target band at 1058 base pair (bp). Based on this result, it could be concluded that Meloni was an excellent melon cultivar because of its ability to overcome the powdery mildew infections naturally. SCAR markers have been used for various purposes, especially to detect resistance genes to plant diseases. The present study had provided information for plant breeders about Meloni as the new melon cultivar that was genetically resistant against powdery mildew infections. Furthermore, Meloni could be proposed as an alternative to native Indonesian superior melon seeds.

Abstract

Lahan kritis berkapur, karst, memberikan cekaman abiotik pada tanaman karena hambatan hidrasi dan nutrisi. Asam absisat (ABA) adalah hormon yang diekspresikan tumbuhan pada kondisi cekaman abiotik. CmBG1 merupakan salah satu gen peregulasi hormon ABA pada melon yang akan terakumulasi saat tumbuhan mengalami cekaman. Tujuan penelitian ini adalah mendeskripsikan pengaruh lahan kritis karst terhadap ekspresi gen CmBG1 secara kualitatif dan kuantitatif melon hasil turunan kultivar TACAPA, yaitu kultivar PT dan AT yang ditanam pada medium tanah karst dari wilayah Agroekosistem II dan III Yogyakarta (Gunungsewu, Dlingo, maupun Sentolo). cDNA library diperoleh dari reverse transcription isolat RNA. cDNA diamplifi kasi dengan primer spesifi k, kemudian dispektrofotometri pada ?260 nm untuk mengetahui konsentrasi gen CmBG1. Ekspresi gen dianalisis dengan real time PCR dengan gen referensi Cm-actin. Uji kualitatif dilakukan dengan elektroforesis gel agarosa 1,5%. Hasil penelitian menunjukkan gen CmBG1 terdeteksi dengan ukuran ±1258 bp pada kultivar PT dan AT. Konsentrasi gen CmBG1 melalui spektrofotometri menunjukkan semua kultivar yang ditanam pada media kontrol memiliki konsentrasi yang lebih rendah bila dibandingkan media tanam dengan perlakuan lahan kritis baik Gunungsewu, Dlingo, maupun Sentolo. Hasil ini sama dengan uji ekspresi gen CmBG1 menggunakan analisis kuantitatif real time PCR. Karsts critical land gives abiotic stresses in plants because of hydration and nutrition disturbances. Abscisic acid (ABA) is a hormone that expressed in the plant in abiotic stress conditions. CmBG1 is one of the regulatory genes encoding hormone ABA in melon plants which is accumulated in stress condition. The purpose of this study was to describe the infl uence of karsts critical land on the genes expression of CmBG1 melon cultivars PT and AT qualitatively and quantitatively. The plants were grown in medium karst land of Agroecosystems II and III of Yogyakarta (Gunungsewu, Dlingo, and Sentolo). Total RNA was extracted from leaf tissue then Reversed Transcriptase (RT-PCR) to collect cDNA library. cDNA was amplifi ed using specifi c primer. Spectrophotometry was conducted in ?260 nm and electrophoresis run in 1.5% agarose gel. Control band and reference gene chosen in Real Time PCR was Cm-Actin. CmBGI band (± 1258 bp) was showed both on PT and AT. Cm-actin was showed band of DNA as ± 445 bp. CmBGI gene concentration in critical land medium treatment which is given greater stress on melons are higher than normal condition. This suggests that the CmBGI gene is expressed more in cultivar PT and AT melons when they are grown under stress condition. This result show similarly when using real time PCR.

Abstract

Melon (Cucumis melo L.) is one of horticultural commodities with good economic prospect for melon farmers because of several advantages. 'Meloni' is result of crossing between ‘Sun Lady-3’ and ‘PI 371795’ by Genetics and Breeding Laboratory, Faculty of Biology, UGM. The 'Meloni' plant produces females flowers that expressed by CmACS-7 gene and it can be expected that melon plants 'Meloni' included in the primitive cultivars, while 'Meloni' is a cultivation cultivar. The aim of this research is to study the relation of 'Meloni' kinship to primitive and modern cultivars. Characterization of the CmACS-7 gene is begun by a total isolation process of RNA from a sample of melon plant flowers and detection using a specific primer. The cDNA bands obtained are sequenced and then phylogenetic analysis is performed based on CmACS-7 gene expression using MEGA 7.0. Result of this study show that the presence of the CmACS-7 gene in female and hermaphrodite flowers of ‘Meloni’, ‘PI 371795’, ‘Sun Lady-3’, and ‘Hikapel’. Phylogenetic tree analyze show that ‘Meloni’ from female plant, ‘PI 371795’ and ‘Sun Lady-3’ have closely relation, while 'Hikapel' separated from other cultivars.
