Plant have been cultivated for a long time, they have been selected and bred by our ancestors for their nutritional qualities, flavor, productivity, storage ability and other valued traits. Crop Wild Relative (CWR) are often neglected because they have not been used for domestication. However, world population growth together with the potentially adverse impact of climate change on agricultural production, calls for greater action to ensure global food security. Indonesian Botanic Garden already ratified Convention on Biodiversity (CBD). One of the programs is the Global Strategy for Plant Conservation (GSPC). The GSPC vision is to stop the plant diversity loss through five objectives and 16 targets for plant conservation to be achieved by 2020. The study was to identify the implementation of target 9 of GSPC by Indonesian Botanic Garden Network. The result showed that there are four reason Indonesian Botanic Garden has participated in target 9 of GSPC, We also provided success stories from other botanic garden to encourage Indonesian Botanic Garden to conserve more Indonesian Crop Wild Relatives (CWR) whilst facilitating their use in crop improvement.

Pangalengan is one of the districts in Bandung Regency which is an agricultural center. One of the potential products in the Pangalengan area is Pangalengan coffee which is well known to foreign countries. However, not all of the village areas in Pangalengan coffee farming have not been made a top priority, so there is still a need for coaching and mentoring as the PPM team of this group is serving together with the PPM team of the Faculty of Social and Political Sciences (FISIP) Unpas in February 2019 in Warnasari Village, Pangalengan District Bandung. The problems identified in the partners are human resources, raw materials, production, products, and marketing. The target or partners in this activity are coffee farmers in the Warnasari Village, Pangalengan. The method of activities includes social mapping related to the identification of problems faced by coffee farmers and FGDs with entrepreneurship material. The results of this period PPM devotion shows a change in the mindset of coffee farmers to turn coffee into a priority or mainstay of the region's commodities, in addition to the awareness related to the increasing spirit of entrepreneurship desiring to process coffee yields with variant forms in the future. The changes in antioxidant compounds and antioxidant activities of melon (Cucumis Melo L.) cultivar Hikapel during postharvest storage at room temperature were evaluated. Melon with three ripening stages (27 DAA, 29 DAA, and 32 DAA) were harvested and stored at 25oC for 20 days. Melon cv. Hikapel were evaluated for their antioxidant compounds such as ascorbic acid, total phenolic (TPC), and total flavonoid content (TFC). Antioxidant capacity was also evaluated using DPPH radical scavenging assay (DPPH-RSA) and ferric reducing power assay (FRPA). The result showed that there were different levels of antioxidant compounds (TPC, TFC, and AAC) and antioxidant activities (DPPH-RSA & FRPA) in different ripening stages of this melon. Antioxidant compounds and antioxidant activity decreased during postharvest storage. In conclusion, melon cv. Hikapel provides various natural antioxidant compounds such as phenolic, flavonoid which can be the main contributors to the overall antioxidant activity of melon cv. Hikapel and its antioxidant properties were influenced by the postharvest storage period.

Latar Belakang: Fasilitas Pelayanan Kesehatan khususnya di Puskesmas merupakan salah satu tempat kerja yang mempunyai risiko terhadap keselamatan dan kesehatan kerja bagi SDM, Pasien, dan Pengunjung. Berdasarkan regulasi “PMK No 52/2018”, pelaksanaan dan pelaporan kegiatan K3 sangat penting untuk menjaga kesejahteraan SDM, pasien dan pengunjung tersebut agar tetap sehat dan produktif. Saat ini sistem pelaporan masih bersifat manual dan lambat dalam mengumpulkan laporan sehingga dibutuhkan sistem pelaporan yang lebih efektif dan efisien yaitu sistem pelaporan berbasis web pada pelaksanaan kegiatan K3 di Puskesmas.Tujuan: Penelitian ini bertujuan untuk mengevaluasi perancangan sistem pelaporan berbasis web pada pelaksanaan kegiatan K3 di Puskesmas.Metode: Jenis penelitian yang digunakan merupakan research and development. Tahapan penelitian terdiri dari analisis kebutuhan dengan wawancara mendalam serta Focus Group Discussion (FGD) yang dihadiri oleh Kepala Puskesmas, Pimpinan serta Regu K3 Puskesmas Tegalrejo Kota Yogyakarta. Analisis kebutuhan selanjutnya digunakan untuk pedoman dalam perancangan sistem pelaporan berbasis website pada penerapan K3 di puskesmasHasil: Sistem pelaporan berbasis web pada pelaksanaan kegiatan K3 di Puskesmas dirancang sesuai dengan hasil analisis kebutuhan. 60% responden menyatakan puas dengan adanya sistem pelaporan berbasis web pada pelaksanaan kegiatan K3 di Puskesmas.Kesimpulan: Pencatatan dan pelaporan pelaksanaan kegiatan K3 di Puskesmas menjadi lebih efektif dan efisien setelah adanya sistem pelaporan berbasis web.Kata kunci: Keselamatan dan Kesehatan Kerja, Puskesmas, Sistem Pelaporan Berbasis Web

The number of published reports of flower buds, anthers and on microspore culture of horticultural plants, especially in soybean is very low. The purpose of this research is to determine soybean microspores development stage based on the length of flower buds. Five local cultivars such as Argomulyo, Grobogan,Wilis, Anjasmoro and the black soybean Malika were used. Flower buds were the main material for microspore culture. Anjasmoro cultivar produces the highest number of flower bud. Anjasmoro soybean cultivar at third stage development, 3.1-3.6 mm length, was selected for microspore culture, based on plant height, length of anther, total of microspore per bud. Anjasmoro cultivar has the most late-uninucleate microspore and the total of shed of Anjasmoro microspore in the B medium with 340C for 4 day is 3820. Results revealed that Anjasmoro soybean cultivars had 354.67±59.67 μm length of anther, 2003±216 microspore per bud and the most mid and late-uninucleat microspore on 2.6-3.6 mm length flower bud. Anjasmoro soybean cultivar can be used for microspore culture.

Begonia is very popular as a foliage plant and some begonia species grow up in pots. Begonia serratipetala and Begonia muricata are Indonesian begonia species that are well suited as indoor potted plants because of their leaf color and shape. In addition, the Bali Botanical Garden's landscape nursery would like to use these begonias for decoration in all buildings. However, it still needs to be studied how long the shelf life of the two begonias is in indoor environments in the highlands without air conditioning (AC). The aim of this study was to investigate the indoor shelf life of B. serratipetala and B. muricata. This research method consists of two steps. The first step was the acclimation phase and the second step was the indoor placement phase. Each begonia was repeated three times. Parameters studied included plant growth (plant height, leaves, leaf width, and leaf length) and environmental factors (sunlight intensity, air temperature, and humidity). The data were analyzed descriptively. Correlation analysis was used to determine the relationship between growth parameters and environmental factors. The result showed that both begonias (B. muricata and B. serratipetala) have similar persistence to indoor begonia species within two weeks, the ideal time for plant replacement

Lahan kritis berkapur, karst, memberikan cekaman abiotik pada tanaman karena hambatan hidrasi dan nutrisi. Asam absisat (ABA) adalah hormon yang diekspresikan tumbuhan pada kondisi cekaman abiotik. CmBG1 merupakan salah satu gen peregulasi hormon ABA pada melon yang akan terakumulasi saat tumbuhan mengalami cekaman. Tujuan penelitian ini adalah mendeskripsikan pengaruh lahan kritis karst terhadap ekspresi gen CmBG1 secara kualitatif dan kuantitatif melon hasil turunan kultivar TACAPA, yaitu kultivar PT dan AT yang ditanam pada medium tanah karst dari wilayah Agroekosistem II dan III Yogyakarta (Gunungsewu, Dlingo, maupun Sentolo). cDNA library diperoleh dari reverse transcription isolat RNA. cDNA diamplifi kasi dengan primer spesifi k, kemudian dispektrofotometri pada λ260 nm untuk mengetahui konsentrasi gen CmBG1. Ekspresi gen dianalisis dengan real time PCR dengan gen referensi Cm-actin. Uji kualitatif dilakukan dengan elektroforesis gel agarosa 1,5%. Hasil penelitian menunjukkan gen CmBG1 terdeteksi dengan ukuran ±1258 bp pada kultivar PT dan AT. Konsentrasi gen CmBG1 melalui spektrofotometri menunjukkan semua kultivar yang ditanam pada media kontrol memiliki konsentrasi yang lebih rendah bila dibandingkan media tanam dengan perlakuan lahan kritis baik Gunungsewu, Dlingo, maupun Sentolo. Hasil ini sama dengan uji ekspresi gen CmBG1 menggunakan analisis kuantitatif real time PCR. Karsts critical land gives abiotic stresses in plants because of hydration and nutrition disturbances. Abscisic acid (ABA) is a hormone that expressed in the plant in abiotic stress conditions. CmBG1 is one of the regulatory genes encoding hormone ABA in melon plants which is accumulated in stress condition. The purpose of this study was to describe the infl uence of karsts critical land on the genes expression of CmBG1 melon cultivars PT and AT qualitatively and quantitatively. The plants were grown in medium karst land of Agroecosystems II and III of Yogyakarta (Gunungsewu, Dlingo, and Sentolo). Total RNA was extracted from leaf tissue then Reversed Transcriptase (RT-PCR) to collect cDNA library. cDNA was amplifi ed using specifi c primer. Spectrophotometry was conducted in λ260 nm and electrophoresis run in 1.5% agarose gel. Control band and reference gene chosen in Real Time PCR was Cm-Actin. CmBGI band (± 1258 bp) was showed both on PT and AT. Cm-actin was showed band of DNA as ± 445 bp. CmBGI gene concentration in critical land medium treatment which is given greater stress on melons are higher than normal condition. This suggests that the CmBGI gene is expressed more in cultivar PT and AT melons when they are grown under stress condition. This result show similarly when using real time PCR.

ABSTRACTBartek is one of local melon varieties which mainly cultivated in Pemalang, Central Java. Bartek has three shapes of fruit variation; Long-Green, Ellipse-Green, and Yellow. Chromosome characterization of the Bartek was investigated to determine the genetic variation. The main purpose of this research was to determine the genetic characters of Bartek including chromosome number, mitosis, cell cycle, and karyotype. Squash method was used for chromosome preparation. The results showed that all of Bartek observed in this study have similar diploid (2n) chromosome number = 24. According to total number of chromosome, Bartek is more related to melon. The mitotic analysis exhibited that the Bartek has same Karyotype formula, 2n = 2x = 24m. According to the R value of the three kind of Bartek (R< 0.27), it has indicated that three kind of Bartek were considered to be originated from same species and one of melon varieties (Cucumis melo L. var. The purpose of this study was to see how the effectiveness of project-based learning with a scientific approach was able to improve students' ability to write explanatory texts. This study uses a quasi-experimental method by using a research design in the form of Nonequivalent Control Group Design. The samples used were two classes, namely class XI MIPA 1 as an experimental class with 36 students receiving project-based learning treatment and class X MIPA 2 as a control class with 36 students receiving problem-based learning treatment. The results of the explanatory text writing test based on observations showed data that project-based learning was more effective in improving students' explanatory text writing skills, this was also reinforced by the data from observations that showed that 95% of students were more motivated to improve their explanatory text writing skills when using project-based learning

Usaha Sabun Herbal sebagai mitra dalam program kemitraan masyarakat (PKM) berada di wilayah Margacinta, Kelurahan Margasari, Kecamatan Buahbatu, Kota Bandung, dan memulai usaha sekitar tahun 2004. Usaha sabun herbal yang dikembangkan oleh Mitra 1 (Bydara) dan Mitra 2 (Marcin) adalah atas dasar kebutuhan para wanita dalam merawat kecantikan dengan bahan-bahan yang alami. Masalah utama mitra adalah belum berkembangnya usaha sesuai harapan pada kedua mitra. Kegiatan program kemitraan masyarakat ini dilaksanakan pada April sampai September 2018. Permasalahan yang dihadapi kedua Mitra meliputi: (1) terhambatnya proses legalitas izin usaha, (2) keterbatasan keahlian SDM di bidang desain produk dan kemasan, (3) terbatasnya kemampuan promosi online maupun offline untuk menjangkau pangsa pasar, (4) terbatasnya peralatan dan perlengkapan produksi. Kondisi ini menyebabkan usaha mitra tidak berkembang. Metode pelaksanaan yang dilakukan adalah pelatihan/workshop desain produk kemasan, dan pelatihan pemasaran/promosi secara online/offline, bimbingan teknis/fasilitasi proses legalitas izin usaha, fasilitasi tambahan bahan baku dan perlengkapan produksi lainnya. Hasil dari kegiatan ini adalah: Proses legalitas usaha sudah memasuki tahap akhir di BPPOM, kedua mitra memiliki tambahan keterampilan dalam membuat kemasan produk, mampu membuat akun e-commerce dan media sosial sebagai media promosi online. Kedua mitra terbantu setelah adanya fasilitasi tambahan bahan baku. Dengan adanya kegiatan PKM ini produktivitas usaha sabun herbal pun meningkat dalam tiga bulan terakhir.
