Abstract

Pesisir Pantai Bocor yang terletak di selatan Kabupaten Kebumen, merupakan salah satu lokasi yang berpotensi sebagai lahan pertanian. Sepanjang Pantai Bocor terbentang gumuk pasir yang berfungsi sebagai penghalang angin dan gelombang. Wilayah tersebut dimanfaatkan sebagai lokasi menanam melon yang menjadi komoditas unggulan daerah. Oleh sebab itu dikembangkan melon ramah lingkungan untuk menjaga kelestarian gumuk pasir. Upaya pengembangan melon ramah lingkungan termasuk dalam Education for Sustainable Development (ESD).  yang didanai UGM. Pogram ini bertujuan untuk pengembangan masyarakat Desa Setrojenar, Kabupaten Kebumen berbasis ESD. Kegiatan program ini antara lain sosialisasi tentang pelestarian gumuk pasir, pengenalan alternatif tanaman pangan yaitu umbi-umbian, penanaman tanaman Casuarina equisetifolia Linn. sebagai barier, dan pendampingan budidaya melon menggunakan potensi lokal oleh petani. Hasil kegiatan antara lain partisipasi masyarakat tinggi, gumuk pasir tidak rusak, tanaman barier dapat tumbuh baik dan petani dapat panen melon dengan kualitas baik. Kegiatan ini diharapkan dapat menjadi role model bagi masyarakat untuk terus mengembangkan budidaya melon dan tetap menjaga kelestarian gumuk pasir.

Abstract

Penge Pakraman village is a traditional village that has the potency to become ecotourism. The emergence of new tourist attractions affects land changes to support tourism activities. This causes the number of plant species in nature to decrease. Plant conservation by utilizing local wisdom is one effort to reduce the decrease of plant species number. The application of Tri Hita Karana and Tri Mandala in Balinese daily life able to support plant conservation activity. The objective of this research was to determine the role of Tri Mandala concept in plant conservation at Pakraman Penge home garden. Data collection methods are carried out through observation plant location with inventory number and name of plants in house sample. Plant use continues with study literature. The results of an inventory of plant diversity in home gardens of Pakraman Penge village recorded 70 species of plants from 16 houses sample. The plant habitus varied from herbs, shrubs until trees. Plant species in the home gardens have functions for ceremonies (51%), medicinal (24%), ornamental (17%), food (6%), and spices (2%). However, based on the location, most plant species were found in madya mandala and the lowest were found in utama mandala. We also found plants with conservation status consist of least concerned (15 species), vulnerable (Dracaena draco), near threatened (Cycas rumphii), endangered (Coffea arabica). Therefore, the application of Tri Mandala concept on Balinese home gardens supports plant conservation and gives economic benefit in individual level

Abstract

This research was conducted to observe the body weight gain and carcass quality of the hybrid chicken derived from the crossing between female F1 Kampung Super and male F1 Kampung-Broiler. The weekly weight gain for seven weeks and carcass quality, including breast yield, protein content, lipid content, moisture, and pH were measured with each group consists of six individuals. The chickens were slaughtered for meat quality measurements on the 7th week. The results showed that the hybrid chickens’ weight at the 7th week (888.22±139.63) was higher than layer (467.06±85.7) and pelung (436.39±42.33). The hybrid chicken’s breast yield (3.20%) was significantly higher than pelung’s (1.66%). These characteristics of hybrid chicken breast muscle lead to the conclusion that hybrid chicken is a potential alternative meat-type chicken with local chicken meat characteristics and has relatively fast growth.

Abstract

Indonesia is an agrarian country whose territory is partly used as an agricultural sector, has especially watermelon (Citrullus lanatus (Thunb.) Matsum & Nakai). At present, the development of watermelons in Indonesia still requires a variety of colors, superior watermelons need to be developed by making triploid orange watermelons. The new watermelon cultivar (F₁) is the result of crossing 'Jelita' (♀) and 'BallyBall' (♂) which are expected to provide a combination of characters that can produce large and sweet fruits. The purpose of this study was to identify ISSR markers that were associated with ploidy levels of horticultural crops, particularly watermelon. The watermelons were growns on the agricultural land of Kebondalem Hamlet, Madurejo Village, Prambanan District, D.I.Y. and analyzed at the Laboratory of Biotechnology PAU, UGM and Laboratory of Genetics and Breeding, Faculty of Biology UGM. Analysis of genetic variation was carried out using the PCR-ISSR method and the DNA bandwidth was calculated using Paint Apps, Microsoft Excel 2013, and Microsoft Word 2013. The results of crossing 'Jelita' and 'King Quality 'is sterile. The result of 'Jelita' with 'BallyBall' produces tillers until sterile harvest time. The results of the analysis of genetic variation using the PCR-ISSR method indicate that the ISSR BI, B3, B5, and CBTC 1 molecular markers cannot be used to distinguish ploidy from the tested watermelons.

Abstract

Citarum watershed is a region of approximately 6,610 km2 in West Java, Indonesia. Citarum watershed has been degraded through historical land use and vegetation clearing. Rehabilitation of Citarum watershed uses Indigofera tinctoria L. that has value as a source of natural blue dye and is considered suitable for the region. Species distribution modelling and Habitat suitability index (SDM/HSI) were undertaken for I. tinctoria. The occurrence and environment data (bioclimatic, topography, and soil type) were input to HSI. Results of the Indigofera tinctoria habitat suitability model in Citarum watershed are very good (0.9–1) for some parts of the Citarum watershed. The medium and high suitability areas were respectively 4.49% and 4.37% of the area were located in the lowlands (Bekasi Regency and Karawang Regency). Prediction based on climate modelling for 2050 and 2070 estimated that the medium-high suitability area of Indigofera tinctoria will be reduced relative to the present.

Abstract

Abstract - Soybean (Glycine max (L.) Merr) is one of the most important food commodity to fulfill the protein necessity in Indonesia. Although Indonesia has many prime soybean seeds, it cannot fulfill the whole need of soybean and always rely on soybean import. This problem can be solved by increasing the quality and productivity of prime soybean seed by polyploidization with colchicine. This research aimed to study ploidy level and phenotype characters of Anjasmoro soybean which was induced by colchicine. The phenotype characters in this research were stomata size, plant height, total pod in one plant, total seed in one plant, weight of 100 seeds, flowering time, and ripening time of soybean. The ploidy level was analyzed with flow cytometry methode. The data was analyzed with one way ANOVA and Duncan test in SPSS 22 program. The result of this research showed that Anjasmoro soybean did not successfully induced by colchicine using concentration 0.01%, 0.02%, 0.025%, 0.05%, 0.075%, 0.1%, 0.15%, 0.2%, and 0.25% with duration of treatment 6, 8, 10, 12, 16, 18, and 24 hours. The treatment with colchicine concentration 0.01% and 0.02% with duration of treatment 10 hours effected the increasing of stomata size, the plant height, and the weight of 100 seeds.Key words : Soybean, polyploidization, colchicine, phenotypeAbstrak - Kedelai (Glycine max (L.) Merr) merupakan salah satu komoditas pangan penting sebagai sumber protein nabati yang kebutuhannya selalu mengalami peningkatan di Indonesia. Meskipun saat ini Indonesia memiliki banyak varietas kedelai unggul, namun Indonesia masih belum mampu mencukupi kebutuhan kedelai nasional sehingga terus bergantung pada impor kedelai. Permasalahan ini dapat diatasi dengan meningkatkan kualitas dan produktivitas varietas kedelai unggul yang sudah ada melalui teknik poliploidisasi dengan kolkisin. Penelitian ini bertujuan untuk mengetahui derajat ploidi dan karakter fenotip pada kedelai Anjasmoro yang diinduksi dengan kolkisin. Karakter fenotip yang diamati adalah panjang dan lebar stomata, tinggi tanaman, jumlah polong per tanaman, jumlah biji per tanaman, berat 100 biji, umur berbunga, dan umur masak kedelai. Analisis derajat ploidi kedelai dilakukan dengan metode flow cytometry. Data yang diperoleh dianalisis dengan uji statistik one way ANOVA dan dilanjutkan dengan uji Duncan taraf 5% menggunakan program SPSS 22. Hasil penelitian menunjukkan bahwa kedelai Anjasmoro belum berhasil terinduksi poliploid dengan perlakuan kolkisin konsentrasi 0,01%, 0,02%, 0,025%, 0,05%, 0,075%, 0,1%, 0,15%, 0,2%, dan 0,25% dengan lama waktu perlakuan 6, 8, 10, 12, 16, 18, dan 24 jam. Perlakuan kolkisin konsentrasi 0,01% dan 0,02% dengan lama waktu perlakuan 10 jam mempengaruhi peningkatan ukuran stomata, tinggi tanaman, dan berat 100 biji kedelai Anjasmoro.Kata kunci : Kedelai, poliploidisasi, kolkisin, fenotip

Abstract

The cultivation of arrowroot (Maranta arundinacea L.) in Indonesia, particularly in Yogyakarta Province has a great potential to be developed. This study aims to determine the genetic variability and analyze the intraspecific relation of arrowroot using Inter Simple Sequence Repeats (ISSR) markers as basic informations for considerations in the character selection of arrowroot cultivation in Yogyakarta. Exploratory survey method was occured in Gunungkidul, Kulon Progo, Sleman, and Bantul Districts to collect cultivar accessions. Accessions were replanted in Sawitsari Research Station. DNA isolation from the leaves of 7 moths old accessions was done using CTAB buffer solution. The DNA fingerprinting analysis was carried out using the result of DNA amplification with 4 ISSR markers. The polymorphism data then used for phenetic analysis using UPGMA algorithm and Baroni-Urbani Busser similarity coefficient to form dendrogram. Total of 5 local cultivars were found, identified as ‘Sili’, ‘Sembowo’,’ Sugo’, ‘Kebo’, and ‘Teropng’, each showed distinct rhizome morphological characters. The ISSR-PCR analysis resulted high polymorphism with 68,17% polymorphism mean and mean of polymorphic band was 6,75. Dendrogram resulted 4 clusters in 80% similarity index. Cluster A, B.I., and B.II.b consisted of ‘Sili’, ‘Teropong’, and ‘Kebo’ cultivars, respectively, while the cluster B.II.a gathered ‘Sugo’and ‘Sembowo’ cultivars.

Abstract

Parfume is one of the cosmetic product and its use is consistent. High prices of cosmetic product cause dependence on imported product. Innovative research is needed to reduce dependence on imported product. Melon is one of the potential horticultural crops to be developed in plant breeding. Gama Melon Parfum cultivars has strong of fragnant so its potentially to be used as parfume material. Research on morphological characters to support of Gama Melon Parfum as superior cultivar. The research aims to determine the level of stability of phenotypic characters and aroma of melon Gama Melon Parfum cultivar. Research methods was cultivated Gama Melon Parfum in Kebun Pendidikan Penelitian dan Pengembangan Pertanian (KP4) UGM Yogyakarta. Furthermore, the phenotypic characters was observed consist of qualitative and quantitative characters. The methods of sampling used Randomize Complete Block Design (RCBD) and quantitative phenotypic characters were analyzed using software PKBT-STAT-2. The result of qualitative characters on Gama Melon Parfum cultivars has oblate shape, small size, yellow-orange skin of colour with 9-10 lobes and longitudinal, white of flesh colour, crunchy texture, bitter taste, zero score of nett (not netted), strong of fragnant, and has spesific qualitative characters called turbin. Quantitative characters on Gama Melon Parfume cultivars consist of average weight of 160 gram; circumference of 21,8 cm; horizontal diameter of 6,89 cm; vertical diameter of 6,44 cm; thick skin of 1 mm, thick flesh of 1,06 cm; the number of seed of 200-300; weight of 100 seed of 1,74 gram; harvest 63-65 day after cultivate; and 17 days of storage time.

Abstract

The study was conducted to determine the stability of MG3 cultivar characters in rainy season based on multilocation test. Used materials were MG3 melon cultivar originating from the Laboratory of Genetics, Faculty of Biology, UGM, and Action 434, Glamour Sakata, MAI, and LADIKA melon cultivars as commercial cultivars. Multilocation test was conducted in March – July 2013 (rainy season) in 3 provinces i.e., East Java, Daerah Istimewa Yogyakarta, and Central Java. Observation of phenotypic characters was conducted at the Laboratory of Genetics, Faculty of Biology, Universitas Gadjah Mada. Qualitative phenotypic characters data of cultivar MG3 and cultivars Action 434, Glamour Sakata, MAI, and LADIKA were compared qualitatively, while the quantitative characters data were analyzed by balanced ANOVA using PKBTSTAT 2.0 software. The results showed that fruit weight, fruit girth, skin thickness, and fruit vertical and horizontal diameter (longitudinal section) has a very significant difference by looking at the interaction of cultivar x locations, while flesh thickness has no significant difference. Qualitative characters were discussed according to its uniformity in three location. It was concluded that melon can also give yield in rainy season. MG3 was unstable for fruit weight and girth, skin thickness, and fruit vertical and horizontal diameter characters, while stable for flesh thickness character. MG3 having uniformity for fruit shape, unripe and ripe skin color, netting intensity, flesh color, flesh texture, and water content characters from three cultivating location, while for netting distribution, cavity shape, flesh flavor, and flesh aroma characters, it have not uniform yet.

Abstract

Plant have been cultivated for a long time, they have been selected and bred by our ancestors for their nutritional qualities, flavor, productivity, storage ability and other valued traits. Crop Wild Relative (CWR) are often neglected because they have not been used for domestication. However, world population growth together with the potentially adverse impact of climate change on agricultural production, calls for greater action to ensure global food security.  Indonesian Botanic Garden already ratified Convention on Biodiversity (CBD). One of the programs is the Global Strategy for Plant Conservation (GSPC). The GSPC vision is to stop the plant diversity loss through five objectives and 16 targets for plant conservation to be achieved by 2020. The study was to identify the implementation of target 9 of GSPC by Indonesian Botanic Garden Network. The result showed that there are four reason Indonesian Botanic Garden has participated in target 9 of GSPC, We also provided success stories from other botanic garden to encourage Indonesian Botanic Garden to conserve more Indonesian Crop Wild Relatives (CWR) whilst facilitating their use in crop improvement.
