Abstract

Escherichia coli O157:H7 with the ability to produce shiga-like toxin was isolated from beef, cattle,chicken, and human feces. Due to its importance to human health, it is necessary to identify the genesencoding the production of shiga-like toxin, stx1 and stx2 respectively to further understand the pathogenesis.Isolation of E. coli was done on Eosin Methylene Blue Agar (EMBA), followed by identification on SorbitolMacConkey Agar (SMAC), latex agglutination test, and H7 antiserum test, respectivelly. The existence ofgenes stx1 and stx2 in E. coli O157:H7 was confirmed molecularly using PCR method with specific primersLP 30/31 and LP 43/44, Stx2 (F)/Stx2 (R) respectively. Escherichia coli O157:H7 was isolated from 22 outof 344 samples (6,4%). Some isolates showed gene stx1 and stx2 was detected in two isolates as indicatedby a 384 bp band (stx1 gene), 584 bp and 1588 bp bands (stx2 gene) respectivelly. The results indicatedthat local isolates E. coli O157:H7 are potential as a zoonoses agent.

Abstract

The research on developing diagnostic method, vaccine, and drugs for coccidiosis has been focused onthe finding of the immunogenic molecule in Eimeria. The identification of this agent will need the antibodywhich can recognize the biomolecule in the antigen. Antibody that has been developed for this purposeshould be analyzed first, and one of the simple methods for analyzing this antibody is through dot blotanalysis. The objective of this research was to analyze the polyclonal antibody which developed from theoocyst, sporocyst, and sporozoite of  E. tenella using dot blot analysis. The antigen for this polyclonalantibody was made from each of the E. tenella stadium by sonication. Fifteen mice, divided into 3 groups,were then injected subcutaneously with each antigen. The sera from these mice were then collected, analyzedby using ELISA and then it will be used for the dot blot analysis. The research result showed that thepolyclonal antibody which has been developed in mice from each antigen can react with the antigen itself.From this result it can be concluded that the developing of this antibody is successful and it can be used forfurther research in immunoproteomic.

Abstract

Epidemiological studies of zoonotic agent Escherichia coli O157:H7 have been analyzed pheneticallyand or phylogenetically. In a phenetic classification, micoorganisms are arranged into groups (phena) onthe basis of high overall similarity using both phenotypic and genotypic methods without judgementaspect of its ancestry or evolutionary. Due to its importance to epidemiological aspect, the study of geneticvariation of isolates origin from some sources need to be conducted in order to trace the routes of infection.A total of 20 samples obtained from some sources i.e clinically human feces, non-clinically human feces,cattle feces, chicken feces, and beef feces were used in this study. The study was started by confirming allof the isolates using O157 latex agglutination test and H7 antiserum test, followed by genomic DNAanalysis by random amplification of polymorphic DNA /RAPD methods. RAPD results were analyzed using a simple matching coeficient (Ssm) and alogorhythm unweighted pair group method using arithmeticaverages (UPGMA) programe. Results showed there were range of genetic DNA from local isolates (75.1–99,6%) which was almost similar to ATCC 43894 control isolate. The highest similarity (99.6%) to ATCC43894 control was showed by SM-7(1) isolate obtained from cattle fecal and KL-68(1), isolate obtainedfrom clinically human fecal. In addition, KL-52(7) obtained from clinically human fecal had high similarity(99.6%) to MK-35 isolate obtained from chicken fecal. On the other hand, DS-21(4) and DS-16(2) isolatesthat were obtained from beef had high similarity (84.9%) to other isolates including ATCC 43894 controlisolate. The highest similarity of E. coli O157:H7 isolates that were obtained from cattle feces, beef, andchicken feces to human feces isolate indicated that there were both cattle and chicken were potentialreservoirs of the zoonotic agen which can be transmitted to human.

Abstract

Coccidiosis is one of the important diseases in poultry industry. In Indonesia the morbidity of thedisease is between 80 to 90%. A rapid and prompt diagnosis would be one of the essential steps ineradication and control of the disease. The objective of this study is to determine the protein profile ofsporozoite of Eimeria tenella isolated in Yogyakarta using sodium dodecyl sulphate polyacrilamide gelelectrophoresis (SDS-PAGE) protein analysis. Protein analysis was performed in 12% polyacrilamide geland further electrophoresis at 100 volts and over-staining with Coomasie brilliant blue. The resultsshowed that the sporozoite of E. tenella isolated in Yogyakarta contained five proteins with molecularweights of 15, 26, 32, 80, and 91 kDa, respectively.

Abstract

The germ plasm variability of Indonesian local chicken is well known and one species which known as“ayam Pelung” originated from West Java is famous for its meat. The present study aimed to observed thephenotype characters i.e. fur color and body weight in a crossbreed hybrids of Pelung and Cemani chickenas a potential meat type breed. Firstly, the breeding between Pelung (one male) and Cemani (4 Females)was performed in semi intensive cage. Secondly, experiment were undertaken using 4 groups of DOC (eachcontain 5 males DOC) : 1) DOC of broiler Cobb 500 vs broiler Cobb 500 ; 2) DOC of Pelung vs Pelung ; 3)DOC of Pelung vs Cemani ; and 4) DOC of Cemani vs Pelung. The animals were kept for 7 weeks (49 days)every week the animals body weight and fur color were recorded. The result showed that there weredifferences in the phenotype characters of between the different crossbreeds. The fur of 5 F1 (femaleCemani vs male Cemani) was black with average body weight 532 ± 39.294 g at week 7. Whilst the fur colorof majority (4/5) F1 (female Pelung vs male Cemani) was black with brown dots with average body weight570 ± 14.445 g at week 7. When Pelung was cross within the same species their F1 fur color was blendedbetween black, brown and whitish with average body weight 652 ± 33.846 g at week 7. It is concluded that,the parent stock in this case the Cemani play a major role in fur color character whereas there ware nodifferences in the body weight at the F1 between the two different parent stock (ie. Pelung, Cemani).

Abstract

Perbandingan metode yang berbeda pada perhitungan keragaman genetik dapat bermanfaat dalam program pemuliaan dan konservasi tanaman. Pada penelitian ini digunakan 16 jagung lokal yang dikoleksi dari pulau Madura. Sebanyak 57 sifat morfologi dan 10 primer RAPD digunakan untuk menilai hubungan kekerabatan berdasarkan karakter morfologi dan penanda RAPD, dan menentukan genotip jagung lokal Madura yang mempunyai sifat untuk dikembangkan dalam program pemuliaan dan konservasi tanaman. Hasil penelitian menunjukkan bahwa berdasarkan karakter morfologi dan penanda RAPD, 16 kultivar jagung lokal di pulau Madura diklasifikasikan dalam 2 grup. Rata-rata kemiripan morfologi (0,74) lebih tinggi dibanding kemiripan berdasarkan penanda RAPD (0,63). Koefisien kemiripan berdasarkan karakter morfologi adalah 0,510,91, sedangkan koefedien kemiripan berdasarkan penanda RAPD adalah 0,390,92. Lima kultivar (Tambin, Delima, Tambin-2, Krajekan dan Duko) mempunyai produksi tinggi, umur genjah dan hubungan kekerabatan agak jauh, sehingga digunakan untuk program pemuliaan dan pengembangan budidaya.

Abstract

Powdery mildews are plant diseases caused by fungi, including Sphaerotheca fuliginea Poll. They cause a decrease on leaves health. Plant resistance to Sphaerotheca fuliginea Poll. might have a relationship to leaf anatomical structure, especially to epidermis. Therefore, the aim of this research was to characterize epidermal leaf structure of resistant and susceptible cultivars. This study used two resistant cultivars (PI 371795 and PMAR), one susceptible cultivar (Action 434) and two other cultivars (Fresh 848 and Sakata 144) which are not known their characteristic to powdery mildews. The fifth leaves (counted from the cotyledon) were collected from melon plants which reached early generative development. Leaf epidermal slides were prepared semi permanently. The results showed that the morphological leaves of resistant cultivars have dendatus leaf edges, while the susceptible one has repandus leaf edges. Anatomically, the leaves of PI 371795 had upper epidermal cells with wavy edges, but not in the others. Both PI 371795 and PMAR 5 had irregular shape of lower epidermal cells with curved edges, while leaf epidermal cells from susceptible cultivar tended to have irregular polygonal upper and lower epidermal cells with straight edges and shorter non-glandular trichomes. PMAR 5 had long glandular trichomes. There was significant difference between cultivars in the average of stomata number, the average of stomata index and trichome number on the lower epidermal tissue, but not on the lower epidermal tissue. In the lower epidermis, PI 371 and PMAR 5 had lower average of stomata number and stomata index compared to Action 434. PMAR 5 had higher number of trichomes than susceptible cultivar (Action 434). Meanwhile, Fresh 848 cultivar tends to have similar characters to resistant ones in terms of leaf edges, lower epidermal shape, stomata number and index. Sakata 144 had similar characters to those the susceptible one in terms on leaf edges, lower epidermal cells shape, stomata number and index, but not in trichome number. It may be assumed that the first protection of melon against S. fuliginea Poll. is located on lower epidermal tissue.

Abstract

Pesisir Pantai Bocor yang terletak di selatan Kabupaten Kebumen, merupakan salah satu lokasi yang berpotensi sebagai lahan pertanian. Sepanjang Pantai Bocor terbentang gumuk pasir yang berfungsi sebagai penghalang angin dan gelombang. Wilayah tersebut dimanfaatkan sebagai lokasi menanam melon yang menjadi komoditas unggulan daerah. Oleh sebab itu dikembangkan melon ramah lingkungan untuk menjaga kelestarian gumuk pasir. Upaya pengembangan melon ramah lingkungan termasuk dalam Education for Sustainable Development (ESD).  yang didanai UGM. Pogram ini bertujuan untuk pengembangan masyarakat Desa Setrojenar, Kabupaten Kebumen berbasis ESD. Kegiatan program ini antara lain sosialisasi tentang pelestarian gumuk pasir, pengenalan alternatif tanaman pangan yaitu umbi-umbian, penanaman tanaman Casuarina equisetifolia Linn. sebagai barier, dan pendampingan budidaya melon menggunakan potensi lokal oleh petani. Hasil kegiatan antara lain partisipasi masyarakat tinggi, gumuk pasir tidak rusak, tanaman barier dapat tumbuh baik dan petani dapat panen melon dengan kualitas baik. Kegiatan ini diharapkan dapat menjadi role model bagi masyarakat untuk terus mengembangkan budidaya melon dan tetap menjaga kelestarian gumuk pasir.

Abstract

Penge Pakraman village is a traditional village that has the potency to become ecotourism. The emergence of new tourist attractions affects land changes to support tourism activities. This causes the number of plant species in nature to decrease. Plant conservation by utilizing local wisdom is one effort to reduce the decrease of plant species number. The application of Tri Hita Karana and Tri Mandala in Balinese daily life able to support plant conservation activity. The objective of this research was to determine the role of Tri Mandala concept in plant conservation at Pakraman Penge home garden. Data collection methods are carried out through observation plant location with inventory number and name of plants in house sample. Plant use continues with study literature. The results of an inventory of plant diversity in home gardens of Pakraman Penge village recorded 70 species of plants from 16 houses sample. The plant habitus varied from herbs, shrubs until trees. Plant species in the home gardens have functions for ceremonies (51%), medicinal (24%), ornamental (17%), food (6%), and spices (2%). However, based on the location, most plant species were found in madya mandala and the lowest were found in utama mandala. We also found plants with conservation status consist of least concerned (15 species), vulnerable (Dracaena draco), near threatened (Cycas rumphii), endangered (Coffea arabica). Therefore, the application of Tri Mandala concept on Balinese home gardens supports plant conservation and gives economic benefit in individual level

Abstract

This research was conducted to observe the body weight gain and carcass quality of the hybrid chicken derived from the crossing between female F1 Kampung Super and male F1 Kampung-Broiler. The weekly weight gain for seven weeks and carcass quality, including breast yield, protein content, lipid content, moisture, and pH were measured with each group consists of six individuals. The chickens were slaughtered for meat quality measurements on the 7th week. The results showed that the hybrid chickens’ weight at the 7th week (888.22±139.63) was higher than layer (467.06±85.7) and pelung (436.39±42.33). The hybrid chicken’s breast yield (3.20%) was significantly higher than pelung’s (1.66%). These characteristics of hybrid chicken breast muscle lead to the conclusion that hybrid chicken is a potential alternative meat-type chicken with local chicken meat characteristics and has relatively fast growth.
