Abstract

Melon (Cucumis melo L.) cv. Gama Melon Parfum (GMP) is the new cultivar from cross-breeding of Natsuno Omoide (NO3) female parent and Miyamauri (MR5) male with phenotypic characteristics that is prominent such as very strong pleasant aroma when ripe. The physiological characteristics will be changed which have an impact on the formation of volatile compounds during fruit development. Therefore, the profile of strong volatile aroma compounds during fruit development to is critical to be identified. The volatile compounds analysis was performed on fruits harvested at 7, 14, 21, 28, and 35 days after pollination and storage at -20°C. Gas chromatography-mass spectrometry (GC-MS) was used for volatile identification. The results showed some volatile compounds changed during fruit development consisted of 8 esters, 2 alcohols, 1 acid, 3 terpenoids, and 3 hydrocarbons. The profile of volatile compounds was dominated by esters, followed by alcohols and acid respectively. Interestingly, the characteristics of the volatile compounds can differentiate between the ripe stage and unripe stage using principal component analysis. The findings of this study can be used to improve the quality aroma of GMP.

Abstract

Pandemi COVID-19 yang terjadi membuat banyak masyarakat mulai merasa jenuh dan bosan karena melakukan aktivitas yang sama setiap harinya. Oleh sebab itu, masyarakat kini mulai mencari aktivitas atau hobi baru guna mengurangi rasa bosan dan menghindari stress seperti berkebun. Namun banyak masyarakat berkebun hanya sekedar mengikuti sebuah tren tanpa mengetahui dan mempelajari bagaimana cara berkebun yang benar. Tujuan dari peneli- tian ini adalah membuat aplikasi berbasis Android yang dapat mengedukasi pengguna dengan memberikan informasi mengenai tanaman hortikultura, serta memudahkan pengguna dalam mengingat dan mencatat jadwal berkebun, agar nantinya kegiatan berkebun ini dapat menjadi rutinitas atau kebiasaan yang positif. Aplikasi ini dibuat menggunakan bahasa pemrograman Dart, Flutter sebagai framework, Visual Studio Code sebagai text editor dan SQLite sebagai database. Metode yang digunakan dalam pengembangan aplikasi ini adalah metode System Development Life Cycle (SDLC) dengan model Waterfall. Berdasarkan hasil pengujian meng- gunakan metode Black Box Testing, aplikasi ini dapat berjalan dengan baik dengan tampilan yang sesuai. Dengan adanya aplikasi ini dapat menampilkan informasi tentang berkebun

Abstract

Lactobacillus has long been known to reconstruct the microbiome of the urogenital organs of healthy people. However, changes in lactobacillus abundance are known occurring in urogenital tract infections (UTI) patients. It is also known that there are Lactobacillus species that have specific genomic characteristics and nutritional requirements that cause them to turn into pathogens. Understanding population characteristics and taxa composition that reconstruct urine microbiome is very important for the diagnosis and treatment of urogenital infections. This review provides an update information of metagenomic sequencing to identify the urinary microbiome profile and the role of lactobacilli in healthy and UTI patients.

Abstract

Menoreh Kuning Durian (Durio zibethinus Murr) cultivar is originated from Dusun Promasan, Banjaroyo Village, Kalibawang Subdistrict, Kulonprogo Regency, Province of Yogyakarta, Indonesia. Durian seller usually claimed their durian to be Menoreh Kuning. Therefore, an accurate method is needed to identify that cultivar. One of the most accurate methods is DNA isolation as the first step. This research was aimed to compare the total DNA profile of Menoreh Kuning Durian using detergent and commercial kit (NucleonTM PhytopureTM Genomic DNA Extraction Kits) qualitatively and quantitatively. The method used in this research was DNA isolation method using detergent and commercial kit (NucleonTM PhytopureTM Genomic DNA Extraction Kits). DNA source came from leaves Menoreh Kuning Durian. DNA pellet then was dissolved in 50 µL TE, after that each of 5 µL sampel was electrophoresized in 1 % of agarose for 30 minutes. The result of eletcrophoresis then was stained with ethidium bromide and finally was observed under transluminator-UV. The results of both isolation methods were compared based on the total DNA profil after electrophoresis, total mass of DNA pellet, and the result of DNA Optical Density. The research result had shown that the DNA of Menoreh Kuning Durian isolated using detergent and commercial kit can be observed under transluminator-UV. The mass of DNA pellet isolated using detergent was 0,004 gram while using kit was 0,0004 gram. The Optical Density results for DNA isolated using detergent were 2,658 AU of Absorbance, Ratio of 1,057, Concentration of 6645,6 µ gr/mL, and Protein of 93,9 milligram/mL. As for the DNA isolated using commercial kit had shown the Optical Density results of 2,760 AU for Absorbance, Ratio of 1,339, concentration of 6900,6 µ gr/mL, and Protein of 54,8 milligram/mL. Based on the research result, can be concluded that the DNA of Menoreh Kuning Durian can be isolated using detergent and commercial kit. The DNA profile isolated using commercial kit had shown a better result than the DNA profile isolated using detergent.

Abstract

Genetic quality improvement in local chickens can be achieved through crossbreeding, which has enhanced productivity and egg quality, satisfying consumer demand. To improve the quality of livestock products, it is crucial to assess the egg quality of chickens. This study evaluated the quality of eggs produced by F2 Mahkota Arab chickens bred from crossbreeding with F1 Mahkota Arab chickens. The research comprised several stages: chicken rearing, tests for egg shape index calculation, yolk index, albumin index, Haugh unit, and eggshell colour observation. The obtained results were compared with those of F1 Mahkota Arab chicken, Arab chicken, and Layer chicken. According to the study's findings, the F2 Mahkota Arab had an egg with a standard egg shape of 70% as per the egg shape index. The F2 Mahkota Arab chicken had a mean yolk index of 0.37±0.00, a mean albumin index of 0.121±0.005, and a mean Haugh unit of 91.99±1.13. The shell colour of their eggs belongs to the orange-white group, and the egg quality, including egg shape index, albumin index and Haugh unit, is higher than other chicken groups. Furthermore, the shell colour of F2 Mahkota Arab chicken eggs is whiter than other chicken groups.

Abstract

Mercury accumulation in the aquatic environment can be highly harmful. The body takes mercury vapor through the lungs, then absorbs mercury metal through the digestive system, and then the blood carries the metal to the brain. Bioremediation is the process of breaking down or converting harmful compounds into non-toxic forms, which can be accomplished through phytoremediation or phycoremediation. The goal of this study was to examine the growth and anatomy of Euglena sp. after being cultured in the mercury-containing FWS-CW waste treatment system. The ability of Euglena sp. and Echinodorus palifolius to bioremediate mercury at different concentration as well as association and non-association treatments. This study was carried out in a bioreactor known as FSW-CW (Free Water Surface-Constructed Wetlands). Plant growth (plant height and number of leaves), chlorophyll content, diameter of root and petiole, metaxylem diameter of root, petiole, and leaves, cortical thickness of root and leaves, and petiole anatomy were all measured. Water temperature, pH, salinity, and light intensity were all measured as environmental parameters. Mercury treatment reduced Euglena density (183.5 cells. mL-1103 in control and 12.6 cells. mL-1103 in 100 ppm mercury treatment) and number of E. palifolius leaves, but not plant height and chlorophyll. Root and petiole diameters were affected by the mercury treatment, petiole diameter decreased unless the concentration was 100 ppm, whereas root diameter actually increased. The diameter of the root metaxylem increased, but the petioles and leaves, as well as the thickness of the root cortex, did not provide a significant response. The growth of E. palifolius was still optimal in the presence of Euglena in mercury-containing medium.

Abstract

Golden Kamper Chicken is a chicken from crossing between pelung and layer chickens that have good meat and egg productivity. Currently, Golden Kamper has reached its fifth generation (F5 Golden Kamper). Phenotype characters of F5 Golden Kamper should be studied to know the character inheritance from the parental, F4 Golden Kamper. This research aims to study the phenotype character of F5 Golden Kamper chickens and to determine the inbreeding coefficient value of F5 Golden Kamper. The research has been done by hybridising F4 Golden Kamper hens and roosters, raising DOC, egg collection, phenotypic data collection, and calculating the inbreeding coefficient value of F5 Golden Kamper chicken. The results showed that F5 Golden Kamper have white and yellow shank, golden-brown and black-brown feather, and single comb. The inbreeding coefficient value obtained is 0,375, shows that individual F5 Golden Kamper chickens have low genetic uniformity in a population.

Abstract

Arabica coffee supplies 60% of world coffee production because has a unique taste as superior quality beverage. Arabica coffee micropropagation can be conducted by somatic embryogenesis technique which produce clonal, fast dan uniform plant. The somatic embryogenesis (SE) process describes the integration of endogenous signals and gene reprogramming, which releases signals to initiate embryogenic processes. The use of endogenous auxin, either alone or in combination with other PGRs or stress, induces differential gene expression, which modifies the genetic program of somatic cells and regulates the transition to each stage during SE development. The RKD4 gene (RWP-RK DOMAIN-4) is a gene that plays a role in early initiation embryo formation and development. The characterization of RKD4 genes in C. arabica is still limited and under explored. The objective of this research is to explore the characteristics of RKD4 gene by comparing the difference and similarity of RKD4 gene in C. arabica and other cultivated plants. The method was initiate by identifying nucleotide sequences from the National Center for Biotechnology Information (NCBI) database. Furthermore, consists of analysis of nucletide alignment, alignment of amino acid sequences, protein analysis, protein motif functions discovery, analysis of phylogenetic tree, protein 2D and 3D-modelling and physiochemical properties. According to the analysis, there were 100 polymorphism points with a total number of mutations of 211 points. The phylogenetic tree show C. arabica L. has a very close relationship with grapes (Vitis vinivera) based on the RKD4 protein, gene structures and protein motifs. There are nine highly conserved motifs found in the protein alignment. C. arabica L. had more methyl jasmonate element responses than A. thaliana. The findings are useful to understand the intitiation of embryo formation mechanisms of C. arabica L and other cultivate plants during propagation through somatic embryogenesis in the long run.

Abstract

Melon plants are challenged with diseases caused by begomovirus. To understand the epidemiology, ecology, and evolution of this genus, information about its temporal and spatial patterns of infection in specific ecosystems is needed. This paper aimed to report begomovirus infections in melon plants cultivated in greenhouse ecosystems under local climactic conditions in the Indonesian lowlands. To achieve this goal, we determined the begomovirus infection rates, plant physiological characteristics, and whitefly number. The data obtained were then analyzed using analysis of variance (ANOVA) and Pearson’s correlation. We documented the symptoms of begomovirus infection in melon plants cultivated in Indonesian lowlands. We also documented begomovirus infections in melon plants, which increased from the first exposure to after the reproductive phase. In different artificial ecosystems, we explained differential patterns of begomovirus infections in melon plants. Finally, we captured the variation in infection conditions found among melon host genotypes.
