Abstract

Background: Beta thalassemia major is a genetic blood disorder inherited by parents thalassemia traits (carrier) characterized by abnormal hemoglobin formation. Abnormalities during the process of hemoglobin formation cause gene changes or mutations that cause symptoms of severe anemia which has a long-term clinical impact on patients. High performance liquid chromatography can be used to determine the description of abnormal hemoglobin analysis, thus suspected diagnosis of thalassemia β major can be established by the clinician as a definitive diagnosis to enhance a more comprehensive diagnosis. Primary theraphy of beta thalassemia major is blood transfusion to support the quality of life regularly throughout the patient's life, transfusion of patients requires measurements such as pre-transfusion hemoglobin levels. Objective: This study aims to determine the description of the results of the analysis of hemoglobin patients with suspected thalassemia beta major at RSUP Dr. M. Djamil Padang. Methods: This study used a cross sectional design. The sampling technique uses total sampling with a total sample of 19 people from 1-35 years old. Patient data was from the suspected diagnosis of medical records established by clinicians at Dr. M. Djamil Hospital Padang. This data was analyzed univariately. Results: The results of this study indicated is mostly female with an average age of 13,74 years (SD ±12.1). This study resulted in a low pretransfusion hemoglobin average of 6.20 g/dL. The results of hemoglobin analysis for beta thalassemia major with average HbA (48,1%), HbA2 (27,3%) and HbF (24,5%). Conclusion: The results of the examination hemoglobin analysis on the HPLC method had results that did not compatible predictions of the suspected diagnosis of beta thalassemia major from the clinician

Abstract

Introduction: Hand dermatitis is a common occupational skin disorder caused by repeated exposure to irritants or allergens. Workers in car and motorcycle wash services are particularly vulnerable due to their frequent contact with detergents and cleaning agents, as well as prolonged exposure to wet work. This study aimed to identify factors associated with hand dermatitis among workers in East Padang District. Methods: A quantitative analytical observational study with a cross-sectional design was conducted among 32 workers selected through simple random sampling. Data were collected through direct interviews using a validated questionnaire. Univariate and bivariate analyses were performed, with chi-square tests used to evaluate associations between hand dermatitis and variables including working period, duration of contact, age, atopic history, personal hygiene, and use of personal protective equipment (PPE). Results: Hand dermatitis was identified in 65.6% of respondents. Bivariate analysis revealed significant associations between duration of contact (p = 0.020), personal hygiene (p = 0.027), and PPE use (p < 0.000) and the incidence of hand dermatitis. No significant associations were found for working period (p = 0.798), age (p = 0.211), or atopic history (p = 0.637). Discussion: Prolonged exposure to irritants, inadequate hygiene practices, and non-use of PPE likely heighten skin barrier disruption, predisposing workers to dermatitis. Conversely, factors such as age and atopic predisposition may play a minor role in this occupational group. Conclusion: Duration of contact, personal hygiene, and the use of PPE are significantly associated with hand dermatitis among car and motorcycle wash workers. In contrast, working period, age, and atopic history show no meaningful relationship. Strengthening preventive practices, remarkably consistent PPE use, is essential.

Abstract

Stunting is a condition of growth failure in toddlers due to chronic malnutrition and chronic infections, including worm infections and giardiasis. Kampung Baru Nan XX Village, Lubuk Begalung District, Padang City, is one of the areas with a high stunting rate. Education related to the prevention and treatment of this infectious disease is needed to improve public understanding in order to break the chain of causes of stunting. This counseling activity aims to improve public knowledge, especially mothers of toddlers, about the relationship between worm infections and giardiasis with stunting and preventive measures. The methods used are interactive lectures and discussions. The counseling involved mothers of toddlers and integrated health post cadres in the area. From the examination of 20 feces samples of toddlers in this area, no positive cases of worm infections and giardiasis were found. The results of this activity showed a good understanding of the participants regarding the importance of environmental sanitation, personal hygiene, and providing nutritious food to children. This counseling is effective in increasing public awareness regarding worm infections and giardiasis as risk factors for stunting. Follow-up activities can be carried out in the form of monitoring and providing nutritional interventions to support sustainable stunting prevention in this region.

Abstract

Urticaria is a skin condition characterized by wheals, which may be accompanied by itching, a burning sensation, or pain, and can occur together with angioedema. House dust mites (HDM) are one of the main aeroallergens capable of triggering IgE-mediated allergic skin reactions. Children living in crowded environments, such as orphanages, are at high risk of HDM exposure. This study aimed to determine the relationship between house dust mite exposure and the occurrence of urticaria and/or angioedema in children at an orphanage in Pauh District, Padang City. This was an observational analytical study with a cross-sectional approach, conducted at an orphanage in Pauh District, Padang City, and in the Parasitology Laboratory of the Faculty of Medicine, Andalas University, from March to October 2025. The sampling technique used was total sampling. Bed dust was collected using a vacuum cleaner, and interviews were conducted using an adapted UAS and ISAAC questionnaire. Chi-square was used to analyze the relationship between the two variables. The results of this study showed that all dust samples (100%) were positive for house dust mites, and identification revealed that the most common species found was Dermatophagoides sp. (57.14%), followed by Acarus sp. (32.14%), Cheyletus sp. (25%), Carpoglyphus sp. (17.86%), Glycyphagus sp. (14.29%), Thyrophagus sp. (14.29%), and Blomia tropicalis (7.14%). The average density of house dust mites was 72.85 mites/gram of dust. The prevalence of urticaria and/or angioedema in children was 25.6%. This study found no significant association between house dust mite density and the occurrence of urticaria and/or angioedema (p = 0.263). The conclusion of this study is that factors such as individual predisposition, environmental conditions, and immune sensitization status are likely to contribute to the development of urticaria and/or angioedema.

Abstract

Hasmiwati, Rusjdi SR, Nofita E. 2018. Detection of Ace-1 gene with insecticides resistance in Aedes aegypti populations from DHF-endemic areas in Padang, Indonesia. Biodiversitas 19: 31-36. Aedes aegypti is distributed widely in West Sumatra as a primary vector of Dengue hemorrhagic fever, especially in Padang City. Synthetic insecticide control is one currently used method to prevent mosquito-borne diseases. The extensive, long-term application of Temephos along with inappropriate dosages, have resulted in the development of resistance in Ae. aegypti populations. Mutation of the Ace-1 gene, encoding an acetyl cholinesterase, is one of the mechanisms that confer resistance to organophosphate (OP). The Temephos resistance status of Ae. aegypti in Padang city has not yet been studied. This study aimed to investigate the resistance status of Ae. aegypti and identify any possible mutation (s) of the Ace-1 gene in Padang city. Ae. aegypti samples were collected in five population in Padang city (Jati (JT), Gunung Pangilun (GP), Lubuk Minturun (LM), Korong Gadang (KG), and Bandar Buat (BB)). The larval susceptibility to Temephos was tested by larval bioassays with Temephos pestanal at 0.02 mg/L dosages. Larval susceptibility was determined by mortality percentage values. The relationship between Ace-1 genotypes and the resistant phenotype was analyzed by percentage of genotype frequency. Out of five populations, assessed by larval bioassays, JT and GP were resistant to Temephos; LM, KG, and BB were tolerant. A total of 50 individuals from larval bioassays were genotyped for Ace-1 gene. Our findings showed that Ace-1 was 495 bp in length. Mutation was not found in the G119S location but in the T506T location. Three alleles in T506T location were detected, including a wild type allele, TT (65.21%), and two mutant alleles, TA (26.08%), AA (8.69%). The use of Temephos showed that some Ae. aegypti populations were resistant, others were tolerant, but no population was vulnerable to Temephos. A novel mutation was detected as substitution in T506T location (ACT>ACA).
