Abstract

The embryogenesis (SE) has important role for genetic engineering of cassava (Manihot esculenta Crantz). However, the success of SE induction depend on plant growth regulator s (PGR)s and treatment enriched in induction media.  This experiment tried to induce callus formation of cassava from several in vitro explants: immature leaf, apical bud, and internode; and to develop somatic embryogenesis of cassava in several media enriched with tyrosine and copper sulphate (CuSO4) added into media enrich with picloram as treatment.  Different response of explants source to callus induction treatment from those three varieties in callus induction as well as friable callus formation were found in this experiment. The best medium to induce varied with variety; MS media supplemented 12 mg/L picloram + 0.5 mg/L CuSO4 was the best for “Adira 4” and  half MS and half GD media supplemented 12 mg/L picloram + 100 mg/L tyrosine for “Malang 6”.  All treatments resulted somatic embryo which developed indirectly and in morphologically normal somatic embryos

Abstract

The CalBsyn gene was previously constructed synthetically to encode Candida antarctica lipase B (CALB). Lipase from CalBsyn gene is slightly different from that of wild type CALB (CALB-wt) where it has three amino acids substitutions at different positions, i.e. V210I, A281E, and V221D, to improve enzyme’s thermostability and catalytic efficiency. The CalBsyn gene was isolated from pJ912-CalBsyn vector by digestion using XhoI restriction enzyme. The CalBsyn gene then was ligated to pGAPZα expression vector and transformed into E. coli TOP10F’ in order to obtain recombinant vector pGAPZα-CalBsyn. The result showed that pGAPZα-CalBsyn recombinant vector was successfully transformed into E. coli TOP10F’ with transformation efficiency of 4.11 x 103 cfu/µg plasmid DNA. The pGAPZα-CalBsyn recombinant plasmid was successfully introduced into Pichia pastoris SMD1168H using electroporation method with transformation efficiency of 1.01 x 102 cfu/µg DNA. Qualitative lipase activity assays showed that transformed P. pastoris secreted recombinant lipase (CALB) and has lipolytic activity; while quantitative lipase activity assays showed that the lipase activity was 63.5 Units/ml in 48 hours. Analysis using SDS-PAGE showed that CALB protein was expressed successfully and the recombinant protein’s molecular size was approximately 45 kDa.

Abstract

   Lactobacillus casei WSP-derived an aspartic protease was sequentially purified by using chromatography gel filtration sephadex G-50. It resulted in a 22.81-fold increase of specific activity (51.5 U/mg) with a final yield of 1.9%. The estimated molecular weight of the purified enzyme was 37 kDa and showed gelatinolytic activity in zymogram assay. The enzyme exhibited optimum activity at 40ºC and pH 6 with casein as the substrate. Enzyme activity was significantly inhibited by pepstatin A (0.5 mM and 1 mM), confirming that this enzyme is a group of aspartic proteases, while other inhibitors such as EDTA, PMSF and iodoacetic acid showed no inhibition effect on the activity of enzyme. The addition of metal ion to the enzyme decreased enzyme activity, indicating the proteolytic enzyme was metal ion- dependent. Denaturant such as DDT tended to increase caseinolytic activity. Furthermore, this enzyme was capable of generating the new peptides from skimmed milk with the size 8 kDa, 10 kDa and 15 kDa. These peptides have potential as antibacterial and antioxidant agents.

Abstract

Evolution cycle of landform in Manado and surrounding area is  caused by  weathering, erosion, transportation, sedimentation, and human factors. These processes have occurred since the land existence, and theywere strongly accelerated   by human  activities.Based on slope level, soil/rock type, vegetation/land covers, and erosion type, the erosion susceptibility in the studied area can be grouped into: high, medium, low,  and no erosion.Very high erosion occurs  on landform  of volcanic cone a and  b, volcanic slope a and b, mountain  dissected volcanic slope and dissected straight  volcanic mountain. High erosion occurs on landform  of solfatara field, fumarola, volcanic foot slope b, volcanic mountain slope elongated  dissected, volcanic slope c, residual  hill and lava flow. Medium erosion occurs on landform of volcanic  foot slope a and alluvium  fan.  Lowerosion occurs on landform of volcanic  foot slope c, intermountain plane and flood plain. Abstract

Penelitian ini bertujuan untuk mengetahui perbedaan berfikir kritis mahasiswa yang proses pembelajarannya menggunakan pembelajaran e-learning dan konvensional pada konsep ekosistem. Metode yang digunakan adalah metode komparatif dengan populasi seluruh mahasiswa biologi semester satu dengan sampel yang diambil adalah kelas 1A dan 1C yang masing-masing sebanyak 30 orang dan sampel diambil dengan teknik cluster random sampling. Pengambilan data dilakukan dengan memberikan soal essay untuk memperoleh data kemampuan berfikir kritis mahasiswa. Teknik analisis data yang digunakan adalah uji perbedaan dua rata-rata (uji-t) dengan taraf signifikan (α) = 5% menggunakan SPSS 21 for Windows. Berdasarkan analisis data dan pengujian hipotesis menunjukan nilai sig.(2 tailed) sebesar 0, 83 > 0,05 sehingga dapat disimpulkan Ho diterima artinya tidak ada perbedaan yang signifikan hasil belajar mahasiswa biologi yang proses pembelajarannya menggunakan model pembelajaran e-learning dan konvensional.Kata kunci : E-learning, konvensional, berfikir kritis

Abstract

Penelitian ini bertujuan untuk mengetahui perbedaan hasil belajar dan kemampuan berpikir kritis mahasiswa biologi yang proses pembelajarannya menggunakan pendekatan e-learning dan konvensional pada konsep sistem pencernaaan makanan. Metode penelitian ini adalah metode komparatif yang menggunakan rancangan posttest control group design dengan populasi seluruh mahasiswa semester I program studi pendidikan Biologi FKIP UNSIL dengan sampel sebanyak 2 kelas yang diambil secara cluster random sampling yaitu kelas 1A sebanyak 30 orang dan kelas 1 C sebanyak 30 orang. Teknik pengumpulan data dilakukan dengan memberikan posttest untuk memperoleh data kemampuan akhir hasil belajar dan rubrik untuk mengetahui kemampuan berpikir kritis mahasiswa, teknik analisis data yang digunakan adalah uji perbedaan dua rata-rata (uji-t) dengan taraf signifikan (α) = 5%. Berdasarkan analisis data dan pengujian hipotesis menunjukan nilai sig. (2 tailed) sebesar 0,690 >0,05 untuk hasil belajar dan sig. (2 tailed) sebesar 0, 83 > 0,05 untuk berfikir kritis, dari data tersebut disimpulkan Ho diterima artinya tidak ada perbedaan yang signifikan hasil belajar dan kemampuan berfikir kritis mahasiswa biologi yang proses pembelajarannya menggunakan model pembelajaran e-learning dan konvensional.Kata kunci: e-learning, pembelajaran konvensional, hasil belajar, kemampuan berpikir kritis

Abstract

Penelitian ini bertujuan untuk mengetahui jumlah konsumsi rata-rata oksigen yang diperlukan untuk bermetabolisme pada hewan invertenrata kelas insekta. Metode eksperimen nyata (True-experimental) dengan menggunakan rancangan acak lengkap (RAL). Perlakuan masing-masing sampel yaitu dengan menggunakan beberapa spesies hewan yang berasal dari ordo yang berbeda diantaranya jngkrik (Gryllus asimilis), belalang (Dissosteira carolina), kecoa (Blattella asahinai), kepik (Leptocorisa acuta) dan capung (Pantala flavescens). Pengumpulan data yang dilakukan dalam penelitian ini dilakukan dengan cara melakukan pengukuran volume terhadap gas oksigen yang dihasilkan pada tabung reaksi pada setiap sampel. Data yang didapat dari hasil penelitian akan dianalisis menggunakan analisis kuantitatif, teknik analisis data yang digunakan adalah uji perbedaan dua rata-rata (uji-t) dengan taraf signifikan (α) = 5%. Berdasarkan analisis data dan pengujian hipotesis menunjukan nilai  sig. (2 tailed) 0,000. < 0,05, dari data tersebut disimpulkan Ho ditolak, artinya Ada perbedaan jumlah konsumsi oksigen (O2) pada respirasi berbagai hewan invertebrata kelas insekta. Dan dilihat dari rata-rata konsumsi oksigen (O2) dapat disimpulkan jangkring menggunakan oksigen paling banyak dan konsumsi paling sedikit adalah kepik. Kata kunci : konsumsi oksigen, respirasi, insekta

Abstract

Fruit flies (Drosophila melanogaster) generally have four phases in their life cycle, namely eggs, larvae, pupae and imago. In general, Drosophila melanogaster experiences a life cycle of 8-11 days at optimal temperatures. At lower temperatures the time required to complete its life cycle is relatively longer and slower which is around 18-20 days. Whereas at higher temperatures adult flies that grow will be sterile. The development period of Drosophila melanogaster in its life cycle is influenced by several factors, namely ambient temperature, food availability, level of maintenance density and light intensity. This study aims to determine the effect of temperature on the life cycle of fruit flies (Drosophila melanogaster). The study will be analyzed using quantitative analysis using randomized block design (RBD) with six temperature difference treatments for each treatment and repeated 4 times so that there are 24 experiments. The results showed the development in the Drosophila melanogaster cycle starting from Egg - Larva (Instar I) - Larva (Instar II) - Larva (Instar III) - Pre Pupa - Pupa - Imago. The life cycle of Drosophila melanogaster has an average of 42.08 hours or 1.75 days up to 79.96 hours or 3.33 days in each treatment. And obtained a fast time during the life cycle of Drosophila melanogaster at a temperature of 30°C is 10.47 days and the longest at a temperature of 18°C is 18.35 days.

Abstract

Lucas and Gibonacci numbers are  two  sequences of numbers derived from a welknown numbers, Fibonacci numbers. The difference between Lucas and Fibonacci numbers only lies on the first and second elements.  The first element in Lucas numbers  is 2 and the second is 1, and nth element,  n ? 3 determined by similar pattern as in the Fibonacci numbers, i.e : Ln = Ln-1  + Ln-2.  Gibonacci numbers  G0 , G1 ,G2 , ...; Gn = Gn-1 + Gn-2 are generalized of Fibonacci numbers, and those numbers are nonnegative integers. If G0 = 1 and G1 = 1, then the numbers are the wellknown Fibonacci numbers, and if G0 = 2 and G1 = 1, the numbers are Lucas numbers. Thus, the difference of those three sequences of numbers only lies on the first and second of the elements in the sequences.  For Fibonacci numbers there are quite a lot identities already explored, including the sum of cubes, but there have no discussions yet about the sum of cubes for Lucas and Gibonacci numbers. Abstract

Self-regulated learning is a constructive process that is active in setting learning goals. This ability should be possessed by every individual to become a successful learner. Another thing that students must have to be successful in the process and learning outcomes are learning motivation. This study aims to determine the correlation between learning motivation with self-regulated learning in grade X MIPA 9 SMA Negeri 1 Tasikmalaya City. This research was conducted in April 2019. The research method used was correlational with a population of all class X MIPA 9 students as many as 30 people. Samples were taken using a saturated sampling technique so that the entire population was sampled. The research instrument used in this study is the Motivated Strategies for Learning Questionnaire (MSLQ), which consists of 2 parts, namely motivational beliefs and self-regulated learning strategies. The data analysis technique used is the bivariate correlation regression test. Based on the results of the study, it can be concluded that there is a strong correlation (R = 0.761; R2 = 0.580) between learning motivation and self-regulated learning. The contribution of learning motivation to self-regulated learning is 58%.
