Abstract

Kemajuan ilmu pengetahuan dan teknologi yang semakin pesat telah mendorong manusia untuk mengatasi segala permasalahan yang timbul di kehidupan. Hampir semua orang tidak bisa lepas dari dunia teknologi, baik itu teknologi sederhana maupun teknologi tinggi. contonya adalah alat-alat elektronik yang beragam dan mempermudah pekerjaan sehari-hari. Dalam kehidupan sehari-hari menjemur adalah salah satu aktifitas yang terkadang sering kita lakukan bukan hanya menjemur pakaian, kasur dll aktifitas menjemur juga sering di lakukan oleh pelaku indutri perumahan contohnya indutri kerupuk, ikan asin, dan sebagainya. Akan tetapi salah satu kendala yang sering terjadi pada saat proses menjemur adalah datangnya hujan yang tiba-tiba, cuaca yang tidak menentu akan menghambat dan menghawatirkan aktifitas menjemur kita pada saat kita tinggal bepergian keluar rumah. Dalam penulisan tugas akhir ini penulis mengamati permasalahan-permasalan yang sering timbul dalam kehidupan sehari-hari dan diangkat sebagai sekripsi untuk mendapatkan penyelesaian masalah tersebut. Dibantu dengan study pustaka. Penelitian ini membahas tentang perancangan penutup jemuran otomatis agar dapat menyelesaikan masalah penjemuran pakaian yang sering mengalami masalah kehujanan.

Abstract

Cowpeas grown in Southwest Maluku have local potential due to their diversity and ability to adapt to drought stress conditions that otherwise cause low productivity. The purpose of this study was to identify of local cowpea varieties of Southwest Maluku that show tolerance to drought. The experimental samples were seven local varieties and threereference cultivars. The drought stress treatment was altered watering periods, i.e., every two (P0) and ten days (P1) from growth to harvest. The results of this research showed that a ten-day watering period significantly decreased plant height, number of leaves, number of root nodules, root and shoot dry weight, photosynthesis rate, stomatal conductance, transpiration rate, relative water content, media water content, chlorophyll content, and number and weight of seeds perplant. Drought stress treatment increased proline content and root length. Correlation analysis showed a significant correlation between plant height with all variables, except for root length, proline content, and weight of seedsper plant. The correlation and cluster analyses showed that the KM7 variety is a drought tolerant genotype among the local cowpea varieties from Southwest Maluku. Therefore, the KM7 variety can be used as plant material in future breeding programs.

Abstract

Isolation and Cloning of cDNA Fragment of Gene Encoding for Multidrug Resistance Associated Protein from M. affine. M. affine can grow well in acid soil with high level of soluble aluminum. One of the important proteins in the detoxifying xenobiotic stress including acid and Al stresses is a multidrug resistance associated protein (MRP) encoded by mrp gene. The objective of this research is to isolate and clone the cDNA fragment of MaMrp encoding MRP from M. affine. By reverse transcription, total cDNA had been synthesized from the total RNA as template. The fragment of cDNA MaMrp had been successfully isolated by PCR by using total cDNA as template and mrp primer designed from A. thaliana, yeast, and human. This fragment was successfully inserted into pGEM-T Easy and the recombinant plasmid was successfully introduced into E. coli DH5α. Nucleotide sequence analysis showed that the lenght of MaMrp fragment is 633 bp encoding 208 amino acids. Local alignment analysis based on nucleotide of mRNA showed that MaMrp fragment is 69% identical to AtMrp1 and 63% to AtMrp from A. thaliana. Based on deduced amino acid sequence, MaMRP is 84% identical to part of AtMRP13, 77% to AtMRP12, and 73% to AtMRP1 from A. thaliana respectively. Alignment analysis with AtMRP1 showed that MaMRP fragment is located in TM1 and NBF1 domains and has a specific amino acid sequence QCKAQLQNMEEE

Abstract

Actin is a major component of the plant cytoskeleton, so all cells contain this protein. Actin is expressed constitutively and is involved in basic housekeeping functions required for cell maintenance. Because of this, it has been frequently used as an internal control to normalize changes in gene expressions analysis. Actually, the information of nucleotide sequence of actin gene of Jatropha curcas L. population IP-2P from Indonesia is not available yet. The objective of this research was to isolate, clone and characterize cDNA of actin genes of J. curcas IP-2P. Three partial actin gene sequences had been successfully isolated by PCR using total cDNA as template, and actin primer designed from conserved region of Arabidopsis thaliana. Nucleotide sequence analysis showed that the length of JcACT fragment is 610, 534, and 701 bp encoding 203, 177, and 234 amino acids respectively. Local alignment analysis based on mRNA sequences shows that JcACT fragment shares 98% similarity with actin mRNA of Hevea brasiliensis and 99% with actin mRNA of Ricinus communis. Based on deduced amino acid sequence, JcACT is 100% identical to actins from Prunus salicina, Gossypium hirsutum, and Betula luminifera. Even though these clones of cDNA are not completed yet, they can be used as reference in J. curcas L. gene expression analysis.

Abstract

Latar belakang : Stroke merupakan penyakit kronik yang membutuhkan waktu perawatan dan penyembuhan yang cukup lama, sehungga menyebabkan pasien merasa tidak berdaya. Terjadinya disbilitas fisik pada pasien stroke menyebabkan timbulnya berbagai masalah fisik maupun psikologis. Perubahan psikologis yang ada tersebut diantaranya adalah terjadinya masalah harga diri rendah dan depresi. Tujuan : Penelitian ini bertujuan untuk mengetahui bagaimana hubungan antara harga diri dengan tingkat depresi pada pasien stroke. Metodologi : Disain penelitian ini adalah kuantitatif dengan pendekatan cross sectional. Responden dalam penelitian ini adalah pasien stroke yang sedang dirawat di RSD Panembahan Senopati Bantul. Cara pengambilan sample dilakukan secara accidental sampling dan didapat sebanyak 30 responden. Metode pengumpulan data dengan menggunakan instrumen Rosenbergs Self Esteem Scale (RSES) untuk mengukur harga diri dan Beck Depression Inventory (BDI) untuk tingkat depresi. Uji statistik korelasi spearman rank digunakan dalam penelitian ini. Hasil penelitian : Hasil penelitian menunjukkan bahwa pasien dengan harga diri tinggi (26,7%) dan sedang sebesar 60 % , sedangkan pasien dengan harga diri rendah (13,3%). Pasien dengan tingkat depresi sedang (50%), dengan tingkat depresi ringan adalah 33,3% dan pasien dengan tingkat depresi berat adalah 16,7%. Hasil penelitian ini menunjukkan ada hubungan yang bermakna antara harga diri dengan tingkat depresi pada pasien stroke dengan nilai p <0,005 dan nilai r=0,377-0,59. Kesimpulan: Simpulan dari penelitian ini adalah ada hubungan yang bermakna antara harga diri dengan tingkat depresi pada pasien stroke di RSD Panembahan Senopati Bantul. Abstract

This study shows that the condition of the well water tested based on physical, chemical and biological parameters to determine the feasibility of the water used by the surrounding community still meets the required quality standard thresholds. Although somewhat polluted because of the mining. Underground water in the karst area of Gunungkidul Regency is safe as raw water for drinking by residents and meets the standards set by the Indonesian Ministry of Health and Government Regulation Number 82 of 2001 concerning Water Quality Management and Water Pollution Control. If used as drinking water, it still has to go through water treatment or be heated to a certain boiling point, because it contains bacteria that may be harmful to humans.

Abstract

In its development, zakat has become one of the Islamic philanthropic institutions that work for the welfare of society and as an effort to assist in realizing the Sustainable Development Goals (SDGs) program. In fact, the contribution of zakat can not be said to be maximal to the SDGs in Indonesia. This is because the collection and distribution of zakat has not been optimal. This study aims to describe the effect of zakat on the SDGs in Indonesia (a study of all provinces on the island of Java). Of the 17 SDGs programs, only 5 programs were tested, namely (1) economic growth (GRDP), (2) poverty-free (JPM), (3) healthy and prosperous life, quality education, and decent work as measured by the Human Development Index ( HDI), (4) reduced inequality (GI), and (5) gender welfare (GPI). The method used is the Panel Vector Error Correction Model (PVECM). The results of this study prove that (1) Zakat in the short and long term has a negative and significant effect on GRDP, (2) Zakat in the short and long term has no effect on JPM, (3) Zakat in the short and long term has no effect on HDI, (4) Zakat in the short term has no effect on GI, but zakat in the long term has a positive and significant effect on GI, (5) Zakat in the short and long term has no effect on GPI.

Abstract

Salah satu kompetensi yang perlu dikuasai oleh pemelajar BIPA dalam belajar bahasa Indonesia adalah kompetensi linguistik. Kompetensi linguistik adalah kemampuan untuk menggunakan kode linguistis secara akurat. Salah satu kemampuan yang perlu dikuasai dalam kompetensi linguistis adalah kompetensi yang berhubungan dengan gramatikal atau tata bahasa. Kemampuan gramatikal atau tata bahasa ini sering dipandang oleh kebanyakan pemelajar BIPA sebagai kemampuan yang lebih sulit dikuasai dibandingkan dengan kemampuan yang lain, seperti membaca, berbicara, menulis, dan menyimak, padahal kemampuan dalam tata bahasa menjadi dasar bagi kemampuan empat keterampilan berbahasa tadi. Begitu juga bagi pengajar BIPA, tata bahasa sering dianggap sebagai bidang yang lebih sulit diajarkan daripada empat keterampilan berbahasa. Tidak jarang pengajar BIPA menghindari penjelasan yang berkaitan dengan aspek ketatabahasaan apalagi jika yang dihadapi adalah pemelajar BIPA yang berada pada tingkat (level) lanjut. Makalah ini hendak menyajikan praktik baik dalam pengajaran tata bahasa BIPA. Diharapkan bahasan ini dapat memberikan wawasan dan inspirasi bagi pengajar BIPA yang mengajarkan tata bahasa atau yang karena tugasnya harus menjelaskan aspek-aspek ketatabahasaan BIPA. Yang dimaksud dengan praktik baik di sini adalah bagaimana cara memahamkan konsep tata bahasa bahasa Indonesia kepada pemelajar BIPA dengan mudah. Pembahasannya akan ditinjau dari segi sifat linearitas bahasa, sifat bahasa yang berpola, konteks, dan teknik penyajian
