Abstract

Abelmoschus manihot L. has been used traditionally as folks medicine plant and it containflavonoid and steroid that has health benefit. The objectives of this research were todetermine anti obesity and hypolipidemic activity of flavonoid and steroid extract fromAbelmoschus manihot L. on white male Wistar induced with high content of protein and lipidfood. Anti obesity test were evaluated by the decreasing of rats weight, whereashypolipidemic test were evaluated by the decreasing of total cholesterol levelsphotometrically using Liebermann-Burchard method. The rats were divided into four groups,namely the negative control (distilled water), extract treatment groups (flavonoids andsteroids), and a positive control group (simvastatin). The percentations of Weight reducingand cholesterol level were analyzed by ANOVA, if there were significant in groups, itcontinued with Duncan Multiple Range Test. The results showed that treatment group possesanti obesity activity, whereas flavonoid extract decrease 7.85% body weight of rats andsteroids extract were 13.70% of body weight. Flavonoids showed hypolipidemic effects bydecrease rats cholesterol levels by 86.45% and steroid extract decrease cholesterol levels by72.53%.Keywords : Abelmoschus manihot L., flavonoid extract, steroid extract, anti obesity,hypolipidemic

Abstract

Isolation and identification flavonoid compounds in beluntas leaf (Pluchea indica L.) have beenconducted. Isolation was carried out by using 96% p.a ethanol solvent of maceration extraction for 7days and partition using n-heksana solvent to obtain concentrated beluntas leaf extract. The extractwas purified by thin layer chromatography using eluent n-butanol : asetic acid : water (BAA) (4:1:5).Isolate 3, result from thin layer chromatography contain flavonoid compound. Ultra Violet - Visiblespectra showed that the flavonoid compound was flavonol, with characteristic wavelengths from 250to 280 nm for band II and 350-385 nm for band I.Keywords: beluntas, flavonoid, Thin Layer Chromatography, UV-Vis Spectrophotometer

Abstract

ABSTRACTVitamin C is one of the nutrients that role as antioxidant and effectively overcome the freeradicals that can damage cells or tissue. Fruit is a good source of vitamin C, such as themango fruit. This study aimed to determine the levels of vitamin C in the mango fruit usingUV-Vis spectrophotometry method and Iodimetry method, and compare the results of bothmethods. Samples were identified as ripe mango. In this study, the levels of vitamin C wereanalyzed by using UV-Vis spectrophotometry and Iodimetry. The results of the analysis ofvitamin C in the mango dodol by using UV-Vis spectrophotometry is 15.88 g/100g, whereasthe content of vitamin C by using Iodimetry was 3.5 g/100g. From these results, showed thelevels of vitamin c by using the spektrofotometry uv-vis method is higher than the iodimetrymethod.Keywords: Vitamin C, mango, UV-Vis Spectrophotometry, Iodimetry.

Abstract

The objective of this research was to determine the species or genus of mercury resistant bacteria culture from mercury polluted waters. Microbiology identification of culture S3.2.2 was done with morphology, physiology and biochemistry test. Molecular analysis was done by amplifying 16S rRNA gene using PCR method. Universal Bact- F1 forward/Uni-B1 Reverse was using as primer. Amplification result was electrophorize with agarose 1,5%, furthermore sequencing was done against 16SrRNA gene. The result shows that culture S3.2.2 were negative grams bacteria, rod shaped, nonmotil, catalase (+), citrat (+), H2S (-), fermentase (+), indol (+) and lisin (+). Blast analysis against nucleotide sequence of 16S rRNA gene possesses 99% similarities with Klebsiella pneumoniae.

Abstract

ABSTRACT Cosmetic whitening is a cosmetic that contains the active ingredient intended to bleach and brighten the skin or whiten the skin. Retinoic acid is used in whitening creams but is prohibited because it can cause dry skin, burning, and teratogenic (defects in the fetus). The purpose of this study was to determine whether whitening creams circulating in the city of Manado contains retinoic acid and to determine the levels of retinoic acid whitening cream found on the face. The samples studied were lightening cream sample A, sample B, sample C, sample D and sample E. The qualitative identification of retinoic acid have been done with Thin Layer Chromatocraphy (TLC) it has been given the dark blood if seen in UV spectrum 254 nm. Quantitative analysis was done by UV spectrophotometric maximum wavelength of 352 nm. The results indicated that three of sample contaided retinoic acid. Retinoic acid concentration in sample C was 0,021%, sample D was 0,026%, sample E was 0,016% and product retinoic acid cream (Vitacid) was 0,053%. Abstract

IDENTIFIKASI BAKTERI ASAM LAKTAT DARI HASIL FERMENTASI SELADA ROMAIN (Lactuca sativa var. longifolia Lam.) MENGGUNAKAN GEN 16S rRNAMaria A. Gani 1) , Trina E. Tallei 2) , Fatimawali 1)1) Program Studi Farmasi FMIPA UNSRAT Manado, 951152) Jurusan Biologi Farmasi FMIPA UNSRAT Manado, 95115 ABSTRACTLactic acid bacteria (LAB) is a group of bacteria that produce lactic acid as the major metabolic end product of carbohydrate fermentation. LAB are highly beneficial because of their probiotic potential properties and as functional starter cultures in food fermentation. This study was aimed to identify the LAB species isolated from romaine lettuce fermented product by using molecular identification method with 16S rRNA gene marker. The fermented product was diluted and spread onto MRS agar supplemented with 1% of CaCO3 and then purified by using streak method. Both isolates were positive Gram bacteria and gave negative results from catalase test. The result of molecular identification showed that LAB from romaine lettuce fermented product have each 99 and 100% similarity to Enterococcus faecium that known have properties as probiotic potential and functional culture starter.Key words: lactic acid bacteria, fermentation, probiotic, Enterococcus faecium, Lactuca sativa var. Longifolia Lam. ABSTRAK Bakteri asam laktat (BAL) merupakan bakteri yang menghasilkan asam laktat sebagai metabolit utamanya dalam fermentasi karbohidrat. BAL merupakan bakteri yang menguntungkan karena memiliki aktivitas probiotik potensial dan dapat berguna sebagai starter dalam proses fermentasi makanan. Penelitian ini bertujuan untuk mengidentifikasi spesies BAL yang diisolasi dari hasil fermentasi selada romain menggunakan metode identifikasi molekuler dengan gen penanda 16S rRNA. Cairan hasil fermentasi diencerkan dan ditebar pada media MRS agar yang telah dicampur dengan CaCO3 1% dan kemudian dimurnikan menggunakan metode streak plate. Kedua isolat merupakan bakteri Gram positif dan menunjukkan hasil negatif pada uji katalase. Hasil identifikasi molekuler menunjukkan bahwa kedua BAL hasil fermentasi selada romain memiliki kemiripan masing-masing 99 dan 100 % dengan Enterococcus faecium yang diketahui memiliki aktivitas probiotik.Kata-kata kunci: bakteri asam laktat, fermentasi, probiotik, Enterococcus faecium, Lactuca sativa var. longifolia Lam.

Abstract

This research aims to describe the antibiotics use in caesarean section patients due to its kind,route of administration, conformity of dose and frequency, and length of time given. It is adescriptive research. The data were collected in retrospective on 129 patients medical recordswhose received antibiotics, both prophylactic antibiotics and therapeutical antibiotics. Dataobtained shows that the third generation of cephalosporin which is ceftriaxone combined withmetronidazole (55.81%) is the most common kind of antibiotic that being used asprophylactic antibiotics and the most common therapeutical antibiotic that being used is thefirst generation of cephalosporin which is cefadroxil combined with metronidazole (53,59%).The prophylactic antibiotic’s route of administration is intravenously (IV) (100%) and oralfor therapeutical antibiotics (100%). The antibiotics doses that are being used bothprophylactic antibiotics and therapeutical antibiotics has been fulfilled the conformity of doseand frequency, with the longest duration of usage the prophylactic antibiotics is 1 day(80,92%).Keywords: antibiotics, surgical wound infections, caesarean section

Abstract

Cosmetic whitening is a cosmetic that contains the active ingredient intended to bleach and brightenthe skin or whiten the skin. Hydroquinone is used in bleaching creams but is prohibited because it cancause skin irritation, skin redness and burning, and can also cause abnormalities in the kidneys, bloodcell cancer, and liver cancer. The purpose of this study is to prove the presence or absencehydroquinone content in various brands of whitening creams circulating in the city of Manado. Bleachcream samples studied were sample A, sample B, sample C, and sample D. The content ofhydroquinone was observed using the method of thin layer chromatography and UVspectrophotometer. The results showed that the four samples, namely Kelly, Quina, Fair Check, andOriens, the hydroquinone was not identified, either using thin layer chromatography or UVspectrophotometer. Abstract

ABSTRACTHepatitis B is a disease caused by the virus called hepatitis B wich characterized by aninflammation that occurs in organs such as the liver. The transmission of Hepatitis B is higherthan the transmission of the HIV virus. This study aimed to investigate the characteristics anduse of the drug in hepatitis B patients in the Government Hospital in Manado. This researchis a descriptive study based on retrospective data collection for 2 years survey from January2011 to December 2012. The respondent of this research involve of 70 patients with HepatitisB medical record. The results showed that based on group age, responden with age group 46-65 has the highest percentage by 31 patients (44.3%), based on gender, males have thehighest percentage of total of 47 patients (67.1%), based on the duration of treatment, thehighest percentage is shown by the patient with 1 day threatment as 21 patients (30%). Basedon therapeutic treatment this research divided it into 2 kind of threatment. The first us singleagent therapy Hepamax consist of 27 patients (38.6%) The second therapy is combinationbetween Hepamax and Curcuma wich consist of 9 patients (12.9%). Abstract

ABSTRACT Copra ashes are ash produced from burning coconut husk in the copra roasting process after more than three roasts. Noodle producers in Manado and Tombatu were  using copra ashes as an additive to noodles because it has benefits as a substitute for borax, which is prohibited from using on food, since it can improve the texture of noodles, as preservatives, improve the color and appearance of noodles. The purpose of this study is to analyze qualitatively and quantitatively the presence of borax compounds in copra ashes. Qualitatively analysis was carried out using the flame and color method, and the quantitative test used the UV-Vis spectrophotometry method at the wavelength of 545 nm and 0.125% curcumin as the color reagent. The study showed positive result on the qualitative test of the color method against turmeric paper whereas turn to brownish red, and negative result on the flame test whereas turn to red greenish fire, on the quantitative test the sample level A=1.459 ppm and sample B=2.092 this indicates that samples A and B were positive contains borax.Keywords: Copra ashes, food additives, borax and spectrophotometry ABSTRAK             Abu kopra adalah abu yang dihasilkan dari pembakaran sabut kelapa dalam proses pemanggangan kopra setelah lebih dari 3 kali pemanggangan. Abu kopra oleh produsen mie di Airmadidi dan di Tombatu digunakan sebagai bahan tambahan  pada mie karena memiliki manfaat sebagai pengganti boraks yang dilarang penggunaannya pada makanan, karena dapat memperbaiki tekstur mie, sebagai  pengawet dan memperbaiki warna dan penampilan mie. Tujuan penelitian ini yaitu menganalisis secara kualitatif dan kuantitatif keberadaan senyawa boraks di dalam abu kopra. Analisis kualitatif dilakukan dengan metode nyala dan warna, dan uji kuantitatif digunakan metode spektrofometri UV-Vis pada panjang gelombang 545 nm dan kurkumin 0,125% sebagai pereaksi warnanya. Penelitian menunjukkan hasil positif pada uji kualitatif metode warna yaitu kertas tumerik menjadi merah kecoklatan, dan hasil negatif pada uji nyala yaitu api berwarna merah kehijauan, pada uji kuantitatif kadar sampel A=1,459 ppm dan sampel B=2,092 ini menunjukkan bahwa sampel A dan B positif mengandung boraks. Kata kunci      : Abu kopra, bahan tambahan makanan, boraks dan spektrofotometri.
