Abstract

The aim of this research was to analyze mercury reduction power from patient urine with teeth amalgam which used for mercury detoxification. The research was descriptive which isolate 5 samples of patient urine from Bahu PUSKESMAS. Bacteria were grow in a media wich consist of 5, 10, 20, 40 and 60 ppm of HgCl2. Bacteria which grow in the highest concentration of mercury were identified and tested it reduction power against HgCl2. The result shows that there were 3 isolates which bacteria can grow with HgCl2 60 ppm namely Isolate U1.3, U3.1, and U3.3. Identification result found that isolate U1.3 and U3.1 were positive gram bacteria, rod, and motil, although U3.3 was negative gram bacteria, rod and not motil. Result of mercury reduction shows that in 24 hours, the three isolates reduce 100% mercury in nutrient broth, it concluded that isolates were highly resistant mecury bacteria. Swartz) is one of the plants which has the antibacterial properties from the zingiberaceae family. Empirically it can treat various diseases such as chest pain, throat inflammation, stomach inflammation, rheumatism, diabetes and skin diseases. White galangal rhizome contains flavonoids, tannins, quinones and terpenoids. This study aims to determine the antibacterial activity of white galangal rhizome extract (Alpinia galanga L. Swartz) and test its inhibition against the growth of Klebsiella pneumoniae bacteria which are resistant to ceftriaxone antibiotics. The white galangal rhizome was extracted by maceration method using 96% ethanol solvent and antibacterial activity was tested using the disc and well method with a concentration difference of 100%, 75%, 50%, 25% and 12.5%. The results showed that white galangal rhizome extract (Alpinia galanga L. Swartz) had antibacterial activity against the bacterium Klebsiella pneumoniae and could inhibit the growth of Klebsiella pneumoniae bacteria. On the disc method for concentrations of 100%, 75%, 50%, 25% and 12.5%, the inhibitors were categorized as medium. Whereas in the well method for concentrations of 100% and 75%, the inhibitors were categorized as strong, for concentrations of 50%, 25%, and 12.5%, which have a medium category.Keywords: White Galangal Rhizome (Alpinia galanga L. Swartz), Klebsiella pneumoniaeABSTRAKRimpang Lengkuas putih (Alpinia galanga L. Swartz) merupakan salah satu tanaman yang memiliki khasiat sebagai antibakteri dari family zingiberaceae. Secara empiris dapat mengobati berbagai penyakit seperti nyeri dada, radang tenggorokkan, radang lambung, rematik, diabetes dan penyakit kulit. Rimpang Lengkuas putih mengandung senyawa flavonoid, tanin, kuinon dan terpenoid. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak rimpang Lengkuas putih (Alpinia galanga L. Swartz) dan menguji daya hambatnya terhadap pertumbuhan bakteri klebsiella pneumoniae yang resisten terhadap antibiotik seftriakson. Rimpang Lengkuas putih diekstraksi dengan metode maserasi menggunakan pelarut etanol 96% dan dilakukan pengujian aktivitas antibakteri dengan menggunakan metode cakram dan sumuran dengan perbedaan konsentrasi 100%, 75%, 50%, 25% dan 12,5%. Hasil penelitian menunjukkan bahwa ekstrak rimpang Lengkuas putih (Alpinia galanga L. Swartz) memiliki aktivitas antibakteri terhadap bakteri Klebsiella pneumoniae dan dapat menghambat pertumbuhan bakteri Klebsiella pneumoniae. Pada metode cakram untuk konsentrasi 100%, 75%, 50%, 25% dan 12,5% memiliki daya hambat yang dikategorikan sedang. Sedangkan pada metode sumuran untuk konsentrasi 100% dan 75% memiliki daya hambat yang dikategorikan kuat, untuk kosentrasi 50%, 25%, dan 12,5% memiliki daya hambat yang dikategorikan sedang. Abstract

ABSTRACTNanoparticles can increase the solubility of compounds, reduce the treatment dose and increase absorption. Flavonoid and phenol compounds in red galangal can inhibit the growth of bacteria that have been resistant to antibiotics. The bioactivity of red galangal compounds is expected to increase when the size of nanoparticles increases antibacterial activity. This study aims to determine the inhibition of the content of Red Galangal nanoparticles (Alpinia purpurata K.Schum) using chitosan on the growth of Klebsiella pneumoniae bacterial urine isolates of UTI antibiotic-resistant Ciprofloxacin. Nanoparticles extracts of red galangal rhizome was made using ionic gelation method, using extract concentration of 0.4%, tested for antibacterial activity using the wells method with a comparison of chitosan 0.4%, acetic acid 5%, red galangal extract 25%. Red galangal rhizome nanoparticles gave antibacterial activity value to the bacteria Klebsiella pneumoniae of 13.5 mm. Therefore, red galangal extracts nanoparticles can inhibit the growth of Klebsiella pneumoniae urine isolate in patients with urinary tract infections with strong inhibitory categories based on Davis and Stout categories.Keywords: Alpinia purpurata K. Schum, Klebsiella pneumonia, Nanoparticles. ABSTRAK Nanopartikel dapat meningkatkan kelarutan senyawa, mengurangi dosis pengobatan dan meningkatkan absorbsi. Senyawa flavonoid dan fenol dalam lengkuas merah mampu menghambat pertumbuhan bakteri yang telah resisten terhadap antibiotik. Sifat bioaktivitas senyawa lengkuas merah diharapkan akan meningkat ketika berukuran nanopartikel untuk meningkatkan aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui daya hambat kandungan nanopartikel rimpang Lengkuas Merah (Alpinia purpurata K.Schum) menggunakan kitosan terhadap pertumbuhan bakteri Klebsiella pneumoniae isolat urin penderita ISK yang resisten antibiotik Ciprofloxacin. Nanopartikel ekstrak rimpang lengkuas merah dibuat dengan menggunakan metode gelasi ionik, dengan menggunakan konsentrasi esktrak sebesar 0,4 %, dilakukan pengujian aktivitas antibakteri mengunakan metode sumuran dengan pembanding kitosan 0,4%, asam asetat 5%, ekstrak lengkuas merah 25%. Nanopartikel rimpang lengkuas merah memberikan nilai aktifitas antibakteri terhadap bakteri Klebsiella pneumoniae sebesar 13,5 mm. Oleh karena itu nanopartikel ekstrak rimpang lengkuas merah dapat menghambat pertumbuhan bakteri Klebsiella pneumoniae isolate urin penderita infeksi saluran kemih dengan kategori daya hambat kuat berdasrkan kategori Davis dan Stout.Kata Kunci: Alpinia purpurata K. Abstract

The accomplishment of this research was to knowing the fito-chemical composition ofGandaria fruit’s juice and its potential as anti-oxidant. The Gandaria fruit’s juice were madeusing the blender device then filtered by filtration paper for extraction of the juice.Determination of fito-chemical is done with color reaction test for identification of alkaloid,fenolik, flavonoid, saponin, steroid and terpenoid. The anti-oxidant test were using freeradicalscavengers DPPH (1,1-difenil-2-pikrilhidrazil) and recorded with Spectrophotometerat 515nm wave length. Result of the research indicates that in Gandaria fruit’s juice has beendetected with saponin and fenolik fito-chemicals. The Gandaria fruit’s juice has the antioxidantactivity in IC50 of 36,4 mg/ml. The verdict and conclusion for the research is,Gandaria fruit’s juice has the composition of fito-chemical (saponin and fenolik) which isvery potential as anti-oxidant.Keywords : Gandaria fruit’s juice, anti-oxidant, DPPH, fito-chemical composition

Abstract

ABSTRACTRhodamine B dye in the form of a crystalline powder green or reddish purple, odorless, anddissolves easily in bright red solution berfluoresan as textile dyes. Rhodamine B is still nofood products were found to contain rhodamine B dye such as crackers, sauces, ice and otherpastries. The purpose of this study is to investigate and determine the levels of rhodamine Bon crackers circulating in the city of Manado. Sampling sites crackers are 4 market in the cityof Manado is Tuminting Market, Market Paal 2, Market Market Bersehati 45 and 45.Samples with ammonia to soak interesting rhodamine B dye using wool yarn, followed byidentification using thin layer chromatography (TLC) and then in detection with UV light 254nm and 366 nm. The reading levels of rhodamine B using UV-Vis spectrophotometry. Theresults showed that of the ten samples tested with three times obtained one positive samplecontaining rhodamine B with an average concentration value of rhodamine B in a sample of45 markets in the sword of 0.28 mg/ml. Abstract

ABSTRACTWhite galangal rhizome (Alpinia galanga (L.) Willd's) scientifically proven has properties as antibacterial, antifungal, anticancer, antitumor, antioxidant and cytotoxic. In nano size, the contact surface area of the particle becomes larger which can increase the amount of active substance which is isolated more so that the antibacterial activity is stronger. This study aims to determine whether the white galangal extract nanoparticles have antibacterial activity against the bacteria Klebsiella pneumoniae and measure the inhibitory ability of white galangal extract nanoparticles against bacterial growth and compared with white galangal ethanol extract. White galangal was extracted using maceration method with ethanol solvent and nanoparticle formulation made of white galangal extract by ionic gelation method. The antibacterial activity test was carried out using agar diffusion method and observations were made 1x24 hours incubation period, with inhibition zones measured using a millimeter ruler. The results showed that the white galangal extract nanoparticles had antibacterial activity to inhibit the bacteria Klebsiella pneumoniae, with the results of inhibition zone measurements of 22.5 mm.Keywords : Antibacterial, Extraction, Klebsiella pneumoniae, Nanoparticles, White galangal. ABSTRAKRimpang lengkuas putih (Alpinia galanga (L.) Willd) memiliki khasiat yang sudah dibuktikan secara ilmiah sebagai antibakteri, antijamur, antikanker, antitumor, antioksidan dan sitotoksik. Dalam ukuran nano, luas kontak permukaan partikel menjadi lebih besar yang dapat meningkatkan jumlah zat aktif yang terisolasi lebih banyak sehingga aktivitas antibakteri semakin kuat. Penelitian ini bertujuan untuk mengetahui apakah nanopartikel ekstrak lengkuas putih memiliki aktivitas antibakteri terhadap bakteri Klebsiella pneumoniae serta mengukur kemampuan daya hambat nanopartikel ekstrak lengkuas putih terhadap pertumbuhan bakteri dan dibandingkan dengan ekstrak etanol lengkuas putih. Lengkuas putih diekstraksi menggunakan metode maserasi dengan pelarut etanol dan dibuat formulasi nanopartikel ekstrak lengkuas putih dengan metode gelasi ionik. Untuk pengujian aktivitas antibakteri dilakukan dengan metode difusi agar dan pengamatan dilakukan 1x24 jam masa inkubasi, dengan zona hambat diukur menggunakan penggaris millimeter. Hasil penelitian menunjukkan bahwa nanopartikel ekstrak lengkuas putih memiliki aktivitas antibakteri untuk menghambat bakteri Klebsiella pneumoniae, dengan hasil pengukuran zona hambat sebesar 22,5 mm.Kata Kunci : Lengkuas putih, Nanopartikel, Antibakteri, Ekstraksi, Klebsiella pneumoniae.

Abstract

ABSTRACT DNA transformation is the process of inserting recombinant DNA into host cells via vector plasmid. The host cell that is often used is TOP-10 Escherichia coli. The transformation method is widely used to transfer plasmids containing genetic material. This study aimed to evaluate the results of plasmid transformation containing the merB gene in the Escherichia coli TOP-10 bacteria. This study initiated with identification of the microbiology of host cells to be used, namely Escherichia coli TOP-10. Escherichia coli TOP-10 host cells were made into competent cells by transforming plasmids containing merB gene into Escherichia coli TOP-10 host cells using the heat shock method. The transformation results were evaluated by observing at the growth of Escherichia coli TOP-10 colonies on agar LB media containing ampicillin antibiotics. Plasmids on Escherichia coli TOP-10 were isolated and analyzed by 1% agarose gel electrophoresis. The results showed that the transformation of plasmids containing merB in Escherichia coli TOP-10 bacteria was successfully carried out as indicated by the growth of Escherichia coli TOP-10 bacteria on LB media agar containing ampicillin and the visualization on agarose gel resulted that the plasmid which carried the merB gene could be transformed in to the E. coli TOP-10 bacteria cell. Keywords: Transformation, Plasmids, merB genes, heat shocks, E. coli TOP-10ABSTRAK Transformasi DNA merupakan proses memasukkan DNA kedalam sel bakteri. Metode transformasi dipakai secara luas untuk mantransfer plasmid yang mengandung bahan genetika. Penelitian ini bertujuan untuk mengevaluasi hasil transformasi plasmid yang mengandung gen merB pada bakteri Escherichia coli TOP-10. Penelitian ini didahului dengan identifikasi secara mikrobiologi bakteri Escherichia coli TOP-10. Bakteri Escherichia coli TOP-10 dibuat menjadi sel kompeten yang digunakan sebagai inang. Selanjutnya dilakukan transformasi plasmid yang mengandung gen merB kedalam sel inang E. coli TOP-10 menggunakan metode heatshock. Hasil transformasi dievaluasi dengan melihat adanya koloni E. coli TOP-10 pada media LB agar yang mengandung antibiotik ampisilin. Plasmid pada E. coli TOP-10 diisolasi dan dianalisis dengan elektroforesis gel  agarose 1%. Hasil penelitian menunjukkan bahwa transformasi plasmid yang mengandung gen merB pada bakteri Escherichia coli TOP-10 berhasil dilakukan, ditunjukkan dengan adanya pertumbuhan bakteri E. coli TOP-10 pada media LB agar yang mengangandung ampisilin dan hasil visualisasi pada agarose gel terlihat bahwa plasmid yang membawa gen merB dapat ditransformasikan ke dalam sel bakteri E. coli TOP-10. Kata kunci : Transformasi, Plasmid, gen merB, heatshock, E. coli TOP-10

Abstract

ABSTRACTWhite galangal rhizome is a plant that has properties such as antifungal and antibacterial. White galangal rhizome contains of active compounds, namely flavonoid, phenol and terpenoid compounds which can inhibit microbes. This study aimed to determine the antibacterial activity of ethanol extracts of white galangal rhizome against Klebsiella pneumoniae urine isolate in patients with urinary tract infections. White galangal rhizome plants were extracted using 96% ethanol solvent. Antibacterial activity test was performed using the disc and wells method. The result showed that ethanol extracts of white galangal rhizome has antibacterial activity against the bacteria Klebsiella pneumoniae. In the disc method with concentrations of 80%, 60%, 40%, 20% and 10% the diameter of the inhibition zone formed are 9,8 mm; 9 mm; 7,8 mm; 7,9 mm and 7,7 mm. Inhibition of the disc method is categorized as medium because the diameter of the zone formed is only around an average of 5-10 mm. Whereas in the wells method for concentrations of 80%, 60%, 40%, 20% and 10% the diameter of the inhibition zone are 11,3 mm; 10,3 mm; 9,3 mm; 6,3 mm and 2,6 mm. The inhibitory ability at concentrations of 80% and 60% is categorized as strong because it has an average of 10-20 mm, for concentrations of 40% and 20% are categorized as medium, because it has an average inhibition zone diameter of 5-10 mm, and for concentration of 10% is categorized as weak because it has a inhibition diameter <5 mm.Keywords: White Galangal Rhizome (Alpinia galanga L. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak etanol rimpang Lengkuas putih terhadap bakteri Klebisella pneumoniae isolat urin pada penderita infeksi saluran kemih. Tanaman rimpang Lengkuas putih di ekstraksi dengan menggunakan pelarut etanol 96%. Uji aktivitas antibakteri dengan metode cakram dan sumuran. Hasil penelitian menunjukkan bahwa ekstrak etanol rimpang Lengkuas putih memiliki aktivitas antibakteri terhadap bakteri Klebsiella pneumoniae. Pada metode cakram dengan konsentrasi 80%, 60%, 40%, 20% dan 10% diameter zona hambat yang terbentuk ialah 9,8 mm; 9 mm; 7,8 mm; 7,9 mm dan 7,7 mm. Penghambatan pada metode cakram dikategorikan sedang karena diameter zona yang terbentuk hanya berkisar pada rata-rata 5-10 mm. Sedangkan pada metode sumuran untuk konsentrasi 80%, 60%, 40%, 20% dan 10% diameter zona hambatnya ialah 11,3 mm; 10,3 mm; 9,3 mm; 6,3 mm dan 2,6 mm. Kemampuan penghambatan pada konsentrasi 80% dan 60% di kategorikan kuat karena memiliki diameter zona hambat rata-rata 10-20 mm, untuk konsentrasi 40% dan 20% di kategorikan sedang, karena memiliki diameter zona hambat rata-rata 5-10 mm, dan untuk konsentrasi 10% di kategorikan lemah karena memiliki diameter zona hambat <5 mm.Kata Kunci: Rimpang lengkuas putih (Alpinia galanga L. Abstract

ABSTRACT Galangal rhizome traditionally used to treat diseases. Red galangal rhizome contains flavonoid compounds, which are thought to be able to inhibit the growth of bacteria. Urinary tract infections are infections that occur due to the proliferation of microorganisms in the urinary tract. This study aims to determine the antibacterial activity of non-polar extracts of red galangal (Alpinia purpurata K.Schum) against the growth of Klebsiella pneumoniae bacterial urine isolates in urinary tract infections. Extraction was carried out by maceration method using n-hexane solvent. The antibacterial activity was testing using the paper disc diffusion method (Kirby and Bauer diffusion). The results showed that the inhibitory activity of extracts with concentrations of 80%, 40%, 20%, and 10% of the average diameter of the clear zone were 11.03 mm, 8.75 mm, 7.1 mm and 6.03 mm. Shows that the inhibition at 80% concentration is strong, while at 40% concentration, 20% is moderate and at 10% concentration is moderate. Based on this study it can be concluded that the non-polar extract of red galangal can inhibit the bacteria Klebsiella pneumoniae urine isolate in urinary tract infections. Keywords: Red Galangal Rhizome, Antibacterial, Inhibition  ABSTRAK Rimpang lengkuas secara tradisional digunakan untuk mengobati penyakit. Rimpang Lengkuas merah mengandung senyawa golongan flavonoid yang diduga mampu menghambat pertumbuhan bakteri. Infeksi saluran kencing merupakan infeksi yang terjadi akibat berkembangbiaknya mikroorganisme di dalam saluran kemih. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari ekstrak non-polar rimpang lengkuas merah (Alpinia purpurata K.Schum) terhadap pertumbuhan bakteri Klebsiella pneumoniae isolat urin pada infeksi saluran kemih. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut n-heksan. Pengujian aktivitas antibakteri menggunakan metode difusi dengan cakram kertas (difusi Kirby dan Bauer). Hasil penelitian menunjukkan bahwa daya hambat ekstrak dengan konsentrasi 80%, 40%, 20%, dan 10% rata-rata diameter  zona beningnya 11,03 mm, 8,75 mm, 7,1 mm, dan 6,03 mm. Menunjukkan bahwa daya hambat pada konsentrasi 80% termasuk kuat, sedangkan pada  konsentrasi  40%,20% termasuk sedang dan pada konsentrasi 10% termasuk  sedang. Berdasarkan penelitian ini dapat disimpulkan bahwa ekstrak non-polar lengkuas merah dapat menghambat bakteri Klebsiella pneumoniae isolat urin pada infeksi saluran kemih. Kata Kunci: Rimpang Lengkuas Merah, Antibakteri, Daya Hambat

Red galangal rhizomes contain a class of flavonoids, phenols and terpenoids, which can be, used as basic ingredients of modern medicines. Flavonoids and phenols are also known to inhibit microbes. Flavonoids can inhibit microbes that have been resistant to antibiotics. This study aims to determine the antibacterial activity of red galangal rhizome extract (Alpinia purpurata K.Schum) and test its inhibitory effect on the growth of Klebsiella pneumoniae bacteria in sputum isolates in patients with pneumonia resistant ceftriaxone antibiotics. Red galangal rhizome plant was extracted by maceration method using 96% ethanol. Antibacterial inhibitory tests were carried out using the disc and well method. The results showed that the red galangal extract of rhizome had a inhibitory effect on the bacterium Klebsiella pneumoniae. On the disc method for concentrations of 100%, 75%, 50%, 25% and 12.5% the diameter of the inhibition zone is 9.6 mm, 8.68 mm, 8.5 mm, 8.5 mm and 7.6 mm. The inhibition ability of bacterial growth on the disc method is categorized as moderate because it has an average inhibition zone diameter of 5-10 mm. Whereas in the well method for concentrations of 100%, 75%, 50%, 25% and 12.5% the diameter of the inhibition zone was 25.8 mm, 20 mm, 15.6 mm, 12.7 mm and 9.7 mm, respectively. The inhibition ability of bacterial growth in the well method for 100% concentration is categorized as very strong because it has an average inhibition zone diameter of >20 mm, for concentrations of 75%, 50% and 25% it is categorized as strong because it has an average inhibition zone of 10- 20 mm and for a concentration of 12.5% it is categorized as moderate because it has an average inhibition zone diameter of 5-10 mm.Keywords: Red galangal rhizome, Alpinia purpurata K.Schum, Klebsiella pneumoniaABSTRAKRimpang Lengkuas merah merupakan tanaman yang memiliki berbagai khasiat di antaranya sebagai antijamur dan antibakteri. Rimpang Lengkuas merah mengandung golongan senyawa flavonoid, fenol dan terpenoid yang dapat digunakan sebagai bahan dasar obat-obatan moderen. Senyawa flavonoid dan fenol juga diketahui dapat menghambat mikroba. Flavonoid dapat menghambat mikroba yang telah resisten terhadap antibiotik. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak rimpang Lengkuas merah (Alpinia purpurata K.Schum) dan menguji daya hambatnya terhadap pertumbuhan bakteri Klebsiella pneumoniae isolat sputum pada penderita pneumonia resisten antibiotik seftriakson. Tanaman rimpang Lengkuas merah di ekstraksi dengan metode maserasi menggunakan pelarut etanol 96%. Uji daya hambat antibakteri dilakukan dengan metode cakram dan sumuran. Hasil penelitian menunjukkan bahwa ekstrak rimpang Lengkuas merah memiliki daya hambat terhadap bakteri Klebsiella pneumoniae. Pada metode cakram untuk konsentrasi 100%, 75%, 50%, 25% dan 12,5% diameter zona hambatnya ialah 9,6 mm, 8,68 mm, 8,5 mm, 8,5 mm dan 7,6 mm. Kemampuan penghambatan pertumbuhan bakteri pada metode cakram di kategorikan sedang karena memiliki diameter zona hambat rata-rata 5-10 mm. Sedangkan pada metode sumuran untuk konsentrasi 100%, 75%, 50%, 25% dan 12,5% diameter zona hambatnya ialah 25,8 mm, 20 mm, 15,6 mm, 12,7 mm dan 9,7 mm. Kemampuan penghambatan pertumbuhan bakteri pada metode sumuran untuk konsentrasi 100% di kategorikan sangat kuat karena memiliki diameter zona hambat rata-rata >20 mm, untuk konsentrasi 75%, 50% dan 25% di kategorikan kuat karena memiliki diameter zona hambat rata-rata 10-20 mm dan untuk konsentrasi 12,5% di kategorikan sedang karena memiliki diameter zona hambat rata-rata 5-10 mm.Kata Kunci:Rimpang Lengkuas merah, Alpinia purpurata K.Schum, Klebsiella pneumonia
