Abstract

This research was conducted to investigate the effect of chorionic gonadotrophin addition into maturation medium on oocyte maturation, fertilization, and embryo development in vitro of Ettawa crossbred. Oocytes were divided into 3 groups, group I: maturation medium without addition of chorionic gonadotrophin (0), group II: 10 μl/10 ml chorionic gonadotrophin was added into maturation medium (1), group III: 20 μl/10 ml chorionic gonadotrophin was added into maturation medium (2). Oocytes were transferred into 50 μl maturation medium, then covered by mineral oil. Oocyte was incubated at 39oC, 5% CO2, 95% humidity for 24 hours for maturation. Matured oocytes were inseminated with frozen semen–thawed concentration 12.5 x 106/ml. Process of fertilization were carried out on incubator 39oC, 5% CO2, 95% humidity for 5 hours. The fertilized oocytes were transferred into 50 μl drop G–1, then incubated at 39oC, 5% CO2, 95% humidity. Embryo development was monitored every 24 hours. Culture medium was changed every 48 hours. G–2 medium used second day after culture. The variables measured involved oocyte maturation, fertilization, and in vitro cleavage rate. The data were analyzed by chi–square, using SPSS 15.0 program. The result showed no significant difference on the percentage of mature oocytes and fertilization rate were 78.0%, 72.8%, 75.0% and 76.6%, 74.5%, 77.8% respectively. But cleavage rate showed significant difference (P≤0.05) withthe values of 40.8%, 11.4%, and 12.2% respectively. Based on the result it could be concluded that chorionic gonadotrophin addition into maturation medium had not increased ettawa crossbred oocytes maturation, fertilization, and in vitro cleavage rate. The best maturation, fertilization, and in vitro cleavage rate were found using maturation medium without any addition of chorionic gonadotrophin.(Key words: Does oocyte, Chorionic gonadotrophin, In vitro maturation, In vitro fertilization, In vitro embryo development)

Abstract

Penelitian ini bertujuan untuk mengetahui hubungan antara kadar hormon testosteron, libido, dan kualitas sperma kambing Bligon, Kejobong, dan Peranakan Etawah (PE). Sampel terdiri dari 3 ekor kambing Bligon, 3 ekor kambingKejobong, dan 3 ekor kambing PE jantan. Kadar testosteron diukur menggunakan metode enzyme-linked immunosorbent assay (ELISA). Tingkat libido diketahui dengan pengamatan waktu pertama mencumbu betina, menaiki betina, sampai ejakulasi. Penampungan sperma menggunakan metode vagina buatan. Data yang diperoleh kemudian ditentukan nilai korelasi antara kadar testosteron dengan libido dan kualitas sperma. Hasil menunjukkan testosteron pagi kambing Bligon berkorelasi terhadap waktu mencumbu betina (R= -0,79 dan Y= 20,91-6,52X), menaiki betina (R= -0,80 dan Y= 20,28-7,16X), ejakulasi (R= -0,81 dan Y= 14,03-7,56X), volume sperma (R= 0,65 dan Y= -0,78+0,03X), dan motilitas spermatozoa (R= 0,70 dan Y= -73,83+1,43X) (P<0,01). Testosteron pagi kambing Kejobong berkorelasi terhadap waktu mencumbu betina (R= -0,75 dan Y= 14,96-0,68X), menaiki betina (R= -0,69 dan Y= 21,59-0,86X), ejakulasi (R= -0,66 dan Y= 28,51-0,83X), dan konsentrasi spermatozoa (R= 0,59 danY= 430,73+12,75X) (P<0,01), sedangkan testosteron sore berkorelasi terhadap waktu mencumbu betina (R= -0,90 dan Y= 21,50-1,06X), menaiki betina (R= -0,90 dan Y= 24,89-1,16X), dan ejakulasi (R= -0,84 dan Y= 34,27-1,14X)(P<0,01). Testosteron sore kambing PE berkorelasi terhadap waktu mencumbu betina (R= -0,72 dan Y= 12,83-7,31X), menaiki betina (R= -0,72 dan Y= 9,11-7,67X), ejakulasi (R= -0,69 dan Y= 2,02-8,25X), viabilitas spermatozoa(R= 0,77 dan Y= -90,99+2,20X) dan konsentrasi spermatozoa (R= 0,72 dan Y= -452,10+9,56X) (P<0,01). Disimpulkan bahwa terdapat korelasi antara kadar hormon testosteron dengan tingkat libido dan kualitas sperma, serta terdapatvariasi korelasi diantara bangsa kambing.(Kata kunci: Korelasi, Testosteron, Libido, Kualitas sperma, Kambing)

Abstract

This experiment was conducted to determine the correlation of progesterone and cortisol levels in plasma between ovulated and non-ovulated Crossbreds Ettawa does. Eight does were used in this experiment and they were divided equally into 2 groups, i.e. group I consisting of 4 normal ovulated does with the average body weight of 37.5 ± 3.109 kg and group II consisting of 4 non-ovulated does with the average body weight of 28 ± 2.160 kg as group II. The estrus cycles of all does were synchronized using intravaginal device of controlled internal drug release (CIDR) accompanied by intra-muscular injection of prostaglandin F2 (PGF2) alpha. Immediately after the onset of estrus, blood samples were collected from jugular every 3 and 6 hours for 72 hours after onset of estrus. The concentration of cortisol and progesterone in plasma was assayed by enzyme linked immunosorbent assay (ELISA). The results showed that the average concentrations of cortisol in ovulated does (90.89±26.22 ng/mL) was higher than in non-ovulated does (42.70±37.18 ng/mL). Similarly, the concentrations of progesterone in ovulated does (0.098±0.0423 ng/mL) was higher than in non-ovulated does (0.093±0.056 ng/mL). It was evident that the change in progesterone level was closely associated with the change in cortisol level in plasma.

Abstract

Transportated animals may subject to a variety of physical stimuli including metabolism, crowding,noise, handling, isolation, agitation, and extreme temperature .The aim of this study was to determine thechanges of serum T4 and T3 concentration, during animals transportation. Six adult Bligon buck with bodyweight ranging from 26-30 Kg were used in this study. Two weeks prior to the experiment, the animalswere given anthelmintic Albendazole to eliminate egg worm. All animals were fed standard diet in theirpen at 10% of their body weight per head daily and commercial concentrate also given everyday. Freshwater was provided ad libitum. All animals were transported around village for 16 hours starting from18.00 pm until 10.00 am in open small truck (3 x 2 m); eye contact each others would be possible. Bloodsamples were withdrawn from jugular vein using vacutainer tubes containing heparin into 1.5 mL glasstubes, then centrifuged at 500 g for 15 minutes. Plasma was collected to be stored at –20° C. The bloodwere collected every 4 hours from 8 hours before transportation ( at 10.00 am, 14.00 pm and 18.00 pm)until the time of arriving after transportation at 10.00 am. Plasma was harvested and stored at –20° Cuntil T3 and T4 concentrations were measured using ELISA method (enzyme linked immunosorbentassay) product DRG, Germany. The result showed that transportation of Bligon bucks for 16 hours havean affect on level of T4 only (P<0.05) and not for T3 concentration (P>0.05) due to physical stimuli such ascrowding, heat stress, noise, handling would be discarded so that the metabolic process was stable. Duringtransportation, decreasing of T4 levels indicated conversion of T4 to T3 to form active hormone.

Abstract

The timing of artificial insemination relies largely on behavioral observation of estrus. The problemfaced is that not all cattle shows signs of estrus significantly, which affect the accuracy of insemination,and therefore,the success rate of Artificial Insemination is less than 50%. Recently, the determination ofestradiol levels as an indicator of estrus is done by observation of physical signs and doing ELISA test,which is expensive and provide longer time. In order to solve these problems, a tool estrus detector is madenamely Electronic Nose (EN). Determination of estrus with EN is cheaper because it does not need to usecomplexmaterials, just only use the samples. Mechanism of action of EN is using a sensor that is vaporized,while animals estrus will emit pheromones that are vaporized. Theaim of this study was to determinewhether the stage of estrus can be detected by using EN. Urine of female Ongole Crossbred which maintainedin Kuwang, district of Cangkringan, Yogyakarta, with BCS of 3 was used in this research. The sample wascollected shortly before injection of dinoprost as estrus synchronization then it repeated when cattle got estrus phase. The urinary sample of the estrus cattle was sensitive to methane, propane, butane, whereasin non-estrus cattle, besides the three of these component (methane, propane, butane), sensor was alsocaptured hydrogen sulfide. Furthermore, our electronic nose had been able to distinguish estrus phase andnon-estrus based on domain area.Thus, the Electronic Nose is very prospect used as a detector of estrus incattle. Hydrogen disulfide could possibly be used as an indicator comparison between cow estrus and nonestrus.

Abstract

A study was carried out to determine the serum estradiol and progesterone concentration, vulvaappearance, and vaginal cytology during estrous cycle in bligon goat. Four female non pregnant bligongoats (Indonesian indigenous) were used in this research. They were clinically healthy and showed normalestrous cycle. Estrous state was examined by using biological method. When the animals showed acceptanceto be mounted by a buck, they were confirmed as the first day of estrous cycle (estrous phase). Collectionof blood sample, preparation of vaginal smears, and measurement of some physical genital parameterswere conducted by standard veterinary clinical methods at day 11, day between 3-5, day between 6-16, andday between 17-19 of estrous cycle. Enzyme linked immunosorbent assay (ELISA) was used to determineserum progesterone and estradiol concentrations. Mean difference of each parameter was analyzed byanalyses of variance (Anova), followed by Tuckey HSD. The serum estradiol concentration ranged between211.25-247.77 pg/ml. Peak of the estradiol was occurred at the first day of estrous phase, namely 247.77pg/ml and to decrease at day 3-16. The serum progesterone concentration was at the level of 0.21-0.70 ng/ml and showed a significant increase at day 6-16 (p<0.05). The specific physical genital changes (swellingvulva, reddish mucous membrane of vulva, and excretion of viscous transparent vaginal discharge) wereidentified as the animals were at the estrous phase. Composition of exfoliative vaginal cytology did notshow any specific and consistent changes throughout the estrous cycle. Superficial and intermediate cellswere very dominant (>60%) during estrous cycle. In conclusion, serum estradiol, progesterone, the vulvaappearance combined with sexual receptivity could be a good estrous predictor for female cycling bligongoat.

Abstract

The objective of this research was to evaluate metabolism of inorganic phosphat (Pi) and calcium (Ca) in kacang goats subjected to 60-d feed restriction and 60-d full feeding (refeeding). Nine adult male goats were divided into three groups used in this study. Restrictions include 40% (group I), 50% (group II), and 60% (group III) of the ration of dry fodder. To obtain the baseline all animals from all groups were fullfeeding from the start of the study (day 0) to day 60 then followed by restriction stage on day 61 to day 120. The next stage is refeeding from day 121 to 180. Pi and Ca levels were analyzed on days 60, 120, and 180, respectively. The results showed that restriction followed by refeeding decreased Ca levels in all three groups and Pi of animals in groups II and III was below the baseline. The level of Pi reach above the baseline were seen only in animals in group I following restriction and refeeding, although there was a tendency to decrease after refeeding. Based on the results of the study it is concluded that the restriction followed by refeeding decreases the level of Pi and Ca in adult male kacang goats. ABSTRAK Tujuan penelitian ini adalah mengkaji metabolisme fosfat inorganik (Pi) dan kalsium (Ca) selama 60 hari restriksi (pembatasan) pakan yang diikuti dengan 60 hari pemberian pakan kembali secara fullfeeding (refeeding) pada kambing kacang jantan dewasa. Sembilan ekor kambing kacang jantan dewasa terbagi menjadi tiga kelompok digunakan dalam penelitian ini. Restriksi meliputi 40% (kelompok I), 50% (kelompok II), dan 60% (kelompok III) dari 3,5% bahan kering dari bobot badan. Tahap awal sebagai baseline dimulai dengan pemberian pakan fullfeeding pada hari ke-0 sampai hari ke-60 dilanjutkan dengan tahap restriksi dimulai pada hari ke-61 sampai hari ke-120. Tahap berikutnya adalah refeeding mulai hari ke-121 sampai dengan hari ke-180. Level Pi dan Ca dianalisis pada hari ke-60, 120, dan 180. Hasil penelitian menunjukkan bahwa restriksi yang diikuti refeeding menurunkan level Ca pada ketiga kelompok dan Pi kelompok II dan III di bawah baseline. Level Pi berada di atas baseline hanya pada kelompok I setelah restriksi dan refeeding, meskipun cenderung menurun setelah refeeding Berdasarkan hasil penelitian disimpulkan bahwa restriksi yang diikuti dengan refeeding menurunkan level Pi dan Ca pada kambing kacang jantan dewasa.

Abstract

Neurotransmitter found in the enteric nervous system that plays an important role in a variety ofenteropathies, including inflammatory bowel disease. Alteration of nitrergic neurons has been reported to bedependent on the manner by which inflammation is caused. This study was performed to determine theprofile of neural nitrergic with Nicotinamide Adenine Dinucleotide Phosphate-diaphorase (NADPH-d)staining in the cecum of broiler infected by Eimeria tenella (E. tenella). Thirty six health free of coccidiosisbroiler one-day old chickens (DOC) used in this study. All of broiler adapted for 7 days with food and drinkgiven ad libitum then divided into three groups, each group consisted of 12 broilers. The 1st group wascontrol only given orally 1.0 ml of distilled water/ heads , while 2nd group was infected with a single dose of5x103 oocysts/ head orally, and 3rd group was infected with a single dose of 2x104 oocysts/ head orally. Onday 7 post infection, all of chickens were fasted for 12 hours then were euthanized and cecum was taken.Lesion score of cecum was assessed. Furthermore, tissue of the coecum was prepared for hisochemicalusing NADPH-d staining to determine morphology, and morhometric of nitrergic neurons. The resultshown that cecum lesion score of group I is 0, group II is +2, group III is +3. Neuron density in the cecum ofgroup I is 2794.96+4.65 neuron/cm2, group II is 4930.36+63.73 neuron/cm2 and group III is 7892.31+44.97 neuron/cm2. Neuron density of group II and III increased signifcantly (p<001) than group I. Nitrergic neurondesity of group III increased significantly (p<0.05) than that of group II. It was concluded that the infectionof E.tenella led to increase the number of neutrl mienteric nitrergic of the cecum. The higher lesion score ofcecum led to increase the number of mienteric nitrergic neuron.
