Abstract

Xylocarpus granatum is an Indonesian plant that has bioactives content of phenols and high antioxidant activity on it. The aim of this research was to determine the optimum condition maceration for Xylocarpus granatum stem as antioxidant and antiglycation. The optimum conditions of maceration were effected by the extraction variables (concentration, sample/solvent ratio, extraction time) were evaluated using surface response method. The optimum condition was determined from the recovery of the respons. The optimum condition of maceration is predicted to be achieved when the solvent concentration is 52.25%, the extraction time is 15.92 hours, sample/solvent ratio is 1 g/9 ml with the response (yield, total phenol content, flavonoid, inhibition for 2,2’-diphenylpicryl hydrazyl, capacity of 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), and inhibition antiglycation were 12.81%, 1.95 mg of gallic acid/g extract, 62.33 μg quercetin/g extract, 41.11%, 0.71 TEAC, and 112.33%, respectively). Optimization extraction conditions shows that the extraction variables have significant effect on respons so it can reduce the extraction time, economic, and produce high bioactivite constituens.

Abstract

The medicinal plants Centella asiatica, Curcuma xanthorrhiza, Guazuma ulmifolia, and Hydrocotyle verticillata are widely used in Indonesian traditional medicine, but little is known about their associated endophytic fungi. This research aimed to study the diversity of endophytic fungi derived from functional parts of these plants and to evaluate their potential as antifungal agents against the plant pathogenic fungus Fusarium oxysporum. A total of 17 isolates of endophytic fungi were obtained: nine from leaves of G. ulmifolia, three each from leaves of C. asiatica and H. verticillata, and two from rhizomes of C. xanthorrhiza. The genus Colletotrichum was found in all plants studied, but each plant was associated with different species. Colletotrichum aeschynomenes was associated with C. xanthorrhiza, C. siamense was associated with C. asiatica, and C. tropicale was associated with G. ulmifolia and H. verticillata. The species Curvularia affinis, Diaporthe tectonae, Lasiodiplodia mahajangana, Parengyodontium album, Talaromyces trachyspermus, and Speiropsis pedatospora were found only in G. ulmifolia; while Didymella coffeae-arabicae and Muyocopron laterale were found only in H. verticillata. The endophytic fungi showed inhibition activity against F. oxysporum with inhibition values of 6.0-78.9%, T. trachyspermus JBd10 and C. affinis JBd14 gave the highest inhibition activity.

Abstract

Endophytic fungi play an important role on mangrove growth and development, however research on the endophytic fungi of the mangrove Rhizophora apiculata is limited. The endophytic fungi produce diverse bioactive compounds involved in the mangrove’s adaptation to varied biotic and abiotic stresses and could have applied uses in agriculture or medicine. The purpose of this research was to isolate endophytic fungi of Rhizophora apiculata mangrove and to study their activity as herbicides on the weed Gomphrena globosa. The fungi were isolated using the surface sterilization method and identified based on morphological characteristics and molecular characteristics using ITS regions of rDNA. Herbicidal activity of the fungal filtrates extracted by ethyl acetate were tested on seed germination and seedling growth of G. globosa. Five fungal isolates were obtained, namely Penicillium citrinum, Diaporthe eucalyptorum, Diaporthe musigena, Colletotrichum queenslandicum, and Diaporthe tectonae. All isolates were able to grow on PDA medium containing 0, 25, 50, 75, and 100 (% v/v) seawater concentrations, but the growth rate varied by species and seawater concentration. In general, all five isolates showed herbicidal activity by delaying seed germination and reducing shoot and root growth. P. citrinum showed the highest herbicidal activity compared to the other isolates. Analysis using Gas Chromatography-Mass Spectrometry of the crude extract of P. citrinum filtrate identified 7 main compounds: 3-Methoxy-2-methyl-cyclohex-2-enone, Cyclohexane-carbohexaldehyde, 6-methyl-3-(1-methylethyl)-2-oxo, Cyclopropane carboxylic acid, 1-(2-propenyl)-1,1-dimethylethyl ester, 2-Hydroxy-4-isopropyl-7-methoxytropone, Beta-Asarone, Oxane, 4-(2-amino-1-hydroxyethyl)-4-(3,4-dimethoxyphenyl)-, and Diisooctyl-phthalate. These compounds should be studied further to determine which ones are responsible for the herbicidal activity.

Abstract

Metabolites from clove (Syzygium aromaticum L.) have been extensively studied for various pharmacological uses, including quercetin. Endophytic bacteria isolated from plant tissues can produce similar secondary metabolites as their host plants. However, little is known about the antioxidant activity of the endophytic bacterial extract from clove leaves. Amongst the potential bacterial groups, the genera of Bacillus have been widely studied as a source of antioxidants. Thus, this study, we examined the potential antioxidant properties of Bacillus sp., isolated from clove leaves. We have successfully isolated ten endophytic bacteria, of which only one isolate (P1U2) was found non-pathogenic based on a hemolytic assay. Based on the 16S rRNA sequence, P1U2 shared the highest similarity to Fictibacillus phosphorivorans. The extract of extracellular metabolites of P1U2 contains quercetin compounds, based on LC-MS analysis. The results of the antioxidant activity test using the DPPH assay showed that the P1U2 extract had an IC50 value of 161 mg/ml. The supplementation of the extract on culture media reduced the ability of S. cerevisiae to decolorize mutagen malachite green and increased cell viability. Furthermore, the addition of 161 ppm extract was able to counteract UV mutagens effects of UV exposure and toxicity of malachite green, significantly increasing the growth of S. cerevisiae. Suggesting the bioactivity of the extract in combating mutagens. To the best of our knowledge, this is the first study to report a quercetin-rich extract derived from endophytic bacteria, F. phosphorivorans. Thus, further study is required to optimize the synthesis of quercetin from isolate P1U2.

Abstract

Coffee extracts are a rich source of potential compounds with numerous biological activities. This study aimed to investigate the potential of chemical compounds derived from robusta coffee extract (Coffea canephora) and its dominant compound, caffeine, to scavenge free radicals, inhibit bacterial growth and biofilm formation, and cytotoxic properties in human breast adenocarcinoma cell line. Chemical constituents of coffee extract were analysed quantitively for total phenolic, flavonoid, and alkaloid contents, along with Liquid Chromatography Quadrupole-Mass Spectrometry (LC/MS–MS) analysis. Antioxidant activity was measured by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis; 3-ethylbenzothiazoline-6-sulfonic acid (ABTS) methods. Antibacterial pair with antibiofilm properties against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, and Staphylococcus aureus were also determined, as well as cytotoxic activity on the MCF7 cell line model. The LC/MS–MS analysis of coffee extracts revealed high levels of caffeine; thus, the caffeine standard is used in all subsequent assays. Notably, robusta coffee extract showed remarkable antioxidant activity and selective inhibition of the growth against gram-positive and negative bacteria, along with the best inhibition of P. aeruginosa biofilm. However, compared with the caffeine standard, robusta coffee extract had lower cytotoxic activity and different bacterial targets in antibacterial and antibiofilm properties. Our results indicate that Robusta coffee extract is potentially a functional food due to its high alkaloid, phenolic, and flavonoid content and antioxidant activity, besides being used for natural compounds against bacterial infections.

Abstract

Purwoceng (Pimpinella pruatjan) is an Indonesian native plant but difficult to cultivate. Hydroponics can be used as a solution for purwoceng cultivation. Hydroponic types and nutrient concentrations can affect purwoceng’s productivity. This study aimed to determine total phenolic content (TPC) and total flavonoid content (TFC), changes in steroid, and antioxidant capacity due to two hydroponic types (nonrecirculating and recirculating drip) and three nutrient concentrations (1,000, 1,500, and 2,000 ppm). Purwoceng was determined for its moisture content, ash content, and dry weight, and extracted with ethyl acetate for gas chromatography-mass spectrometry analysis and with ethanol for TPC, TFC, and antioxidant capacity. The data were analyzed statistically and grouped using principal component analysis (PCA) and heat map analysis (HMA). Nonrecirculating drip with high nutrient concentration at the aerial part was the best treatment to produce high TPC, TFC, and antioxidant capacity. There were no significant changes in steroid content. PCA showed that purwoceng extracts could be grouped in to two groups, i.e. aerial and root groups. HMA showed that purwoceng extracts could be distinguished from the abundance of palmitic acid and stigmasterol. The antioxidant capacity of purwoceng was directly proportional to the TPC value and also related to the presence of phytol, stigmasterol, and palmitic acid.

Abstract

Actinobacteria are Gram-positive bacteria that can produce various secondary metabolites, including pigments that have potential as antioxidants. This study assessed the characteristics of yellowish-orange pigmented actinobacteria. The antioxidant activity of their extract pigment was tested by DPPH and analyzed by LC-MS. The identification results based on the 16S rRNA gene showed that the HRA isolate had 99.91% similarity value to Streptomyces gramineus JR-43, the HVA isolate had 99.22% similarities value to S. tendae ATCC 19812 and the HVB isolate had 81.18% similarities value to Rhodococcus ruber DSM 43338. Meanwhile, AGM 2.2 and 2.3 isolates had similarities to S. xanthophaeus NRRL B-5414 with similarity values of 98.95% and 99.82%, respectively. The Inhibitory Concentration (IC50) of actinobacterial crude extract pigments ranges from 53.38 µg/ml to 184.38 µg/ml. The HVB isolates with the major compound luteolin, have the highest antioxidant activity. The crude extract pigment of HVB isolates may consider rich in luteolin and has potency as an antioxidant.

Abstract

Identifying floral resources used by honey bees and stingless bees is essential for sustainable beekeeping and understanding pollination ecology. This study compared the diversity of floral resources utilized by the honey bee Apis cerana and stingless bees Geniotrigona thoracica and Heterotrigona itama based on pollen in honey. Bee colonies were in the same Flora Nauli Beekeeping area, Pematang Siantar, North Sumatra, Indonesia, ensuring equal access to similar surrounding floral resources. Honey samples were collected, and pollen grains were extracted, acetolysed, and analyzed by counting 1,200 grains to determine frequency classes. Pollen diversity was calculated using the Shannon–Wiener index (H′). Stingless bee honey contained more diverse pollen than A. cerana, which only contained pollen from the Arecaceae family. Heterotrigona itama and G. thoracica honey contained pollen from 20 and 11 plant families, respectively. Each species had a distinct predominant pollen type (>45%): Cocos nucifera (A. cerana), Sapotaceae type (G. thoracica), and Casuarinaceae type (H. itama). A higher pollen diversity index was also observed in H. itama (1.05-1.83) than in G. thoracica (0.34–1.64) and A. cerana (0.66). These results indicate that stingless bees are more generalist than honey bees, highlighting their ecological role in supporting pollination networks in tropical agroecosystems.
