Abstract

Strawberries were a lot of holticulture commodities consumed by the people of Indonesia. Strawberry consumption continues to increase over time in Indonesia but not matched by production. This it because the strawberry is native to subtropical climate so that production in Indonesia limited in the plateau regional still affected by the tropical climate. Therefore, it is necessary to increase the production of both quality and quantity of strawberries, one with plant breeding through the induction of colchicine. The objective of this research were to determine the differences in phenotypic characters of strawberry plants with the control and induction results to determine the length of time and the induction colchicine 0,05% and 0,01% on the Festival cultivar of strawberry most effective. Research were done by inducing with 0,05% and 0,01% colchicine in leaves, roots 24 hours and 36 hour, the root and leaves of 24 and 36 hour. The observed for 6 months long leaf growth, leaf width,number of leaves, stem circumference, plant height, bloom vast flower and volumes of the fruit. The results indicate that the induction of the leaf growth at the optimal parameter leaf lenghth, leaf width and number of leaves. While 24-hour induction of root growth at optimum height and volume of fruitplants, then root induction at 36 hour to flowers bloom. Induction in root and leaves 36 hour optimum growth on trunck circumference. From this study it could be concluded that there are differences in the character of the strawberry crop cultivar Festival phenotype induction results colchicine 0,05% and 0,01% with control. Induction for 36 hours and induction on leaves was most effective for induction of strawberry cultivar Festival.Keywords: character phenotype, colchicine, festival

Abstract

Wonosadi forest is managed forests by indigenous forest communities and is located in the hamlet of Duren and Sidorejo, Beji village, subdistrict Ngawen, Gunung Kidul regency, Yogyakarta. One of the biological richness contained in Wonosadi forest is very valuable natural orchids and need to be preserved. Management to the preservation of nature in the woods Wonosadi orchids can be done through natural orchid protection against infectious pathogens, including viruses. Odontoglossum ringspot virus (ORSV) is one type of orchid viruses infect most widely reported and has spread widely in the world, including in Indonesia. ORSV can cause severe damage to the orchid. Therefore, it is necessary to study to detect ORSV that infect wild orchids in the forest Wonosadi molecular basis for detecting the presence and pattern of infection ORSV as well as possible to reduce the severity of disease caused by infection with the virus. The purpose of this study is to assess the natural orchid ORSV infection in molecular Wonosadi forest that can be used as a database in an effort to protect against natural orchid Wonosadi forest. In the long term, the results of this study are very useful to support conservation efforts and determine appropriate conservation efforts for wild orchids in the forest Wonosadi. The initial phase of the study conducted a survey and collection of samples of wild orchids in the forest leaves that show symptoms of infection Wonosadi ORSV. Furthermore, molecular analysis performed with gene amplification Coat Protein ORSV using Reverse engineering Trancriptase Polymerase Chain Reaction (RT-PCR).Keywords: orchid soil, ORSV, RT-PCR, Wonosadi forest

Abstract

Melon (Cucumis melo L.) is one of favorite fruits for Indonesian. The fruit is sweet with good odor, and has a high nutrient content. Efforts to increase the production of melon can be reached by improving cultivation technology along with seeds that have better quality, such as resistance to pests and disease, and high production. The low production of melon in farmer fields is often caused by use of local varieties with less improvement technology in crop management. Faculty of Biology has developed a new melon cultivars called Gama Melon Basket. Test the stability of melon cultivars at various enviromental condition have been conducted, except for karst land. Therefore, the stability test of Gama Melon Basket phenotype character cultivated in karst land of Porok Beach of Gunungkidul Yogyakarta is required. The purpose of research was to study stability and uniformity of phenotypic character on Gama Melon Basket cultivar cultivated in karst land of Porok Beach, Gunungkidul, DIY and comparison of its phenotypic characters using modern, intermediate-modern, and traditional methods of cultivation. The research was carried out by cultivating Gama Melon Basket at Coastal Land of Porok Beach with modern, intermediate-modern and traditional cultivation methods. Every week phenotype characters of plants were observed until harvest. Results of quantitative characters of Gama Melon Basket showed that intermediate-modern cultivation method produced highest melon production compare to other methods.The result of Gama Melon Basket using intermediate-modern method is 1552 grams for average weight, fruit circumference is 46,19 cm, fruit diameter is 14,3 cm, fruit length is 14,08 cm, skin thickness is 0,6 cm and thick meat is 0,62 cm. The results revealed that intermediate-modern method is more advisable for melon cultivation in karst land of Porok Beach compared with other methods. Karst Land of Porok Beach has a potential for melon cultivation and production in Gunungkidul.Keywords: Gama Melon Basket, Gunungkidul, karst, phenotype, Porok beach

Abstract

Red ginger (Zingiber officinale Roxb. var. rubrum Rosc.) is one of the herb plants that often being used as a spice, traditional medicines, sweets, drinks, and material export commodities. The ginger’s rhizome is the most common because it is good for health freshness, and spice for food. The spicy from the ginger comes from ketone compounds ‘gingerol’. This research aims to produce polyploidy crops in red ginger by observing the changes of phenotype characters on the leaves, stems and rhizome resulted from colchicine induction. This research was based on experimental method with Randomized design with two factors, which were colchicine concentration (0,05%, 0,1% and 0,2%) and incubation time (6, 12 and 24 hours). Ploidy level was observed using flow cytometry technique. The data were analyzed using Analysis of Variance (ANOVA) and statistical data processing was done using F test at α = 5%. If the result shows any significant differences then was followed with Duncan Multiple Range Test (DMRT). The results showed that the change in the ploidy degree of red ginger was seen in K1W2 (0,05 % concentration of incubation time 12 hours) with increasing in ploidy from the diploid (2n) to mixoploid (2n= 2x+4x).

Abstract

Maize (Zea mays L.) is one of important food in Indonesia. Java Downy Mildew is a plant disease caused by Peronosclerospora spp. This specific fungus attacks corn plants, especially the leaves. The purpose of this study was to examine maize cultivars resistance to Java Downy Mildew.  This research was conducted at the land owned by Agrotechnology Innovation Center (PIAT) UGM located in Kalitirto Village, Berbah Subdistrict, Sleman Regency, Special Region of Yogyakarta. The main ingredients used in this study included maize with 7 different cultivars: Gama GS, Gama SG, Bisi 816, Pioneer 21, Lagaligo, Talenta, and Pulut. This research was conducted using Randomized Block Design with 7 treatments and 2 replications. The results showed that Talenta, Lagaligo and Bisi-816 were resistant to downy mildew. Pioneer-21 quite resistant to downy mildew. Cultivars Gama GS, Gama SG and Pulut were not resistant to downy mildew. This study provides new information related to the infection of downy mildew especially in Yogyakarta.

Abstract

Butternut pumpkin (Cucurbita moschata 'Butternut') is a horticultural commodity with great potential to be developed as food and medicine. Nutrients and unique shape cause butternut pumpkin to be popular. To supply of butternut pumpkin, required an increase in production which has superior and stable characters. This research aims to determine the stability phenotypic characters of butternut pumpkin in Sleman, Yogyakarta using quantitative data analysis of variance (One Way ANOVA) and qualitative analysis through descriptive phenotype. Based on the results, butternut pumpkin have a uniform phenotype characters but unstable in fruit’s shape and size of plant. Butternut pumpkin plants have 3 different shapes of pear or paprika form including dumbbell shape and gooseneck (pyriform), crispy texture, pale yellow (RHS greyed grange group 65) peel of fruit, strong orange (RHS orange group 24) flesh of fruit, 3-5°brix level of sweetness, long shelf life, and short harvest age (70-90 HST). The cultivated butternut pumpkin in PIAT UGM farmland has greater productivity of 3.1±0.99 kg/plant.

Abstract

Melon (Cucumis melo L.) usually used as a fresh fruit, which contained high vitamin C and in Indonesia melon is one of the favorite fruit. Indonesia is a country that has a high biodiversity. One of the potential strategies in order to improve productivity and quality of crops competitiveness is through plant breeding. This study used melon Aramis from PT East West Seed Indonesia hybrid melon from crosses between 7319 (F) with 7347 (M). The research aims to determine the phenotype character of melon cultivar Aramis and to detect powdery mildew resistance gene. The research is conducted in Pusat Inovasi Agro Teknologi (PIAT UGM) Yogyakarta and at the Laboratory of Genetics and Breeding Faculty of Biology, Universitas Gadjah Mada. Resistance genes detection is conducted by Sequence Characterized Amplified Regions method (SCARs) using PCR and showed negative result of Aramis cultivar on detection of gene resistance to powdery mildew.

Abstract

GK-Bro (Gallus gallus Linnaeus, 1758) is a hybrid chicken from crossing between ♀ F2 Broiler ♂ F2 Golden Kamper that have variation of phenotypic character. To meet the market’s demand, GK-Bro must have prime phenotypic character in both quantity and quality. This research aims to examine the phenotype character that occur in GK-Bro by the character of feet morphometry, crest’s shape, feather’s color, and feet’s color. The parental chicken are crossed in Innovation and Agriculture Centre of Gadjah Mada University (Indonesia) and produce 11 chickens with five males and six females filial. The filial are maintained intensively and qualitative data were observed at the end of seven weeks, classified based on the percentage of the phenotypic class. Quantitative characters that observed were tibia and femur length after seven weeks, using control pelung, broiler, and F1 broiler. Quantitative data showed that the average length of the tibia and femur of GK-Bro is lower when compared to the control but the ratio of feet morphometry is closer to pelung. Qualitative data classified crest form in one class, feather’s color in 4 classes and feet’s color in 3 class.

Abstract

On the recent classification of native Indonesian chicken, there are 31 breeds of chicken. Highly circulated chicken breed in Indonesia is fast-growing broiler that includes several strains such as Cobb 500, Hubbard, Hybro, Isa Hyline, and Hisex. Fast-growing broiler chickens have rapid growth, with excessive fat deposition on chicken carcass which implicates a health problem and unfavorable meat quality. Gama Ayam Research Team conducted selective breeding which produces two new chicken breed Kamper and Kambro. Further selective breeding then crossbred female F1 Kamper and male BC1 (backcross I) Kambro. In this research morphological traits, productive performance and genotyping of PPAR gene related to fat deposition gene and blood lipid content in Gama Ayam were identified. Based on data analysis hybrid chickens morphological traits it concluded that Gama Ayam has a significant variation based on feather color. The productive performance was determined with the feed-conversion ratio (FCR) value which was 3.17. Genotyping of PPAR gene resulted in four polymorphisms that formed 14 haplotypes groups. Based on blood lipid content analysis of cholesterol content, triglycerides content, HDL and LDL, Gama Ayam have significantly lower content of cholesterol (107.05 mg/dl), LDL (44.71 mg/dl) and triglycerides (22.41 mg/dl). PPAR gene polymorphisms are not correlated with blood lipid content in Gama Ayam. A significantly strong correlation between PPAR gene polymorphism on the bodyweight of Gama Ayam at 49-days-old. Triglyceride level, cholesterol level and LDL level in Gama Ayam were lower than broiler chicken. Further research with a larger population size and sex classification of hybrid chicken must be conducted to validate the results.

Abstract

In this study the genetic resource of Pelung chicken from Cianjur, West Java, Indonesia, was exploited. Pelung chicken has a higher body weight growth, unique meat flavor, and superior posture, compared with other indigenous breeds. Kamper chicken line selective breeding program was conducted, to increase the performance of Pelung breed by crossing with Layer Lohmann Brown-Classic. The Layer Lohmann Brown-Classic is an imported laying-type breed, which is widely known for its reproductive performance, based on the egg productivity. This study aims to use quantitative genetic method in estimating the commercial and reproductive traits' performance of Kamper chicken line. Based on commercial, phenotypic and reproductive traits, the progenies in Kamper chicken line have significant improvements, compared to the parental cross of Pelung and Layer Lohmann Brown-Classic. The quantitative genetic method was used in describing and underlying some phenomenon, in the selective breeding program. The results showed that the phenotypic and reproductive types of progenies in Kamper chicken, have significant improvements compared to the parental crossing of Pelung breed and Layer Lohmann Brown-Classic. Although quantitative genetic method is utilized in basic breeding program with significant precision and rapidness, it is only used in the preliminary study, for the advanced type. Therefore, the addition of quantitative trait loci (QTL), provide a more thorough genetic examination, and play a role in selective breeding program.
