Abstract

Tea is one of the most popular beverages in the world. Produced from Camellia sinensis leaves, tea has been studied to provide health benefits due to the content of important metabolites. This study aimed to investigate the catechin contents, antioxidant, and antibacterial activities of Indonesian tea varieties, namely green tea, black tea, and white tea. Tea infusion was prepared by extracting 1 g of each sample into 10 mL of distilled water and incubated at 80 °C for 60 min. The catechin and epigallocatechin gallate (EGCG) contents of tea extracts were determined using high-performance liquid chromatography (HPLC). Antioxidant activity was measured using the free radical method with 2,2-diphenyl-1-pycrylhidrazyl (DPPH), while antimicrobial activity was assesed using paper disc diffusion assay. The results indicated that green tea had the highest contents of catechin (646 ± 17.14 mg/L) and EGCG (997.8 ± 36.72 mg/L), and antioxidant activity with IC50 of 5.65 µg/mL. Furthermore, green tea and white tea extracts showed inhibitory activity against Gram-positive bacteria such as Micrococcus luteus, Bacillus subtilis, and Staphylococcus aureus whereas black tea had no activity against all bacterial strains tested. Generally, we concluded that white tea and green tea contributed to the higher content of catechins and exhibited strong antioxidant and antibacterial properties.

Abstract

Pectinases are a group of an enzyme that break down pectin, a polysaccharide that is found in plant cell walls. Today, the application of pectinolytic enzymes plays an important role in food technology for the maceration of fruits and vegetables, including for the extraction and clarification of juice. This research aimed to produce pectinase enzyme for clarifying of apple juice. A microbial culture was selected from cocoa bean fermentation samples and identified as Bacillus sp.. The highest enzyme activity was investigated after 48 hours of incubation. Citrus pectin as the carbon source and peptone as the nitrogen source was found as the best component for pectinase production. The optimum condition of pectinase activity was observed at pH 5, temperature 40 °C and the crude enzyme had the higher activity at one hour storage. Apple juice was treated with the enzyme at different concentrations (0%, 0.5%, 1%, 2%, 4%). Apple juice clarification was evaluated for its percent clarity and viscosity. The result showed that enzyme treatment at 4% in apple juice promoted juice clarification and decreased pH and viscosity. In conclusion, the quality of apple juice can be improved by enzymatic treatment using pectinase.

Abstract

Trichoderma is a well-known candidate to be promoted as cellulase producer for the hidrolysis of lignocellulose that contain in woody biomass. The number of trichodermal isolates in our laboratory collected from diverse ecosystem types in Indonesia increases significantly during the last 5 years. It is our aim to assess the cultures for its ability in producing cellulase. Sixty-six trichodermal isolates used in this experiment are obtained from Biotechnology Culture Collection (BTCC), Research Center for Biotechnology, Indonesian Institute of Sciences (LIPI). The 31 isolates were isolated from District of Liwa (South Sumatra, Indonesia) and the 35 isolates from District of Maros (South Sulawesi, Indonesia). The isolates were screened qualitatively, 7 isolates from Liwa and 12 isolates from Maros showed cellulolytic activity. From the results of quantitative test, two strains (ID08-T004 and ID08-T63) showed the higher cellulolytic activity among the selected strains, 133.5 and 133.5 U/ml, respectively. These extracellular enzymes were characterized their temperature and pH optimum. The temperature optimum for both enzymes was the same, 50C, with activity 213.6 U/ml for enzyme extracted from ID08-T004 and 197.3 U/ml for enzyme from ID08-T0063. The pH optimum was pH 5 of ID08-T004 with activity 137.7 U/ml and pH 6 for ID08-T063 with activity 75.0 U/ml. The enzymes from ID08-T004 and ID08-T063 were stable in their temperature and pH optimal condition even after 90 minutes incubation with activity 179.0 U/ml and 86.7 U/ml, respectively. The enzyme stability was approximately 150 minutes for both enzymes in the temperature and pH optimum.

Abstract

Biogrouting is a process that transforms soil or sand into calcarenite or sandstone by bacterial calcium carbonate precipitation . The mechanisms of this process are that the bacteria hydrolyze urea catalyzed by urease, and with the existence of dissolved Ca2+, a solid crystalline calcite or calcium carbonate (CaCO3) will be produced. The main advantage of biogrout is that soil or sand can be strengthened without interfering with the hydraulics of the treated soil or sand and without excavation or replacement. In this study, the isolation and identification of bacterial calcium carbonate precipitation , and characterization of urease produced by bacteria were conducted. In the isolation method, the enrichment method using urea as abacterial carbon source was carried out. The formation of crystalline calcite was observed by a light microscope. The urease enzyme activity was determined by the 3, 5-Dinitrosalicylic acid (DNS) method. The molecular identification of isolates was analyzed by the determination of 16S rRNA gene sequencing. As a result, 19 calcium carbonate precipitation have been isolated from soils, sands, water, and rocks collected from several areas in Indonesia. They showed calcite formation in a medium with urea. Molecular identification of isolates with high urease enzyme activity revealed that the isolates belong to the group Bacilli. The highest enzyme activity produced by ID10-U004 is 374.94 U/mL. The preliminary biogrouting experiment was also conducted using isolated strain, and the reaction was able to make the sand solid and stiff.

Abstract

Islamic boarding schools not only function as religious educational institutions, but also have a strategic role in producing economically independent students. This study aims to analyze the role of the Al Azhar Azzayyadiyah Islamic Boarding School Cooperative in developing students' entrepreneurial skills. This study uses a qualitative approach, with a case study approach, data collected through observation, in-depth interviews, and documentation. The results show that the Islamic boarding school cooperative functions as an experiential learning vehicle that equips students with technical skills (sewing, batik, knitting, and marketing), while also developing soft skills such as discipline, communication, leadership, and responsibility. These findings also show an increase in students' self-efficacy in managing businesses, in contrast to previous studies that highlighted students' weak self-confidence. This study proves that Islamic boarding school cooperatives are not only economic units, but also strategic educational tools in empowering students. These findings emphasize the importance of integrating cooperative programs into non-formal education to produce independent students who are ready to compete in the business world. This research contributes to the development of religious community-based entrepreneurship education theory. In practice, this research strengthens the position of Islamic boarding schools as agents of local economic development through systematic and sustainable empowerment of students.

Abstract

Modernity has brought about profound transformations in the understanding and practice of education. The dominance of instrumental rationality, technological advancement, and an increasing emphasis on efficiency and performance have driven education toward technocratic and pragmatic approaches, potentially diminishing its meaning as a process of holistic human formation. Within this context, the role of educators extends beyond fulfilling professional and administrative demands to confronting increasingly complex ethical tasks related to moral responsibility within pedagogical relationships. This article aims to critically examine the concept of educational ethics and to analyze the moral responsibility of educators in the modern era through the perspective of critical philosophy of education. The study employs a philosophical literature review using normative, hermeneutic, and reflective–critical approaches to both classical and contemporary educational thought. The findings indicate that educational ethics constitutes an ontological and axiological foundation that significantly shapes the direction and quality of pedagogical practice. Educators should not be viewed merely as technical implementers of curricula, but rather as reflective moral subjects responsible for safeguarding the dignity of learners as human beings. This article argues that strengthening educational ethics is an urgent necessity as a critical response to the crises of values, dehumanization, and alienation within modern educational practices, and it contributes to the development of a more humane and meaningful philosophical framework for education.

Abstract

This study examines the impact of religious moderation course services—namely course materials, learning strategies, and learning environment—on students' moderate behaviour, mediated by their attitudes. The research was conducted at UIN Walisongo with a sample of 215 students who had taken the course "Islam and Religious Moderation." The collected data were analyzed using regression analysis, F-test, t-test, and path analysis with the assistance of SPSS version 23. The findings reveal varying relationships among course materials, learning strategies, learning environment, attitudes, and moderate behaviour. Simultaneously, course materials, learning strategies, and learning environment significantly influence students' moderate attitudes. However, only learning strategies do not significantly affect students' moderate attitudes when examined individually. Furthermore, in different patterns, course materials, learning strategies, learning environment, and moderate attitudes collectively influence students' moderate behaviour, yet only learning strategies individually influence moderate behaviour. Additionally, only course materials and learning environment affect moderate behaviour through students' moderate attitudes. These findings underscore the importance of enhancing course services to foster students' moderate attitudes and behaviours, both directly and indirectly.
