Abstract

Metabolic syndrome is a group of several risk factors consist of hypertension, dyslipidemia, glucose intolerance disorder, obesity, and has a relationship with CVD pathomechanism. WHO was report in 2003 more than 300 million adult in the world suffered from obesity. The role of adipose tissue in infectious diseases and adipocyte quality changes after exposure to pathogen has not been revealed. On the other hand, the prevalence of infectious diseases in developing country is still high. Previous studies proved a relationship between infection of Toxoplasma gondii and tissue dysfunction. Profilin is a membrane component of Toxoplasma gondii that able to stimulate expression of proinflammation cytokines through TLR-11 upregulation, and predicted able to trigger the adipocyte dysfunction. The aim of this study are to knows the effect of profilin exposure in vitro on subcutaneous adipocyte to the expression of IL-6 and TNF-α. The measurement of IL-6 and TNF-a levels was done by ELISA technique. The result showed profilin exposure doses 5, 15 and 25 ng / ml increasing IL-6. Profilin exposure cause adipocyte dysfunction through the increasing proinflammatory adipocytokine levels as pathomechanism of metabolic syndrome associated with adipose tissue.

Abstract

Indonesia is considered as a mega-biodiversity country that has a massive amount of vascular and non-vascular plants. The tropical environment condition of Indonesia could support the growth of macrofungi. Information about edible macrofungi from the Forest of Lombok Island is based on limited data. This research aims to characterize the edible macrofungi at Kerandangan Protected Forest & Natural Park, West Lombok Regency, Indonesia. This research was a descriptive and explorative study. The edible mushrooms were observed through the Cruise method by following the particular track inside the forest. The sample found in the forest then documented and evaluated. A morphological analysis procedure was performed to assess the profile and similarity between the microscopic evaluations with the mushrooms identification book. In this study, we also offered a phylogeny analysis based on morphological characters similarity. The Dendogram tree was reconstructed using PAST 3.0. software. The result showed that there are eight species of edible mushrooms found that were group into Basidiomycota, namely, Termitomyces clypeatus, Termitomyces umkowaan, Termitomyces sp.1, Pleorotus flabelatus, Pleurotus ostreatus, Coprinus desimenatus, Tremella fuciformis, and Polyporus sp. All of them involved in Basidiomycota. Based on characterization and identification, the species are grouped into three Ordos and five Families. Most of the edible macrofungi found at Kerandangan Protected Forest & Natural Park growth at soil and deadwood.

Abstract

Porterweed (Stachytarpheta jamaicensis (L.) Vahl) contain many fl avonoids compound. It is a potential candidate to supress the inflammatory process through down regulation of TNF-α level as a potent pro-infl ammatory cytokine in chronic infl ammation cases. The aim of this research was to improve the effect of Porterweed leaf extract to the TNF-α level and considered the optimum dosage in wistar rats with chronic infl ammation model. Twenty fi ve rats was divided into negative control group, positive control group, and treatment groups with dosage 50, 100 and 150 mg/kg BW. Data obtained through microscopical histologic observation on aortic organafter immunohistochemistry staining and was analyzed descriptively. The result revealed that Porterweed leaf extract is able to inhibit the increasing of TNF-α level. This phytochemical substance is predicted capable of preventing the oxidation reaction progession ofcyclooxygenase-2 (COX-2) to produce prostaglandins. The decreasing of prostaglandin level causes a negative feedback to TNF-α production and iniciating the declining level of TNF-α. Thus, the Porterweed leaf extract indicates supress chronic infl ammatory process through down regulation of TNF-α and prostaglandin activity in optimum dose 150 mg/kg bw.

Abstract

Hepatocellular carcinoma (HCC) is one type of cancer that is included in the fifth order of cancer that causes death in the world. The highest incidence of liver cancer is found in men. Liver function related to the metabolism of various endogenous and exogenous substances is prone to chronic inflammatory stimulation, one of which is hepatitis C virus (HCV) infection. TGF-β1, NOX4, and TNF-α are three potential genes responsible for regulating chronic inflammation and fibrogenesis. This study aims to describe the profile of potential fibrogenesis and inflammation genes that play a role in the progression of HCC due to chronic hepatitis C. This type of research is a descriptive study with a quantitative approach that was carried out using Quantitative Real-Time PCR analysis. The results of serological tests in the male gender group showed that the average fasting body glucose level was 85.22 mg/dL, total cholesterol 242 mg/dL, triglyceride 158.44 mg/dL, HDL-C 18.56 mg/dL, and LDL-C 172 mg/dL. In the female gender group, the data obtained on average levels of fasting blood glucose 82.33l mg.dL, total cholesterol 350 mg/dL, triglyceride 181 mg/dL, HDL-C 29 mg/dL, and LDL-C 190 mg/dL. All gender groups also showed an increase in AST and ALT levels that had exceeded normal limits. The expression of fibrosis-related genes, namely TGF-β1 was 5.8x higher in male patients than female patients. The NOX4 gene also showed almost the same thing, namely, its expression level was 1.2 times higher in male patients. From the expression of the proinflammatory cytokine TNF-, it is also known that in male patients, the expression level is 2x times higher than in female patients. The difference in expression levels in the two groups involved a repressive mechanism of estrogen. Increased expression levels of TGF-β1, NOX4, and TNF- genes are thought to involve the activity of the HCV core protein, which then induces several signaling pathways such as NF-κB and SMAD3. From these results, the expression levels of TGF-β1, NOX4, and TNF- as the main regulators of chronic inflammation and fibrosis can describe the progression of HCC and can be used as initial data on the profile of inflammation and fibrogenesis in HCC cases that have previously been exposed to HCV infection.

Abstract

Diethylnitrosamine (DEN) is a metabolic product of various foods and beverages in which there are preservatives in the form of nitrate and nitrite. Nitrosamines are found in various products such as alcoholic beverages, processed meats, cigarettes, and cosmetic products. DEN is toxic and carcinogenic to organs such as the liver, lungs, kidneys, skin, stomach, and blood. In addition to giving DEN compounds, there are also CCl4 compounds as tumor promoter agents known as genotoxic substances that can increase the risk of genetic errors and stimulate cells to become malignant neoplasms. Administration of DEN and CCl4 induction can damage various organs of the body, one of which is the kidney. In a healthy body, angiogenesis plays a role in wound healing and the formation of new tissue. However, angiogenesis also contributes to carcinogenesis or the uncontrolled growth of cancer cells, and is malignant when the body is contaminated with toxic substances. Therefore, to determine the effect of the administration of DEN and CCl4 observations were made using the immunohistochemical method, to see differences in the histology of the kidneys of experimental animals, the positive control group, and the negative control group based on the VEGF angiogenesis factor. From these results, there are significant differences. In the negative group, there was no necrosis and high-value VEGF expression, but in the positive group, there was necrosis and a decrease in the value of VEGF expression compared to the negative control group.
