Abstract

Acceleration of wood weathering in the mangrove area is possible with the role of cellulolytic bacteria. Cellulolytic bacteria producing cellulase enzymes are needed by the animal feed and agriculture industries. Mangroves in Muntok Subdistrict undergo a process of adaptation to environmental changes due to tin mining, including microorganisms. Exploration of new species and strains of cellulolytic bacteria in miningaffected mangroves is needed to enrich the collection and its potential use for human needs. This study aims to identify cellulolytic bacteria in mangrove weathered wood in Muntok District, West Bangka Regency through isolation, screening, and biochemical characterization. Three sampling locations, namely Sukal Mangrove, Peltim Mangrove, and Tembelok Mangrove result 22 bacterial isolates with 11 isolates showing the ability of cellulose degradation in qualitative tests using Lugol. The biochemical characterization of bacterial isolates with the greatest cellulose degradation was directed towards Citrobacter freundii and Vibrio alginolyticus in samples of Sukal Mangrove and Actinomyces bovis in isolates from Peltim Mangrove.

Abstract

Dunaliella sp. adalah mikroalga halotoleran yang banyak dijumpai pada lingkungan hipersalin. Mikrolaga ini mengandung berbagai substansi yang berharga seperti karetonoid, gliserol, lipid, protein dan vitamin sehingga banyak dibudidayakan. Salah satu faktor utama yang memengaruhi pertumbuhan Dunaliella sp. yang dibudidayakan adalah ketersediaan senyawa di dalam media kulturnya. Salah satu senyawa yang dapat menstimulus pertumbuhan mikroalga adalah vanadat (VO43-). Penelitian ini menganalisis pengaruh konsentrasi vanadat dalam media kultur terhadap pertumbuhan Dunaliella sp. Konsentrasi vanadat yang digunakan adalah 0,002 mg/L; 0,02 mg/L; 0,2 mg/L; dan 0 mg/L (kontrol). Hasil penelitian ini menunjukkan bahwa keberadaan vanadat dalam media kultur memengaruhi secara signifikan pertumbuhan Dunaliella sp. di mana pertumbuhan terbaik didapatkan pada pemberian vanadat dengan konsentrasi 0,002 mg/L. Berdasarkan hasil penelitian ini, vanadat dalam media kultur dapat digunakan sebagai penstimulus pertumbuhan Dunaliella sp.Dunaliella sp. is halotolerant microalgae commonly found in hypersaline waters. This microalgae contains various valuable substances such as carotenoids, glycerol, lipid, protein, and vitamins, and thus, widely cultivated. One of the primary factors that influence the growth of cultured Dunaliella sp. is the availability of certain nutrient/chemical compounds in its culture media. One of the compounds that can stimulate the growth of microalgae is vanadate (VO43-). This study analyzed the effects of vanadate concentration in the culture media on the growth of Dunaliella sp. The vanadate concentrations used were 0.002 mg/L, 0.02 mg/L, 0.2 mg/L, and 0 mg/L as the control concentration. The results of this study suggested that the existence of vanadate in the culture media significantly affected the growth of Dunaliella sp. in which the best growth of Dunaliella sp. was obtained by the treatment with 0.002 mg /L of vanadate. This study recommends that the vanadate with concentration of 0.002 mg/L should be used in the culture media of Dunaliella sp.

Abstract

Desa Sedayu Lawas merupakan salah satu sentra produksi garam di Lamongan. Potensi tambak garam di desa ini cukup besar hanya saja produksi garam yang ada sebagian besar masih sangat tergantung kondisi cuaca. Salah satu inovasi dalam memproduksi garam rakyat untuk mengatasi masalah cuaca tersebut adalah dengan menggunakan teknologi Prisma Rumah Kaca.  Inovasi ini memanfaatkan rumah kaca dan plastik geomembran dalam proses kristalisasi garam. Tujuan dari penelitian ini adalah untuk menganalisis kualitas garam dari hasil produksi metode Prisma Rumah Kaca di desa Sedayu Lawas. Parameter yang diukur dalam penelitian ini adalah suhu, salinitas, kandungan Mg, kandungan Ca, kandungan NaCl dan kadar air. Hasil penelitian ini menunjukkan kalau kadar salinitas air pada kolam penampungan air muda adalah 23-24 ppt dan pada kolam penampungan air tua adalah 34-42 ppt. Nilai rata-rata NaCl, Mg dan Ca pada air muda secara berturut-turut adalah 115.000 mg/L, 313,6 mg/L dan 202,3 mg/L. Hasil pengujian air tua menunjukkan nilai rata-rata NaCl, Mg dan Ca secara berturut-turut adalah 129.333,3 mg/L, 313,6 mg/L dan 214,3 mg/L. Hasil uji kualitas garam menunjukkan kadar NaCl atas dasar berat basah (adbb) sebesar 87,56%, kadar Mg sebesar 2,15%, kadar Ca sebesar 3,45% dan kadar air sebesar 5,86%. Hasil penelitian ini mengindikasikan kalau kualitas garam yang dihasilkan melalui metode Prisma Rumah Kaca di desa Sedayu Lawas termasuk dalam kategori Kualitas K1 berdasarkan SNI 4435:2017.

Abstract

One of the main potentials of marine resources in Indonesia is salt. Salt production locations are located in various regions of Indonesia. Aceh Province is one of the leading salt-producing areas on the island of Sumatra. Salt production in Aceh faces various challenges, such as the production system and salt production locations' suitability. Therefore, to develop salt production in Aceh, analysis of the production system and suitability of salt production sites are fundamental. This study aims to analyze the production system and the suitability of salt production locations in North Aceh Regency and East Aceh Regency as salt production centers in Aceh Province. This research uses a combination of qualitative and quantitative methods (mixed method) with a descriptive approach. Production system analysis is carried out through direct observation and in-depth interviews with the stakeholders (government and salt farmers). Analysis of the salt location's suitability level was carried out using the Salt Suitability Index (SSI) method. This study shows that the production system in North Aceh and East Aceh districts uses three production methods consisting of boiling seawater, solar evaporation, and a combination of boiling and solar evaporation. The main obstacle to salt production in Aceh is the weather, such as high rainfall and fluctuations throughout the year. Analysis of the suitability of the location of salt production shows the value of the SSI at the salt ponds site in the North Aceh and the East Aceh Regencies is 80% which indicates that the locations are sufficiently appropriate as a salt production site. The results of this study show that the application of the Continuously Dynamic Mixing (CDM) method in Greenhouse Salt Tunnel (GST) technology may be used to improve the quality and quantity of salt production in Aceh.

Abstract

Water contamination is one of the main environmental problem presently. One of the contaminants that become serious problem in aquatic ecosystems is Cr(VI) which is toxic for living organisms. Therefore, technology to solve this problem is urgent. Biosorption is one of the alternative technologies that inexpensive and environmentally friendly. The present study analyzed the biosorption of Cr(VI) using streamer biofilm. Biofilm is a predominant habitat for most microbes in aquatic ecosystems. The sample of biofilms used in this study was streamer biofilm collected from the river. The results show that the streamer biofilm had ability to adsorb Cr(VI). The adsorption of Cr(VI) are fitted well to the Langmuir adsorption isotherm model. The maximum adsorbed amount of Cr(VI) is estimated to be around 8.33 mg/g and the adsorption equilibrium constant is around 0.02 L/mg. The results of the present study indicate that the streamer biofilm is a promising alternative biosorbent for the biosorption of water pollutant such as Cr(VI).

Abstract

Cantang grouper fish (Epinephelus fuscoguttatus-lanceolatus) is a result of hybridized fish between a female of tiger grouper fish (Epinephelus fuscoguttatus) and male of kertang grouper (Ephinephelus lanceolatus). In the development of cultivation, there are many problems, one of them infected with the class of virus Nodaviridae, namely Viral Nervous Necrosis (VNN). Fish had a defense against cellular immunity against the VNN virus by β-actin. The aims of this research to explore the crude extract of Spirulina sp as a β-actin inducer for the anti-inflammatory immune system in grouper fish against VNN attack. The method used in this research is experiment methods.  Crude extracts of Spirulina sp (33 μg/ml) were conducted by feeding orally to groupers, and VNN infections were conducted by feeding the already positive VNN meat. Detection of VNN using RT-PCR, however β-actin detection using PCR, and IHC in the organ of Cantang grouper fish. The results showed that the percentage of DAB value of control fish (14.0%), fish treated with Spirulina sp (25.7%), fish treated with VNN (31.9%), and fish treated with Spirulina sp extract and VNN infection (32.4%). The percentage of DAB values were indicated by the detection of the target gene β-actin. Immunity in fish increases with the addition of Spirulina sp. The increased β-actin expression may also be used as an indicator of a grouper's body defense against VNN infection.

Abstract

One of the important commodities for the Indonesian people produced from the marine sector is salt. The variety of salting problems has more or less affected the competitiveness of people's salt in each region in Indonesia. This condition clearly affects the welfare of salt farmers, because the selling price of salt received by farmers tends to be low. Tuban Regency is a coastal area that contributes to the achievement of salt production targets in East Java. Geographically, Tuban Regency is in the middle of salt processing areas, namely Surabaya-Gresik and Pati-Rembang, where these areas are also salt producing areas as well. Seeing this phenomenon, this study was conducted to analyze what factors affect the competitiveness of local salt in Tuban Regency. This study used primary data with a total of 90 respondents conducted randomly or random sampling. The analysis tool used is factor analysis. The research location was carried out in the salt production center area in Tuban regency, namely in Palang district and Tambakboyo district. From the results of the analysis, the results were obtained that there were five (5) factors that affect the competitiveness of local salt in Tuban Regency from the most influential to the factors that had the least influence, namely (1) Government Assistance, (2) Supporters, (3) Production Time and Government Regulations, (4) Social, and (5) Production and Post-Production.

Abstract

Catfish is an aquaculture commodity that has great potential to be developed in various regions in Indonesia. The success of catfish cultivation is primarily determined by creating a suitable environment for catfish. One way to create a suitable environment for catfish cultivation is the Recirculation Aquaculture System (RAS). In applying RAS, the presence of organic matter in the pond is controlled so the pond water can be reused. The presence of organic matter in pond water is determined mainly by the activity of bacteria in the water. These bacteria are actively involved in the pond's residual organic matter degradation process. Therefore, the total number of bacteria in the water can be one of the essential parameters in the RAS. The method widely used to calculate the abundance of bacteria is counting the number of bacteria based on bacterial culture method. However, the bacteria that can be cultured are only a small part of the bacteria in the water, so the abundance value obtained does not reflect the actual bacterial population. Therefore, it is necessary to analyze the abundance of bacteria using a method closer to the actual abundance value, such as the environmental DNA (eDNA) analysis. This study aims to analyze the abundance of bacteria in water in catfish aquaculture with RAS using the eDNA method. In addition, this study also analyzes water quality data (temperature, pH, Dissolved Oxygen). This study's results indicate that the water temperature value in catfish ponds ranges from 28.0 to 29.0 °C, the average pH value is 7.7, and the dissolved oxygen is between 5.7 - 6.2 mg/L. The water quality analysis results indicate that RAS can maintain optimum conditions in the catfish cultivation process. This study also showed that the total microbial abundance value at the beginning of cultivation was 1.68 × 107 cells/ml, and on day 30, it was 3.6 × 106 cells/ml. The dynamic of bacterial densities in this study may indicate that this system can maintain the stability of the microbial community.

Abstract

Kegiatan ini dilakukan dengan menginduksikan metode Continuously Dynamic Mixing (CDM) yang diterapkan dalam Greenhouse Salt Tunnel (GST) untuk mengkreasi produksi garam di Pantai Selatan Kabupaten Malang. Mitra kegiatan ini adalah Kelompok Usaha Garam Rakyat (Kugar) Bajulmati Sejahtera yang merupakan kelompok usaha garam pertama di Kabupaten Malang. Mitra memiliki masalah terkait dengan teknis produksi garam untuk menghasilkan garam yang berkualitas baik (NaCl > 95%). Indikator keberhasilan kegiatan pengabdian kepada masyarakat ini adalah: 1) teknologi GST di lokasi mitra sudah menggunakan metode CDM untuk produksi garam; 2) peningkatan pengetahuan dan keterampilan mitra dalam pengaturan air dalam metode CDM dalam GST; dan 3) Mitra dapat memproduksi garam berkualitas K-1 dengan peningkatan kapasitas produksi lebih dari 50%. Kegiatan pengabdian kepada masyarakat ini telah dapat mengkreasi produksi garam untuk pertama kali di Pantai Selatan Kabupaten Malang.

Abstract

Potential of resource commodities contained in the waters of the Bali Strait needs to be studied for the level of utilization and the level of sustainability. This research aimed to analyze the conditions for the utilization of lemuru fish resources commodities and the sustainability status of lemuru fisheries in the Bali Strait based on four aspects, namely ecological, economic, social, and technological dimensions. Commodity condition of resources is carried out with Catch Per Unit Effort (CPUE) analysis, while the sustainability status of resource commodities uses Rappid Appraisal for Fisheries (RAPFISH) analysis. Data used in this study is based on data from PPN Pengambengan, Bali and PPP Muncar, Banyuwangi. The questionnaire was distributed to 55 respondents with purposive sampling techniques. The results of the analysis on the CPUE of the commodity condition of lemuru fish resources in 2013-2017 experienced a decrease and the occurrence of over-fishing in the Bali Strait. Meanwhile, based on the status of sustainability, it shows that the status is less sustainable in a multidimensional manner with a value of 36.47.
