Abstract

One of the most severe environmental problems is heavy metal pollution such as Cr(VI) in aquatic ecosystems. Cr(VI) is toxic and carcinogenic heavy metals pollutants that persistent and easily accumulated through the food web. Accumulation of Cr(VI) concentration in the aquatic ecosystem gradually increases with the increases of industrial activity. Therefore, Cr(VI) concentration in aquatic ecosystems should be monitored continuously, and thus, suitable and accuracy monitoring technologies are necessary. This study analyzed the utilization of biofilm as a biomonitoring agent of Cr(VI) in the Badek River, Malang City. The concentration of Cr(VI) inside the biofilm and the surrounding water of biofilm were investigated. The results revealed that the concentrations of Cr(VI) inside the biofilms were hundred times higher than the surrounding water. The biofilm may accumulate Cr(VI) from the surrounding water resulting in the Cr(VI)-rich microenvironment inside the biofilm. The adsorbed Cr(VI) is retained mostly in the interstitial water of the biofilm matrices. The findings in this study suggested that the biofilm can be used as a potential biological agent to monitor of Cr(VI) pollutant in the river ecosystems.

Abstract

Koi merupakan ikan hias favorit di pasar nasional dan internasional. Proses budidaya untuk menghasilkan koi dengan kualitas terbaik masih menjadi kendala, salah satunya adanya infeksi parasit Myxobolus sp. yang dapat menyebabkan kematian massal dalam waktu singkat. Myxobolus yang menyerang jaringan ikan dapat menyebabkan kerusakan prah pada jaringan. Banyak cara dilakukan oleh pembudidaya Ikan Koi untuk menghindari infeksi Myxobolus, yaitu dengan pengobatan secara alami dan menggunakan bahan kimia. Penelitian ini bertujuan untuk mengetahui efek hematologi dan respon imun pada Ikan Koi yang terinfeksi parasit Myxobolus dengan pengobatan dimilin. Metode yang digunakan adalah metode eksperimental dengan perlakuan kontrol ikan sehat (A), Ikan koi yang terinfeksi oleh Myxobolus sp. tanpa perlakuan dimilin (B), ikan koi terinfeksi Myxobolus sp dengan perlakuan dimilin 0,02 mg (C), dan ikan yang terinfeksi Myxobolus sp dengan perlakuan dimilin 0,01 mg (D). Pengambilan darah ikan dilakukan dengan metode punksi pembuluh darah bagian caudal dan dilakukan pengamatan hematologi, serta dilakukan pengamatan imunohistokima dengan melihat persentase Diaminobenzidine (DAB) pada ikan. Hasil rata-rata pada perhitungan hematologi leukosit dan eritrosit tertinggi  ditemukan pada ikan yang terinfeksi Myxobolus sp tanpa perlakuan dimilin (B), dengan eritrosit 2.913.334 sel / mm3 dan leukosit 197.184 sel / mm3. Hasil respon imun berdasarkan pengamatan imunohistokimia didapatkan nilai DAB perlakuan A adalah 16%, nilai DAB perlakuan B 42,7%, nilai DAB perlakuan C 25,6%, dan nilai DAB perlakuan D 31,4%. Kesimpulan penelitian ini adalah pemberian dimilin dapat mempengaruhi respon hematologi dan dapat mempengaruhi respon imun pada ikan yang dapat ditunjukkan dengan Interleukin-6 pada ikan dengan hasil DAB immunoratio. Koi is a favorite ornamental fish in the national and international markets. Cultivation process to produce koi with the best quality is still have problem, one of the problem is parasitic infection of Myxobolus sp. which can cause mass death in a short time. Myxobolus attacks fish tissue can cause severe damage to tissue. Many ways were used by koi fish farmers to avoid Myxobolus infection which  with natural treatment and using chemicals drug. This study aims to determine the hematological effects and immune respons in the koi fish infected with the Myxobolus and treated with dimilin. Experimental method was used in this research with control treatment of healthy fish (A), koi fish infected by Myxobolus sp. without treatment (B), koi fish infected by Myxobolus sp with dimilin treatment 0.02 mg (C), and koi fish infected by Myxobolus sp with dimilin treatment 0.01 mg (D). fish Blood samples were collected with puncture of caudal vessel method and then haematological observations to blood samples, and immunohistokima observations by looking percentage of Diaminobenzidine (DAB) in fish. Highest hematological Results of leukocytes and erythrocytes were found in treatment B, with erythrocytes result 2,913,334 cells / mm3 and leukocyte 197,184 cells / mm3. Immune response result based on immunohistochemical observations with DAB value of treatment A is 16%, treatment B is 42.7%, treatment C is 25.6%, and treatment D is 31.4%. Conclusion in this study that dimilin treatment can affect hematological response and immune response in fish which can be shown with Interleukin-6 in fish with DAB immunoratio results.

Abstract

The study aimed to obtain isolate of cellulolytic bacteria from leaf litter on mangrove in Bangka Island. Sampling was conducted on mangroves in Sungailiat, Bangka and Tukak Sadai, South Bangka district. The isolation was carried out using 1% enriched agarmedia of Carboxymetyl Cellulose (CMC). The bacterial isolates were tested with cellulolytic growth on 1% enriched CMC agar medium and lugol added at 72 hours. The clear zone resultingindicates cellulose degradation ability. The positive isolate of cellulolyticwas identified by biochemical tests. 1 of 5 isolates in Sungailiat positive cellulolytic mangroves and identified as Bacillus subtilis. 2 of 5 isolates in mangrove Tukak Sadai is positive as a cellulolytic bacteria and identified as Staphylococcus saproviticus and Bacillus cereus.

Abstract

Cellulose as an abundant source of glucose in Indonesia requires acceleration of decomposition utilizing cellulolytic bacteria.  Cellulolytic bacteria can be obtained from the isolation of mangrove organic matter, such as sediments. Muntok Sub-district is one of the regions with the most tin mining in West Bangka Regency also has mangroves in the coastal area. Exploration of cellulolytic bacteria in mangroves with different environmental characteristics encourages researchers to find new bacterial strains that produce cellulase enzymes with new properties. Thirteen isolates were successfully isolated from three locations. Tembelok mangrove sediments produced Seven bacterial isolates, Peltim Mangrove samples produced three isolates and from Sukal Mangrove three isolates were obtained. Seven isolates showed clear zones in the Lugol test and three isolates including were gram-positive bacteria. Molecular test with 16S rRNA analysis showed TBL1 isolate has 85% similar identity of  Vibrio parahaemolyticus strain HY3 and TBL2 isolate has 98% similar identity of Bacillus amyloliquefaciens strain HS8. Bacillus amyloliquefaciens potential to further study as cellulose degrading bacteria for feed ingredients.

Abstract

Bioremediation is one of kind method which use of bacteria to degrade of hydrocarbon to be another compound. This study aims to get isolation of indigenous bacteria and potential test with Gordonia terrae on diesel oil bioremediation. The water sample was taken from Tanjung Perak Port of Surabaya, Indonesia where contaminated with diesel oil. Analysis of in-situ water quality parameters were consist of Dissolved oxygen, temperature, pH, salinity. Strain isolate bacteria were isolated by Bushnell-Hass medium. The bacterial density result shows in IN6 shows that bacterial density increased up to 9 × 108 cells/ml at day 2 incubation and decrease up to 1.79 × 108 cells/ml at final incubation, IN8 increased up to 8.6 × 108 cells/ml, then decrease up to 3.57 × 108 cells/ml. While, EX8 increased up to 9.1 × 108 cells/ml, and then decrease up to 3.06 × 108 cells/ml. While, results of degradation diesel oil hydrocarbon show treatment IN6, TPH values showed a decrease in the initial hydrocarbon concentration of 30 ppm to 16 ppm. IN8 TPH value shows a decrease in the initial hydrocarbon concentration of 30 ppm to 16 ppm. EX8 TPH value shows a decrease in the initial hydrocarbon concentration of 30 ppm to 24 ppm. In this experiment was found that Bacillus cereus was effectively degrading of diesel oil and can be agent bacterial for bioremediation.

Abstract

The bioremediation system can be improved by using specific bacterial potential as oil-degrading bacteria which microorganisms can use hydrocarbons as a carbon source for their metabolic processes. The aim of this study is to identify R. erythropolis on degradation oil contamination and to obtain indigenous bacteria as new agent bacteria on bioremediation of oil contamination. The polluted water samples from used oil were taken from PPN Prigi, Trenggalek, East Java, Indonesia. The parameters considered were the detection and characterization of indigenous bacteria that degraded used oil. The density of bacteria was analyzed in the interval time of days 0, 2, 4, 6 and 7 and TPH was analyzed at final incubation. The results of this study indicate that the effectiveness of reducing oil concentration was used in testing the potential of bacteria from the highest was Pseudomonas aeruginosa which as indigenous bacteria isolated from water contaminated sites with application cell rate 1×108 cells/ml. It reduced of oil concentration up to 53%, and 1×106 cells/ml reduced oil concentration up to 47%. While, R. erythropolis with application cell rate 1×108 cells/ml reduced 47%. This result was found that Pseudomonas aeruginosa was effectively removed of oil concentration.

Abstract

Aquatic sediment has a role for heavy metals sink. Understanding the geochemical fractions of heavy metals in the aquatic sediment can predict the mobility and reactivity of heavy metals fractions that can induce environmental problems. The aim of this study was to determine the distribution of geochemical fractions of copper and zinc in aquatic sediment of Nyolo spring. Sediments samples were taken from five locations along the spring. Geochemical fractions of copper and zinc were extracted using BCR (Community Bureau of Reference) sequential extraction method. The concentration of these metals in each extract was analyzed using AAS. The result showed that overall the sediment consisted of zinc at a higher concentration than copper. Amongst the geochemical fractions, copper and zinc were found dominantly at an oxidizable fraction. As the first fraction, Zn was more bioavailable than copper for biological uptake. Furthermore, based on the Risk Assessment Code (RAC) value, the aquatic sediment of Nyolo Spring was low risk to the Zn and Cu contaminations at most sites, except for Zn in Site III showed a medium risk. This metals fraction will potentially available for organism uptake with the changing of pH sediment or overlying water

Abstract

Penelitian ini mengambarkan pola pemanfaatan pariwisata terumbu karang di Taman Wisata Perairan (TWP) Gita Nada dan mengestimasi nilai manfaat ekosistem terumbu karang dengan pendekatan biaya perjalanan sebagai dasar rujukan perencanaan pengembangan kawasan pariwisata di Kabupaten Lombok Barat. Penelitian dilakukan di Taman TWP Gita Nada Sekotong Lombok pada bulan Januari-Maret 2020. Pengumpulan data primer dilakukan dengan cara wawancara menggunakan kuisoner dan observasi segala aktivitas pariwisata yang ada di TWP Gita Nada. Data sekunder dikumpulkan dengan cara penelusuran literatur pada hasil penelitian terdahulu serta publikasi yang dilakukan oleh instansi terkait. Metode analisis yang digunakan adalah Zona Travel Cost Method (ZTCM). Perairan TWP Gita Nada memiliki kombinasi perairan dangkal dengan tipe fringing reefs dan letak TWP Gita Nada yang berbatasan dengan Selat Lombok. Atraksi wisata yang ditawarkan di TWP Gita Nada adalah wisata pantai dan bahari. TWP Gita Nada dengan luas terumbu karang sebesar 1279 ha memiliki nilai manfaat pariwisata Rp3.004.031.073/ha dengan jumlah total pengunjung per 1000 penduduk pada kedua zona adalah sebanyak 51.228 orang. Berdasarkan model fungsi permintaan pariwisata TWP Gita Nada pengembangan kedepan adalah wisata alam yang dikemas menjadi wisata edukasi yang fokus pada anak muda dengan minat belajar tinggi. Perbaikan aksesibiltas dan peningkatan kualitas sarana dan prasarana yang memadai akan menambah daya tarik TWP Gita Nada, dan kedepan lokasi wisata harus mampu memberikan jaminan 2H yaitu healthy dan hygiene.Title: Benefit Value of Coral Reef Ecosystem Tourism in The Marine Park Gita Nada Sekotong LombokThis study describes the use patterns of coral reef tourism in Marine Park Gita Nada. It estimates the benefit value of coral reef ecosystem with travel cost approach as a reference for planning the development of tourism areas in West Lombok Regency. The research conducted at Marine Park Gita Nada Sekotong Lombok in January to March 2020. Primary data were collected by interview questionnaires and observations of entire tourism activities in Marine Park Gita Nada. Secondary data were collected by literature review on the results of previous research and publications of related agencies. The research used zona travel cost method (ZTCM) analysis.The waters of Marine Park Gita Nada is a combination of shallow water with fringing reef circulation, and Marine Park Gita Nada is located in the border of Lombok Strait. Marine Park Gita Nada offers beach and marine attraction, and coral reefs cover 1279 ha in the area. Marine Park Gita Nada has a tourism benefit value of Rp3,004,031,073/ha with total number of visitors in both zones are 51,228 people per 1000 inhabitants. Based on tourism demand function, the future development for Marine Park Gita Nada would be educational nature-based tourism focusing on young people with high learning interests. Improvement of accessibility and quality of infrastructure will attract more tourists to TWP Gita Nada, and in the future it must guarantee the healthy and hygiene (2H) of the tourism park.

Abstract

Bakteri selulolitik memiliki kemampuan degradasi selulosa dan membuat karbohidrat lebih mudah dicerna bagi ternak. Penelitian ini bertujuan untuk mengidentifikasi bakteri selulolitik di tanah, kayu lapuk dan daun dari mangrove pulau Bangka melalui skrining dan analisis gen 16s rRNA. Penelitian dilakukan dari September 2017 hingga Maret 2018. Isolat TSL7 dan TSS4 dari Mangrove Tukak Sadai dan SLS5 dari Mangrove Sungailiat yang memiliki kemampuan mendegradasi selulosa terbesar berdasarkan hasil skrining menjadi isolat yang diidentifikasi pada gen 16S rRNA untuk diurutkan dan analisis BLAST. Analisis gen bank menunjukkan kedekatan dengan Pseudomonas aeruginosa dengan query cover 37% - 87%. Hasil query cover rendah dihasilkan karena sekuens yang sesuai pendek. Kontaminasi dan perubahan komunitas karena seleksi lingkungan dengan adanya logam berat dapat menjadi penyebab dominasi Pseudomonas aeruginosa. Pseudomonas aeruginosa yang bersifat patogen tidak disarankan untuk diaplikasikan sebagai pendegradasi selulosa pakan ternak.
